Abstract
본 발명은 세포 손상을 조기에 진단함에 따라 암 치료효과에 대한 조기 판정과 체내 독성을 신속하게 평가할 수 있어 양의 생체 내 진단과 치료를 위한 프로브의 후보물질로서 유용하게 사용될 수 있는 인간 피브로넥틴 도메인 Ⅲ 기본 골격의 신규 칼레티큘린 특이적 결합 단백질 및 그의 용도를 제공한다.
Description
본 발명은 신규 칼레티큘린 특이적 결합 단백질 및 그의 용도에 관한 것으로서, 더 상세하게는 인간 피브로넥틴 도메인 Ⅲ 기본 골격의 신규 칼레티큘린 특이적 결합 단백질 및 그의 용도에 관한 것이다.
분자영상기법인 핵의학(nuclear medicine) 영상은 각종 생화학적 물질의 생체 내 분포를 영상화하여 인체 내의 생리적 지표들을 정량적으로 측정할 수 있어 생화학 또는 병리현상의 규명과 질병 진단, 치료 후 예후 판정, 치료계획 등에 유용하게 이용되고 있다. 또한 그 생화학적, 물리적 원리상 핵의학 영상기기가 제공해주는 해부학적인 정보가 부족하고 해상도가 낮으므로 우리 몸의 구조를 높은 해상도를 가지고 보여주는 X선 CT 또는 MRI 등과 융합한 융합영상기기가 중요한 역할을 차지하고 있고 그러한 관점에서 핵의학영상진단기기 시장 규모는 앞으로 계속 성장할 것이다. 특히 PET/CT의 수요 증가가 시장 성장을 주도하고 있으며 이는 새로운 진단/치료 기술에 사용할 수 있는 방사성화합물의 개발 시 세계적인 고부가가치 산업으로 성장할 수 있음을 의미한다. 그러나 현재 임상에서 암의 치료효과 판정에 사용될 수 있는 방사성의약품은 2-deoxy-2-[18F]fluoro-D-glucose ([18F]FDG)밖에 없는 상황이나, 치료효과를 확인하기까지 수개월의 시간이 필요하며, 종양이 아닌 부위의 섭취(근육, 염증, 중추신경계)로 인해 그 사용이 굉장히 제약적이다. 따라서 수 일 내에 치료효과 및 독성을 평가할 수 있는 목적의 방사성화합물을 개발의 필요성이 대두되고 있다. 이와 관련하여 한국 공개특허 제2020-0001971호는 흑색종 치료용 방사성 화합물 및 그의 용도에 대해 개시하고 있다.
그러나 상기 선행기술의 경우, 흑색종 표적능이 향상된 치료용 방사성 화합물에 관한 것으로 암 치료효과 조기판정 및 체내 독성을 동시에 평가를 할 수 있는 물질에 대한 연구는 전무하다.
본 발명은 상기와 같은 문제점을 포함하여 여러 문제점들을 해결하기 위한 것으로서, 세포 손상을 조기에 진단함에 따라 암 치료효과에 대한 조기 판정과 체내 독성을 신속하게 평가할 수 있는 인간 피브로넥틴 도메인 Ⅲ 기본 골격의 신규 칼레티큘린(CRT) 특이적 결합 단백질 및 그의 용도를 제공하는 것을 목적으로 한다. 그러나 이러한 과제는 예시적인 것으로, 이에 의해 본 발명의 범위가 한정되는 것은 아니다.
본 발명의 일 관점에 따르면, 인간 피브로넥틴 도메인 Ⅲ(Fn3)의 BC 루프 및 FG 루프 중 적어도 어느 하나 이상에 칼레티큘린 결합 펩타이드가 삽입된, 칼레티큘린에 특이적으로 결합하는 재조합 모노바디가 제공된다.
본 발명의 다른 일 관점에 따르면, 상기 재조합 모노바디에 항암 화합물 또는 항암 단백질이 공유결합 또는 비공유결합에 의해 결합된 펩타이드 컨쥬게이트가 제공된다.
본 발명의 다른 일 관점에 따르면, 상기 재조합 모노바디 또는 상기 펩타이드 컨쥬게이트를 유효성분으로 함유하는, 암 치료용 약학적 조성물이 제공된다.
본 발명의 다른 일 관점에 따르면, 상기 재조합 모노바디에 조영용 화합물이 공유결합 또는 비공유결합에 의해 결합된, 펩타이드 컨쥬게이트가 제공된다.
본 발명의 다른 일 관점에 따르면, 상기 펩타이드 컨쥬게이트를 유효성분으로 함유하는, 암 진단 또는 조영용 조성물이 제공된다.
