Abstract
티모신 베타 4의 융합 단백질을 포함하는 혈관 신생 촉진용 조성물에 관한 것으로, 일 양상에 따른 티모신 베타 4의 융합 단백질은 매우 효과적으로 세포막을 통과하여 혈관신생을 유도할 뿐만 아니라 세포 사멸을 억제할 수 있으며, 상처 치유를 원활히 유도할 수 있으므로 혈관 기타 손상 조직의 재생이나 허혈성 질환 또는 당뇨성 족부궤양 등의 치료제로서 유용하게 사용될 수 있다.
Description
티모신 베타 4의 융합 단백질을 포함하는 혈관 신생 촉진용 조성물에 관한 것이다.
티모신 β4(Thymosinβ4; Tβ4)는 혈관신생 (Angiogenesis) 뿐만 아니라, 세포의 이동, 분화, 성숙, 염증조절 등 치료 약물로서 긍정적인 효과가 다수 보고된 바 있다. 티모신 β4가 그 기능을 제대로 발휘하기 위해서는 세포 내부로 전달되어야 하나, 세포표면에는 특이적 수용체가 없으며, 그 크기나 생화학적 성질 등에 의해 세포막을 통과하기 어려우므로 실제 보고된 바와 같은 이론적 효과를 달성하기 어렵다는 문제점이 있다.
한편, 목적 단백질을 세포 내로 이동시키기 위한 운반체 펩티드로서 단백질수송도메인(protein transducing domain; PTD)으로 알려져 있는 TAT 펩티드가 개발된 바 있다. 그러나, 선행 연구 결과에 따르면, 세포도입이 용이하지 않은 단백질에 단백질수송도메인을 융합시킨다고 해서 융합된 모든 단백질이 세포 내로 도입된 후 원활한 활성을 나타낸다고 단정할 수는 없다. 즉, Tat 형질도입부위와 결합시킨 단백질이 세포 내로 도입은 되었으나 활성을 나타내지 않는다는 연구결과가 다수 보고된 사실이 있다 (Sengoku, T. et al. Experimental Neurology 188(2004) 161-170, Falnes P.O. et al. Biochemistry 2001 Apr 10;40(14):4349-4358, Daniele Peroni et. al., Neuroscience letters 421(2007) 110-114 등).
따라서, 티모신 β4를 세포 내로 성공적으로 운반하되, 티모신 β4의 본래의 활성을 유지 또는 보다 향상시킬 수 있는 재조합 단백질에 대한 필요성이 요구되는 실정이다.

상기 “티모신 베타 4 (Thymosinβ4; Tβ4)"는 흉선의 추출물로 처음 발견되었으며, 43개의 아미노산으로 구성되어 분자량이 5KDa으로 비교적 작은 단백질이며 적혈구를 제외한 거의 모든 세포에 존재한다. 상기 Tβ4는 본래 액틴을 조절하는 단백질로써 G-액틴에 결합하여 액틴의 중합체 합성을 억제하며 내피세포의 분화 및 이동, 신생 혈관 생성을 유도하는 것으로 알려져 있다. 본 발명에 있어서, Tβ4는 세포의 분열, 분화 및 이동기작을 조율하는 역할을 하며, 혈관 신생 유도, 세포 증식, 세포 이동 촉진, 세포 사멸 억제 등을 통해 손상된 조직의 재생 및 성장을 촉진하는 효능을 나타내는 한, 그의 아미노산 서열은 특별히 제한되지 않는데, 상기 Tβ4의 전체 아미노산 서열을 사용할 수도 있고, 이의 변이된 아미노산 서열을 사용할 수도 있으며, 이의 일부 단편을 사용할 수도 있다. 상기 Tβ4의 구체적인 아미노산 서열 또는 그를 코딩하는 유전자의 염기서열 정보는 NCBI의 GenBank 등 공지의 데이터베이스에서 얻을 수 있다. 상기 Tβ4는 구체적으로는 서열번호 2의 아미노산 서열로 표시되는 펩타이드일 수 있다.
용어 “융합 단백질”은 단백질 또는 펩타이드가 이종의(heterologous) 다른 단백질 또는 펩타이드에 결합되도록 인위적으로 합성된 단백질로서, 구체적으로 상기 세포 투과성 펩타이드 및 상기 Tβ4를 포함할 수 있다. 보다 구체적으로 상기 세포 투과성 펩타이드는 서열번호 1의 아미노산 서열로 구성된 펩타이드일 수 있으며, 상기 Tβ4는 서열번호 2로 구성된 펩타이드일 수 있다.
일 구체예에 있어서, 상기 세포 투과성 펩타이드는 티모신 베타 4의 N-말단에 결합된 것일 수 있다.
상기 융합 단백질은 이에 포함되는 각 도메인의 아미노산 서열과 하나 이상의 아미노산 잔기가 상이한 서열을 가지는 펩타이드를 포함할 수 있다. 분자의 활성을 전체적으로 변경시키지 않는 펩타이드에서의 아미노산 교환은 당해 분야에 공지되어 있다. 가장 통상적으로 일어나는 교환은 아미노산 잔기 Ala/Ser, Val/Ile, Asp/Glu, Thr/Ser, Ala/Gly, Ala/Thr, Ser/Asn, Ala/Val, Ser/Gly, Thy/Phe, Ala/Pro, Lys/Arg, Asp/Asn,Leu/Ile, Leu/Val, Ala/Glu, Asp/Gly 간의 교환이다. 또한, 아미노산 서열상의 변이 또는 수식에 의해서 단백질의 열, pH등에 대한 구조적 안정성이 증가하거나 단백질 활성이 증가한 단백질을 포함할 수 있다.
