Abstract
본 발명은 보다 효율적인 고형암 치료를 위하여, 종양 항원에 특이적으로 결합하는 모노바디가 L-아스파라기나제에 연결된 신규 융합단백질 및 그의 용도를 제공한다.
Description
본 발명은 신규 항암제에 관한 것으로서, 더 상세하게는 종양 항원을 표적으로 하는 신규 항암제에 관한 것이다.
종양 항원(tumor antigen)은 암세포에서 특이적으로 발현되거나 정상세포에 비해서 과발현되는 분자를 의미한다. 이러한, 종양 항원들은 암세포의 증식과 전이 등과 밀접한 관련이 있는 경우가 많기 때문에 이들 종양 항원의 기능을 차단할 수 있는 항체와 같은 분자들을 투여할 경우 종양조직의 성장이나 전이가 억제될 수 있다. 선택적으로, 상기 항체 등 종양 항원에 대해 특이적으로 결합할 수 있는 분자의 종양 항원 특이적 결합능력을 이용하여, 항암제나 치료용 방사성 핵종을 공유결합 또는 킬레이터와의 배위결합 등 비공유결합을 이용하여 종양 항원 표적화 분자에 연결하여 암세포를 선택적으로 공격할 수 있는 표적 항암제가 개발되고 있다. 또한, 최근에는 T 세포 또는 NK 세포 등 선천성 면역세포의 선천성 면역기능(innate immunity)을 강화시킬 수 있도록 종양 항원에 특이적인 항체 단편(예컨대 scFv 등)을 TCR의 세포내 신호전달 도메인과 연결하여 제조된 융합단백질인 키메라 종양 수용체(chimeric antgen receptor, CAR)를 T 세포나 NK 세포에 형질도입한 후 세포를 항암 치료제로 사용하는 치료전략이 채택되고 있다. 예컨대, 유럽특허공보 EP12102374B1은 PMSA에 특이적으로 결합하는 단일클론 항체 및 상기 항체 또는 상기 항체에 세포독소(cytotoxin)가 결합된 컨쥬케이트를 항암 치료제의 제조에 사용하는 용도를 개시하고 있다.
한편, L-아스파라기나제(L-ASNase)는 L-아스파라긴을 가수분해하여 아스파르트산과 암모니아를 생성하는 반응을 촉매하는 효소로 백혈병 유발 마우스의 암 세포를 신속하고 거의 완벽하게 제거하는 것을 발견한 이후, 1978년 FDA의 승인을 받아 급성림프구성백혈병(acute lymphoblastic leukemia, ALL) 및 비호지킨 림프종(non-Hodgkin's lymphoma)등 특정 타입을 포함한 주로 혈액을 통해 감염되는 종양의 치료에 사용되었다. 그 이유는 아스파라긴 합성에 있어 T-림프구성 백혈병 암세포는 아스파라긴의 세포외 풀(extracellular pool)에 의존해야 하는 아스파라긴 합성효소의 감소된 발현으로 인해 아스파라긴을 합성할 수 없으므로 외부 소스로부터 아스파라긴을 흡수해야 하는 약점이 있기 때문이다. 따라서 아스파라긴에서 아스파테이트로 분해시키는 L-아스파라기나제(L-ASNase)에 노출되면 영양분으로 공급되는 아스파라긴의 결핍으로 단백질 합성이 금지되고, T-림프구성 백혈병 암세포는 사멸하게 된다. 그러나 혈액암에 대하여 잘 작동하는 L-아스파라기나제는 고형암에 대하여는 잘 작동하지 않는 것으로 보고되었는데, 이는 혈액암과 달리 고형암의 경우 단백질의 약물의 전달이 용이하지 않은 점 때문인 것으로 추정되고 있다. 이러한 L-아스파라기나제의 종양조직 전달효율을 증가시키기 위해 본 발명자들은 L- 아스파라기나제를 표면에 제시할 수 있도록 형질전환된 약독화 살모넬라 균을 개발하여 고형암에 대한 항암활성을 보고한 바 있다(대한민국 특허 제10-1750007호).
그러나 상기 선행기술의 경우, 약독화되긴 하였으나 살아있는 세균을 사용한다는 점에서 인체 안전성 문제가 해소되지 않은 단점을 가지고 있다. 따라서 보다 안전하면서도 기존의 재조합 단백질의 문제점인 종양조직으로의 약물전달 효율을 강화한 새로운 아스파라기나제 기반의 항암제의 개발이 절실히 요구되고 있다.
본 발명은 상기와 같은 문제점을 포함하여 여러 문제점들을 해결하기 위한 것으로서, 종양 항원을 표적화하는 아스파라기나제 기반의 신규 항암제를 제공하는 것을 목적으로 한다.
