Abstract
본 발명은 MSH6 억제제를 유효성분으로 포함하는 암 예방 또는 치료용 조성물에 관한 것으로, 본 발명에 따른 MSH6 억제제는 간세포암종를 포함한 다양한 암종 세포에서 종양 세포 성장, 증식을 억제하였고, 특히 간세포암종에서는 이동 및 침습, 종양 형성 활성을 억제하고, MSH6의 발현을 증가시키는 경우 DNMT1의 발현을 증가시키고, KLF4의 발현을 감소시키는 것을 확인함으로써, 이를 암의 예방 및 치료용 약학적 조성물로 이용할 수 있다.
Description
본 발명은 MSH6 억제제를 유효성분으로 포함하는 암 예방 또는 치료용 조성물에 관한 것이다.
MSH6 (MutS homolog6, GTBP)는 DNA mismatch repair 단백질로 잘 알려져 있다. MSH6은 MSH2 (MutS homolog2) 와 hetero dimer를 이루게 되며, 이는 DNA의 복제 과정 중 mismatch 부분을 인식하여 교정을 시작하게 하는 인자의 역할을 하게 된다.
HDACs (Histone deacetylases)는 종종 보조억제자 (corepressors)나 다중-단백질 전사복합체 (multi-protein transcriptional complexes)들에 의해 유전자 프로모터에 붙을 수 있으며, 그곳에서 DNA에 직접 결합하지 않고 크로마틴 (chromatin) 변형을 통해 전사를 조절한다. 암호화된 사람 HDACs는 18개가 있으며, 이들은 클래스 I (HDAC 1, 2, 3 및 8), 클래스 II (HDAC 4, 5, 6, 7, 9 및 10), 클래스 III (SIRT 1-7), 및 클래스 IV (HDAC11) 효소들로 분류된다. 히스톤 아세틸화 효소 (acetyltransferases) 및 HDACs 모두 세포 증식, 분화 및 세포주기 조절에 관여한다는 사실이 알려져 있다. 또한, HDACs의 병리학적 활성 및 조절감소 (deregulation)가 암, 면역질환, 및 근이영양증 (muscular dystrophy)과 같은 여러 질병들을 야기할 수 있다는 사실이 보고되었다. HDAC6는 HDACs의 클래스 IIb 패밀리 멤버이고, 미세소관 (MTs)과 관련 있는 세포질내 탈아세틸화효소 (cytoplasmic deacetylase)로 작용하며, 알파-튜뷸린 (α-tubulin)을 탈아세틸화시킨다.
DNMT1 (DNA (cytosine-5)-methyltransferase 1)은 DNA의 특이적인 CpG 구조에 methyl기를 전달하는 효소이다. Cancer Genome Atlas 프로젝트에서 DNMT family는 암에서 확인된 127개의 돌연변이 된 유전자 중 하나이다. DNMT1은 다양한 암에서 과발현 되어있으며 이는 치료적 타겟이 될 수 있음이 연구된 바가 있다.
KLF4 (Kruppel-like factor 4)는 세포 증식, 분화, 사멸과 관련된 유전자의 전사 활성 또는 저해 두 방향으로 모두 작용할 수 있는 zinc finger 단백질이다. KLF4는 점막 상피세포의 세포분열 후에 세포의 성장 억제와 분화에 관여하여 주로 성장이 억제되는 세포의 핵에서 과발현되고, p53 의존성으로 cyclin-dependent kinase inhibitor 인 p21WAF1/Cip1을 활성화시켜 G1/S에서 세포주기를 억제한다.
본 발명의 목적은 MSH6 (MutS homolog6) 유전자 또는 단백질에 대한 억제제를 유효성분으로 포함하는 암 예방 또는 치료용 약학적 조성물을 제공하는 데 있다.
또한, 본 발명의 다른 목적은 MSH6 유전자, 이의 단백질 또는 상기 단백질을 구성하는 펩타이드 수준을 측정하는 제제를 포함하는 암 진단용 조성물을 제공하는 데 있다.
또한, 본 발명의 다른 목적은 암 예방 또는 치료용 물질 스크리닝 방법을 제공하는데 있다.
상기와 같은 목적을 달성하기 위해, 본 발명은 MSH6 (MutS homolog6) 유전자 또는 단백질에 대한 억제제를 유효성분으로 포함하는 암 예방 또는 치료용 약학적 조성물을 제공한다.
이어서, 본 발명은 MSH6 유전자, 이의 단백질 또는 상기 단백질을 구성하는 펩타이드 수준을 측정하는 제제를 포함하는 암 진단용 조성물을 제공한다.
마지막으로, 본 발명은 (a) MSH6 유전자 또는 이의 단백질을 포함하는 세포에 분석하고자 하는 시료를 접촉시키는 단계; (b) 상기 MSH6 유전자의 발현량, 단백질의 발현량 또는 단백질의 활성을 측정하는 단계; 및 (c) 상기 (b) 단계의 측정 결과, MSH6 유전자의 발현량, 단백질의 발현량 또는 단백질의 활성이 감소되는 경우에 상기 시료는 암의 예방 또는 치료용 물질로 판정하는 단계;를 포함하는 암 예방 또는 치료용 물질 스크리닝 방법을 제공한다.
본 발명에 따른 MSH6 억제제는 간세포암종를 포함한 다양한 암종 세포에서 종양 세포 성장, 증식을 억제하였고, 특히 간세포암종에서는 이동 및 침습, 종양 형성 활성을 억제하고, MSH6의 발현을 증가시키는 경우 DNMT1의 발현을 증가시키고, KLF4의 발현을 감소시키는 것을 확인함으로써, 이를 암의 예방 및 치료용 약학적 조성물로 이용할 수 있다.
도 1은 본 발명의 일 실시예에 있어서, TCGA의 33개 종류의 암종에서 MSH6의 발현이 높은 암종을 식별과정을 간단히 나타낸 도이다.

본 발명은 MSH6 (MutS homolog6)의 유전자 또는 단백질에 대한 억제제를 유효성분으로 포함하는 암 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 상기 MSH6 유전자는 인간 2번 염색체 상에 존재하는 것일 수 있고, 바람직하게는 인간 2번 염색체 47,783,145-47,806,954에 위치하는 것일 수 있다(Genome Reference Consortium: GRCh38.p13, NCBI Reference Sequence: NC_000002.12, Gene ID: 2956).
본 발명에서 사용되는 용어 “MSH6 (MutS homolog6)”는 DNA mismatch repair 단백질로, MSH2 (MutS homolog2) 와 hetero dimer를 이루게 되며 DNA의 과오염기쌍 수복에 관여하는 복합체를 형성하는 단백질 중 하나이다.
또한, 상기 억제제는 MSH6에 특이적인 siRNA, shRNA, miRNA, 안티센스 올리고뉴클레오타이드, 앱타머, 항체 및 이들의 조합으로 이루어진 군으로부터 선택되는 것일 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 MSH6에 특이적인 siRNA는 서열번호 1의 염기서열로 표시되는 것일 수 있으나, 이에 제한되는 것은 아니다
또한, 상기 MSH6의 발현 억제제는 DNMT1의 발현을 억제시키거나, KLF4의 발현을 증가시키는 것일 수 있다.
본 발명에서 사용되는 용어 “DNMT (DNA methyltransferase)”는 DNA에 메틸기(-CH3)를 전달하여 결합시키는 역할을 하는 효소이다. 비정상적인 DNA 과메틸화(hypermethylation)는 대부분의 암세포에서 일어나는 공통적인 현상으로 알려져 있으며, 따라서 DNMT의 활성을 저해하면 암 억제 유전자에 대한 과메틸화를 억제하여 암 억제 유전자가 정상적으로 재발현 되어 암세포의 증식을 억제할 수 있다.
본 발명에서 사용되는 용어 “DNMT1 (DNA (cytosine-5)-methyltransferase 1)”은 DNA 복제 후 메틸화 패턴을 유지하는 주요 효소로, genomic DNA의 시토신 뉴클레오티드에 메틸기를 전달하는 역할을 한다. 본 발명에서는 암세포에서 KLF4 프로모터의 메틸화를 유도하여 KLF4의 발현을 억제하는 역할을 한다.
일 구현예에서, DNMT1은 DNMT family 중 DNMT3A와 DNMT3B와 상호작용할 수 있다.