본 발명의 다른 일 관점에 따르면, 상기 암 진단 또는 조영용 조성물을 개체에 투여하는 단계; 및 상기 조성물에 의한 영상을 수득하는 단계를 포함하는 종양 진단방법이 제공된다. 이 때, 상기 개체는 인간이거나 인간을 제외한 포유동물일 수 있다.
본 발명의 다른 일 관점에 따르면, 상기 암 진단 또는 조영용 조성물을 암에 걸린 개체에 투여하는 단계; 상기 조성물에 의한 영상을 수득하는 단계; 및 상기 조성물에 의한 신호가 나타난 지역을 절제하는 단계를 포함하는 인간을 제외한 포유동물의 수술 중 암 조직 조영방법이 제공된다.
상기한 바와 같이 이루어진 본 발명의 인간 피브로넥틴 도메인 Ⅲ 기본 골격의 신규 칼레티큘린 특이적 결합 단백질은 암 치료반응의 조기 평가 및 독성 영향 평가에 매우 효과적이므로 종양의 생체 내 진단과 치료를 위한 프로브의 후보물질로서 유용하게 사용될 수 있다. 물론 이러한 효과에 의해 본 발명의 범위가 한정되는 것은 아니다.

본 문서에서 사용되는 용어 "피브로넥틴(fibronectin)"은 혈장 및 세포외 기질(extracellular matrix, ECM)에 존재하는 당단백질로, 두 개의 비대칭 베타-쉬트(anti-parallel beta-sheet) 구조의 Ⅰ, Ⅱ 및 Ⅲ 형의 도메인들이 다수 연결된 구조를 가진다. Ⅰ 및 Ⅱ 형 도메인은 구조 내 이황화 가교가 있으나 Ⅲ 형의 도메인(FnⅢ)은 없는 것이 특징이다.

아울러, 본 발명의 일 실시예에 따른 약학적 조성물은 0.1 mg/kg 내지 1 g/kg의 용량으로 투여될 수 있으며, 더 바람직하게는 0.1 mg/kg 내지 500 mg/kg의 투여량으로 투여된다. 한편, 상기 투여량은 환자의 나이, 성별 및 상태에 따라 적절히 조절될 수 있다.
본 발명자들은 Fn3 도메인의 각 루프 위치(BC, DE, FG)에 Int-α(서열번호 15), Hep-I(서열번호 16) 두 가지 CRT 펩타이드를 치환하여 재조합 CRT-monobody를 제조하였고 In vivo 이미징을 위해 CRT-3, CRT-4, FN3-DGR 유전자 C-말단에 Rluc8 유전자를 추가하여 제조하였다. 상기 재조합 CRT-monobody의 발현, 활성 및 결합력 등을 분석한 결과 칼레티큘린 결합력이 높고 생체 내 안정성도 우수한 인간 피브로넥틴 도메인 Ⅲ 기본 골격의 신규 칼레티큘린 특이적 결합 단백질을 개발하였다(도 1).
이하, 실시예를 통하여 본 발명을 더 상세히 설명한다. 그러나 본 발명은 이하에서 개시되는 실시예에 한정되는 것이 아니라 서로 다른 다양한 형태로 구현될 수 있는 것으로, 이하의 실시예는 본 발명의 개시가 완전하도록 하며, 통상의 지식을 가진 자에게 발명의 범주를 완전하게 알려주기 위해 제공되는 것이다.
