제1도는 고정된 여러가지 농도의 정착제 존재하에 동일한 양의 안티-Leu 3a(PE)로 착색시킨 용균되지 않은 전체 혈액세포의 착색 부피에 대한 10g형광성의 평균값을 도식화한 것이다.
제2도)는 안티-Leu 3a(PE) 및 Pandex D2.21μ자가 형광 미세입자를 포함하는 희석제로 착샛시킨 용균되지 않은 전체 혈액세포에 과향의 일차 형광트리거(trgger)에 의해 기록된, 10g형광 1에 10g형광 2의 윤곽선 그래프이고 B)는 A)에서 고정된 이트(gate)에서 세포및 입자에 대한 10g형광 1의 막대그래프이다.
제3도는 안티-Leu 3a(PE)로 착색시킨 3개의 개별적인 샘플의 2회 반복 실시한 용균되지 않은 전체 혈액 세포내의 미세입자당 CD4+임파구의 절대수를 막대 그래프로 나타낸 것이다.
Claims (3)
- (a)각 세포군을 표지할 수 있는 하나 혹은 그 이상의 세포표식자와 기지수의 형광성 미세입자를 포함하는 희석제를 함유하는 시험관에 샘플을 첨가하고, (b) 형광 트리거는 모든 미세입자 및 측정하고자 하는 세포군을 필수적으로 포함하도록 조정하고, (c)하나 또는 그 이상의 형광 게이트는 각각의 세포표식자 및 미세입자들을 구분할 수 있도록 조정하고, (d)(c)단계에서 형광트리거와 같거나 초과하는 세포수를 측정하고, 그리고, (e)(d)단계로부터 각 형광게이트에서 미세입자당 세포수를 계산하고 미세입자의 농도를 곱하여 결과적으로 단위 부피당세포의 절대수를 계산하는 단계들로 구성된, 전체 혈액 혹은 골수 샘플내에 있는 하나 혹은 그 이상의 세포군의 절대수를 플로싸이토메터에 의하여 측정하는 방법.
- (a) 등장 완충용액, 세포를 면역 형광 표식으로 표지될 수 있도록 하는 하나의 면역 형광 표식자, 정착제 및 두개의 형광 채널에서 자기 형광성을 지니고 더 나아가 첫번째 채널과 두번째 채널에서 자기 형광성 미세입자의 비율이 상기의 첫번째와 두번째 채널에서 면역 형과 표식자에 의해 방사되는 형광 비율과 구분될 수 있도록 기지 농도의 미세입자로 구성된 희석제를 포함하는 시험관에 샘플을 첨가하고, (b)두번째 형광 채널에서 형광 트리거는 필수적으로 측정하고자 하는 모든 미세입자 및 세포군을 포함하도록 조정하고, (c)플로싸이토메트리에 의해 두번째 형광 채널에서 형광 트리거와 같거나 초과하는 모든 미세입자 및 면역 형광으로 표지된 세포를 측정하고, (d) 첫번째 및 두번째 형광채널에서 형광 게이트는 면역 형광 표식자 및 미세입자 사이에서 구분이 되도록 조정하고, 그리고 (e)(d)단계로부터 미세입자당 세포의 수를 계산하고 여기서 미세입자의 농도를 곱하면 결과적으로 단위 부피당 세포의 절대수를 계산하는 단계들로 구성된, 비용균된 전체 혈액샘플내에 있는 한 세포군의 절대수를 플로싸이토메터를 이용해 측정하는 방법.
- (a) 피코에리트린에 결합된 안티-CD4단일항체, 파라포름알데히드 및 기지 농도의 2㎛자기 형광성 미세입자를 포함하는 희석제가 들어있는 시험관에 샘플을 첨가하고, (b)두번째 형광채널에서 형광트리거는 최소한 99%의 모든 미세입자 및 DC4+임파구를 포함하도록 조정하고, (c)첫번째와 두번째 형광채널에서 형광게이트는 표지된 CD4+임파구와 결합한 피코에리트린 형광성과 미세입자의 자기 형광성사이에 구분이 되도록 조정하고, (d)(c)단계의 CD4+임파구 및 미세입자의 수를 측정하고, 그리고 (e)(d)단계의 미세입자당 CD4+임파구의 수를 계산한 후 미세입자의 농도를 곱하여 결과적으로 단위 부피당 CD4+임파구의 절대수를 계산하는 단계들로 구성된, 전체 혈액 샘플내의 CD4+임파구의 절대수를 플로싸이토메터에 의해 측정하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