본 발명의 일 관점에 따르면, 인간 피브로넥틴 도메인 Ⅲ(Fn3)의 BC 루프 및 FG 루프 중 적어도 어느 하나 이상에 칼레티큘린(calreticulin) 특이적 결합 펩타이드가 삽입되어 상기 칼레티큘린에 특이적으로 결합하는 재조합 모노바디의 C-말단에 L-아스파라기나제에 연결된, 융합단백질이 제공된다.
본 발명의 다른 일 관점에 따르면, 상기 융합단백질을 유효성분으로 포함하는, 고형암 치료용 약학적 조성물이 제공된다.
본 발명의 다른 일 관점에 따르면, 치료적으로 유효한 양의 상기 약학적 조성물을 고형암에 걸린 개체에 투여하는 단계를 포함하는, 상기 개체의 고형암 치료방법이 제공된다.
본 발명의 다른 일 관점에 따르면, 방사선 조사와 동시에 또는 방사선 조사 전후 치료적으로 유효한 양의 상기 융합단백질 또는 상기 약학적 조성물을 고형암에 걸린 개체에 투여하는 단계를 포함하는, 상기 개체의 고형암 치료방법이 제공된다.
상기한 바와 같이 이루어진 본 발명의 신규 융합단백질은 기존에 L-아스파라기나제에 의해 치료가 쉽지 않았던 고형암에 대하여 효과적인 치료가 가능한 매우 유용한 물질로서, 암 특히 특정 종양 항원의 고발현을 특징으로 하는 암에 대한 치료제로 사용이 가능하다. 물론 이러한 효과에 의해 본 발명의 범위가 한정되는 것은 아니다.

도 2a는 본 발명의 PASylated CRT 표적 L-아스파라기나제 융합단백질의 개략적인 구조를 나타내는 개요도이다. 상기 L-ASNase는 각각 N- 및 C-말단에 모노바디 및 PAS200 태그와 결합되었다. CRT3 및 CRT4 모노바디는 Fn3 백본의 BC 및 FG 루프에 CRT-표적서열[Int-α(KLGFFKR, 서열번호 15), Hep-I(GQPMYGQPMY, 서열번호 16)]을 포함한다. 각 모노바디의 DE 루프는 야생형 Fn3(Wt)의 것과 동일하다. 재조합 단백질은 C-말단에 His6 태그 서열이 있고 #DGRLP는 FG 루프에서 RGD 대신 DGR(Scrambled monobody)이 있는 Wt Fn3 단백질인 #DGR 모노바디를 포함하는 음성 대조군이다.

본 발명의 일 실시예에 따른 약학적 조성물은 비경구 투여로 상기 개체에 투여될 수 있으며, 상기 비경구 투여는 정맥내 투여(intravenous injection), 복강내 투여(intraperitoneal injection), 근육내 투여(intramuscular injection), 피하 투여(subcutaneous injection), 뇌내 투여(intraventricular injection), 두 개내 투여(intracranial injection), 뇌척수액내 투여(intracerebrospinal injection) 또는 종양내 투여(intratumoral injection)일 수 있다.
본 발명의 일 실시예에 따른 약학적 조성물은 제약상 허용되는 담체를 비롯한 불활성 성분을 추가로 포함한다. 본 명세서에서 사용된 "제약상 허용된 담체"란 조성물, 구체적으로 의약 조성물의 활성 물질을 제외한 성분을 지칭하는 용어이다. 제약상 허용되는 담체의 예로는 결합제, 붕해제, 희석제, 충진제, 활택제, 가용화제 또는 유화제 및 염이 포함된다.
아울러, 본 발명의 일 실시예에 따른 약학적 조성물은 0.1 mg/kg 내지 1 g/kg의 용량으로 투여될 수 있으며, 더 바람직하게는 0.1 mg/kg 내지 500 mg/kg의 투여량으로 투여된다. 한편, 상기 투여량은 환자의 나이, 성별 및 상태에 따라 적절히 조절될 수 있다.