실시예 1: Monobody 발현 컨스트럭트의 제조
본 발명자들은 본 발명의 모노바디(Monobody) 발현 컨스트럭트 제조하였다. 구체적으로 Fn3 도메인의 각 루프 위치에 칼레티큘린에 특이적으로 결합하는 펩타이드(Int-α, Hep-I)를 조합 후 결합시켜 칼레티큘린(Calreticulin) 특이적 결합하는 단백질을 제조하였다. 먼저, 하기 표 1과 같이 삽입된 유전자를 합성(마크로젠, 한국)하여 제한효소 Nhe1 부위를 포함하도록 제작된 정방향 프라이머(서열번호 17) 및 제한효소 BamH1 부위를 포함하도록 제작된 역방향 프라이머(서열번호 18)를 이용하여 유전자를 증폭하였다(도 2). 그 후, 상기 증폭 산물에 제한효소 Nhe1 및 BamH1을 넣어 절단하고 정제하였으며, 유전자 증폭 산물을 수득한 뒤에 이를 동일한 제한효소로 절단한 pETh 플라스미드(PLoS One. 2017 Jul 7;12(7):e0180786)에 도입하여 pETh_CRT1, pETh_CRT2, pETh_CRT3, pETh_CRT4, pETh_CRT5, pETh_CRT6 플라스미드를 제조하였다. 또한 대조군으로 인테그린(integrin) 결합을 방지하기 위해 야생형 FN3 스캐폴드의 FG 루프에 있는 RGD(arginine-glycine-aspartate) 서열을 스크램블(DGR)서열로 치환하여 pETh_DGR 플라스미드를 제조하였다(도 2). 이어서, In vivo 이미징을 위해 Rluc8 유전자의 정방향 프라이머(서열번호 19) 및 역방향 프라이머(서열번호 20)를 이용하여 유전자를 증폭 후 정제하여 유전자 증폭 산물을 얻은 뒤에 BamH1 제한효소로 절단한 pETh_CRT3, pETh_CRT4, pETh_DGR 플라스미드에 gibson assembly 방법으로 도입하여 pETh_CRT3-R, pETh_CRT4-R, pETh_DGR-R 플라스미드를 제조하였다(도 3). 상기 제조한 플라스미드를 E.coli BL21(DE3) 균주에 형질전환시킨 후, 50 ㎍/ml의 카나마이신(Kanamycin)이 포함된 LB 고체 배지를 이용하여 상기 각각의 형질전환 균주를 밤새 배양하였고 이후 생성된 콜로니를 카나마이신이 포함된 LB 액체 배지로 배양하여 실험에 사용하였다. 상기 제조한 CRT-monobody 구성 정보를 하기 표 1에, 프라이머 서열정보를 하기 표 2에 요약하였다.

실시예 2: 재조합 CRT-monobody의 단백질 발현 수준
본 발명자들은 상기 실시예 1에 따라 제조한 재조합 CRT-monobody(CRT1, CRT2, CRT3, CRT4, CRT5, CRT6) 및 Scrambled monobody(DGR)의 단백질 발현 수준을 조사하였다. 먼저, 상기 재조합 대장균 콜로니를 카나마이신이 포함된 LB 액체배지에 밤새 배양한 후 새로운 LB 배지를 사용하여 1:100의 비율로 희석하였다. 그 후, 추가 배양을 통해 OD600 값이 0.5 내지 0.7에 도달할 때, 상기 배양액에 최종 농도가 0.2 mM인 IPTG(Isopropyl β-D-1-thiogalactopyranoside)를 첨가하였고 200 rpm 및 37℃의 조건으로 진탕 배양기에서 배양하였다. 상기 배양된 재조합 대장균은 OD4 기준으로 SDS-PAGE 샘플 버퍼를 첨가하여 95ㅀ에서 10분간 끓인 후 SDS-PAGE에 로딩하여 단백질 발현정도를 확인하였다.
그 결과, IPTG에 의한 유전자 과발현이 유도됨을 확인하였으며 발현된 유전자는 가용성을 가짐을 알 수 있었다(도 4).
실시예 3: CRT-monobody 정제
본 발명자들은 상기 실시예 2의 방법으로 재조합 균주를 배양한 후, PBS로 세척하여 상층액을 제거한 뒤 용해 완충액(50 mM NaH2PO4, 300 mM NaCl, 10 mM imidazole, pH 8.0)로 재현탁하였다. 그 후, 소니케이터(Sonicator)를 이용하여 5 초/10 초(온/오프 정지) 10분 조건으로 상기 재현탁한 균주를 파쇄한 다음 파쇄된 균주를 원심분리(10,000 x g, 40 분, 4도)하여 상층액을 Ni-NTA 스핀 컬럼(Qiagen)에 로딩하였고 원심분리(890 x g, 1분)하여 CRT-monobody를 Ni-NTA에 결합 시켰다. 그 후, 비결합 단백질은 세척 버퍼(50 mM NaH2PO4, 300 mM NaCl, 20 mM imidazole, pH 8.0)를 컬럼(column)에 로딩하여 제거하였고 용리 완충액(50 mM NaH2PO4, 300 mM NaCl, 500 mM imidazole, pH 8.0) 300 μl를 컬럼에 로딩하여 히스택(his tag)-단백질을 용출시켰다. 상기 용출된 단백질은 투석막 키트(66380, Thermo Fisher Scientific)를 이용하여 이미다졸(imidazole)을 제거한 뒤 SDS-PAGE 샘플 버퍼를 첨가하여 95ㅀ에서 10분간 끓인 후 SDS-PAGE에 로딩하여 단백질 정제정도를 확인하였다.
그 결과, 정제한 모든 단백질이 95% 이상 순도로 정제되어 실험에 이용하였다(도 5).