본 발명에서 사용되는 용어 “KLF4 (Kruppel-like factor 4”는 특정 전사인자와 상호작용하여 전사를 활성화 할 수 있고 종양억제제로 잘 알려져 있다.
일 구현예에서, KLF4는 MSH2, DNMT1과 상호작용 할 수 있다.
본 발명에서 사용되는 용어 “발현 억제"란 표적 유전자의 (mRNA로의) 발현 또는 (단백질로의) 번역 저하를 야기하는 것을 의미하며, 바람직하게는 이에 의해 표적 유전자 발현이 탐지 불가능해지거나 무의미한 수준으로 존재하게 되는 것을 의미한다.
또한, 상기 조성물은 종양 세포의 성장, 증식, 이동 및 침습을 억제하는 것일 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 암은 간암, 폐암, 두경부암, 결장암, 육종 (sarcoma), 위암 및 쓸개관암으로 이루어진 군으로부터 선택된 것일 수 있으며, 바람직하게는 간암, 더욱 바람직하게는 간세포암종 (hepatocellular carcinoma)일 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 암은 KLF6 (Kruppel-like factor 6), HDAC6 (Histone deacetylase 6) 또는 KLF4 (Kruppel-like factor 4)의 발현이 저해되거나, HDAC2 (Histone deacetylase 2), MSH6 (mutS homolog 6) 또는 DNMT1 (DNA (cytosine-5)-Methyltransferase 1)의 발현이 증가된 환자의 간암일 수 있고, 상기 간암은 간세포암종일 수 있고, 바람직하게는 stage Ⅰ, Ⅱ, Ⅲ, ⅣA 또는 ⅣB 병기 (phase)의 간세포암종일 수 있으며, 더욱 바람직하게는 초기 병기보다 치료가 어려운 stageⅢ 내지 Ⅳ 병기의 간세포암종일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 사용되는 용어 “암”은 세포 자체의 조절 기능에 문제가 생겨 정상적으로는 사멸해야 할 비정상 세포들이 과다 증식하여 주위 조직 및 장기에 침입하여 덩어리를 형성하고 기존의 구조를 파괴하거나 변형시키는 상태를 의미하며, 악성 종양과 동일한 의미로 사용된다.
본 발명에서, 사용된 용어 "예방"이란 본 발명에 따른 조성물의 투여에 의해 암의 발생, 발달 및 재발을 억제 또는 지연시키는 모든 행위를 의미한다.
본 발명에서 사용된 용어 "치료"란 본 발명에 따른 조성물의 투여로 암 및 이로 인한 합병증의 증세를 호전시키거나 이롭게 변경하는 모든 행위를 의미한다. 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자라면, 대한의학협회 등에서 제시된 자료를 참조하여 본원의 조성물이 효과가 있는 질환의 정확한 기준을 알고, 개선, 향상 및 치료된 정도를 판단할 수 있을 것이다.
본 발명의 조성물의 치료적으로 유효한 양은 여러 요소, 예를 들면 투여방법, 목적부위, 개체의 상태 등에 따라 달라질 수 있다.
본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에서 사용되는 용어 "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미하며, 유효용량 수준은 환자의 건강상태, 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와 순차적으로 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여, 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명에서 "개체"는 암을 예방 또는 치료를 목적으로 하는 개체이면 특별히 한정되지 않고, 인간을 포함하는 동물, 예를 들어 비-영장류 (예를 들면, 소, 돼지, 말, 고양이, 개, 래트 및 마우스) 및 영장류 (예를 들면, 원숭이, 예를 들어 사이노몰구스 (cynomolgous) 원숭이 및 침팬지)를 비롯한 포유동물을 나타낸다. 때에 따라서는 인간을 제외하는 개체일 수 있다.
본 발명에 따른 조성물은 약학적으로 유효한 양의 MSH6 억제제를 단독으로 포함하거나 하나 이상의 약학적으로 허용되는 담체, 부형제 또는 희석제를 포함할 수 있다. 상기에서 약학적으로 유효한 양이란 면역질환의 증상을 예방, 개선 및 치료하기에 충분한 양을 말한다. 상기에서 "약학적으로 허용되는"이란 생리학적으로 허용되고 인간에게 투여될 때, 통상적으로 위장 장애, 현기증과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 조성물을 말한다.
또한, 약학적으로 허용 가능한 담체를 포함하는 조성물은 경구 또는 비경구의 여러 가지 제형일 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 상기 담체, 부형제 및 희석제로는 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로오스, 메틸 셀룰로오스, 미정질 셀룰로오스, 폴리비닐 피롤리돈, 생리식염수, 메틸히드록시벤조에이트, 프로필히드록시 벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유, 덱스트린, 칼슘카보네이트, 프로필렌글리콜 및 리퀴드 파라핀으로 이루어진 군에서 선택된 하나 이상일 수 있으나, 이에 한정되는 것은 아니며, 통상의 담체, 부형제 또는 희석제 모두 사용 가능하다. 상기 성분들은 상기 유효성분인 MSH6 억제제에 독립적으로 또는 조합하여 추가될 수 있다.
또한, 경구 투여를 위한 고형제제에는 정제환제, 산제, 과립제, 캡슐제 등이 포함될 수 있으며, 이러한 고형제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제, 예를 들면, 전분, 탄산칼슘, 수크로오스 (sucrose) 또는 락토오스 (lactose), 젤라틴 등을 섞어 조제될 수 있다. 또한, 단순한 부형제 이외에 스테아린산 마그네슘, 탈크 등과 같은 윤활제들도 사용될 수 있다. 경구투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁용제, 유제, 동결건조제제, 좌제 등이 포함된다. 비수성용제, 현탁용제로는 프로필렌글리콜, 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔 (witepsol), 마크로골, 트윈 (tween) 61, 카카오지, 라우린지, 글리세롤, 젤라틴 등이 사용될 수 있다.
또한, 본 발명의 약학적 조성물은 정제, 환제, 산제, 과립제, 캡슐제, 현탁제, 내용액제, 유제, 시럽제, 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제 및 좌제로 이루어진 군으로부터 선택되는 어느 하나의 제형을 가질 수 있다. 좌제의 기제로는 위텝솔 (witepsol), 마크로골, 트윈 (tween) 61, 카카오지, 라우린지, 글리세롤, 젤라틴 등이 사용될 수 있다.
본 발명의 약학적 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여 (예를 들어, 정맥 내, 피하, 복강 내 또는 국소에 적용)할 수 있으며, 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설율 및 질환의 중증도 등에 따라 그 범위가 다양하다.
본 발명의 약학적 조성물은 암의 예방 또는 치료 효과를 위하여 단독으로, 또는 수술, 호르몬 치료, 약물 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
이어서, 본 발명은 MSH6 유전자, 이의 단백질 또는 상기 단백질을 구성하는 펩타이드 수준을 측정하는 제제를 포함하는 암 진단용 조성물을 제공한다.
또한, 상기 암은 간암, 폐암, 두경부암, 결장암, 육종 (sarcoma), 위암 및 쓸개관암으로 이루어진 군으로부터 선택된 것일 수 있으며, 바람직하게는 간암, 더욱 바람직하게는 간세포암종 (hepatocellular carcinoma)일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 사용되는 용어 "진단"은 특정 질병 또는 질환에 대한 한 객체의 감수성 (susceptibility)을 판정하는 것, 한 객체가 특정 질병 또는 질환을 현재 가지고 있는 지 여부를 판정하는 것, 특정 질병 또는 질환에 걸린 한 객체의 예후 (prognosis) (예컨대, 전-전이성 또는 전이성 암 상태의 동정, 암의 단계 결정 또는 치료에 대한 암의 반응성 결정)를 판정하는 것, 또는 테라메트릭스 (therametrics) (예컨대, 치료 효능에 대한 정보를 제공하기 위하여 객체의 상태를 모니터링 하는 것)을 포함한다.
또한, 상기 유전자 수준을 측정하는 제제는 유전자에 특이적으로 결합하는 프라이머 또는 프로브일 수 있으나, 이에 제한되는 것은 아니다.
