Abstract

피검시료중의 T세포에서 cDNA 중의 T세포 항원 리셉터 유전자를 PCR법에 의해 증폭하고, 이어서 증폭된 유전자를 싱글 스트렌드 콘포메이션 폴리모르피즘(SSCP)법에 의해 해석함으로서 피검시료에 직접하여 있는 T세포 클론에 특이적인 T세포 항원 리셉터 유전자의 존재를 검출할 수 있다.

Claims (7)

  1. T세포를 함유하는 피검시료로부터 T세포 유래의 cDNA를 함유하는 DNA 혼합물을 조제하고; 이 cDNA의 혼합물을 T세포 항원 리셉터의 β사슬을 코드하는 DNA 단편을 프라이머로서 사용하는 폴리메나제 체인 리액션(PCR)에 걸고, 이 cDNA의 혼합물중의 T세포 항원 리셉터의 β사슬을 코드하는 유전자를 증폭하고; 이어서 증폭된 유전자를 싱글 스트렌드 콘포메이션 폴리모르피즘(SSCP)법에 의한 해석에 걸고, T세포 항원 리셉터의 β사슬을 코두하는 유전자로 피검시료중에 직접하여 있는 T세포 클론에특이적인 유전자의 존재를 검출하는; 것으로 이루어진 T세포 항원 리셉터 유전자 발현의 검출방법.
  2. 제1항에 있어서, 피검시료가 면연응답이상이 의문되는 환자, 바이러스성 질환환자, 담암환자 또는 간염환자의 체액 또는 조직에서 얻어진 것인 검축방법.
  3. 제1항 또는 제2항에 있어서 PCR의 프라이머로서, T세포 항원 리셉터의 β사슬 V영역을 코드하는 DNA단편 및 β사슬 C영역을 코드하는 DNA 단편을 사용하는 검출방법.
  4. 제3항에 있어서, PCR의 프라이머로서 사용하는 T세포 항원 리셉터의 β의 V영역을 코드하는 DNA 단편이 Vβ1에서 Vβ20의 서브타입에서 선택된 영역을 코드하는 DNA 단편인 검출방법.
  5. 제1항 내지 제4항의 어느 한항에 있어서, SSCP법에 의한 해석을 글리세롤을 첨가한 가교도가 낮은 아크릴아미드 겔을 사용하여 수행하는 검출방법.
  6. 제1항 내지 제5항의 어느 한 항에 있어서, 피검시료중에 직접하여 있는 T세포 클론에 특이적인 유전자로서 T세포 항원 리셉터 β사슬 D영역 및/또는 β사슬 J영역을 코드하는 DNA 배열부분에 특이적인 배열을 갖는 T세포 클론과 그의 유전자의 존재를 검출하는 검출방법.
  7. 제1항 내지 제6항의 어느 한 항에 있어서, SSCP법에 의한 해석을 T세포 항원 리셉터의 β사슬 J영역을 코드하는 DNA 단편을 프로우브로서 사용하는 하이브리다이제이션법에 의해 수행하는 검출방법.