상기 융합 단백질 또는 상기 융합 단백질을 구성하는 단백질은 당해 분야에 공지된 화학적 단백질 합성방법으로 제조하거나, 상기 융합 단백질을 코딩하는 유전자를 PCR(polymerase chain reaction)에 의해 증폭하거나 공지된 방법으로 합성한 후 발현벡터에 클로닝(cloning)하여 발현시켜서 제조할 수 있다.
본 발명의 융합 단백질은 상기 세포 투과성 펩타이드 및 상기 Tβ4 사이에 링커 펩타이드를 포함할 수 있다. 구체적으로 상기 융합 단백질은 상기 세포 투과성 펩타이드가 상기 Tβ4의 N-말단에 공유결합을 통해 직접적으로 연결될 수도 있고, 링커(Linker)를 통해 간접적으로 연결될 수도 있다. 상기 링커는 구체적으로 글라이신, 알라닌, 루이신, 이소루이신, 프롤린, 세린, 트레오닌, 아스파라긴, 아스파르트산, 시스테인, 글루타민, 글루탐산, 리신, 아르기닌산 등의 아미노산을 사용하여 연결시킬 수 있고, 보다 구체적으로 발린, 루이신, 아스파르트산, 글라이신, 알라닌, 프롤린 등의 아미노산을 여러 개 사용하여 연결시킬 수 있으며, 보다 더 구체적으로 유전자 조작의 용이성을 고려하여 글라이신, 발린, 루이신, 아스파르트산 등의 아미노산을 1개 내지 5개씩 연결하여 사용할 수 있다. 구체적으로, 본 발명의 융합 단밸질은 서열번호 3의 아미노산 서열로 구성된 펩타이드일 수 있다.
용어 “혈관신생(angiogenesis)"은 혈관이 새로 형성되는 과정, 즉, 새로운 혈관이 세포, 조직 또는 기관 내로 발생하는 것을 의미한다. 이러한 혈관신생 과정은 1) 혈관생성 성장인자(angiogenic growth factor)가 기존 혈관에 휴지상태로 존재하는 혈관내피세포에 존재하는 수용체를 활성화시키고, 2) 활성화된 혈관 내피세포가 주변의 기저막 및 세포외기질을 분해하는 단백질분해효소를 분비하기 시작하여, 3) 혈관내피세포가 기존 혈관벽으로부터 벗어나서 혈관생성인자(angiogenic factor)를 분비하는 조직을 향해 이동하고 증식함으로써, 이루어질 수 있다.
일 구체예에 있어서, 상기 혈관 신생 촉진용은 피부판 재생용, 인공 피부 이식용, 및 이식용 혈관 제조용으로 이루어진 군에서 선택된 하나 이상의 용도일 수 있다. 상기 "인공 피부"는 피막보호를 목적으로 체액의 누출을 방지하고 세균 침입을 저지하기 위해 사용되는 피복제를 의미하며, 건조 돈피 외에 콜라겐, 키틴 혹은 합성 고분자를 소재로 제조된 것일 수 있다.
다른 양상은 서열번호 1의 아미노산 서열을 갖는 세포 투과성 펩타이드가 티모신 베타 4 (Thymosinβ4; Tβ4) 단백질에 결합된 융합 단백질을 포함하는, 혈관 신생 의존성 질환의 예방 또는 치료용 약학적 조성물을 제공한다. 상기 약학적 조성물에 있어서, 세포 투과성 펩타이드, 티모신 베타4 단백질, 융합 단백질, 혈관 신생에 대해서는 전술한 바와 같다.
용어 “혈관 신생 의존성 질환”은 혈액 공급이 원활하게 이루어지지 않거나 혈관 신생이 제대로 이루어지지 않는 증상을 수반하는 질환, 즉, 혈관신생 부족으로 인한 질환을 의미할 수 있다. 일 양상에 따른 융합 단백질의 혈관 신생 촉진 효과에 의하여 예방 또는 치료될 수 있는 질환이라면 그 종류가 특별히 제한되지 않으나, 일 구체예에 있어서, 상기 혈관 신생 의존성 질환은 허혈성 질환, 상처, 화상, 건선, 만성궤양, 심혈관 출혈, 뇌출혈, 욕창, 당뇨, 망막증, 미숙아 망막증, 연령관련 황반변성증, 녹내장, 당뇨성 족부궤양, 및 폐고혈압으로 이루어진 군에서 선택된 어느 하나일 수 있다.
상기 허혈성 질환은 뇌허혈, 심장허혈, 당뇨병성 혈관심장질환, 협심증, 심근경색, 심부전, 심근비대증, 망막허혈, 허혈성 대장염, 허혈성 급성 신부전증, 허혈성 뇌졸중, 뇌혈관성 치매, 뇌외상 및 신생아 저산소증으로 구성되는 군으로부터 선택된 어느 하나일 수 있다.
용어 “상처”는 생체가 손상된 상태를 의미하며, 생체 내부 또는 외부 표면을 이루는 조직, 예를 들 피부, 근육, 신경조직, 뼈, 연조직, 내부기관 또는 혈관조직이 분단 또는 파괴된 병리학적 상태를 포괄한다. 상기 상처는 비-치유 외상성 상처, 방사선조사에 의한 조직의 파괴, 찰과상 (abrasion), 골괴저, 열상 (laceration), 결출상 (avulsion), 관통상 (penetrated wound), 총상 (gunshot wound), 창상, 만성창상, 절상, 동상, 타박상 (contusion or bruise), 피부궤양, 피부건조, 피부 각화증, 갈라짐, 터짐, 피부염, 피부 사상균증에 의한 통증, 수술상, 혈관질환 상처, 각막창상 등의 상처, 욕창, 와창, 당뇨성피부 미란과 같은 당뇨병 및 순환불량에 관련된 상태, 만성궤양, 성형수술 후 봉합부위, 척추상해성 상처, 부인과적 상처, 화학적 상처 및 여드름으로 이루어진 군에서 선택된 어느 하나일 수 있고, 개체의 어떠한 부분에 대한 손상이 포함될 수 있으나, 이에 제한되지 않는다.