또한, 상기 유전자 수준의 측정은 역전사 중합효소연쇄반응 (RT-PCR), 경쟁적 중합효소 연쇄반응, 실시간 중합효소 연쇄반응, Nuclease 보호 분석 (RNase, S1 nuclease assay), in situ 교잡법, DNA 마이크로어레이 이용법, 노던 블롯, 웨스턴 블롯, ELISA (Enzyme Linked Immuno Sorbent Assay), 방사선 면역분석법, 면역 확산법, 면역 전기영동, 조직 면역염색, 면역침전 분석법, 보체 고정분석법, FACS, 질량분석법 (Mass spectrometry) 및 단백질 마이크로어레이 이용법으로 이루어진 군에서 선택된 1종 이상의 방법으로 수행되는 것일 수 있으나, 이에 제한되지 않는다.
상기 MSH6 단백질은 인간 2번 염색체 47,783,145-47,806,954에 위치하는 핵산 서열로 번역되는 아미노산으로 이루어진 전체 또는 일부 펩타이드로 구성될 수 있으며, 상기 MSH6 단백질의 기능적 동등물 및 변이체를 포함한다. 또한, 상기 MSH6 단백질을 구성하는 펩타이드는 MSH6 단백질의 단편을 의미하는 것으로, MSH6 단백질을 구성하는 펩타이드라면 제한되지 않고 포함된다.
상기 단백질의 기능적 동등물이란, 단백질 분자의 활성을 전체적으로 변경시키지 않는 단백질 및 폴리펩티드를 포함하는 것으로 기능적으로 동일한 작용을 할 수 있는 단백질이 본 발명의 범위에 포함된다. 상기 변이체란, MSH6 천연 아미노산 서열과 하나 이상의 아미노산 잔기가 결실, 삽입, 비보전적 또는 보전적 치환 또는 이들의 조합에 의하여 상이한 서열을 가지는 단백질을 의미한다. 예를 들면 상기 아미노산 서열에서 하나 또는 수개의 아미노산이 결실, 치환 또는 부가된 것, 상기 아미노산 서열과 95 % 이상, 바람직하게는 98 % 이상, 보다 바람직하게는 99 % 이상의 동일성을 갖는 것 등을 말한다. 여기서 "동일성"이란, 2개의 아미노산 서열에 갭을 도입하거나, 도입하지 않고 가장 높은 일치도가 되도록 정렬시켰을 때에, 상기 갭의 수를 포함한, 한쪽 아미노산 서열의 전체 아미노산 잔기수에 대한 다른 쪽의 아미노산 서열의 동일한 아미노산 잔기수의 비율 (%)을 말한다. 또한, "수개"란, 2 내지 10의 정수, 예를 들면 2 내지 7, 2 내지 5, 2 내지 4, 2 내지 3의 정수를 말한다. 천연 변이체의 구체예로는 SNP (일염기 다형) 등 의 다형에 기초하는 변이체나 스플라이스 변이체 등을 들 수 있다. 상기 치환은 보존적 아미노산 치환인 것이 바람직하다. 보존적 아미노산 치환이면, 상기 아미노산 서열을 갖는 MSH6과 실질적으로 동등한 구조 또는 성질을 가질 수 있기 때문이다. 보존적 아미노산이란, 서로 비극성 아미노산 (글리신, 알라닌, 페닐알라닌, 발린, 류신, 이소류신, 메티오닌, 프롤린, 트립토판) 및 극성 아미노산 (비극성 아미노산 이외의 아미노산), 하전 아미노산 (산성 아미노산 (아스파라긴산, 글루탐산) 및 염기성 아미노산 (아르기닌, 히스티딘, 리신)) 및 비하전 아미노산 (하전 아미노산 이외의 아미노산), 방향족 아미노산 (페닐알라닌, 트립토판, 티로신), 분지상 아미노산 (류신, 이소류신, 발린) 및 지방족 아미노산 (글리신, 알라닌, 류신, 이소류신, 발린) 등이 알려져 있다.
또한, 아미노산 서열상의 변이 또는 수식에 의해서 단백질의 열, pH등에 대한 구조적 안정성이 증가하거나 단백질 활성이 증가한 단백질을 포함할 수 있다.
또한, 상기 단백질 또는 단백질을 구성하는 펩타이드 수준을 측정하는 제제는 상기 단백질 또는 단편 펩타이드에 특이적인 항체, 안티센스 RNA (antisense RNA), 앱타머 (Aptamer) 및 화합물로 이루어진 군에서 선택된 어느 하나일 수 있으나, 이에 제한되지 않는다.
본 발명에서 사용되는 용어 "항체"는 항원성 부위에 대해서 지시되는 특이적인 단백질 분자를 의미한다. 본 발명의 목적상, 항체는 MSH6 단백질에 대해 특이적으로 결합하는 항체를 의미하며, 다클론 항체, 단클론 항체 및 재조합 항체를 모두 포함한다.
또한, 상기 항체는 2개의 전체 길이의 경쇄 및 2개의 전체 길이의 중쇄를 가지는 완전한 형태뿐만 아니라, 항체 분자의 기능적인 단편을 포함한다. 항체 분자의 기능적인 단편이란 적어도 항원 결합 기능을 보유하고 있는 단편을 뜻하며, Fab, F (ab'), F (ab') 2 및 Fv 등이 있다.
또한, 상기 다클론 항체는 당업자에 알려진 방법에 따라 면역원으로 상기 유전자에 의해 발현된 단백질 또는 그 단편을 외부 숙주에 주사함으로써 제조할 수 있다. 외부 숙주는 마우스, 랫트, 양 또는 토끼와 같은 포유동물을 포함한다. 면역원은 근내, 복강내 또는 피하 주사방법으로 주사되며, 일반적으로 항원성을 증가시키기 위한 보조제 (adjuvant)와 함께 투여된다. 외부숙주로부터 정기적으로 혈청을 채취하여 향상된 역가 및 항원에 대한 특이성을 보이는 혈청을 수거하거나 이로부터 항체를 분리 정제한다.
또한, 상기 단일클론 항체는 당업자에 알려진 융합에 의한 불멸화된 세포주 생성기술에 의해 제조될 수 있다. 예를 들어, 상기 유전자에 의해 발현된 단백질을 마우스에 면역화 시키거나 펩타이드를 합성하여 소혈청 알부민과 결합시켜 마우스에 면역화 시킨다. 마우스에서 분리된 항원-생산 B 임파구를 인간 또는 마우스의 골수종 (myeloma)과 융합하여 불멸화된 하이브리도마 (hybridoma)를 생성하며, 상기 하이브리도마 세포를 가지고 간접적인 효소 결합면역흡착 분석법 (enzyme-linked immunoabsorbent assays, ELISA)을 사용하여 모노클노날 항체의 생성 여부를 확인하고 양성 클론을 택하여 배양한 후 항체를 분리정제하거나 랫트의 복강에 주입한 후 복수를 채취함으로써 단일클론 항체를 제조할 수 있다.
또한, 상기 단백질 수준의 측정은 웨스턴 블롯, 엘라이자 (enzyme linked immunosorbent assay, ELISA), 방사선면역분석 (Radioimmunoassay, RIA), 방사 면역확산법 (radioimmunodiffusion), 오우크테로니 (Ouchterlony) 면역 확산법, 로케트 (rocket) 면역전기영동, 조직면역 염색, 면역침전 분석법 (Immunoprecipitation Assay), 보체 고정 분석법 (Complement Fixation Assay), 유세포분석 (Fluorescence Activated Cell Sorter, FACS), 단백질 칩 (protein chip)으로 이루어진 군에서 선택된 1종 이상의 방법으로 수행되는 것일 수 있으나, 이에 제한되지 않는다.
나아가, 본 발명은 생물학적 시료에서 MSH6의 발현 수준을 확인하는 단계; 및 정상 시료와 MSH6의 발현 수준을 비교하는 단계;를 포함하는 간암의 진단 또는 예후 예측을 위한 정보 제공 방법을 제공한다.
또한, 상기 방법은 MSH6가 정상 시료에 비해 1.5배 이상 과발현되어 있는 경우, 간암에 걸릴 위험이 높은 것으로 판단하는 단계;를 추가로 포함할 수 있다.
또한, 상기 생물학적 시료는 조직, 세포, 전혈, 혈청, 혈장, 타액, 객담, 뇌척수액 또는 뇨와 같은 시료 등을 포함할 수 있다.