실시예 4: CRT-monobody의 칼레티큘린 결합력 분석
본 발명자들은 상기 정제된 CRT-monobody의 칼레티큘린 결합력을 검증하기 위하여 암 세포에 CRT-monobody를 처리하여 결합된 정도를 공초점 현미경을 이용하여 관찰하였다. 구체적으로 암세포인 CT26(결장암) 세포를 챔버 슬라이드(154534PAK, Thermo Fisher Scientific)에 웰(well)당 1x105 세포를 분주하여 C02 인큐베이터에서 밤새 배양하였다. 그 후, 상기 각 세포에 칼레티큘린 표면 유도를 위해 항암제인 미토산트론(mitoxantrone, 25μM)를 처리하고 대조군으로 젬시타빈(gemcitabine, 15μM)를 처리하고 4시간동안 배양하였으며 PBS로 세척한 뒤 3%의 파라포름알데히드(paraformaldehyde)를 10분간 반응시켜 세포를 고정하였다. 그 후, 블록 완충액(PBS에서 3% BSA)을 1시간 동안 처리하여 난결합 항체(antibody)를 방지하였으며, 본 발명의 CRT-monobody(10 ㎍/ml)를 각 웰에 처리 후 1시간 반응시켜 칼레티큘린과 결합을 유도하였다. 이어서, CRT-monobody 확인을 위해 항-히스택 1차 항체(1:5000, PBS에서 3% BSA)를 처리하였고 칼레티큘린 확인을 위해 CRT 항체(1:500, PBS에서 3% BSA)를 처리하였으며 이미징을 위해 Alexa 488 2차 항체(1:500, PBS에서 3% BSA)를 처리하였다. 또한 세포 막(cell membrane)을 구분하기 위해 Alexa 555 conjugated WGA-555 (1:2000, 3% BSA in PBS)를 처리하였고 DAPI-Glod-antifade를 처리하여 마운팅하였으며 상기 제작된 슬라이드는 어두운 조건에서 냉장보관하였다.
그 결과, 항암제를 처리하지 않은 세포과 젬시타빈 처리한 세포는 세포막에 칼레티큘린 노출이 없음을 확인하였으며 CRT-monobody가 세포에 결합하지 않음을 확인하였다. 그러나 미토산트론을 처리한 세포는 세포막에 칼레티큘린이 노출되었으며 이 세포에 DGR 단백질은 세포표면에 결합하지 않았으나 CRT-monobody는 세포표면에 결합함을 확인할 수 있었다. 특히, CRT3, CRT4 단백질이 다른 CRT-monobody 보다 칼레티큘린이 세포막에 노출된 세포에 강하게 결함함을 알 수 있었다(도 6 내지 8). 이후 실험은, 결합력이 가장 우수한 CRT3와 CRT4을 선정하였으며 in vivo 실험을 위해 Rluc8 유전자를 융합하여 실험에 이용하였다.
실시예 5: 재조합 CRT-monobody-Rluc8의 단백질 발현 수준
본 발명자들은 상기 실시예 1에 따라 제조한 재조합 CRT-monobody-Rluc8 (CRT3-R, CRT4-R) 및 Scrambled monobody-Rluc8(DGR-R)의 단백질 발현 수준을 조사하였다. 실시예 2와 같이 배양한 재조합 대장균 배양액을 OD4 기준으로 SDS-PAGE 샘플 버퍼를 첨가하여 95ㅀ에서 10분간 끓인 후 SDS-PAGE에 로딩하여 단백질 발현정도를 확인하였다.
그 결과, IPTG에 의한 유전자 과발현이 유도됨을 확인하였으며 발현된 유전자는 가용성을 가짐을 알 수 있었다. 또한, CRT-monobody-Rluc8 단백질의 절단된 (Truncated) 사이즈가 없어 안정적으로 단백질 구조를 유지함을 확인하였다(도 9).
실시예 6: 리포터 단백질 활성 분석
본 발명자들은 리포터 단백질의 활성 분석을 위해 상기 실시예 2에서 배양된 균주(CRT3-R, CRT4-R, DGR-R) 1 ml을 PBS에 재현탁시켰다. 그런 다음, 상기 재현탁된 균주에 기질로 에탄올에 희석된 50 ㎕의 코엘렌테라진(Coelenterazine, 1 μg/ml)을 첨가한 후, NightOWL Ⅱ LB 983 In Vivo 이미징 시스템(Berthold technologies, GmbH & Co. KG, 독일)을 사용하여 1초 노출 시간의 조건으로 루시퍼라아제 활성 값을 이미징화하였다.
그 결과, 코엘렌테라진을 첨가하기 전에 발광을 확인 할 수 없었던 재조합 균주들은 기질 첨가 후 발광 시그널을 확인하였다. 이는 Rluc8이 융합된 monobody가 루시퍼라아제 활성을 나타냄을 시사하는 것이다(도 10).
