Abstract

바이오센서는 질산염 또는 아질산염이온 존재하에 생화학 반응을 생성할 수 있는 생화학물질과 질산염 또는 아질산염이온 존재하에 생화학물질에 의해 생산된 반응으로 직간접적으로 생산된 변환성 출력물을 감지할 수 있도록 배열된 변환기로 구성되고 이때, 생화학물질은 질산염 또는 아질산염이온의 환원효소와 질산염 또는 아질산염의 환원에 의해 산화될 수 있는 공-인자로 구성되고 생화학물질은 공-효소의 산화에 의해 직간접적으로 광자를 생산하는 광발광 성분을 포함하고 변환기는 광감지기로 구성된다.

질산염 또는 아질산염이온 감지용 바이오센서(BIOSENSORS FOR DETECTING NITRATE OR NITRITE IONS)

제1도는 본 발명에 따른 바이오센서 배열의 측면도이고,
제2도는 본 발명에 따른 또 다른 바이오센서의 측면도이고,
제3도는 본 발명에 따라 구체화된 바이오센서에서 이용되는 생화학반응 관련 일반 과정을 도식적으로 나타낸 것이고,
제5도는 본 발명에 따라 구체화된 바이오센서에서 이용되는 생화학 과정의 예를 도식적으로 나타낸 것이다.

Claims (8)

  1. 바이오센서는 질산염 또는 아질산염이온 존재하에 생화학 반응을 생성할 수 있는 생화학물질과 질산염 또는 아질산염이온 존재하에 생화학물질에 의해 생산된 반응으로 직간접적으로 생산된 변환성 출력물을 감지할 수 있도록 배열된 변환기로 구성되고 이때, 생화학물질은 질산염 또는 아질산염이온의 환원 효소와 질산염 또는 아질산염의 환원에 의해 산화될 수 있는 공-인자로 구성되고 생화학물질은 공-효소의 산화에 의해 직간접적으로 광자를 생산하는 광발광 성분을 포함하고 변환기는 광감지기로 구성된 것을 특징으로 하는 바이오센서.
  2. 제1항에 있어서, 고정된 효소 조성물은 질산염 또는 아질산염 환원효소 일정량과 산화된 상태와 환원된 상태를 가지는 공-인자 일정량과 산화상태의 공인자하에 산화반응을 유도할 수 있는 공-효소 일정량으로 구성되고 이때 공-인자는 환원효소에 의한 환원반응으로 환원상태에서 산화상태로 전환할 수 있고, 따라서 공-인자는 환원효소에 의해 추가 환원반응을 허용하기 위해 환원상태의 공인자의 공급을 유지하기 위해 공인자는 환원상태로 전환하게 되고. 추가로 제2공-효소에 의해 유도되는 산화반응 정도와, 산화된 상태에 있는 공-인자의 형성을 측정하는 제2공-효소에 의한 산환정도를 측정하기 위한 수단이 포함되어 환원 효소에 의해 환원된 질산염 또는 아질산염의 양을 측정하게 되고, 감지수단은 제2공-효소에 의해 유도된 산화반응의 결과로써 직간접적으로 방출되는 광자를 감지하기 위한 광감지기로 구성된 것을 특징으로 하는 바이오센서.
  3. 제2항에 있어서, 바이오센서는 질산염 환원반응에 의해 생산된 아질산염이 아질산염 환원효소에 의해 추가 환원될 수 있도록 질산염 공-효소와 아질산염 공-효소 모두가 결합된 고정된 효소 조성물을 포함하는 것을 특징으로 하는 바이오센서.
  4. 제1항 내지 3항 중 어느 한 항에 있어서, 산화될 공-인자는 환원효소에 의해 제공되는 환원반응과 같이 NADP+로 산화될 니코틴아미드 효소 NADPH로 구성된 것을 특징으로 하는 바이오센서.
  5. 제1항 내지 4항중 어느 한 항에 있어서, 광자는 공-효소 화학반응에서 작용하는 하나이상의 다른 공-효소를 경유하여 생산되는 것을 특징으로 하는 바이오센서.
  6. 제1항 내지 5항중 어느 한 항에 있어서, 광자는 NADPH와 함께 생화학반응에서 이용된 효소 루시퍼라제에 의해 생산되는 것을 특징으로 하는 바이오센서.
  7. 제2항 내지 6항중 어느 한 항에 있어서. NADP+의 생산은 알코을 디하이드로게나제의 존재하에 유기알코올이 이에 상응하는 알데히드로의 산화반응에 의해 감지되는 것을 특징으로 하는 바이오센서.
  8. 제7항에 있어서, 알데히드는 반응을 촉매하는 효소 루시퍼라제와 함께 산소존재하에 환원된 플라빈 FMNH2와의 반응에 의해 감지되는 것을 특징으로 하는 바이오센서.