본 발명자들은 종래 Fn3 도메인의 각 루프 위치(BC, DE, FG)에 Int-α(서열번호 15), Hep-I(서열번호 16) 두 가지 CRT 펩타이드를 치환하여 재조합 CRT-monobody를 제조하였고 이렇게 제조된 항-CRT 모노바디가 칼레티큘린에 특이적으로 결합함을 확인하였다. 이어서, 상기 항-CRT 모노바디를 칼레티큘린 과발현 종양에 대한 치료제로 활용하기 위해 항암 단백질인 L-아스파라기나제를 항-CRT 모노바디의 C-말단에 연결한 융합단백질을 제조하였다(도 1). 상기 융합단백질이 칼레티큘린을 과발현하는 암세포에 대한 살상능력이 높음을 확인함으로써 본 발명을 완성하였다. 뿐만 아니라, 본 발명자들은 항-CRT 모노바디 외에도 다른 종양 항원인 EphA2에 특이적으로 결합할 수 있는 항-EphA2 모노바디를 L-아스파라기나제에 연결한 융합 단백질을 제조하여 이 역시 EphA2를 과발현하는 종양에 대하여 항암활성을 나타냄을 확인함으로써, 본 발명의 종양 항원 특이적 모노바디-L-아스파라기나제 융합단백질이 항암치료 특히 고형암 치료에 있어서 매우 효과적인 플랫폼임을 확인할 수 있었다.
구체적으로, 세균성 L-ASNase는 다른 항암제와 병용하여 백혈병 치료에 널리 사용되었다(N. Verma, et al., Critical reviews in biotechnology, 27(1), 45-62. 2007). 그러나 고형 종양에 대한 적용을 확장하기 위해 고안된 임상 시험은 종양 조직에 대한 제한된 축적으로 인해 실패했다(F. Chiarini, et al., Biochimica et Biophysica Acta (BBA)-Molecular Cell Research, 1863(3), 449-463. 2016). 상기 효소에 표적 활성을 부여하기 위한 시도는 표적 펩타이드와의 결합으로 수행되었다. 예를 들어, 표적-특이적 scFv와 결합된 L-ASNase가 설계되었다(L. Guo, et al., Biochemical and Biophysical Research Communications, 276(1), 197-203. 2000). 그러나 단백질이 E. coli에서 발현되었을 때 대부분은 봉입체에 위치하고 있었다. T-CAN(targeted-catalytic nanobody)이 최근에 보고되었는데(M. Maggi, et al., Cancers, 13(22), 5637. 2021), 상기 재조합 단백질은 기능적 효소 활성을 나타내는 L-ASNase의 단일 서브유닛과 결합된 나노바디를 함유하고 있고 CD19-발현 ALL 세포와 결합한다. 그러나 상기 단백질 역시 대장균에서도 봉입체로 발현되었다. scFv 및 나노바디와 같은 단백질은 면역글로불린 접힘 구조를 가지고 있기 때문에 E. coli에서 봉입체로부터 기능성 단백질을 생성하기 위해서는 별도의 재접힘 공정 등으로 인해 고비용 공정을 필요로 한다(A. Singh, et al., Microbial cell factories, 14(1), 1-10. 2015).
본 발명자들은 CRT-표적 모노바디(CRT3LP 및 CRT4LP)와 결합된 PASylated L-ASNase가 E. coli에서 가용성 및 기능적 형태로 발현됨을 보여주었다. 실제로 이러한 결과는 CRT 모노바디에 결합된 Rluc8에 대해 얻은 결과와 일치했다. 모노바디와 PAS200 태그를 L-ASNase와 결합하면 몇 가지 장점이 있다. L-ASNase는 사량체 단백질이기 때문에 CRT3LP와 CRT4LP는 N-말단에 4개의 모노바디 모이어티를 가지고 있다. 이와 일관되게 CRT에 대한 친화성은 단일 모노바디보다 4배 더 높았다. 아울러 C-말단에서의 PASylation은 이들의 용해도를 증가시켰다. 따라서 이들 결합체는 E. coli에서 비-PASylated 결합체보다 훨씬 더 높게 발현되었고 생체 내에서 더 긴 반감기를 보였다. 이는 VEGFR2-표적 모노바디인 PASylated adnectin의 결과와 일치했다. 생체 내 반감기 시간의 증가는 부분적으로 N-말단 모노바디 모이어티에 의한 효소의 보호로 인한 것일 수 있다. L-ASNase는 mTORC1 신호 전달을 억제하고 G1 단계에서 세포 주기 정지를 유발하여 세포 사멸을 유도했다.