실시예 7: 재조합 CRT-monobody-Rluc8의 칼레티큘린 결합력 분석
본 발명자들은 상기 실시예 3과 같이 CRT3-R, CRT4-R, DGR-R를 정제하여, CRT-monobody-Rluc8 단백질의 칼레티큘린 결합력을 검증하기 위하여 암세포에 CRT-monobody-Rluc8 단백질을 처리하여 결합된 정도를 공초점 현미경을 이용하여 관찰하였다. 구체적으로 암세포인 CT26(결장암), MC38(대장암), Hela(자궁암), MDA-MB-231(유방암) 세포를 챔버 슬라이드(154534PAK, Thermo Fisher Scientific)에 웰(well)당 1x105 세포를 분주하여 C02 인큐베이터에서 밤새 배양하였다. 그 후, 상기 각 세포에 칼레티큘린 표면 유도를 위해 항암제인 독소루비신(25 μM) 및 미토산트론(25 μM)를 처리하고 대조군으로 젬시타빈(15 μM)을 처리하였으며 이를 4시간동안 배양하였고 PBS로 세척한 뒤 3%의 파라포름알데히드(paraformaldehyde)를 10분간 반응시켜 세포를 고정하였다. 그 후, 블록 완충액(PBS에서 3% BSA)을 1시간 동안 처리하여 난결합 항체(antibody)를 방지하였으며, CRT-monobody(10 ㎍/ml)를 각 웰에 처리 후 1시간 반응시켜 칼레티큘린과 결합을 유도하였다. 이어서, CRT-monobody 확인을 위해 항-히스택 1차 항체(1:5000, PBS에서 3% BSA)를 처리하였고 칼레티큘린 확인을 위해 CRT 항체(1:500, PBS에서 3% BSA)를 처리하였으며 이미징을 위해 Alexa 488 2차 항체(1:500, PBS에서 3% BSA)를 처리하였다. 또한 세포 막(cell membrane)을 구분하기 위해 Alexa 555 conjugated WGA-555 (1:2000, 3% BSA in PBS)를 처리하였고 DAPI-Glod-antifade를 처리하여 마운팅하였으며 상기 제작된 슬라이드는 어두운 조건에서 냉장보관하였다.
그 결과, CT26(결장암), MC38(대장암), Hela(자궁암), MDA-MB-231(유방암) 세포에 항암제를 처리하지 않은 경우와 젬시타빈 처리한 경우는 세포막에 칼레티큘린 노출이 없음을 확인하였으며 CRT-monobody가 세포에 결합하지 않음을 확인하였다. 그러나 미토산트론과 독소루비신을 처리한 세포는 세포막에 칼레티큘린이 노출되었으며 상기 세포에 DGR-R 단백질은 세포표면에 결합하지 않았으나 CRT3-R과 CRT4-R 단백질은 세포표면에 결합함을 확인하였다(도 11 내지 22).
실시예 8: 유세포 분석(Flow Cytometry)
본 발명자들은 본 발명의 일 실시예에 따라 정제한 CRT-monobody-Rluc8의 칼레티큘린 결합력을 보다 정확하게 확인하기 위하여 암 세포와 CRT-monobody-Rluc8를 반응시킨 후 결합 정도를 세포수 단위로 측정하였다. 이를 위해 우선 CT26, MC38, Hela, MDA-MB-231 세포를 배양한 후 상기 실시예 5와 동일한 방법(젬시타빈, 독소루비신 및 미토산트론)으로 칼레티큘린 세포 표면발현을 유도하였다. 먼저, PBS로 세척한 뒤 부착된 세포를 수득하였고 3%의 파라포름알데히드를 10분간 반응시켜 세포를 고정하였으며 블록 완충액(PBS에서 3% BSA)를 1시간 동안 처리하여 난결합 항체를 방지하였다. 그 후, CRT-monobody(5 ㎍/ml)를 각 웰에 처리하여 1시간 동안 반응시켜 칼레티큘린과 결합을 유도하였다. 이어서, CRT-monobody 확인을 위해 항-히스택 1차 항체(1:5000, PBS에서 3% BSA)를 처리하였고 칼레티큘린 확인을 위해 CRT 항체(1:500, PBS에서 3% BSA)를 처리하였으며 Alexa 488 2차 항체(1:500, PBS에서 3% BSA)를 처리하였다. 마지막으로 CRT-monobody와 결합된 세포를 확인하기 위해 유세포 분석기기(BD Bioscience)를 이용하여 세포의 형광 시그널을 확인하였다.