마지막으로, 본 발명은 (a) MSH6 유전자 또는 이의 단백질을 포함하는 세포에 분석하고자 하는 시료를 접촉시키는 단계; (b) 상기 MSH6 유전자의 발현량, 단백질의 발현량 또는 단백질의 활성을 측정하는 단계; 및 (c) 상기 (b) 단계의 측정 결과, MSH6 유전자의 발현량, 단백질의 발현량 또는 단백질의 활성이 감소되는 경우에 상기 시료는 암의 예방 또는 치료용 물질로 판정하는 단계;를 포함하는 암 예방 또는 치료용 물질 스크리닝 방법을 제공한다.
또한, 상기 암은 간암, 폐암, 두경부암, 결장암, 육종 (sarcoma), 위암 및 쓸개관암으로 이루어진 군에서 선택되는 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 사용되는 용어, “후보물질”은 MSH6 유전자의 발현 또는 MSH6 단백질의 활성에 영향을 미치는지 여부를 검사하기 위하여 스크리닝에서 이용되는 미지의 물질을 의미한다. 상기 후보물질은 이에 제한되는 것은 아니나, 화학물질, 펩타이드 및 천연 추출물을 포함할 수 있다. 본 발명의 스크리닝 방법에 의해 분석되는 후보물질은 단일 화합물 또는 화합물들의 혼합물일 수 있으며, 합성 또는 천연 화합물의 라이브러리로부터 얻을 수 있다.
또한, 상기 (b) 단계는 MSH6 유전자 발현량, MSH6 단백질 발현량, MSH6 단백질의 작용을 받은 다른 단백질, MSH6 단백질과 다른 단백질과의 상호작용, MSH6 단백질과 다른 단백질과의 복합체 및 MSH6 단백질의 작용을 받은 다른 단백질의 복합체로 이루어진 군에서 선택된 어느 하나 이상을 검출 또는 측정할 수 있으나, 이에 제한되지 않는다.
또한, 상기 MSH6 유전자 또는 단백질의 발현량 측정은 mRNA의 발현 수준 또는 유전자에 의해 코딩되는 단백질 수준을 확인함으로써 알 수 있다. mRNA의 양은 프라이머 쌍 또는 프로브를 이용하여 확인할 수 있으며, 이를 위한 분석 방법으로는 RT-PCR, 경쟁적 RT-PCR (Competitive RT-PCR), 실시간 RT-PCR (Real-time RT-PCR), RNase 보호 분석법 (RNase protection assay, RPA), 노던 블랏팅 (Northern blotting), DNA 칩 등이 있으나 이로 제한되는 것은 아니다. 상기 유전자에 의해 코딩되는 단백질의 양은 상기 단백질에 대하여 특이적으로 결합하는 항체를 이용하여 단백질의 양을 확인할 수 있으며, 이를 위한 분석 방법으로는 웨스턴블랏, ELISA (enzyme linked immunosorbent assay), 방사선면역분석 (RIA: Radioimmunoassay), 방사면역확산법 (radioimmunodiffusion), 오우크테로니 (Ouchterlony) 면역 확산법, 로케이트 (rocket) 면역전기영동, 조직면역염색, 면역침전 분석법 (Immunoprecipitation assay), 보체고정분석법 (Complement Fixation Assay), FACS, 단백질 칩 (protein chip) 등이 있으나, 이에 제한되는 것은 아니다.
또한, 상기 (c) 단계는 는 상기 측정 결과, 상기 후보물질 중에서 MSH6 단백질의 발현 또는 활성을 감소시키는 후보물질을 선택하는 단계로서, 상기 후보물질에 의하여 MSH6 유전자의 발현 또는 MSH6 단백질의 활성이 하향 조절 (downregulation) 되는 것을 측정하여 암의 예방 또는 치료용 물질로 판정할 수 있다.
본 발명의 스크리닝 방법은 다양한 방식으로 실시할 수 있으며, 특히 당업계에 공지된 다양한 결합 분석 (binding assay)에 따라 고성능 (high throughput) 방식으로 실시할 수 있다. 본 발명의 스크리닝 방법에 있어서, 후보물질 또는 MSH6 단백질은 검출가능한 표지 (detectable label)로 표지될 수 있다. 예를 들어, 상기 검출가능한 표지는, 화학적 표지 (예컨대, 바이오틴), 효소 표지 (예컨대, 호스래디쉬 퍼옥시다아제, 알칼라인 포스파타아제, 퍼옥시다아제, 루시퍼라아제, β-갈락토시다아제 및 β-글루코시다아제), 방사능 표지 (예컨대, C14, I125, P32 및 S35), 형광 표지 (예컨대, 쿠마린, 플루오레세인, FITC (fluoresein Isothiocyanate), 로다민 (rhodamine) 6G, 로다민 B, TAMRA (6-carboxy-tetramethylrhodamine), Cy-3, Cy-5, Texas Red, Alexa Fluor, DAPI (4,6-diamidino-2-phenylindole), HEX, TET, Dabsyl 및 FAM), 발광 표지, 화학발광 (chemiluminescent) 표지, FRET (fluorescence resonance energy transfer) 표지 또는 금속 표지 (예컨대, 금 및 은)이다. 검출가능한 표지가 표지된 MSH6 단백질 또는 후보물질을 이용하는 경우, MSH6 단백질과 후보물질 사이의 결합 발생 여부는 표지로부터 나오는 신호를 검출하여 분석할 수 있다. 예를 들어, 표지로서 알칼린 포스파타아제가 이용되는 경우에는, 브로모클로로인돌일 포스페이트 (BCIP), 니트로 블루 테트라졸리움 (NBT), 나프톨-AS-B1-포스페이트 (naphthol-AS-B1-phosphate) 및 ECF (enhanced chemifluorescence)와 같은 발색반응 기질을 이용하여 시그널을 검출한다. 표지로서 호스 래디쉬 퍼옥시다아제가 이용되는 경우에는 클로로나프톨, 아미노에틸카바졸, 디아미노 벤지딘, D-루시페린, 루시게닌 (비스-N-메틸아크리디늄 니트레이트), 레소루핀 벤질 에테르, 루미놀, 암플렉스 레드 시약 (10-아세틸-3,7-디하이드록시페녹사진), YR (p-phenylenediamine-HCl 및 pyrocatechol), TMB (tetramethylbenzidine), ABTS (2,2-Azine-di[3-ethylbenzthiazoline sulfonate]), o-페닐렌디아민 (OPD) 및 나프톨/파이로닌과 같은 기질을 이용하여 시그널을 검출한다. 택일적으로, 후보물질의 MSH6 단백질로의 결합 여부는 상호작용물 (interactants)의 표지 없이 분석할 수도 있다. 예를 들어, 마이크로피지오미터 (microphysiometer)를 이용하여 후보물질이 MSH6 단백질에 결합하는지 여부를 분석할 수 있다. 마이크로피지오미터는 LAPS (light-addressable potentiometric sensor)를 이용하여 세포가 그의 환경을 산성화하는 속도를 측정하는 분석 도구이다. 산성화 속도의 변화는, 후보물질과 MSH6 단백질 사이의 결합에 대한 지시자로 이용될 수 있다 (McConnell et al., Science 257:19061912 (1992)).
이하, 본 발명의 실시예를 첨부된 도면을 참고하여 보다 상세하게 설명하도록 한다. 그러나, 하기의 실시예는 본 발명의 내용을 구체화하기 위한 것일 뿐, 이에 의해 본 발명이 한정되는 것은 아닐 것이다.
실시예 1. 암 억제 유전자인 MSH6의 발현을 large cohort RNA seq data에서 확인

TCGA (The Cancer Genome Atlas)에서 얻을 수 있는 RNA sequencing 데이터에서 33개의 암종 중 MSH2, MSH6의 발현이 정상 환자 대비 암 환자에게서 1.5배 이상 증가한 암종 및 clinical relevance를 가지고 있는 암종을 확인하였다.
그 결과, MSH2의 경우 12개의 암종(LIHC, SARC, UCEC, CHOL, CESC, LUSC, STAD, ESCA, BLCA, LUAD, COAD, HNSC)에서 정상 환자 대비 암 환자에서 발현량이 1.5배 증가하였으며, 그 중 LIHC, SARC, UCEC는 overexpressed 되었을 때 survival에 영향을 주는 것으로 나타났다 (도 1a 및 표 1).
MSH6의 경우 10개의 암종(LIHC, LUAD, LUSC, CHOL, ESCA, HNSC, SARC, STAD, COAD, GBM)에서 정상 환자 대비 암 환자에서 발현량이 1.5배 증가하였으며, 그 중 LIHC, LUAD는 MSH6가 overexpressed 되었을 때 survival에 영향을 주는 것으로 나타났다 (도 1a 및 표 2).
