Claims (40)
- 적어도 하나의 펜던트 설프하이드릴 그룹을 함유하는 단백질 용액을 설프하이드릴 그룹과 단단하게 결합하는 방사선 핵종의 방사선 금속이온의 용액과 접촉시키고, 방사선라벨된 단백질의 생성된 용액을 회수하는 단계를 포함함을 특징으로 하는, 단백질의 직접 방사선라벨링 방법.
- 제1항에 있어서, 단백질 용액이 저분자량이 티올 화합물을 실질적으로 함유하지 않는 방법.
- 제1항에 있어서, 적어도 하나의 펜던트 설프하이드릴 그립을 함유하고, 또한 디설파이드 그룹을 환원시키고 설프하이드릴 그룹을 안정화시키기 위한 저분자량의 분자 또는 이온을 함유하는 단백질의 용액을 사이징(sizing) 컬럼에 통과시켜 저분자량의 분자 및 이온을 제거하고, 생성된 단백질 용액을 회수함으로써 단백질 용액을 제조하는 방법.
- 제1항에 있어서, 단백질이 항체 또는 항체 단편인 방법.
- 제4항에 있어서, 단백질이 좀더 고분자량의 항체 또는 항체 단편 전구체내의 디설파이드 결합을 환원시킴으로서 제조된 항체 단편인 방법.
- 제5항에 있어서, 항체 단편이 2가 F(ab)2또는 F(ab)2전구체 단편의 환원적 분해에 의해 제조된 1가 Fab-SH 또는 Fab`-SH단편인 방법.
- 제5항에 있어서, 항체 단편이 완전한 면역 글로불린의 환원적 분해에 의해 제조된 1가 Fab`c-SH 경/중쇄 단편인 방법.
- 제6항에 있어서, 방사선핵종이 Tc-99m인 방법.
- 제6항에 있어서, 방사선핵종이 Re-186인 방법.
- 제6항에 있어서, 방사선핵종이 Re-188, Cu-67, Hg-197, Pb-203 또는 Bi-212인 방법.
- 제8항에 있어서, 접촉반응을 환원된 과테크네튬염 이온에 대한 트랜스킬레이터의 존재하에 수행하는 방법.
- 제8항에 있어서, 방사선라벨된 생성 단백질을 방사선 금속이온과 단단히 결합하는 외인성 리간드와 접촉시키고, 안정화되고 방사선라벨된 생성 단백질을 회수하는 방법.
- 제1항에 있어서, 단백질 용액이 설프하이드릴 그룹을 환원시키거나 안정화시키기 위한 저분자량의 분자량이 분자 또는 이온을 함유하고, 언급된 용액을 설프하이드릴 그룹과 단단히 결합하는 방사선핵종의 방사선 금속이온의 용액이 적재된 분자 사이징 겔 컬럼에 적용시키며, 언급된 컬럼을 용출시킨 후, 저분자량의 분자 및 비결합된 방사선 금속이온이 실질적으로 함유되지 않은 생성된 용액을 회수하는 방법.
- 제13항에 있어서, 사이징 겔 컬럼이 추가로 방사선 금속이온에 대한 트랜스킬레이터를 함유하는 방법.
- 제13항에 있어서, 컬럼을 방사선 금속이온에 대한 트랜스킬레이터의 용액으로 용출시키는 방법.
- 제13항에 있어서, 단백질이 항체 또는 항체단편인 방법.
- 제16항에 있어서, 항체 또는 항체 단편이 부분적으로 환원된 완전한 면역글로블린, 부분적으로 환원된 F(ab`)2, 부분적으로 환원된 F(ab)2, Fab`-SH, Fab-SH 또는 Fabc`-SH인 방법.

- 제24항에 있어서, 단백질이 항체 또는 항체단편인 방사선라벨된 단백질.
- 제25항에 있어서, 라벨된 Fab`또는 Fab 단편인 방사선라벨된 항체 또는 단편.
- 제25항에 있어서, 방사선 금속이 Tc-99m인 방사선 라벨된 항체 또는 단편.
- 제26항에 있어서, 방사선 금속이 Tc-99m인 방사선 라벨된 항체 단편.
- 제1항의 방법에 의해 라벨된 방사선 금속-라벨된 단백질.

- 제36항에 있어서, Fab-SH 또는 Fab`-SH가 동결 건조된 키트.