본 발명에서 L-ASNase로 처리된 종양 세포는 성장 억제를 보였지만 ecto-CRT를 감지하지 못했다. 따라서 CRT3LP 및 CRT4LP의 L-ASNase 활성에 의해 유발된 세포사멸 및 처리된 세포는 ecto-CRT를 유도하지 않았다. 이는 CRT3LP 및 CRT4LP가 외부 CRT에 노출된 종양 세포만을 죽일 수 있음을 나타낸다. 이는 CRT3LP 및 CRT4LP가 불규칙한 점선 패턴으로 DOX-처리된 종양에 국한되었음을 보여주는 면역조직화학 결과와 일치했다. 고용량의 CRT3LP(20 IU)는 항암제로 처리한 마우스에서 갑작스러운 사망을 일으켰다. 이에 대한 이유는 첫 번째, 정제된 단백질의 내독소 오염으로 생각되었다. 단백질 정제를 위해 His6 태그 친화성 및 크기 배제의 이중 크로마토그래피 절차를 사용했다. 이는 단백질에서 내독소를 완전히 배제할 수 있다. 이런 오염을 배제하기 위한 많은 프로토콜이 있었다(S.J. Wakelin, et al., Immunology letters, 106(1), 1-7. 2006). 두 번째 가능성은 CRT3LP에 의해 유발된 급성 면역 반응이었다. 박테리아의 L-ASNase는 면역 반응을 유도할 수 있다(A.M. Lopes, et al., Critical reviews in biotechnology, 37(1), 82-99. 2017). 모노바디는 인간 Fn3에서 유래되었습니다. 또한 PAS200 태그는 인공적으로 합성되었으며 면역 반응을 유발할 수도 있습니다(PEG와 유사). 마지막 가능성은 CRT3LP의 L-ASNase 활성에 의해 생성된 암모니아이다. 많은 양의 암모니아는 세포 pH 변화, 미토콘드리아 막 파괴 및 ATP 고갈과 같은 생체 내 부작용을 유발했다. 고형 종양에 대해 CRT3LP 및 CRT4LP를 테스트했지만 ALL(급성림프구성백혈병)과 같은 혈액 암에 임상적으로 적용할 수 있다. 현재 PEGylated L-ASNase는 화학적 결합으로 인해 PEG 모이어티의 수가 명확하게 정의되지 않았지만 혈액암에 사용되었고 PEGylated L-ASNase는 혈액 종양에 대한 표적 특이성을 가지고 있지 않았다. 혈액암에 대한 화학요법에 결합할 때 CRT3LP 및 CRT4LP는 더 나은 CRT 표적화로 인해 L-ASNase 단독보다 더 효율적일 수 있다.
"모노바디와 결합된 PASylated L-ASNase"의 개념은 다양한 고형 종양으로 확장될 수 있다. 많은 모노바디 및 기타 결합 펩타이드는 종양 표적에 특이적으로 결합한다. 모노바디 모이어티의 변화는 L-ASNase에 새로운 표적 특이성을 제공할 수 있다. 또한 특정 아미노산을 분해하는 다양한 효소를 이용하여 암환자를 대상으로 한 임상시험이 진행되어 왔다(H. Komuro, et al., Bioengineering, 9(2) 56. 2022). 다른 치료용 단백질(예: 독소, 사이토카인 및 면역원)은 신약 개발을 위해 L-ASNase 대신 사용될 수 있다.
본 발명자들은 ICD-유도 화학요법으로 치료된 고형 종양에서 항암제로서 PASylated CRT-표적 L-ASNase의 효능을 관찰했다. 혈액암에 대한 항암제로 사용되는 효소인 L-ASNase는 표적 특이적 모노바디와의 결합을 통해 고형 종양에 적용될 수 있다. 더 구체적으로 설명하면, 아스파라긴을 분해하는 박테리아 효소인 L-아스파라기나제(L-ASNase)는 급성림프구성백혈병(ALL)과 같은 악성 조혈암을 치료하기 위해 여러 화학 약물과 함께 일반적으로 사용되었다. 대조적으로, 상기 효소는 시험관 내에서 고형 종양 세포의 성장을 억제하는 것으로 알려져 있지만 생체 내에서는 효과적이지 않다. 본 발명자들은 종래 발명에서 2개의 새로운 모노바디(CRT3 및 CRT4)가 면역원성 세포사멸(ICD) 동안 종양 세포 및 조직에 노출된 칼레티큘린(CRT)과 특이적으로 결합한다고 보고했다. 그 후, 본 발명에서는 C-말단(CRT3LP 및 CRT4LP)에서 N-말단 및 PAS200 태그에서 모노바디와 결합된 L-ASNase를 설계했다. 상기 단백질은 L-ASNase 형태를 방해하지 않는 4개의 모노바디 및 PAS200 태그 모이어티를 보유할 것으로 예상되었다. 상기 단백질은 PASylation이 없는 것보다 E. coli에서 3.8배 더 높게 발현되었다. 상기 정제된 단백질은 예상보다 훨씬 더 큰 겉보기 분자량으로 용해도가 높았다. CRT에 대한 상기 단백질의 친화도(Kd)는 약 2 nM으로 모노바디보다 4배 더 높았다. 효소 활성(~6.5 IU/nmol)은 L-ASNase(~7.2 IU/nmol)와 유사했으며 열 안정성은 55℃에서 크게 증가했다. 반감기는 마우스 혈청에서 > 9시간으로, L-ASNase보다 약 5배 더 길었다(~1.8시간). 또한, CRT3LP 및 CRT4LP는 in vitro에서 종양 세포에 노출된 CRT와 특이적으로 결합하여, ICD 유도 약물(독소루비신 및 미토잔트론)로 처리한 CT-26 및 MC-38 종양 보유 마우스에서 종양 성장을 유의하게 억제했지만 비-ICD 유도 약물(젬시타빈)로는 그러지 않았다. 모든 데이터는 PASylated CRT-표적 L-ASNase가 ICD 유도 화학 요법의 항암 효능을 향상시켰음을 나타낸다. 따라서 상기 L-ASNase는 고형 종양을 치료하기 위한 잠재적인 항암제로 면역원성 세포사멸-유도 화학요법에 의한 고형암 갖는 마우스에서 PASylated 칼레티큘린-표적 L-ASNase의 항암 효과를 확인하였다(도 1).
