Abstract

본 발명은 효모로부터 재조합 인체종양괴사인자(TNF-α)를 정제하는데 있어서, 부분적으로 정제된 조 종양괴사인자를 셀루파인 설페이트(cellufine-sulfate)를 담체로 사용한 친화성 크로마토그래피를 통하여 고순도로 정제하는 방법에 관한 것이다.

제1도는 본 발명에 따른 재조합 인체 종양괴사자인자의 정제과정에 있어서 음이온 교한수지를 이용하여 분리한 크로마토 그래피의 패턴이다. 제2도는 본 발명에 따른 정제과정에 있어서 셀루파인 셀페이트를 이용하여 분리한 크로마토그래피의 패턴이다. 제3도는 본 발명에 따른 정제과정에 있어서 S-300를 이용하여 분리한 크로마토그래피의 패턴이다.

Claims (3)

  1. 조 종양 괴사인자로 부터 셀루파인 설페이트(cellufine sulfate) 친화성 크로마토그래피를 이용하여 재조합 종양괴사인자(TNF-α)를 정제하는 방법.
  2. 제1항에 있어서, 조 종양 괴사인자는 가)세포 분쇄액을 암모니움 설페이트 분별 침전시키는 과정, 나)페닐-세파로즈 크로마토 그래피, 다)음이온 교환 수지 크로마토그래피 등의 단계를 포함하는 정제과정을 통하여 수득된 것임을 특징으로 하는 방법.
  3. 제1항에 있어서, 재조합 종양괴사인자(TNF-α)는 재조합 인체, 원숭이 또는 쥐의 종양괴사인자인 것을 특징으로 하는 방법.
