Abstract

본 발명은 인 혈장 또는 동결 혈장을 융해한 후 원심 분리하여 침전(Cryoprecipitate)을 제거한 혈장으로부터 이온 교환 수지법과 헤파린 흡착겔을 이용하여 구연산 나트륨 완충액 및 염화칼슘 용액으로 용출함을 특징으로 하는 혈액 응고 제 2 인자 및 제 9 인자를 고순도로 얻는 제조 방법에 관한 것이다.
본 발명은 인 혈장으로부터 혈액 응고 인자를 정제함에 있어서, 이온 교환수지법과 친화성 크로마토그래피를 이용하여 염화칼슘을 함유한 완충액으로 용출함을 특징으로 한다.
본 발병에 의하여 순수하게 정제된 제 9 인자는 혈우병 B의 치료제로, 제 2인자는 혈액 응고제로 쓰이는 트롬빈의 생산원료로 사용되어질 수 있다

Claims (8)

  1. 인 혈장으로 부터 혈액 응고 인자를 정제함에 있어서, 이온 교환수지법과 친화성 크로마토그래피를 이용하여 염화칼슘을 함유한 완충액으로 용출함을 특징으로 하는 제 2 인자 및 제 9 인자의 정제 방법.
  2. 제 1 항에 있어서, 이온 교환수지로 DEAE-겔 또는 QAE-겔을 사용하는 것을 특징으로 하는 방법.
  3. 제 1 항에 있어서, 친화성 크로마토그래피로 헤파린 흡착겔을 사용함을 특징으로 하는 방법.
  4. 제 1 항에 있어서, 완충액이 구연산 나트륨 완충액과 트리스 완충액임을 특징으로 하는 방법.
  5. 제 2 항에 있어서, QAE-겔로 부터 10-50mM의 염화칼슘을 함유한 용출액으로 혈액 응고 인자를 용출함을 특징으로 하는 방법.
  6. 제 3 항에 있어서, 헤파린 흡착겔로 부터 2.5-5.0mM의 염화칼슘을 함유한 용출액으로 혈액 응고 인자를 용출함을 특징으로 하는 방법.
  7. 제 4 항에 있어서, 구연산 나트륨 완충액의 농도가 10mM 내지 50mM임을 특징으로 하는 방법.
  8. 제 4항에 있어서, 트리스 완충액의 농도가 10mM 내지 50mM임을 특징으로 하는 방법.