용어 “조직 재생"은 조직 외부 및 내부 원인에 의한 손상에 대해 조직의 회복 과정을 의미한다. 상기 외부 원인에 의한 손상은 자외선, 외부 오염 물질, 창상, 외상 등을 들 수 있으며, 상기 내부 원인에 의한 손상은 스트레스 등일 수 있다
상기 "당뇨성 족부궤양"은 당뇨병의 가장 흔한 합병증으로 당뇨병 환자의 50% 이상이 발병한다. 당뇨병 환자의 혈관에 존재하는 고농도의 혈당에 의해 세포 내 일부 기능이 손상되면서 다리 부위의 미세혈관과 신경세포가 죽는 합병증을 의미한다.
상기 "당뇨 망막증"은 당뇨병으로 인한 모세혈관 벽의 병변(기저막의 비후, 벽세포의 감소, 내피 세포의 과도한 증식)으로 모세혈관이 좁아지고 결국 폐색되어 망막 미세혈관의 순환 장애가 나타나 발생하는 합병증을 의미한다.
일 양상에 따른 개량형 TAT-thymosin β4 융합 단백질은 매우 효과적으로 세포막을 통과할 수 있고, thymosin β4 본래의 혈관신생 활성을 상실하지 않을 뿐 아니라, 혈관 형성 효과가 크다고 알려진 VEGF와 유사하거나 보다 우수한 혈관 신생 효과가 있으면서, 세포 사멸을 억제 및 조직 재생을 유도할 수 있으므로 혈관 기타 손상 조직의 재생이나 허혈성 질환 등의 치료에 유용하게 사용될 수 있다.
용어 “예방”은 상기 약학적 조성물의 투여로 혈관 신생 의존성 질환을 억제 또는 지연시키는 모든 행위를 의미할 수 있다.
용어 "치료"는 상기 약학적 조성물의 투여로 질환의 증세가 호전되거나 이롭게 되는 모든 행위를 의미한다.
본 발명의 약제학적 조성물은 목적하는 투여 경로에 적합한 형태로 제형화된다. 투여 경로의 예는 비경구 투여, 이를 테면, 정맥내, 근육내, 피내, 피하, 경구, 비강내(이를 테면, 흡입), 경피(이를 테면, 국소), 점액막을 통해 그리고 직장을 통한 투여를 포함하나 이에 한정되지 않는다. 일 특정 구체예에서, 조성물은 정맥내, 피하, 근육내, 비강 또는 국소 투여에 적합한 약제학적 조성물을 형성하기 위하여 제형화된다. 보통, 정맥내 투여에 적합한 조성물은 멸균 등장 수성 완충액 중 용액이다. 필요에 따라, 조성물은 용해제 및 국소 마취제, 이를 테면, 에르고타민을 더 포함하여, 주사 부위에 통증을 줄일 수 있다.
본 발명의 조성물이 국소로 투여된다면, 조성물은 연고, 크림, 피부 패치, 로션, 젤, 샴푸, 스프레이제, 에어로졸, 용액, 에멀젼 또는 본 분야의 당업자에게 공지된 다른 형태로 제형화될 수 있다
스프레이제를 제외한 국소 투여 형태의 경우, 점착성 내지 반고체 또는 고체 형태가 보통 이용되며, 이는 국소적용에 적합한 담체 또는 하나 이상의 부형제를 포함하고, 물보다 더 큰 점성을 지닌다. 적합한 제형은 용액, 현탁액, 에멀젼, 크림, 가루, 광택제, 연고 등을 포함하나, 이에 한정되지 않고, 필요에 따라, 멸균하거나 삼투압과 같은 그의 특성을 변화시키기 위한 보조제(이를 테면 보존제, 안정화제, 습윤제, 완충액 또는 염)와 혼합된다. 다른 적합한 국소 투여 형태는 에어로졸을 포함하며, 여기서 활성 성분과 조합된 고체상 또는 액상 담체는 바람직하게 혼합물로, 혹은 휘발성 압력(이를 테면 압축 가스, 이를 테면 프레온)을 함유하는 스퀴즈 병으로 패키징된다. 필요에 따라, 가습제 또는 습윤제가 약제학적 조성물 및 투여 형태로 첨가될 수 있다. 이러한 부가적 성분의 예는 본 분야에 공지이다.

용어 "투여하는", "주입하는", "주사하는" 및 "이식하는"은 상호교환적으로 사용될 수 있으며, 일 구체예에 따른 조성물의 원하는 부위로의 적어도 부분적 국소화를 초래하는 방법 또는 경로에 의한 개체내로의 일 구체예에 따른 조성물의 배치를 의미할 수 있다. 일 구체예에 따른 조성물의 융합 단백질 또는 융합 단백질 성분의 적어도 일부를 생존하는 개체내에서 원하는 위치로 전달하는 임의의 적절한 경로에 의해 투여될 수 있다.

상기 약학적 조성물은 약학적 유효량으로 투여될 수 있으며, 약학적 유효량은 적용부위, 처리회수, 처리시간, 제형, 환자의 상태, 보조제의 종류 등에 따라 변할 수 있다. 상기 약학적 조성물의 투여량은 투여 경로, 환자의 연령, 성별, 체중, 환자의 중증도, 상처종류, 적용부위, 처리회수, 처리시간, 제형, 환자의 상태, 보조제의 종류 등의 여러 관련 인자에 비추어 결정되는 것이므로 상기 유효량은 어떠한 측면으로든 본 발명의 범위를 제한하는 것으로 이해되어서는 아니된다.