이하, 실시예를 통하여 본 발명을 더 상세히 설명한다. 그러나 본 발명은 이하에서 개시되는 실시예에 한정되는 것이 아니라 서로 다른 다양한 형태로 구현될 수 있는 것으로, 이하의 실시예는 본 발명의 개시가 완전하도록 하며, 통상의 지식을 가진 자에게 발명의 범주를 완전하게 알려주기 위해 제공되는 것이다.
재료 및 방법
세포주 및 시약
본 발명에서 사용한 마우스 결장암 CT-26 및 MC-38 세포주는 각각 American Type Culture Collection(ATCC, USA) 및 Kerafast社, USA에서 구입하였다. DMEM 배지는 미국 GIBCO에서 구입하였고 태아 소 혈청(FBS) 및 인산염 완충 식염수(PBS)는 미국 Gibco/Thermo Fisher Scientific社에서 구입했으며 페니실린/스트렙토마이신 용액은 미국 Sigma-Aldrich社에서 구입했다. 아스파라기나제 활성 분석 키트와 재조합 칼레티큘린 단백질(rCRT)은 미국 Abcam社에서 구입했고 파라포름알데히드(PFA)는 한국의 Biosesang社에서 구입했다. 또한 L-ASNase는 미국 Prospec社에서 Cell Counting Kit-8(CCK-8)은 미국 Enzo Life Sciences社에서 구입했다. 본 발명에 사용된 모든 항체는 하기 표 1에 요약하였다.

PASylated L-ASNase를 전달하는 발현 벡터의 구축
PAS200 조각(ASPAAPAPASPAAPAPSAPA)10은 종래 기술을 참조하였고(J. Breibeck, et al., Biopolymers, 109(1), e23069. 2018) E. coli 코돈 사용에 기반하여, 아미노산 서열을 EMBOSS Backtranseq 프로그램(EMBL's European Bioinformatics Institute, UK)에 의해 역번역하여 유전자를 산출하였다. EcoR1 및 BamH1 사이트에 해당하는 서열은 각각 5' 및 3' 말단에 추가되었고 PSA200 유전자 중간에 아미노산의 돌연변이 없이 Pst1 사이트를 생성하기 위해 일부 뉴클레오티드를 변경했다. 상기 유전자 염기서열을 기반으로 EcoR1-PAS100-Pst1 및 Pst1-PAS100-BamH1 두 조각을 화학적으로 합성하였다(Macrogen, Korea). 단편은 상응하는 제한 효소로 분해되었고, EcoR1 및 BamH1로 분해된 플라스미드 pETh-E1-EGFP와 삼중 결찰되었다(M.A. Kim, et al., PloS one, 12(7), e0180786. 2017). 생성된 플라스미드를 pETh-E1-PAS200으로 명명하였다. 전체 길이의 PAS200 단편은 pETh-E1-PAS200에 대한 BglII-G4S-PAS200-F(서열번호 30) 및 pET-R(서열번호 31) 프라이머를 중합효소 연쇄 반응(PCR)의 주형으로 사용하였다. T4 폴리뉴클레오티드 키나제로 인산화한 후, 단편을 pBluescript II SK(+)(Agilent Technologies, USA)에 클로닝하고 EcoRV로 분해하였으며 알칼리 포스파타아제(Thermo Fisher Scientific, USA)로 탈인산화했다. 생성된 플라스미드를 pBS-BglII-G4S-PAS200-BamH1로 명명하였다. E. coli L-ASNase를 포함하는 플라스미드 pASN은 최현이 박사(전남대학교 의과대학)로 부터 수득하였다(K. Kim, et al., Molecular Therapy-Oncolytics, 2, 15007. 2015). 상기 L-ASNase 유전자 단편을 pASN에 대한 LASP-EcoR1-F(서열번호 32) 및 LASP-BamH1-R(서열번호 33) 프라이머를 주형으로 사용하여 PCR 증폭하였다. 