그 결과, CT26(결장암), MC38(대장암), Hela(자궁암), MDA-MB-231(유방암) 세포에 항암제를 처리하지 않은 경우와 젬시타빈 처리한 경우는 세포막에 칼레티큘린 노출이 없음을 세포마다 형광 시그널로 확인하였으며 본 발명의 CRT-monobody 처리 후 시그널 변화가 없는 것으로 보아 세포에 결합하지 않음을 확인하였다. 그러나 미토산트론과 독소루비신을 처리한 세포는 세포막에 칼레티큘린이 노출되었으며 상기 세포에 DGR-R 단백질은 시그널 변화가 없어 세포 표면에 결합하지 않았으나 CRT3-R 및 CRT4-R 단백질은 Alexa 488 형광 시그널이 감지되어 세포표면에 결합하는 것으로 나타났다(도 23 내지 26).
실시예 9: 생체 내 안정성 확인
본 발명자들은 본 발명의 CRT-monobody-Rluc8 단백질이 체내에서 얼마나 안정성을 갖는지 확인하기 위해 마우스의 혈청에 상기 단백질을 시간 별로 처리 후 Rluc8 활성을 확인하였다. 구체적으로, 마우스의 혈액을 채취 후 냉장조건에서 상기 혈액을 응고시킨 다음 원심분리하여 혈청을 분리하였다. 그 후, 상기 마우스 혈청(150 ㎕)에 정제된 CRT-monobody-Rluc8(60 ㎍)(CRT3-R, CRT4-R) 및 DGR-R을 시간 별로 (0, 4, 12 및 24시간) 처리하였고 10 ㎕의 코엘렌테라진(40 ㎍/ml)을 처리하여 Rluc8 활성을 측정하였다. 상기 조건에서 끓인 혈청을 양성 대조군으로, 혈청을 음성 대조군으로 사용하였다.
그 결과, 혈청에 처리한 CRT3-R, CRT4-R 및 DGR-R은 4시간이 지나도 85% 이상 활성이 유지하였고 12시간이 지나도 Rluc8 활성이 50% 유지하는 것으로 나타났다. 상기 결과는 본 발명의 CRT-monobody-Rluc8 단백질이 생체 내에서 매우 안정적임을 시사하는 것이다(도 27 내지 29).
실시예 10: 칼레티큘린에 대한 특이성 확인
본 발명자들은 본 발명의 CRT-monobody-Rluc8가 세포 표면의 칼레티큘린(CRT)에만 특이적으로 결합하는지 확인하기 위해 경쟁적결합분석(Competitive binding analysis)을 수행하였다. 우선 CT26, MC38, Hela, MDA-MB-231 세포를 배양한 뒤 미토산트론(Mitoxantrone, 20 μM)을 처리하여 세포 표면에서 칼레티큘린 발현을 유도하였다. 그 후 약물처리된 암세포를 PBS로 세척하였고 부착된 세포를 수득한 후 3%의 파라포름알데히드를 10분간 반응시켜 상기 세포를 고정하였으며 블록 완충액(3% BSA in PBS)을 1시간 동안 처리하여 난결합 항체를 방지하였다.
그 후, CRT-monobody-Rluc8(100 ㎍/well)를 처리하였고 1시간 반응하여 칼레티큘린과 결합을 유도하였다. 또한 10 ㎍/well의 칼레티큘린 단백질(ab15729, abcam)을 약물처리된 암세포에 미리 처리한 후 CRT-monobody-Rluc8(100 ㎍/well)를 처리하여 1시간 동안 반응시켜 칼레티큘린과 결합을 유도하였다. CRT-monobody-Rluc8 확인을 위해 항-히스택 1차 항체(1:5000, PBS에서 3% BSA)를 처리하였고 Alexa 488 2차 항체(1:500, PBS에서 3% BSA)를 처리하였다. 마지막으로 CRT-monobody와 결합된 세포를 확인하기 위해 유세포 분석기기(BD Bioscience)를 이용하여 세포의 형광 시그널을 확인하였다.
그 결과, 세포막에 칼레티큘린 노출을 유도한 CT26(결장암), MC38(대장암), Hela(자궁암), MDA-MB-231(유방암) 세포에 CRT3-R과 CRT4-R 단백질이 강하게 결합하여 형광 시그널이 강하게 감지되었으나, 칼레티큘린 단백질을 함께 처리한 그룹은 형광 시그널이 약화됨을 확인하였다. 상기 결과는 CRT3-R과 CRT4-R 단백질이 재조합 칼레티큘린 단백질에 결합하여 세포막의 칼레티큘린에 결합하는 농도가 낮아짐에 따라 상기 CRT3-R과 CRT4-R 단백질이 칼레티큘린에 특이적으로 결합함을 시사하는 것이다(도 30 및 31).