다른 양상은 서열번호 1의 아미노산 서열을 갖는 세포 투과성 펩타이드가 티모신 베타 4 (Thymosinβ4; Tβ4) 단백질에 결합된 융합 단백질을 이를 필요로 하는 개체에 투여하는 단계를 포함하는 혈관 신생 의존성 질환의 치료 방법을 제공한다. 상기 치료 방법에 있어서, 세포 투과성 펩타이드, 티모신 베타4 단백질, 융합 단백질, 혈관 신생, 혈관 신생 의존성 질환에 대해서는 전술한 바와 같다.
상기 개체는 인간 및 포유동물을 예로 들 수 있고, 구체적으로는 인간, 원숭이, 마우스, 래트, 토끼, 양, 소, 개, 말, 돼지 등을 포함할 수 있다.
일 양상에 따른 티모신 베타 4의 융합 단백질은 매우 효과적으로 세포막을 통과하여 혈관신생을 유도할 뿐만 아니라 세포 사멸을 억제할 수 있으며, 상처 치유를 원활히 유도할 수 있으므로 혈관 기타 손상 조직의 재생이나 허혈성 질환 또는 당뇨성 족부궤양 등을 비롯한 다양한 혈관 신생 의존성 질환의 치료제로서 유용하게 사용될 수 있다.
도 1은 개량형 TAT-thymosin β4의 세포내 도입 여부를 대조군인 thymosin β4와 비교 평가하기 위하여 thymosin β4 항체를 사용하여 면역형광염색을 수행한 결과이다.
도 2는 HUVEC 세포주에 개량형 TAT-thymosin β4를 다양한 농도로 처리한 후 HUVEC tube formation test를 수행하여 혈관형성능력을 평가한 결과로, 도 2a는 공초점 현미경으로 획득된 세포의 이미지를 그래픽 처리하여 튜브의 형성 정도를 확인한 결과; 도 2b는 도 2a의 결과를 정량화한 그래프이다.
도 3은 섬유아세포 세포주에 개량형 TAT-thymosin β4를 다양한 농도로 처리한 후 세포의 이동능력과 증식 정도를 현미경 관찰과 ImageJ software를 통하여 시간 경과에 따라 평가한 결과이다.
도 4는 피부와 유사한 구조를 갖도록 제작된 피부각질세포(keratinocyte), 섬유아세포 (fibroblast), 및 상피세포(epithelial cell)로 구성된 3중층 세포 모델과 양성대조군인 중간엽줄기세포에 cell punch를 이용하여 세포 손상을 유도한 후 개량형 TAT-thymosin β4를 다양한 농도로 처리하여 세포의 재생과 회복능을 평가한 결과로, 도 4a는 각 세포별 손상 영역의 감소 정도를 현미경으로 관찰한 결과; 도 4b는 도 4a의 결과를 정량화한 그래프이다.
도 5는 개량형 TAT-thymosin β4와 양성 대조군인 VEGF를 녹인 Matrigel을 마우스 주입한 후 형성된 matrigel plug 내의 hemoglobin 농도를 측정한 결과이다.

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1. 개량형 TAT-thymosin β4 융합 단백질의 제조
(1.1)
재조합 벡터 및 재조합 균주의 제작
하기 표 1에서와 같이 인간 유래 티모신 베타 4 (thymosin β; Tβ4) 단백질의 전체 서열의 일측에 세포투과성 펩티드(cell penetrating peptide) 서열이 결합된 융합 단백질을 코딩하는 폴리뉴클레오티드 서열을 포함하는 재조합 발현 벡터를 제조하였다.
구체적으로, 프라이머를 이용하여 PCR을 통해 세포투과성 펩티드와 티모신 베타 4가 융합된 형태의 DNA 절편을 획득하고, 이를 pBluescript Sk2+에 삽입하여 플라스미드 발현 벡터를 제조하였다. 상기 제조한 벡터를 E. coli (RBC Bioscience, New Taipei City, Taipei)에 형질전환하고, 해당 DNA가 잘 삽입되었는지 확인한 후 유전자 염기서열을 분석하였고, 발현된 융합단백질을 회수하여 아미노산 서열을 분석한 결과 아래 표의 서열번호 3과 같이 나타났다.

(1.2) 융합단백질의 발현 및 분리 정제
상기 제작된 발현벡터를 E.coli에 도입하여 수득된 형질전환체를 배양한 다음, 배양액을 여과하여 Sepharose FF 컬럼에 적용함으로써, 최종적으로 개량형 TAT-Tβ4 융합 단백질(서열번호 3)을 생산하였다.
구체적으로, 생산은 발효기를 통해 진행되었고, 융합단백질의 효율적인 발현을 위해 1mM IPTG를 처리하였다. 배양 종료 후 12g 의 균체(wet cell)를 60ml 라이시스 버퍼 (Lysis buffer, 30mM Sodium phosphate, 0.5% Triton X-100, 0.1M NaCl, 5mM EDTA, pH 7.0)에 현탁하였다. 현탁액에 소니케이터(SONOSMASHER from UL SSO HI-TECH, Korea)를 처리하여 균체를 파쇄한 후, 원심분리하여 (9,000rpm, 50 min., Mega21R from Hanil, KOREA), 수용성 세포추출액인 상등액을 회수하였다. 회수한 상등액을 'Sepharose Fast Flow resin (Merk-Millipore, Burlington, VT, USA)' 컬럼에 주입한 후, UV값이 평형화될 때가지 'A buffer'를 흘려주고, 0~60%의 'B buffer'를 사용하여 용출한 후, SDS-PAGE와 BCA assay로 분석하였다.