상기 단편을 EcoR1 및 BamH1로 절단하고, pBS-BglII-G4S-PAS200-BamH1의 EcoR1-BglII 부위에 클로닝하였고 생성된 플라스미드를 pBS-LASP-PAS200으로 명명하였다. CRT-표적 모노바디(CRT3 및 CRT4) 및 이들의 이소타입 대조군(#DGR) [pETh-CRT3, pETh-CRT4 및 pETh-Fn3(DGR)]에 대한 발현 벡터는 종래 기술을 참조하였다(Y. Zhang, et al., Cancers, 13(11), 2801. 2021). 상기 모노바디 유전자를 T7(서열번호 34) 및 94old(GC)-R(서열번호 35) 프라이머로 각각의 상응하는 플라스미드를 주형으로 PCR 증폭한 후 Nhe1 및 EcoR1로 절단하였다. LASP-PAS200 단편은 pBS-LASP-PAS200을 EcoR1 및 BamH1으로 절단한 후 제조하였다. 각각의 모노바디 및 LASP-PAS200 유전자 단편을 Nhe1 및 BamH1로 분해된 pETh-E1-PAS200으로 삼중 결찰시켰고 생성된 플라스미드를 pETh-CRT3-LASP-PAS200, pETh-CRT4-LASP-PAS200 및 pETh-Fn3(DGR)-LASP-PAS200으로 명명하였다. 이들은 각각 CRT3LP, CRT4LP 및 #DGRLP로 명명된 PASylated L-ASNase 단백질을 암호화하는 유전자를 가지고 있다. 그 결과, 모노바디, L-ASNase 및 PAS200 태그의 유전자는 (G4S)2 링커를 통해 연결되었고 C-말단에 His6 태그를 포함하였다. 상기 기술된 모든 플라스미드는 시퀀싱(Macrogen, Korea)에 의해 확인되었다. 상기 클로닝에 사용한 프라이머의 핵산 서열을 하기 표 2에 요약하였다.

재조합 단백질의 정제 및 특성화
재조합 단백질은 종래 기술을 참조하여 정제하였다(Y. Zhang, et al., Cancers, 13(11), 2801. 2021). 요약하면, 발현 벡터로 형질전환된 대장균 BL21(DE3)(Invitrogen, CA)을 50 μg/mL 카나마이신을 함유하는 LB 배지에서 37℃에서 밤새 배양하였다. 박테리아를 50 μg/mL 카나마이신을 함유하는 신선한 LB 배지 500 mL에 접종(1/100 희석)했다. 37℃에서 3시간 배양한 후 박테리아를 0.5 mM 최종 농도의 isopropyl-β-D-thiogalactopyranoside(IPTG)로 처리했다. 4시간 동안 더 배양한 후, 박테리아 펠릿을 4℃에서 5분 동안 8,000 xg로 원심분리하여 수확하였고 100 μg/mL의 라이소자임 용액을 포함하는 얼음처럼 차가운 용해 완충액[50mM NaH2PO4, 300 mM NaCl 및 10 mM 이미다졸(pH 8.0)]에서 30분 동안 재현탁했고 얼음 위에서 부드럽게 초음파 처리했다. 원심분리 후 상등액은 His GraviTrap 컬럼(GE Healthcare, USA)을 사용하여 정제하였고 과량의 이미다졸은 PD-10 탈염 칼럼(GE Healthcare, USA)으로 제거하였다. 그 후, 단백질을 PBS에 분산시키고 필요할 때까지 4℃에서 보관했다.
CRT3LP 구조의 컴퓨터 모델링
CRT3 모노바디의 구조는 AlphaFold를 사용하여 구축하였다(J. Jumper, et al., Nature, 596(7873), 583-589. 2021). 시각화를 위해 가장 순위가 높은 구조가 선택되었다. CRT3LP의 구조는 L-ASNase 테트라머(PDB 2HIM)의 결정 구조에 CRT3 모노바디의 4개 단위를 중첩하여 구축되었다. 모든 단백질 구조는 PyMOL을 사용하여 렌더링 및 분석되었다(W.L. DeLano, et al., Protein Crystallogr, 40(1), 82-92. 2002).