또한, GENT2를 활용한 array data 분석 결과, MSH6의 발현이 14개의 암종에서 증가되어 있는 것을 확인하였다 (도 1b).

실시예 2. 다양한 암종에서 MSH6의 종양 형성에 대한 특성 확인
여러 암종에서 인비트로 종양형성 (in vitro tumorigenesis)을 확인하기 위해, si-MSH6 (서열번호 1)을 이용하여 MSH2 및 MSH6의 발현이 높은 암종 중 LIHC, LUAD, LUSC, CHOL, ESCA, HNSC, SARC, STAD, COAD에 대한 MTT assay, BrdU assay 및 cell viability assay를 수행하였다.
구체적으로, 다양한 암세포주에 si-MSH6의 형질전환을 위해 12-well plate에 40% confluency로 시딩(seeding)한 후 si-MSH6를 형질전환하여 MSH6를 knockdown하였다.
MTT assay를 위해, 세포를 12-웰 플레이트에 분주하고 si-MSH6을 트랜스펙션한 뒤, MTT [3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] 용액 (Sigma) 0.5 mg/ml과 5% CO2, 37℃ 에서 1시간 동안 인큐베이션한 뒤, VICTOR3 Multilabel 플레이트 리더기 (PerkinElmer, Waltham, MA)를 이용하여 흡광도를 측정하였다. 그 결과, MSH6를 knockdown 하였을 때 growth가 저해되는 것을 확인하였다 (도 2).
또한, BrdU assay를 위해 si-MSH6가 형질전환된 다양한 암세포주에 5-bromo-2’-deoxyuridine (BrdU) 시약을 6시간 동안 처리하였으며, 제조업체의 프로토콜에 따라 BrdU cell proliferation assay kit (Millipore)를 사용하여 수행하였다. 그 결과, MSH6를 knockdown 하였을 때 Proliferation이 저해되는 것을 확인하였다 (도 3).
또한, cell viability assay를 위해, si-MSH2 또는 si-MSH6가 형질전환된 다양한 세포들을 6-웰 플레이트에 분주하고 72시간 동안 배양한 뒤, 트립신으로 세포를 수득한 뒤 트립판 블루 용액 (Sigma, St. Louis, MO)으로 염색하였다. 염색한 뒤, 세포를 헤모사이토메터 (Marienfeld Superior, Lauda-Koonigshofen, Germany)로 계수하여 세포 생존을 확인하였다. 그 결과, MSH6를 knockdown 하였을 때 Cell number가 줄어드는 것을 확인하였다 (도 4). 여기서 DLD1 cell line의 경우 세포 수가 줄어들지 않았지만 MSH2 null cell이기 때문에 유의미하지 않다.
실시예 3. 간암에서 발현이 증가한 MSH6 검증
3-1. HCC에서의 MSH6 발현 확인
많은 암종 중 사망률 3위에 해당하는 간암에서 MSH6의 발현을 추가적으로 분석하기 위하여 TCGA, ICGC, GSE77314, Catholic_mLIHC(GSE114564) data를 분석하였다.
HCC(hepatocellular carcinoma)에서 서브유닛 유전자의 발현 수준을 확인하기 위해, TCGA_LIHC(The Cancer Genome Atlas liver hepatocellular carcinoma project), ICGC_LIRI(International Cancer Genome Consortium liver CancerRIKEN, JP) 및 NCBI의 GEO(Gene Expression Omnibus) 데이터베이스 (Accession Numbers: GSE77314 및 GSE114564)에서 데이터를 얻었다. TCGA datasets HTSeq-FPKM의 레벨 3 mRNA 발현 데이터는 [log2(fpkm+1)] 값으로 유전자 발현 수준을 평가하는데 사용하였다.
그 결과, MSH6의 발현이 Non tumor 대비 tumor에서 증가되었음을 확인하고, 그 중 TCGA, ICGC, GSE77314 data set의 Paired sample에서도 발현이 올라가 있는 것을 확인하였다(도 5A).
이어서, TCGA data set에서 survival 분석(캐퓰란-메이어 생존 분석)을 진행 하였을 때 MSH6의 발현이 높은 환자들의 경우 survival이 좋지 않은 것을 확인하였다(도 5B).
3-2. 40 matched-pair 분석
HCC에서 MSH6의 비정상적인 발현을 확인하기 위해, 40 비교-쌍 (matched-pair) (HCC 조직 vs 비 암성 주변 간 조직)의 HCC 조직과 13가지의 HCC 세포주 (MIHA, L-02, Hep3B, Huh7, PLC/PRF/5, SNU182, SNU354, SNU368, SNU387, SNU398, SNU423, SNU449, SNU475) 에서 총 RNA를 TRIzol 시약 (Invitrogen, Carlsbad, CA)을 이용하여 분리한 후, cDNA를 Tetro cDNA 합성 키트 (Bioline, London, UK)를 이용하여 합성하였다. 합성한 cDNA를 가지고 qRT-PCR을 진행하였다. qRT-PCR은 SensiFASTTM NoROX Kit (Bioline)로 수행되었다. 또한 같은 HCC 조직과 HCC 세포주에서 총 단백질을 추출하여 Western blot을 시행하여 MSH6의 발현도 확인하였다.
상기 40 비교-쌍의 HCC 조직 및 이에 상응하는 비 암성 주변 간 조직은 한국인체유래자원은행(National Biobank)에서 입수하였으며, 헬싱키 선언에 따라 각 피험자로부터 서면 동의를 얻고, 가톨릭대학교 의과대학 성의캠퍼스의 IRB (Institutional Review of Board)에서 승인을 받았다 (승인번호: MC19TESI0016). 그 결과, 40명의 HCC 환자들 중 24명 (60.0%)에게서 비-암성 간 조직에 비해 MSH6의 현저한 과발현이 나타나는 것을 qRT-PCR을 통해 확인하였다 (도 5C).
또한, 인체유래자원은행에서 분양 받은 간암 환자의 정상 간 조직, 간암 조직의 Protein을 추출하여 Western blot을 통해 MSH6 단백질의 발현을 확인하였을 때 Tumor 조직에서 발현이 증가되어있는 것을 확인하였다 (도 5G).
3-3. 간암세포주에서 MSH6 발현 확인
다양한 간암세포주에서 MSH6 발현 수준을 확인하기 위해, MSH6에 대한 qRT-PCR 및 Western blot을 수행하였다. 그 결과, 정상 간세포 (MIHA) 대비 MSH6 mRNA 및 단백질의 발현이 증가되어 있는 것을 확인하였다(도 5D, E).
추가적으로, MSH6 발현이 증가된 간암에서 MSH6 억제에 따른 효과를 확인하기 위해 MSH6의 발현이 높은 간암 세포주 (SNU182, SNU475)에서 MSH6를 knockdown 한 후, MTT assay와 BrdU assay를 수행하였다. 그 결과, MSH6가 증가된 간암세포주에서 MSH6를 억제하는 경우 Cell growth 및 Cell proliferation이 감소하는 것을 확인하였다 (도 5F).
실시예 4. MSH6의 암 발생 과정에 대한 특성 확인
4-1. 종양 세포 증식에 대한 효과 확인
간암에서의 MSH6의 종양 형성 특성을 확인하기 위하여, si-MSH6가 형질전환된 SNU182 및 SNU475에 대하여 Clonogenic assay를 수행하였다.
Clonogenic 분석은 음성 대조군 siRNA(si-Cont)와 si-MSH6 transfection 세포를 6-well plate(1000 cells/well)에 시드하였다. 12일 후, 콜로니를 상온에서 1% 파라포름알데히드로 30분간 고정하고, 상온에서 0.5% 크리스탈 바이올렛으로 1시간 동안 염색하였다. 염색된 콜로니는 상업용 소프트웨어(Clono 카운터)를 사용하여 계산되었다.
그 결과, MSH6의 발현이 증가되어 있는 간암 세포주 (SNU182, SNU475)에서 MSH6를 knockdown 했을 때 Colony formation이 줄어드는 것을 확인하였다(도 6A).