실험예 1. 개량형 TAT-thymosin β4의 세포내 도입여부 확인
실시예 1에서 제조된 개량형 TAT-thymosin β4의 세포 내 도입 여부를 확인하기 위하여 thymosin β4 항체를 사용하여 면역형광염색을 실시하여 thymosin β4가 세포질에 위치하는지를 확인하였다.
구체적으로, 인간혈관내피세포 (HUVEC, 1x104 cells/well) 를 24 well plate에 배양한 후 10 μg/mL의 농도로 개량형 TAT-thymosin β4와 thymosin β4를 1시간 동안 처리하였다. 배양액을 제거하고, PBS로 세척한 후 4% 파라포름알데히드(Biosesang, Gyeonggi-do, Korea)로 상온에서 20분 동안 고정하였다. 고정된 세포들을 PBS 내 0.1% Triton X-100 (Sigma)로 10분 동안 투과처리 하였다. 이후, 세포를 PBS로 2회 세척한 후, 차단 용액 (DAKO North America Inc., Carpinteria, CA, USA)으로 1시간 동안 상온에서 차단하였고, 일차 항체와 함께 2시간 동안 상온에서 인큐베이션하였다. 이후, 세포를 PBS로 2회 세척한 후, 형광 표지된 이차 항체와 1시간 동안 상온에서 인큐베이션하였다. 이 때 사용된 항체는 다음과 같다: Anti-thymosin β4 (ab14335, Abcam, Cambridge, MA, USA). 사용된 이차 항체는 다음과 같다: thymosin β4에 대한 Alexa Fluor 488-conjugated anti-rabbit IgG (1:200, Invitrogen, Carlsbad, CA, USA). 모든 처리 후 공초점 현미경(Carl Zeiss LSM 880, Oberkochen, Germany)을 사용하여 면역형광이미지를 얻었다.
도 1은 개량형 TAT-thymosin β4의 세포내 도입 여부를 대조군인 thymosin β4와 비교 평가하기 위하여 thymosin β4 항체를 사용하여 면역형광염색을 수행한 결과이다.
그 결과, 도 1에 나타난 바와 같이, 개량형 TAT-thymosin β4는 대조군인 thymosin β4에 비해 세포 내부로의 도입이 크게 증가한 것을 확인하였다.
실험예 2. 개량형 TAT-thymosin β4의 혈관 신생(angiogenesis) 및 조직 재생 효과의 확인
(2.1) 개량형 TAT-thymosin β4 처리에 따른 혈관내피전구세포의 튜브 형성능 (tube formation) 증가 확인
실시예 1에서 제조된 개량형 TAT-thymosin β4의 처리에 따른 혈관내피전구세포의 혈관 형성 정도를 확인하기 위하여, HUVEC 튜브 형성능 테스트 (HUVEC tube formation test)를 수행하였다.
구체적으로, 실험 수행 전 Matrigel을 20 ug/ml 농도로 24웰 플레이트에 분주하여 37℃ 인큐베이터에서 30분 coating을 한다. 이후에 24 웰 플레이트에 인간 혈관 내피세포인 Huvec (human umbilical vein endothelial cells) 세포주 7.5x104 개를 분주하고, 개량형 TAT-thymosin β4를 다양한 농도로 처리한 후, 이를 37℃ 인큐베이터에서 4시간 동안 배양한 후, calcein AM 염색을 수행하여 공초점 현미경을 이용하여 관찰하였다. 또한, 튜브 형성능은 획득된 세포의 이미지를 그래픽 처리하였을 때 나타나는 튜브의 형성 정도를 ImageJ software를 통하여 평가하였다. 양성 대조군으로 혈관형성효과가 크다고 알려진 VEGF를 사용하였다.
도 2는 HUVEC 세포주에 개량형 TAT-thymosin β4를 다양한 농도로 처리한 후 HUVEC tube formation test를 수행하여 혈관형성능력을 평가한 결과로, 도 2a는 공초점 현미경으로 획득된 세포의 이미지를 그래픽 처리하여 튜브의 형성 정도를 확인한 결과; 도 2b는 도 2a의 결과를 정량화한 그래프이다.
그 결과, 도 2a 및 도 2b에 나타난 바와 같이, 개량형 TAT-thymosin β4는 4 ug/ml 내지 13 ug/ml의 농도에서 우수한 튜브 형성능을 나타내었고, 특히, 10ug/ml의 농도로 처리하였을 때, 양성 대조군인 VEGF와 유사한 정도로 혈관내피전구세포의 튜브 형성능이 증가됨을 확인하였다. 이는 개량형 TAT-thymosin β4 처리가 혈관내피전구세포의 혈관 형성능을 촉진시킬 수 있어 혈관 신생을 유도할 수 있음을 의미한다.
(2.2) 개량형 TAT-thymosin β4 처리에 따른 세포의 이동 증가 및 생장 촉진 확인
실시예 1에서 제조된 개량형 TAT-thymosin β4가 섬유아세포의 이동 및 생장을 촉진시켜 세포의 재생 및 회복을 유도할 수 있는지 확인하기 위하여, 스크래치 상처 치유 분석 (Scratch wound healing assay)을 실시하여 세포 이동 능력을 평가하였다.
구체적으로, 세포를 접종하기 전 60 mm 세포 배양접시 표면에 Poly-L-lysine (50 ug/mL) 을 37℃ 인큐베이터에서 30분 coating을 한 후에 섬유아세포 (fibroblast cell) 를 60 mm 세포배양 접시에 4×105 cells/well의 농도로 24시간 배양하였다. 단층으로 세포가 증식된 것을 확인한 후, 200 ul tip으로 세포 중심을 긁어낸 후, 실시예 1에서 제조된 개량형 TAT-thymosin β4를 농도 별로 첨가하여 24시간 동안 배양하고, 손상 영역의 감소와 세포증식 정도를 관찰하였다. 개량형 TAT-thymosin β4를 첨가하지 않은 PBS 처리군을 대조군으로 사용하였다.
