Claims (14)
- 바실러스(bacillus) 290-3(DSM 5850)에서 얻어진 r-시클로덱스트린 그릴코실전이효소.
- 자연미생물기원(origin)에서 가급적 다량으로 취득할 수 있음을 특징으로 하는 제1항에 r-시클로덱스트린 글리코실전이효소의 아미노-종결영역내의 변경유도체.
- 제1항에 의한 r-시클로덱스트린 글리코실전이효소의 유전자, 즉 바실러스 1-1에서 얻어진 r-CGTase의 대응아미노산을 엔코팅하는 DNA영역에 의해 5′영역에서 치환되는 유전자에 의해 엔코팅(encoding)함을 특징으로 하는 r-시클로덱스트린 글리코실전이효소.
- 제1항 내지 제3항중 한항에 의해 얻어질 수 있는 시클로덱스트린 글리코실전이효소를 엔코팅(encoding)하는 발현 플라스미드(expression plasmds).
- (a) r-CGTase의 분비용으로 친알칼리성 전분분해 세균을 선별시켜 (b) 이들 세균을 분류학적으로 특성화하고, (c) 그 세균에서 얻어진 r-CGTase를 정제하여 생화학적으로 특성화하며, (d) r-CGTase의 유전자를 클로닝(cloning) 및 시 싱(sequencing)하고, (e) 이 유전자를 충분히 성장한 단백질의 5′영역을 엔코팅한 영역에서 변경시키며, (f) 이와같이 변경한 유전자를 조절할 수 있는 프로모터로 조절하고 세균에 도입시켜 그 유전자를 그 세균으로 증식시켜 그 유전자생성물이 분비될 수 있도록 하며, (g) 유도 및 발현 후에 배지상증액에서 효소를 취득함을 특징으로하는 r-시클로덱스트린 글리코실전이효소의 취득방법.
- 제5항에 있어서, 상기 공정 (e)에서 5′영역을 엔코팅하는 영역은 또다른 r-CGTase의 대응영역을 엔코딩하는 영역에 의해 대치시킴을 특징으로하는 상기 방법.
- 제6항에 있어서, 사용한 또다른 r-CGTase의 대응영역을 엔코딩하는 그 영역은 바실러스 1-1의 대응영역을 엔코딩하는 영역임을 특징으로 하는 상기 방법.
- r-시클로덱스트린 글리코실전이효소의 조절할 수 있는 발현이 가능하도록 신호펩티드를 사용하여 r-시클로덱스트린 글리코실전이효소를 코딩하는 유전자가 유전자 공학적적인 변경을 하여, 그 변경유전자를 분비 돌연변이체(secretor mutants)로 발현함을 특징으로 하는 r-시클로덱스트린 글리코실전이효소의 제조방법.
- 제8항에 있어서, 사용되는 r-시클로덱스트린 글리코실전이효소를 코팅하는 유전자가 제1항에 의한 r-시클로덱스트린 글리코실전이효소의 유전자임을 특징으로 하는 상기방법.
- 제2도에 나타낸 아미노산 시퀀스(sequence)를 엠코딩하는 DNA.
- 제2도의 아미노산75에서 시작하는 아미노산 시 스를 포함하는 r-시클로덱스트린 글리코실전이효소를 엔코딩하는 DNA.
- 아미노산 198에서 아미노산214이내 및/또는 아미노산 238에서 아미노산 247이내 및/또는 아미노산 267에서 아미노산 275이내 및/또는 아미노산 336에서 아미노산 343이내를 포함하는 제2도의 아미노산 영역을 포함하는 r-시클로덱스트린 글리코실전이효소를 엔코딩하는 DNA.
- 1항 내지 3항중 하나이상의 항에 의한 r-CGTase를 사용함을 특징으로하는 r-시클로덱스트린의 제조방법.