4-2. 종양 세포의 세포 사멸에 대한 효과 확인
si-MSH6 트랜스펙션에 의한 세포 사멸 및 세포주기 조절의 변화를 확인하였다. 세포 사멸은 Annexin V-FITC 아포토시스 검출 키트 I(BD Biosciences, San Jose, CA)를 사용하여 Apoptosis 수준을 측정하였다. 구체적으로, SNU182, SNU475세포들을 si-MSH6로 트랜스펙션하고 48시간 배양 후, 트립신-EDTA으로 세포를 분리하고, PBS로 세척하였다. 1 × binding buffer로 재부유하고 1X105 개의 세포를 함유하도록 100 μl를 5 ml 배양 튜브로 옮겼다. 그 후, 5 μl Annexin V-FITC 및 5 μl PI (propidium iodide) 용액을 첨가하고 세포들을 상온에서 암조건으로 15분 동안 인큐베이션하였다. 인큐베이션 후, 1x 바인딩 버퍼 400 μl를 각 튜브에 첨가하고 사멸 분획을 FACS CantoTMflow cytometer(BD Biosciences)로 검출하였다. 또한, 상기 si-MSH6가 형질전환된 SNU182, SNU475 세포에서 Apoptosis marker인 Caspase-3 및 Cleaved PARP의 발현 수준을 확인하였다.
그 결과, MSH6를 knockdown 했을 때 Apoptosis가 증가하는 것을 FACS analysis를 통해 확인하였으며(도 6B), 마찬가지로 단백질 발현 분석 결과 PARP 및 caspase-3는 감소하고 Cleaved PARP가 증가되는 것으로 나타났으며, 이를 통해 세포 사멸에 관여하는 PARP 및 caspase-3가 활성화되는 것을 확인하였다 (도 6C).
4-3. 종양 세포 이동 및 칩습에 대한 효과 확인
상피-간엽 전이 (Epithelial-tomesenchymal transition, EMT)가 세포 성장과 더불어 암 진행의 핵심 과정으로 제시되어 왔으므로, HCC 세포에서 MSH6를 억제하고 스크래치 상처 치유 분석을 수행하였다. 구체적으로, SNU182 및 SNU475 세포를 si-MSH6로 트랜스펙션한 뒤 24시간 동안 인큐베이션하고, 트립신-EDTA로 세포들을 떼어내어 6-웰 플레이트에 웰당 1X106개로 분주하였다. 다음날, 세포 단일층을 멸균된 마이크로피펫 팁으로 긁어냈다. 스크래치 직후 초기 스크래치의 간격과 24시간 후의 간격을 IX71 photomicrograph (Olympus, Tokyo, Japan)를 이용하여 촬영하였다.
그 결과, SNU182 및 SNU475 세포 모두에서 MSH6를 knockdown하였을 때 상처 치유 효과가 현저히 감소된 것을 알 수 있었다 (도 6D).
또한, 변형된 Boyden chamber assay (BD Biosciences, San Jose, CA) 및 transwell invasion assay를 통해 암세포의 이동 및 침습 특성을 확인하였다. transwell invasion assay는 transwell plate 및 세포 배양 인서트(BD Bioscience)를 사용하였다. 구체적으로, Matrigel(BD Biosciences)는 coating buffer(0.01 M Tris, 0.7% NaCl, pH 8.0)와 함께 0.3 mg/ml 농도로 희석되었으며, 100 μl Matrigel은 세포배양 인서트 상부에 코팅되었다. 37 ℃에서 1시간 동안 인큐베이션한 후, 인서트의 표면에 웰당 1.5×105 cell (motility assay) 또는 1.0×105 cell (invasion assay)을 serum-free medium와 함께 시딩(seeding)하고 lower transwell은 chemoattractant로서 2% FBS를 포함하도록 하였다. 플레이트를 37 ℃에서 12시간 동안 인큐베이션한 후, Diff-Quik staining kit (Sysmex, Kobe, Japan).를 이용하여 염색하였다. 세포 이미지는 Axiovert 200 도립 현미경 (Zeiss, Oberkochen, Germany)으로 X200 배율로 확인하였다.
그 결과, SNU182 및 SNU475 세포 모두에서 MSH6를 knockdown하였을 때Migration, Invasion이 현저히 감소하는 것으로 나타났다 (도 6E).
상기 결과들을 추가적으로 검증하기 위해 EMT marker에 대한 웨스턴 블롯을 수행하였으며, 그 결과 N-Cadherin, Fibronectin, Slug가 감소하는 것을 확인하였다 (도 6F).
4-4. 종양 세포의 세포주기 조절 변화 효과 확인
세포주기 조절의 변화를 확인하기 위해서, 세포들을 si-MSH6로 트랜스펙션하고 48시간 배양 후, 트립신-EDTA으로 세포를 분리하였다. 분리된 세포는 70% 에탄올에 고정된 PBS로 세척되었으며, 3 mg/ml RNase A, 50 μg/ml propidium iodide (PI, Sigma) 및 1% Triton X-100를 포함하는 200μl PBS로 재부유되었다. 세포는 37℃에서 암조건으로 45분 동안 인큐베이션하였다. 인큐베이션 후, 염색된 세포 분획은 FACS CantoTM flow cytometer (BD Biosciences, San Jose, CA, USA)를 이용하여 검출하였다.
그 결과, MSH6의 발현이 증가되어 있는 간암 세포주 (SNU182, SNU475)에서 MSH6를 knockdown 했을 때 G1/S arrest를 FACS analysis를 통해 확인하였다 (도 7A).
상기 결과들을 추가적으로 검증하기 위해 Cell cycle marker에 대한 웨스턴 블롯을 수행하였으며, 그 결과 p27의 증가와 CDK4, Cyclin D1, CDK2, Cyclin E의 감소를 확인하였다 (도 7B).
상기 결과들을 통해, MSH6의 비정상적 발현이 HCC의 항-사멸 및 전이 특성에 기여하는 것을 알 수 있었다.
실시예 5. MSH6의 종양유전자 특성 확인
MSH6의 oncogenic function을 확인하기 위해, MSH6의 발현이 감소되어 있는 정상 간 세포주 (MIHA)와 간암 세포주 (Huh7)에서 Transient transfection을 통해 MSH6를 Overexpression 한 후, MTT assay 및 BrdU assay를 수행하였다.
그 결과, MSH6가 감소된 정상 간 세포주 및 간암 세포주에서 MSH6를 과발현시키는 경우 Cell growth 및 Cell proliferation이 증가하는 것을 확인하였다 (도 8A).
이어서, MSH6의 발현이 감소되어 있는 정상 간 세포주 (MIHA)와 간암 세포주 (Huh7)에 MSH6를 지속적으로 과발현하는 Stable cell line을 제작하였다.
구체적으로, ‘Geneticin’ 약물에 내성이 있어 selection이 가능한 MSH2 플라스미드를 MSH2의 발현이 감소되어있는 정상 간 세포주 (MIHA)와 간암 세포주 (Huh7)에 transfection 하여 Stable cell line을 제작하였다. Transfection 후 Geneticin 약물을 처리하게 되면 플라스미드가 도입된 cell들만 살아남게 되어 selection 할 수 있고, 살아남은 cell들만을 계대 배양하여 플라스미드가 도입된 cell들만 배양할 수 있게 하였다. 이렇게 selection된 세포들은 지속적으로 MSH2를 발현되게 된다.
그 결과, 상기 결과와 마찬가지로 Cell growth 및 Cell proliferation이 증가하는 것을 확인하였다 (도 8B).
실시예 6. MSH6의 표적 분자 탐색
6-1. MSH6의 표적 분자 탐색
Lynch syndrome에서 MSH6와 hetero dimer를 이루는 MSH2는 Promoter 부분에 methylation이 되어있다고 알려져 있다. 이를 간암에서도 확인을 하기 위해 TCGA, ICGC data set에서 MSH6와 DNA methylation에 관여하는 DNMT family 와의 correlation을 확인하였다.
그 결과, TCGA_LIHC에서 MSH6과 DNMT1, DNMT3A, DNMT3B는 유의미한 양의 상관관계 (r=0.724, r=0.594, r=0.460, P<0.0001)을 가지고 있었고 이는 ICGC_LIRI_JP의 RNA seq 데이터에서도 동일한 결과 (r=0.586, r=0.479, r=0.337, P<0.0001)를 나타내어 MSH6는 DNMT family인 DNMT1, DNMA3A, DNMT3B와 강한 positive correlation을 가지고 있음을 확인할 수 있었다 (도 9A). 마찬가지로, MSH6를 knockdown 하였을 때에도 DNMT1, DNMT3A, DNMT3B의 단백질 발현이 줄어드는 것을 확인하였다 (도 9B).