도 3은 섬유아세포 세포주에 개량형 TAT-thymosin β4를 다양한 농도로 처리한 후 세포의 이동능력과 증식 정도를 현미경 관찰과 ImageJ software를 통하여 시간 경과에 따라 평가한 결과이다.
그 결과, 도 3에 나타난 바와 같이, TAT-thymosin β4 처리군에서 대조군에 비해 세포 이동이 활발히 이루어 진 것을 확인하였고, 특히, 10ppm (10ug/ml)의 농도에서 24시간째에 세포 이동이 완료되는 것을 확인하였다. 이는 개량형 TAT-thymosin β4 처리가 섬유아세세포의 이동과 증식을 촉진시킬 수 있어 세포의 재생 및 회복을 유도할 수 있음을 의미한다.
(2.3) 개량형 TAT-thymosin β4 처리에 따른 피부세포의 재생 및 회복 확인
상처 치유/조직 재생을 위해서는 세포 이동이 중요한 역할을 하는 바, 실시예 1에서 제조된 개량형 TAT-thymosin β4의 처리에 따라 피부를 구성하는 세포들인 피부각질세포(keratinocyte), 섬유아세포 (fibroblast), 및 상피세포(epithelial cell)의 이동이 촉진되는지 확인하기 위하여, 피부와 유사한 3중층 세포 모델을 제작하여 개량형 TAT-thymosin β4을 처리하고 반경 상처 치유 분석 (radius wound healing assay)을 실시하여 세포의 재생과 회복능을 평가하였다.
구체적으로, 세포를 접종하기 전 60 mm 세포 배양접시와 transwell 표면에 Poly-L-lysine (50 ug/mL) 을 37℃ 인큐베이터에서 30분 coating을 하였다. 이후 가장 아래층이 되는 배양접시에는 피부상피세포를, 중간층의 transwell에는 섬유아세포를, 가장 위층의 transwell에는 피부각질세포를 각각 4×105 cells/well의 농도로 넣어, 24시간 배양하였다. 각 층의 세포가 단층으로 증식된 것을 확인한 후, cell punch를 이용하여 세포 중심부를 뚫어주고, 각 농도별로 개량형 TAT-thymosin β4를 첨가하여 24시간 동안 배양하였다. 각 세포별 (피부 층별) 손상 영역의 감소와 세포증식 정도를 현미경으로 관찰하고, ImageJ software를 통하여 분석하였다.
도 4는 피부와 유사한 구조를 갖도록 제작된 피부각질세포(keratinocyte), 섬유아세포 (fibroblast), 및 상피세포(epithelial cell)로 구성된 3중층 세포 모델과 양성대조군인 중간엽줄기세포에 cell punch를 이용하여 세포 손상을 유도한 후 개량형 TAT-thymosin β4를 다양한 농도로 처리하여 세포의 재생과 회복능을 평가한 결과로, 도 4a는 각 세포별 손상 영역의 감소 정도를 현미경으로 관찰한 결과; 도 4b는 도 4a의 결과를 정량화한 그래프이다.
그 결과, 도 4a 및 도 4b에 나타난 바와 같이, 피부를 구성하는 세포들인 피부각질세포(keratinocyte), 섬유아세포 (fibroblast), 및 상피세포 (epithelial cell) 모두에서 4 ug/ml 내지 20 ug/ml의 농도에서 증식이 활발히 이루어진 것을 확인하였고, 특히 10ug/ml의 농도에서 우수한 증식 능력을 나타냄을 확인하였다. 양성대조군인 중간엽줄기세포의 경우, 개량형 TAT-thymosin β4의 농도에 상관없이 우수한 증식 능력을 나타내었다. 이는 개량형 TAT-thymosin β4 처리가 피부조직과 유사한 구조에서 피부세포의 재생 및 회복을 촉진시킬 수 있어 상처 치유/조직 재생을 유도할 수 있음을 의미한다.
(2.4) 동물 모델에서 개량형 TAT-thymosin β4 처리에 따른 신생 혈관 형성 증가 확인
실시예 1에서 제조된 개량형 TAT-thymosin β4의 혈관신생 효과가 in vivo에서도 나타나는지 확인하기 위하여, 매트리겔 플러그 분석 (Matrigel Plug assay)을 실시하였다.
구체적으로, Matrigel (BD Biosciences, San Jose, CA, USA)과 60 U/mL heparin (Sigma-Aldrich, CA, USA)을 혼합한 후, TAT-thymosin β-4 (n=3)와 양성대조군인 VEGF (n=3)를 각각 혼합하여 matrigel 용액을 제조한 후, 상기 용액을 실험동물인 마우스 (6주령, BALB/c nude mouse, 오리엔트 바이오)의 등쪽 허벅지 부위에 피하주사로 주입한 후 2주 후에 형성된 matrigel plug를 꺼내어 혈관생성여부를 육안으로 관찰하였고, 신생혈관이 일어난 matrigel plug는 drabkin's reagent를 처리하여 hemoglobin 농도를 측정함으로써 혈관생성효과를 정량화하였다.
도 5는 개량형 TAT-thymosin β4와 양성 대조군인 VEGF를 녹인 Matrigel을 마우스 주입한 후 형성된 matrigel plug 내의 hemoglobin 농도를 측정한 결과이다.
그 결과, 도 5에 나타난 바와 같이, 양성대조군인 VEGF 투여군의 hemoglobin은 5.25±0.64 ug/mg로 측정되었으며, TAT-thymosin β4 투여군은 5.84±0.82 ug/mg로 hemoglobin이 측정되어, 개량형 TAT-thymosin β4는 양성 대조군인 VEGF보다 조직 내 혈관 형성이 더 활발히 일어남을 확인하였다.