실시예 11: 종양 동물 모델에서 칼레티큘린 특이성 확인
본 발명자들은 종양 마우스 모델에서 본 발명의 CRT-monobody-Rluc8의 칼레티큘린 특이성을 확인하였다. 구체적으로 약물 처리된 종양 동물 모델에서 칼레티큘린에 대한 CRT-monobody-Rluc8 결합을 확인하기 위해 CT26 암 세포를 Balb/c 마우스의 옆구리에 피하 주사하여 종양 동물 모델을 제조한 후 미토산트론(3 mg/kg)을 이용하여 종양 세포의 표면에 칼레티큘린을 유도하였으며, CRT-monobody-Rluc8(60 ㎍/100 ㎕)를 주입 후 6시간 뒤에 종양을 적출하고 50 ㎕의 코엘렌테라진(2 mg/ml)를 처리하여 종양 조직에 있는 루시퍼라아제 활성을 확인하였다.
그 결과, DGR-R 단백질을 처리한 종양조직은 Rluc8 발광 시그널이 거의 없는것에 비해 CRT3-R과 CRT4-R 단백질을 처리한 종양조직은 Rluc8 발광 시그널이 나타남을 확인하였다. 상기 결과는 본 발명의 CRT3-R과 CRT4-R 단백질에 의해 소동물 모델에서 약물처리된 종양세포를 이미징화 할 수 있음을 시사하는 것이다(도 32).
결론적으로, 본 발명의 인간 피브로넥틴 도메인 Ⅲ 기본 골격의 신규 칼레티큘린 특이적 결합 단백질은 칼레티큘린 결합력이 높고 생체 내 안정성도 우수하므로 암 치료반응의 조기 평가 및 독성에 대한 영향평가의 수행을 위한 프로브의 후보물질로서 유용하게 사용될 수 있다.
본 발명은 상술한 실시예를 참고로 설명되었으나 이는 예시적인 것에 불과하며, 당해 기술분야에서 통상의 지식을 가진 자라면 이로부터 다양한 변형 및 균등한 다른 실시예가 가능하다는 점을 이해할 것이다. 따라서 본 발명의 진정한 기술적 보호 범위는 첨부된 특허청구범위의 기술적 사상에 의하여 정해져야 할 것이다.

- 인간 피브로넥틴 도메인 Ⅲ(Fn3)의 BC 루프 및 FG 루프 중 적어도 어느 하나 이상에 칼레티큘린 결합 펩타이드가 삽입된, 칼레티큘린에 특이적으로 결합하는 재조합 모노바디.
- 제1항에 있어서,
상기 칼레티큘린 결합 펩타이드가 상기 BC 루프 및 FG 루프 모두에 삽입된, 재조합 모노바디. - 제1항 또는 제2항에 있어서,
상기 칼레티큘린 결합 펩타이드는 서열번호 15 또는 16으로 기재되는 아미노산 서열로 구성되는, 재조합 모노바디. - 제1항에 있어서,
서열번호 7 내지 12로 구성되는 군으로부터 선택되는 아미노산 서열로 구성되는, 재조합 모노바디. - 제1항 또는 제2항의 재조합 모노바디에 항암 화합물 또는 항암 단백질이 공유결합 또는 비공유결합에 의해 결합된 펩타이드 컨쥬게이트.
- 제5항에 있어서,
상기 항암 화합물은 독소루비신(doxorubicin), 사이클로포스파아마이드(cyclophosphamide), 메클로레타민(mecholrethamine), 우라무스틴(uramustine), 멜파란(melphalan), 클로라부실(chlorambucil), 이포스파미드(ifosfamide), 벤다무스틴(bendamustine), 카르무스틴(carmustine), 로무스틴(lomustine), 스트렙토조신(streptozocin), 부설판(busulfan), 다카바진(dacarbazine),테모졸로마이드(temozolomide), 티오테파(thiotepa), 알트레타민(altretamine), 듀오카르마이신(duocarmycin), 시스플라틴(cisplatin), 카르보플라틴(carboplatin), 네다플라틴(nedaplatin), 옥사리플라틴(oxaliplatin), 사트라플라틴(satraplatin), 트리플라틴 테트라나이트레이트(triplatin tetranitrate), 5-플루오로우라실(5-fluorouracil), 6-머캅토퓨린(6-mercaptopurine), 카페시타빈(capecitabine), 클라드리빈(cladribine), 클로파라빈(clofarabine), 시스타르빈(cystarbine), 플록스유리딘(floxuridine), 플루다라빈(fludarabine), 겜시타빈(gemcitabine), 하이드록시우레아(hydroxyurea), 메토트렉세이트(methotrexate), 페메트렉세드(pemetrexed), 펜토스타틴(pentostatin), 티오구아닌(thioguanine), 캠토테신(camptothecin), 토포테칸(topotecan), 이리노테칸(irinotecan), 에토포사이드(etoposide), 테니포시드(teniposide), 미토산트론(mitoxantrone), 