SDS-PAGE 및 웨스턴 블롯 분석
정제된 단백질의 박테리아 펠렛을 5x 샘플 버퍼(ElpisBio, Korea)와 혼합하고 12% SDS-PAGE 겔(10 μg/웰)에 로딩하였다. 전기영동으로 분리한 후 Coomassie blue R-250 염색액(Enzynomics, Korea)으로 단백질을 가시화하였다. 박테리아에서의 단백질 발현 및 재조합 단백질의 순도를 평가하기 위해 웨스턴 블롯팅을 수행하였다. 요약하면, SDS-PAGE 젤의 단백질을 4℃에서 100분 동안 120 전압으로 니트로셀룰로스 멤브레인으로 옮겼다. 그런 다음 상기 멤브레인을 실온에서 1시간 동안 5% 탈지유를 포함하는 Tris-완충 식염수-0.1% Tween®20(TBS-T) 용액에서 배양했다. 상기 세척된 멤브레인을 HRP(horseradish peroxidase)가 결합된 항-토끼 2차 항체(1:2000 희석)와 함께 1시간 동안 배양하였다. LAS-3000 화학발광 검출 시스템(Fuji, Japan)을 사용하여 면역블롯된 단백질을 검출하였다.
질량 분석
재조합 단백질의 분자량은 광주과학기술원(GIST) 중앙연구시설(GCRF, Korea)에서 Autoflex speed MALDI-TOF/TOF 질량분석기로 측정하였다.
제타 전위 측정
재조합 단백질의 제타 전위 값은 제타 전위 및 입자 크기 분석기 ELS-Z 시리즈(Otsuka Electronics, 일본)로 측정하였다. 25℃에서 정제된 샘플[800 μL의 증류수(pH 7.2)에 50 μg의 단백질]에 대해 3회 측정을 수행했다.
결합 친화성
CRT에 대한 재조합 단백질의 친화성은 ELISA(enzyme-linked immunosorbent assay)로 측정하였다(J.D. Beatty, et al., Journal of immunological methods 100(1-2), 173-179. 1987). 요약하면, 100 μL의 PASylated CRT-표적 L-ASNase를 4℃에서 밤새 96-웰 마이크로플레이트(0 - 10 μM/웰)에 코팅했다. 결합되지 않은 단백질은 웰에서 흡인하여 제거되었고 10 μM rCRT 단백질 100 μL를 각 웰에 첨가하고 실온에서 2시간 동안 배양하였다. rCRT를 제거한 후, 토끼 항-CRT 항체(1:500 희석)를 각 웰에 첨가하고 실온에서 2시간 동안 배양하였다. 상기 항체는 흡인에 의해 제거되었고, 상기 웰은 T-PBS로 5회 세척되었다. 그런 다음, 비오틴-결합 항-토끼 2차 항체(1:1000 희석)를 첨가하고 1시간 동안 배양하였다. T-PBS로 세척한 후, 100 μL의 아비딘-HRP(1:250 희석)을 각 웰에 첨가하고 실온에서 30분 동안 배양하였다. 흡인 및 5회 세척 후, 100 μL의 1x3,3',5,5'-테트라메틸벤지딘(TMB) 완충액(Thermo Fisher Scientific, USA)을 각 웰에 첨가하여 황색 반응 생성물을 형성하였다. 15분 후, 각 웰에 0.5M H2SO4 용액 50 μL를 첨가하여 발색 반응을 중단시켰다. SpectraMax M2 마이크로타이터 플레이트 판독기(Molecular Devices, USA)에 의해 450 nm(OD450)에서 광학 밀도 값으로 비색 제품의 양을 평가했다.
L-ASNase 활성 분석
재조합 단백질의 L-ASNase 효소 활성은 OxiRed 프로브(Abcam, USA)를 사용하는 아스파라기나제 활성 분석 키트로 측정하였다. 1 국제 단위(IU)는 25℃에서 분당 1.0 μmol의 Asp를 생성하는 효소의 양으로 정의되었다.
열 안정성 분석
간략하게, 200 μL의 PASylated 재조합 단백질 또는 L-ASNase(1 mg/mL)를 열 순환기(Finepcr, Korea)에서 55℃의 조건으로 30-180분 동안 배양했다. 단백질의 잔류 효소 활성은 상기한 바와 같이 아스파라기나제 활성 분석 키트를 사용하여 측정하였다. 각 인큐베이션 시간에서의 잔류 활성(%)은 시간 0에서의 샘플 활성에 상대적으로 계산되었다.