(2.5) 혈관신생 작용 기전 확인
개량형 TAT-thymosin β4의 혈관신생 작용 기전을 확인하기 위하여 단백질 분석실험인 웨스턴 블랏을 수행하였다.
구체적으로, 개량형 TAT-thymosin β4를 농도별(0, 5, 10, 15, 20 ug/ml)로 HUVEC 세포에 24시간 처리한 후 방사면역침전 분석 완충액(radioimmunoprecipitation assay buffer; RIPA)으로 용해시켰다. 용해시킨 단백질은 BCA assay를 통하여 정량을 하고, 단백질 20 ug을 10 % SDS-PAGE 겔을 사용하여 분리하고 0.22-μm 나이트로셀룰로오스(nitrocellulose; NC) 막으로 옮겼다. 막은 5 % 탈지 우유 블로킹 완충액에서 실온에서 1 시간 동안 인큐베이션한 다음 4℃에서 1차 항체 (1:1000 dilution, 3% bovine serum albumin 용액)와 함께 밤새 인큐베이션하였다. 이 때 사용된 항체는 다음과 같다: Anti-F-actin (ab205, Abcam), Anti-VEGFA (sc-7269, Santacruz), Anti-VEGFR2 (SAB4501645, Sigma), Anti-phospho VEGFR2 (PS1013, Merk), Anti-Angiopoietin1 (AB3120, Millipore), Anti-Angiopoietin2 (AB3121, Millipore), Anti-phospho AKT (9271, CST), Anti-AKT (9272, CST), Anti phospho-ERK (9102, CST), Anti-ERK (9101, CST), Anti-CD31 (ab28364, Abcam), Anti-β-actin (A5441, Sigma). Tris-완충 식염수 및 Tween 20(Tris-buffered saline and Tween 20; TBST)으로 세척한 후, 2차 항체를 실온에서 1 시간 동안 막에 첨가하였다. 막은 웨스턴 블랏 ECL 용액(Supex)으로 전개하여 화학발광이미지를 획득하였고, 나타난 band를 ImageJ software를 통해 분석하였다. β-actin은 면역블로팅 대조군으로 사용되었다.
도 6은 HUVEC 세포주에 개량형 TAT-thymosin β4를 다양한 농도로 처리한 후 웨스턴 블랏을 수행하여 VEGFR2 신호전달 경로에 관여하는 단백질인 VEGFR2, p-VEGFR2, ang1, ang2의 상대적 양을 측정한 결과로, 도 6a는 band image; 도 6b는 band density를 측정 및 정량화한 그래프이다.
그 결과, 도 6a 및 도 6b에 나타난 바와 같이, HUVEC tube formation 실험에서 가장 혈관형성효과가 우수했던 10ug/ml의 농도에서 VEGFR2의 인산화가 가장 증가하였고, 혈관형성의 마커인 Ang1과 Ang2의 발현 역시 10ug/ml의 농도에서 가장 강력하였다. 이는 개량형 TAT-thymosin β4의 혈관형성작용이 VEGFR2 의 인산화를 통한 전통적인 VEGFR2 신호전달 경로를 통해 이루어지는 것을 의미한다.
도 7은 HUVEC 세포주에 개량형 TAT-thymosin β4를 VEGFR2 신호전달경로의 억제제(bevacizubam; B, sorafenib; S)와 함께 처리한 후 웨스턴 블랏을 수행하여 VEGFR2 신호전달 경로에 관여하는 단백질인 VEGFA, p-VEGFR2, t-VEGFR2, p-AKT, t-AKT, p-ERK, t-ERK, CD31의 양을 측정한 결과로, 도 7a는 세포 밖으로 분비된 단백질의 양을 확인한 band image; 도 7b는 분비되지 않은 세포 내 단백질의 양을 확인한 band image; 도 7c는 band density를 측정 및 정량화한 그래프이다.
그 결과, 도 7a 내지 도 7c에 나타난 바와 같이, 개량형 TAT-thymosin β4 처리시에 세포에서 세포 밖으로 분비하는 VEGFA의 양이 증가한 것을 확인하였다. 이 때 증가한 VEGFA의 분비량은 VEGF 억제제인 bevacizubam과 sorafenib에 의해 대조군 수준으로 다시 감소함을 확인하였다. 이는 분비되지 않은 세포내 VEGFA 농도에서도 마찬가지였으며, 도 6a 및 도 6b의 결과와 마찬가지로 인산화된 p-VEGFR2 역시 개량형 TAT-thymosin β4 처리군에서 많이 인산화되었고, VEGF 억제제 처리군에서 인산화가 줄어든 것이 확인되었다. 또한, 신호전달경로의 하부에 위치한 AKT와 ERK의 인산화 역시 개량형 TAT-thymosin β4에 의해 증가하였고, VEGF 억제제에 의해 감소함이 관찰되었다. 혈관분화의 마커인 CD31 역시 마찬가지의 양상을 보였다. 이러한 결과는 도 6a 및 도 6b에서 확인된 개량형 TAT-thymosin β4의 혈관형성작용이 VEGFR2의 신호전달경로를 통해 이루어진다는 점을 뒷받침한다.
도 8은 상기 단백질 분석 결과를 통해 도출한 개량형 TAT-thymosin β4에 의한 혈관형성 작용의 세포내 분자생물학적 신호전달경로를 나타낸 모식도이다.
도 8에 나타난 바와 같이, 개량형 TAT-thymosin β4 단백질은 세포 내로 도입되어 VEGFA를 세포 외부로 분비하고, 세포 외부의 VEGFA가 세포의 VEGFR2 신호전달경로를 촉진하여 ERK 및 AKT를 인산화를 촉진시켜 혈관형성효과를 촉진함을 알 수 있다.
