지방세포에 제조예와 비교예 각각을 50 ng/ml 농도로 처리하여 4일 동안 배양하였다. Fat cells were treated with each of Preparation Examples and Comparative Examples at a concentration of 50 ng/ml and cultured for 4 days.
마우스 유래 3T3-L1 전 지방세포는 100mm, 35mm culture dish 와 96 well multi plast 에 각각 seeding 한 후 growth medium (high glucose DMEM+ 1% P/S + 10% FBS)에서 100% confluence 될 때 까지 배양하였다. 이후, 2일 후 지방세포 분화 유도 물질 (1 ug/mL insulin, 1uM Dex, 0.5 mM IBMX)를 함유한 growth medium을 사용하여 전지방세포를 지방세포로 분화 유도하였다.Mouse-derived 3T3-L1 pre-adipocytes were seeded in 100mm and 35mm culture dishes and 96 well multi plast, respectively, and then cultured in growth medium (high glucose DMEM + 1% P/S + 10% FBS) until 100% confluence. Then, after 2 days, differentiation of preadipocytes into adipocytes was induced using growth medium containing adipocyte differentiation-inducing substances (1 ug/mL insulin, 1uM Dex, 0.5mM IBMX).
배양한 세포는 dPBS로 세척하여 tri-solution을 이용하여 3T3-L1 세포에서 총 RNA를 추출 및 분리하였다. 5 ug RNA를 TOP scriptTMRT DRY MIX를 이용하여 complementary DNA (cDNA)를 합성하였다. Ampi GeneTM qPCR Green Mix Hi-ROX와 primer가 포함된 혼합물 9 uL와 cDNA 1 ul를 polymerase chain reaction (PCR) 사이클을 30회 사용하였다. PCR 반응 조건은 95℃ 에서 10분, 95℃에서 30초, annealing 온도에서 30초, 72℃에서 30초 후 다시 95℃에서 30초로 돌아가는 사이클을 반복하고 72℃에서 5분을 수행하였다. 중합효소 반응에 사용된 primer 정보는 표 7에 나타내었다.Cultured cells were washed with dPBS, and total RNA was extracted and isolated from 3T3-L1 cells using tri-solution. Information on the primers used in the polymerase reaction is shown in Table 7.

배, 귤, 포도, 딸기 각각에 대하여 상기 제조예 및 비교예와 동일한 방법으로 추출물을 제조한 후, 실험예 1 내지 6과 동일한 실험을 수행하였다.Extracts were prepared from each of pears, tangerines, grapes, and strawberries in the same manner as in the Preparation Examples and Comparative Examples, and then the same experiments as in Experimental Examples 1 to 6 were performed.

제조예와 비교예에 의한 세포 독성을 확인하기 위해 MTT 분석법을 이용하여 3T3-L1세포에서 확인하였다. 2일 동안 다양한 농도(10ng/ml, 50ng/ml, 100ng/ml)에서 세포 독성 여부를 알아본 결과 모든 추출물에 대하여 독성이 나타나지 않았다.