하지만 MSH6의 promoter 부분의 methylation을 Methylation specific PCR을 통해 확인한 결과, HCC cell line (L-02, MIHA, SNU-182, HepG2, SNU387) 에서는 MSH6의 methylation이 확인되지 않았다 (도 9C).
따라서, 다른 기작으로 MSH6이 DNMT family의 발현을 조절한다고 가정하여 DNMT1, DNMT3A, DNMT3B의 promoter에 MSH6가 binding 하는지 확인하기 위해 ChIP assay (Pierce Agarose ChIP kit; Thermo Fisher Scientific, Waltham, MA)를 진행하였다. Cross-linked DNA는 IgG 컨트롤로 normalize하여 MSH6 항체를 이용하여 측정하였다. DNMT family의 promoter를 확인할 수 있는 프라이머를 제작하여 바인딩을 확인하였다. 그 결과 DNMT1의 프로모터 부분에만 MSH6가 바인딩 하는 것을 확인할 수 있었다 (도 9D).
6-2. MSH6 억제제의 DNMT 억제 활성 확인
MSH6과 DNMT1의 promoter 위치에 binding 하는 것을 확인하였기 때문에 MSH6이 Transcription factor로서 작용하여 DNMT1의 발현을 조절한다는 것을 유추할 수 있었다. 따라서 MSH6에 의해 DNMT1의 전사가 조절되는지 확인하기 위하여 Promoter luciferase assay를 진행하였다. SNU182, SNU475 세포주의 cDNA로부터 DNMT1의 프로모터를 PCR로 증폭한 뒤 pGL3_basic 벡터 (Promega, Madison, WI)의 Xho1/Bgl2 위치에 클로닝하여 리포터 벡터 pGL3_basic_DNMT1을 제작하였다. 그 후 pGL3_basic_DNMT1과 si-MSH6 (서열번호 1)을 트랜스펙션하여 루시퍼레이즈 리포터 분석을 진행하였으며 이의 결과를 음성 대조군인 대조군 siRNA 처리군 (si-N.C)의 결과와 비교하였다. 그 결과, SNU-182, SNU-475 세포주 모두에서 si-MSH6 처리로 인해 대조군에 비해 루시퍼레이즈 활성이 현저히 감소한 것을 확인할 수 있었다 (도 9E).
실시예 7. 간암에서 MSH6-DNMT1-KLF4 상관관계 확인
DNMT1이 KLF4의 promoter를 메틸화하여 발현을 억제한다고 알려져 있다. 따라서, 간암에서도 DNMT1과 KLF4의 상관관계를 확인하기 위해 MSH6를 과발현 또는 억제한 후, KLF4 methylation specific PCR을 진행하여 확인하였다.
그 결과, MSH6를 overexpression 했을 때 DNMT1이 증가하게 되면서 KLF4의 promoter methylation이 증가함을 확인하였으나 (도 10A), 반대로 MSH6를 knockdown 했을 때 MSH6가 감소하며 DNMT1의 발현이 감소하게 되면서 KLF4의 promoter methylation이 감소함을 확인하였다 (도 10B).
또한, MSH2 및 MSH6에 의한 DNMT1, KLF4의 발현 변화를 단백질 수준에서 확인하기 위해 MSH2, MSH6를 과발현 또는 억제한 후, MSH2, MSH6, DNMT1 및 KLF4에 대한 western blot을 수행하여 단백질 발현 수준을 평가하였다.
그 결과, 상기 도 10A, B와 마찬가지로 MSH6를 overexpression 하였을 때 DNMT1의 발현이 증가하게 되면서 KLF4의 promoter methylation이 증가하게 되고 그로인해 KLF4의 단백질 발현이 증가함을 확인하였다(도 10C). 반대로 MSH6를 knockdown 했을 때 MSH6가 감소하여 DNMT1의 발현이 감소하게 되면서 KLF4 promoter methylation이 감소하면서 KLF4의 단백질 발현이 증가하는 것을 확인하였다 (도 10D).
실시예 8. 간암에서 HDAC2/KLF6-HDAC6-MSH6 상관관계 확인
8-1. HDAC6 수준에 따른 MSH6의 발현 변화
간암에서 MSH2가 HDAC6에 의해 ubiquitination 되면서 발현이 감소하는지 확인하기 위해, TCGA, ICGC, GSE77314 data에서 HDAC6의 발현변화에 따른 MSH2와 서로 dimer 역할을 하는 MSH6의 발현을 확인하였다.
그 결과, HDAC6의 발현이 떨어진 sub-group (TGCA n=34, ICGC n=98, GSE77314 n=34)에서 MSH6의 발현은 tumor에서 모두 올라가 있음을 확인하였다 (도 11A).
또한, HCC 세포주(SNU182, SNU387, SNU475)에서 HDAC6를 과발현 또는 억제한 후, HDAC6 및 MSH6에 대한 western blot을 수행하여 단백질 발현 수준을 평가하였다.
그 결과, HDAC6를 과발현하는 경우 MSH6 단백질 발현이 감소하였으며, 반대로 HDAC6를 knockdown하는 경우 MSH6 단백질 발현이 증가하는 것을 확인하였다 (도 11B, C).
이러한 결과는 HDAC6와 MSH6는 음의 상관관계를 가지며, HDAC6 수준에 따라 MSH6의 발현이 조절된다는 것을 의미한다.
8-2. HDAC2/KLF6-HDAC6 상관관계 확인
HDAC6는 promoter 부분에 Gcbox를 가지고 있는데 Gcbox는 transcription factor가 binding 할 수 있는 부위이고 HDAC2, KLF6가 binding 할 수 있다고 알려져 있다. 따라서 HCC (hepatocellular carcinoma) 환자에서 HDAC2, HDAC6 및 KLF6 유전자의 발현 수준을 확인하여 HDAC2-HDAC6, KLF6-HDAC6, KLF6-HDAC2 correlation을 분석하였다.
그 결과, HDAC6와 HDAC2, KLF6와 HDAC2는 negative correlation, HDAC6와 KLF6는 positive correlation을 가지고 있음을 확인하였다 (도 12A).
HDAC2-HDAC6 및 KLF6-HDAC6 상관관계를 추가적으로 검증하기 위해 HDAC2의 발현이 적고 HDAC6, KLF6의 발현이 높은 정상 간세포주 (MIHA)와 HDAC2의 발현이 높고 HDAC6, KLF6의 발현이 낮은 간암세포주 (Hep3B)에서 HDAC2 또는 KLF6 발현을 조절하여 단백질 및 mRNA 발현 수준 변화를 확인하였다.
그 결과, HDAC2가 과발현되는 경우 HDAC6의 발현이 감소하고, HDAC2가 knockdown되는 경우 HDAC6의 발현이 증가하는 것으로 나타났다 (도 12B, D). 또한, KLF6가 과발현되는 경우 HDAC2의 발현이 감소하고 HDAC6의 발현이 증가하였으며, KLF6가 knckdown되는 경우 HDAC2의 발현이 증가하고 HDAC6의 발현이 감소하는 것으로 나타났다 (도 12C, E).
8-3. KLF6 또는 HDAC2 억제제의 HDAC6 억제 활성 확인
KLF6 또는 HDAC2에 의해 HDAC6의 전사가 조절되는지 확인하기 위하여 Promoter luciferase assay를 진행하였다. 도 12F는 HDAC6의 promoter luciferase assay를 위한 벡터 구조를 간단히 나타낸 것이다.
그 결과, 정상세포인 MIHA 세포주에서 KLF6를 knockdown하거나 HDAC2를 과발현시켰을 때 Promoter vector의 루시퍼레이즈 활성이 줄어드는 것을 확인하였고, 간암세포인 Hep3B 세포주에서 KLF6를 과발현시키거나 HDAC2를 knockdown하였을 때 Promoter vector의 luciferase activity가 증가하는 것을 확인하였다 (도 12G).