실험예 3. 개량형 TAT-thymosin β4의 세포 사멸(apoptosis) 억제 효과의 확인
상기 실험예 2.5에서 개량형 TAT-thymosin β4에 의한 혈관형성작용이 VEGFR2 신호전달경로에 의하여 일어나는 것을 확인하였고, 이 과정에서 AKT의 인산화 역시 증가하는 것을 확인하였다. AKT의 인산화는 혈관형성작용에도 관여하지만 세포사멸 신호도 억제하는 것으로 알려져 있기 때문에, 세포 사멸에 관련된 단백질들을 추가로 분석하기 위해 웨스턴 블랏을 수행하였다.
웨스턴 블랏은 실험예 2.5에서와 동일한 방법으로 Apoptosis Western blot Cocktail 항체 (ab136812, Abcam)를 사용하여 수행하였다.
도 9는 HUVEC 세포주에 개량형 TAT-thymosin β4를 농도별로 처리한 후 웨스턴 블랏을 수행하여 VEGFR2 세포사멸 관련 단백질인 cleaved PARP, claeaved caspase3, Procaspase 3의 양을 측정한 결과로, 도 9a는 band image; 도 9b는 band density를 측정 및 정량화한 그래프이다.
그 결과, 도 9a 및 도 9b에 나타난 바와 같이, 개량형 TAT-thymosin β4 처리시 세포사멸신호 과정에서 증가하는 것으로 알려진 cleaved PARP와 cleaved caspase3가 모두 감소하는 것을 확인하였고, 특히, 10ug/ml 농도에서 세포사멸 억제 효과가 우수한 것을 확인하였다. 이는 본 발명의 융합 단백질이 혈관 세포의 사멸을 억제할 수 있는 바, 창상 또는 질환에 의한 혈관 손상이나 괴사로부터 혈관내피 세포를 보호하여 세포의 증식 및 회복을 촉진시킴으로써 조직 재생을 원활히 유도할 수 있음을 의미한다.
실험예 4. Thymosin β4 (TMSB4X)와 VEGF (VEGFA)의 String database 분석
Thymosin β4 (TMSB4X)의 혈관신생 작용 기전을 확인하기 위하여 단백질-단백질 간의 상호작용을 분석할 수 있는 string database (https://string-db.org/)를 이용하여 VEGF (VEGFA)와의 연관성, 상호관계를 분석하였다.
도 10은 단백질-단백질 간의 상호작용을 분석할 수 있는 string database를 이용하여 thymosin β4 (TMSB4X)와 VEGF (VEGFA) 간의 혈관신생관련 기전을 분석하여 그림으로 나타내고 연관성을 중간 신뢰도 지수 (Medium confidence score)를 기반으로 분석한 결과로, 도 10a는 중간 신뢰도 지수 0.400 이상 20 이하의 연관성을 그림으로 표현한 결과; 도 10b는 그 중에서 높은 지수를 추려서 연관성이 높은 단백질을 추려서 그린 결과이다.
그 결과, 도 10a 및 도 10b에 나타난 바와 같이, thymosin β4는 VEGFR2 신호전달경로를 통해 VEGF의 혈관신생신호를 증가시켜 혈관신생을 촉진시키는 작용을 하는 것으로 확인되었다. 이는 thymosin β4가 VEGFR2 신호전달경로를 촉진시킬 수 있는 물질임과 동시에 EGF, VEGF보다 상위 신호전달체계를 통해 더 효율적인 혈관신생을 할 수 있는 물질임을 의미한다.

- 서열번호 1의 아미노산 서열을 갖는 세포 투과성 펩타이드가 티모신 베타 4 (Thymosinβ4; Tβ4) 단백질에 결합된 융합 단백질을 포함하는, 혈관 신생 촉진용 조성물.
- 청구항 1에 있어서, 상기 세포 투과성 펩타이드는 티모신 베타 4의 N-말단에 결합된 것인, 혈관 신생 촉진용 조성물.
- 청구항 1에 있어서, 상기 혈관 신생 촉진용은 피부판 재생용, 인공 피부 이식용, 및 이식용 혈관 제조용으로 이루어진 군에서 선택된 하나 이상의 용도인 것인, 혈관 신생 촉진용 조성물.
- 서열번호 1의 아미노산 서열을 갖는 세포 투과성 펩타이드가 티모신 베타 4 (Thymosinβ4; Tβ4) 단백질에 결합된 융합 단백질을 포함하는, 혈관 신생 의존성 질환의 예방 또는 치료용 약학적 조성물.
- 청구항 4에 있어서, 상기 혈관 신생 의존성 질환은 허혈성 질환, 상처, 화상, 건선, 만성궤양, 심혈관 출혈, 뇌출혈, 욕창, 당뇨, 망막증, 미숙아 망막증, 연령관련 황반변성증, 녹내장, 당뇨성 족부궤양, 및 폐고혈압으로 이루어진 군에서 선택된 어느 하나인 것인, 혈관 신생 의존성 질환의 예방 또는 치료용 약학적 조성물.
- 청구항 5에 있어서, 상기 허혈성 질환은 뇌허혈, 심장허혈, 당뇨병성 혈관심장질환, 협심증, 심근경색, 심부전, 심근비대증, 망막허혈, 허혈성 대장염, 허혈성 급성 신부전증, 허혈성 뇌졸중, 뇌혈관성 치매, 뇌외상 및 신생아 저산소증으로 구성되는 군으로부터 선택된 어느 하나인 것인, 혈관 신생 의존성 질환의 예방 또는 치료용 약학적 조성물.
- 청구항 4에 있어서, 상기 세포 투과성 펩타이드는 티모신 베타 4의 N-말단에 결합된 것인, 혈관 신생 의존성 질환의 예방 또는 치료용 약학적 조성물.