파클리탁셀(paclitaxel), 도세탁셀(docetaxel), 이자베필론(izabepilone), 빈블라스틴(vinblastine), 빈크리스틴(vincristine), 빈데신(vindesine), 비노렐빈(vinorelbine), 에스트라머스틴(estramustine), 메이탄신(maytansine), DM1 (mertansine, 메르탄신), DM4, 돌라스타틴(dolastatin), 아우리스타틴 E(auristatin E), 아우리스타틴 F(auristatin F), 모노메틸 아우리스타틴 E(monomethyl auristatin E), 모노메틸 아우리스타틴F(monomethyl auristatin F) 및 이들의 유도체로 이루어진 군으로부터 선택되는 어느 하나 이상인, 펩타이드 컨쥬게이트. - 제5항에 있어서,
상기 항암 단백질은 아스파라기네이즈(asparaginase), 단백질 독소, 암항원에 특이적인 항체 또는 상기 항체의 단편, 종양억제 단백질(tumor suppressor protein), 또는 항혈관생성 인자(antiangiogenic factor)인, 펩타이드 컨쥬게이트. - 제7항에 있어서,
상기 종양억제 단백질은 VHL(von HippelLindau), APC(Adenomatous polyposis coli), CD95(cluster of differentiation 95), ST5(Suppression of tumorigenicity 5), YPEL3(Yippee like 3), p53, ST7(Suppression of tumorigenicity 7) 또는 ST14(Suppression of tumorigenicity 14)인, 펩타이드 컨쥬게이트. - 제1항 또는 제2항의 재조합 모노바디 또는 제5항의 펩타이드 컨쥬게이트를 유효성분으로 함유하는, 암 치료용 약학적 조성물.
- 제9항에 있어서,
상기 암은 폐암, 위암, 간암, 골암, 췌장암, 담낭암, 담관암, 피부암, 두경부암, 피부 흑색종, 자궁암, 난소암, 직장암, 대장암, 결장암, 유방암, 자궁 육종, 나팔관 암종, 자궁내막 암종, 자궁경부 암종, 질 암종, 외음부 암종, 식도암, 후두암, 소장암 또는 갑상선암인, 조성물. - 제1항 또는 제2항의 재조합 모노바디에 조영용 화합물이 공유결합 또는 비공유결합에 의해 결합된, 펩타이드 컨쥬게이트.
- 제11항에 있어서,
상기 조영용 화합물은 형광단백질, 형광분자, 양전자 방출 단층촬영(positron emission tomography, PET) 조영용 방사성 동위원소 함유 조영제, 단일 광자 방출 단층촬영(single photon emission computer tomography, SPECT) 조영용 방사성 동위원소 함유 조영제, 또는 자기공명영상화(magnetic resonance imaging, MRI) 조영제인, 펩타이드 컨쥬게이트. - 제12항에 있어서,
상기 형광단백질은 녹색형광단백질(green fluorescent protein, GFP), 황색형광단백질(yellow fluorescent protein, YFP), 적색형광단백질(red fluorescent protein, RFP), 주황형광단백질(orange fluorescent protein, OFP), 청록색형광단백질(cyan fluorescent protein, CFP), 청색형광단백질(blue fluorescent protein, BFP), 원적색형광단백질(far-red fluorescent protein) 또는 테트라시스테인 모티프(tetracystein motif)인, 펩타이드 컨쥬게이트. - 제11항의 펩타이드 컨쥬게이트를 유효성분으로 함유하는, 암 진단 또는 조영용 조성물.
- 제14항에 있어서,
상기 암은 폐암, 위암, 간암, 골암, 췌장암, 담낭암, 담관암, 피부암, 두경부암, 피부 흑색종, 자궁암, 난소암, 직장암, 대장암, 결장암, 유방암, 자궁 육종, 나팔관 암종, 자궁내막 암종, 자궁경부 암종, 질 암종, 외음부 암종, 식도암, 후두암, 소장암 또는 갑상선암인, 조성물. - 제14항의 암 진단 또는 조영용 조성물을 개체에 투여하는 단계; 및
상기 조성물에 의한 영상을 수득하는 단계를 포함하는 인간을 제외한 동물의 종양 진단방법. - 제14항의 암 진단 또는 조영용 조성물을 암에 걸린 개체에 투여하는 단계;
상기 조성물에 의한 영상을 수득하는 단계; 및
상기 조성물에 의한 신호가 나타난 지역을 절제하는 단계를 포함하는 인간을 제외한 포유동물의 수술 중 암 조직 조영방법.
