Abstract

본 발명은 간단하고, 실제적인 절차로 인간 소변의 트롬보모둘린을 정제하는 것을 가능하게 하며, 낮은 함량의 트롬보모둘린을 갖는 출발 물질로부터도 높은 함량의 트롬보모둘린의 경우에보다 더 큰 정제율로 정제하는것을 가능하게 한다.

제1도는 히드록시아파타이트 컬럼에 흡착된 인간 소변의 트롬보모둘린을 함유하는 출발 물질(할당 번호 1)을 인산나트륨의 직선 농도 구배로 용출 시켰을 때의 실시예 1에서 수득된 트롬보모둘린의 용출 경향이다.

Claims (5)

  1. 흡착제로서 히드록시아파타이트를 사용하는 흡착 크로마토그래피를 수행함으로 인간 소변의 트롬보모둘린을 정제함을 특징으로 하는 트롬보모둘린의 정제 방법.
  2. 인간 소변의 트롬보모둘린을 함유하는 출발 물질을 히드록시아파타이트가 충전된 컬럼에 부어, 트롬보모둘린을 흡착시키고, 이어서 히드록시아파타이트로부터 분리함으로 이루어지는 방법임를 특징으로 하는 트롬보모둘린의 정제 방법.
  3. 제2항에 있어서, 트롬보모둘린을 함유하는 출발 물질은 원료 인간 소변, 황산암모늄을 사용한 분획을 통하여 수득된 농축물 또는 음이온 교환크로마토그래피 또는 트롬빈 결합 친화성 크로마토그래피에 의하여 수득되는 예비 정제된 물질임을 특징으로 하는 방법.
  4. 제2항에 있어서, 흡착은 pH 5-8에서 수행되며, 분리는 인산의 농도 구배에서 수행됨을 특징으로 하는 방법.
  5. 제1항 또는 제2항에 있어서, 정제는 이온 교환 크로마토그래피, 트롬빈 결합 친화성 크로마토그래피 및 겔 투과 크로마토그래피의 절차중 하나 또는 그들의 조합으로 수행됨을 특징으로 하는 방법.