8-4. HDAC2 및 KLF6의 경쟁적 결합 확인
HDAC2와 KLF6가 HDAC6의 promoter에 경쟁적으로 binding 하는지 확인하기 위해 HDAC2의 발현이 낮고 HDAC6, KLF6의 발현이 높은 정상 간세포주 MIHA에서 HDAC2를 overexpression 하고 KLF6를 knockdown 한 후, HDAC2와 KLF6를 각각 Immunoprecipitation을 진행하여 HDAC2와 KLF6에 binding 되어있는 HDAC6 promoter의 양을 상대적으로 계산하였다.
그 결과, HDAC6의 promoter에 존재하는 세 개의 Gcbox region을 각각 봤을 때 공통적으로 HDAC2를 과발현하거나 KLF6를 knockdown 했을 때에는 HDAC2에 binding 하는 HDAC6 promoter의 양은 증가하였고, KLF6에 binding 하는 HDAC6 promoter의 양은 감소하는 것을 확인하였다 (도 13A).
이어서, HDAC2의 발현이 높고 HDAC6, KLF6의 발현이 낮은 간암세포주 Hep3B에서 KLF6를 overexpression 하고, HDAC2를 knockdown 한 후, HDAC2와 KLF6를 각각 Immunoprecipitation을 진행하여 HDAC2와 KLF6에 binding 되어있는 HDAC6 promoter의 양을 상대적으로 계산하였다.
그 결과, 상기 도 13A의 결과와 반대로 HDAC2를 knockdown 하거나 KLF6를 과발현했을 때에는 HDAC2에 binding 하는 HDAC6 promoter의 양은 감소하였고, KLF6에 binding 하는 HDAC6 promoter의 양은 증가하는 것을 확인하였다 (도 13B).
이러한 결과는 KLF6 및 HDAC2가 각각 발현을 조절하였을 때 binding 하는 HDAC6 promoter의 양이 변하는 것으로 보아 KLF6와 HDAC2가 HDAC6의 promoter에 경쟁적으로 결합한다는 것을 의미한다.
또한, 이전의 Promoter luciferase assay 결과를 통해 KLF6와 HDAC2가 HDAC6의 발현 조절을 확인하였기 때문에 KLF6 및 HDAC2가 경쟁적으로 HDAC6의 promoter에 binding 하여 HDAC6의 발현을 조절한다는 것을 의미한다.
실시예 9. HDAC6-MSH6-DNMT1-KLF4 신호전달 활성화의 암-예방 효과 확인
9-1. HDAC6-MSH6-DNMT1-KLF4 신호전달 활성화의 암-예방 효과의 인비보 확인
상기의 내용들을 인비보에서 확인하기 위해, H-ras 형질전환 마우스 (Ras-Tg 마우스) (Laboratory of Human Genomics, Korea Research Institute of Bioscience and Biotechnology, Daejeon, Korea)에 14주령일 때부터 매주 pcDNA3.1_HDAC6 50 ug (Turbofect in vivo Transfection Reagent (Invitrogen, Carlsbad, CA)로 정맥 주사), si-MSH2, si-MSH6 또는 si-DNMT1을 0.25 mg/kg (Invivofectamine 3.0 (Invitrogen)으로 정맥 주사)을 간 특이적으로 전달하였다. 그 후 16주, 18주, 20주 및 22주령에 ultrasound machine (Philips, Amsterdam, Nederland)로 초음파 영상을 찍고, 23주령차에 간을 수득하여 HCC를 확인하였다. 음성 대조군 (N.C) 마우스 간의 HCC는 16주령부터 초음파로 감지되었으며 5마리 중 5마리 모두 대형 및 다중 HCC로 발달되었다. 그러나, HDAC6를 과발현한 마우스에서는 20주령에 HCC가 검출되었고 (1마리), MSH6 억제군 (si-MSH6) 및 DNMT1 억제군 (si-DNMT1) 은 5마리 중 두 마리 내지 세 마리에서 작은 HCC가 발달되었다 (도 14A).
또한, 웨스턴 블롯 분석 결과, si-MSH6 군의 인접 정상 간에서 DNMT family 발현이 감소되고, KLF4 발현이 증가된 것을 확인하였다 (도 14B).
이를 Mouse Xenograft 모델에서도 동일하게 확인하였다 (도 15A). Xenograft 모델은 면역이 억제된 Nude mouse에 mouse HCC cell line (Hepa1-6)을 1 X 106 개씩 20% matrigel과 함께 옆구리에 피하주사하였다. Injection 후 2일 간격으로 확인하여 그 크기를 확인하였고 (도 15B) 2주 후 그 무게를 재어 확인하였다 (도 15C).
그 결과, si-MSH6 군은 대조군(N.C)과 비교하여 tumor growth가 현저히 줄어드는 것을 확인하였다.
이상에서 살펴본 바와 같이, 본 발명의 구체적인 실시예를 상세하게 설명되었으나, 본 발명의 사상을 이해하는 당업자는 동일한 사상의 범위 내에서 다른 구성요소를 추가, 변경, 삭제 등을 통하여, 퇴보적인 다른 발명이나 본 발명 사상의 범위 내에 포함되는 다른 실시예를 용이하게 제안할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다. 본 발명의 범위는 상술한 상세한 설명보다는 후술하는 특허청구의 범위에 의하여 나타내어지며, 특허청구의 범위의 의미 및 범위 그리고 그 균등 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.

Claims (11)
- MSH6 (MutS homolog6)의 유전자 또는 단백질에 대한 억제제를 유효성분으로 포함하는 암 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서,
상기 억제제는 MSH6에 특이적인 siRNA, shRNA, miRNA, 안티센스 올리고뉴클레오타이드, 앱타머, 항체 및 이들의 조합으로 이루어진 군으로부터 선택되는 것을 특징으로 하는, 암 예방 또는 치료용 약학적 조성물.
- 제2항에 있어서,
상기 MSH6에 특이적인 siRNA는 서열번호 1의 염기서열로 표시되는 것을 특징으로 하는, 암 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서,
상기 암은 간암, 폐암, 두경부암, 결장암, 육종 (sarcoma), 위암 및 쓸개관암으로 이루어진 군으로부터 선택된 것인, 암 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서,
상기 암은 KLF6 (Kruppel-like factor 6), HDAC6 (Histone deacetylase 6) 또는 KLF4 (Kruppel-like factor 4)의 발현이 저해되거나, HDAC2 (Histone deacetylase 2), MSH6 (mutS homolog 6) 또는 DNMT1 (DNA (cytosine-5)-Methyltransferase 1)의 발현이 증가된 환자의 암인 것을 특징으로 하는, 암 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서,
상기 MSH6의 발현 억제제는 DNMT1 또는 KLF4의 발현을 억제하는 것을 특징으로 하는, 암 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서,
상기 조성물은 종양 세포의 성장, 증식, 이동 및 침습을 억제하는 것을 특징으로 하는, 암 예방 또는 치료용 약학적 조성물.
- MSH6 유전자, 이의 단백질 또는 상기 단백질을 구성하는 펩타이드 수준을 측정하는 제제를 포함하는, 간암, 폐암, 두경부암, 결장암, 육종 (sarcoma), 위암 및 쓸개관암으로 이루어진 군으로부터 선택된 어느 하나의 암 진단용 조성물.
- 제8항에 있어서,
상기 유전자 수준을 측정하는 제제는 유전자에 특이적으로 결합하는 프라이머 또는 프로브를 포함하는 것인, 암 진단용 조성물.
- 제8항에 있어서,
상기 단백질 수준을 측정하는 제제는 상기 단백질 또는 단편 펩타이드에 특이적인 항체, 안티센스 RNA, 앱타머 및 화합물로 이루어진 군에서 선택된 어느 하나를 포함하는 것인, 암 진단용 조성물.
- (a) MSH6 유전자 또는 이의 단백질을 포함하는 세포에 분석하고자 하는 시료를 접촉시키는 단계;
(b) 상기 MSH6 유전자의 발현량, 단백질의 발현량 또는 단백질의 활성을 측정하는 단계; 및
(c) 상기 (b) 단계의 측정 결과, MSH6 유전자의 발현량, 단백질의 발현량 또는 단백질의 활성이 감소되는 경우에 상기 시료는 암의 예방 또는 치료용 물질로 판정하는 단계;를 포함하는,
간암, 폐암, 두경부암, 결장암, 육종 (sarcoma), 위암 및 쓸개관암으로 이루어진 군으로부터 선택된 어느 하나의 암 예방 또는 치료용 물질 스크리닝 방법.
