DPPH 자유 라디칼 소거 활성은 금화규 발효물 0.4㎖에 상기 DPPH 반응용액 4.6㎖을 넣어 잘 혼합한 후 암실에서 30분간 반응시켜 분광광도계(Hitachi U-2900, Japan)로 528 nm에서 흡광도를 측정한 후 아래 식에 대입하여 산출하였다. 이때, 합성 항산화제 0.05% BHT(Butylated hydroxytoluene)를 양성 대조군으로 이용하였다.The antioxidant activity of fermented Geumhwagyu was analyzed according to the method of Blois et al. 리폭시게나아제(lipoxygenase)는 정상적인 세포에서는 활성이 미비하지만, 염증이 유발되면 류코트리엔(leukotrienes) 물질의 증가로 인해 활성이 증가한다. 금화규 발효물 100㎕에 200mM 보레이트 완충액(borate buffer, pH 9.0) 1㎖을 넣은 후, 기질인 리놀렌산(linoleic acid, 0.6 mM/㎖ in ethanol) 2㎖을 추가하여 혼합한 후 234 nm에서 보정값을 측정하였다. 이후, 상기 금화규 발효물에 10,000unit/㎖의 리폭시게나아제 100㎕를 넣고 상온에서 10분간 반응시킨 후 234 nm에서 흡광도를 측정하였다. 양성 대조군으로는 0.1% 인도메타신(indomethacin, IDM)을 사용하였으며, 리폭시게나아제 억제 활성(%)은 다음과 같은 식에 의해 산출되었다. 본 발명의 금화규 발효물의 피부 주름개선 활성을 분석하기 위하여 엘라스타아제(elastase) 저해 활성을 측정하였다. 엘라스타아제에 특이적인 기질인 N-succinyl-Ala-Ala-Ala-p-nitroanilide 1,300㎕에 본 발명의 금화규 발효물 100㎕와 0.12M Tris-HCl(pH 8.0) 100㎕를 혼합하여 96웰 플레이트에 분주하고, 엘라스타아제를 20초 간격으로 100㎕씩 분주하여 실온에 10분 반응시킨 뒤, 20초 간격으로 410 nm에서 흡광도를 측정한 후 하기식에 대입하여 엘라스타아제 저해 활성을 산출하였다. 양성 대조군으로는 엘라스타아제 저해제로 알려진 0.1% 우르솔산(ursolic acid)을 사용하였다.In order to analyze the skin wrinkle improvement activity of the fermented Geumhwagyu of the present invention, the elastase inhibitory activity was measured.