유세포 분석
세포를 10% FBS 및 100U의 스트렙토마이신/페니실린이 첨가된 DMEM 배지에서 5% CO2의 대기조건으로 37℃에서 배양하였다. 세포사멸을 유도하기 위해 각 항암제[3 μM methotrexate(MTX), 25μM doxycycline(DOX) 또는 15 μM gemcitabine(GEM)]로 세포를 처리하였다(Y. Zhang, et al., Cancers, 13(11), 2801. 2021). 4시간 배양 후, 상기 세포를 긁어서 접시에서 분리하였다. 상기 세포(105)를 1% PFA로 15분 동안 고정한 다음 얼음 위에서 100 nM의 PASylated 재조합 단백질로 1시간 동안 처리했다. 0.1% Tween-20(T-PBS)을 포함하는 차가운 PBS 완충액으로 5회 세척한 후, 상기 세포를 항-His 태그 단클론 항체(1:1,000 희석)와 함께 1시간 동안 배양한 다음, Alexa Fluor 488-결합된 2차 항체(5 μg/mL)로 1시간 동안 처리하였다. FACSCanto II Flow Cytometer(BD Biosciences, USA)로 형광 신호를 검출하였고 차단 실험을 위해 세포를 먼저 항암제와 함께 4시간 동안 배양하고 1% PFA로 고정시켰다. 그런 다음 세포를 항-CRT 항체(1:400 희석)와 함께 1시간 동안 배양하여 ecto-CRT를 마스킹했다. 세포에 대해 PASylated 재조합 단백질로 염색한 후 상기 설명한 것과 동일한 절차를 수행했다.
공초점 면역형광 영상 분석
세포 상의 ecto-CRT에 결합된 PASylated CRT-표적 L-ASNase는 종래 문헌을 참조하여 공초점 면역형광 현미경으로 가시화되었다(Y. Zhang, et al., Cancers, 13(11), 2801. 2021). 커버-글래스에서 밤새 배양한 후, CT-26 및 MC-38 세포(104)를 37℃에서 4시간 동안 항암제로 처리하였다. 이어서, 상기 세포를 1% PFA로 15분 동안 고정하고, 100 nM PASylated 재조합 단백질로 얼음 위에서 1시간 동안 염색한 후, 항-His 태그 단클론 항체(1:1,000 희석) 및 Alexa Fluor 488-결합된 2차 항체로 1시간 동안 처리하였다(5 μg/mL). 세포막을 Alexa Fluor 555-결합 밀 배아 응집소(WGA)(1:5,000 희석)로 염색했다. 각 단계 사이에 샘플을 차가운 T-PBS로 세척했다. 마지막으로 상기 세포를 4',6-diamidino-2-phenylindole(DAPI) (Thermo Fisher Scientific, USA)과 함께 배양하여 핵을 염색했다. 면역형광 신호는 LSM510 공초점 현미경(ZEISS, Jena, Germany)을 사용하여 이미지화하고 데이터는 ZEN-LSM 이미징 소프트웨어(ZEISS, Jena, Germany)로 분석했다.
세포 생존력
CT-26 및 MC-38 세포(웰당 104개)를 96-웰 플레이트의 3개의 웰에 파종하고 37℃에서 배양하였다. 다음날 배양 배지를 상기 농도의 항암제를 포함하는 신선 배지 100 μL로 교체하였다. 37℃에서 4시간 동안 배양한 후 배양액을 제거하고 세포를 PBS로 3회 세척하였다. 그런 다음 상기 세포를 100 μL의 L-ASNase 또는 PASylated 재조합 단백질(DMEM 배지에서 1 IU/mL)과 함께 24시간 동안 배양했다. 마지막으로 CCK-8 시약(10 μL/웰)을 37℃에서 3시간 동안 첨가하고 각 샘플의 색상 양을 OD450에서 측정했다.
생체 내 약동학 분석
6-주령의 암컷 BALB/c 마우스(Orient Company, Korea)를 무작위로 다른 처리군(n = 3)으로 배정하고 10 IU의 PASylated 재조합 단백질을 정맥 주사했다. 그런 다음, 지정된 시간(0 - 52시간)에 안구에서 혈액 샘플을 채취하고 40분 동안 4℃에서 방치했다. 12,000 rpm에서 20분 동안 원심분리한 후 혈청 분획을 얻어 추가 분석 전에 -80 ℃에 보관했다. 혈청에서 PASylated 재조합 단백질은 상기 설명한 ELISA 방법을 사용하여 평가되었다. 혈청 재조합 단백질의 양은 효소 활성으로 전환되었다.
생체 내 항암 효능 분석
CT-26 및 MC-38 종양 세포(100 μL PBS 중 106개)를 각각 6-주령의 BALB/c 및 C57BL/6 암컷 마우스(Orient company, Korea)의 우측 옆구리에 피하 이식하였다. 종양이 약 100 mm3에 도달했을 때 종양-보유 마우스에게 항암제(15 mg/kg GEM, 10 mg/kg DOX 또는 MTX 2 mg/kg)를 2일마다 3회 복강 내 주사했다. 동시에 상기 마우스에게 본 발명의 PASylated 재조합 단백질을 2일마다 5회 정맥 주사했다. 각 종양의 길이(L), 너비(W) 및 높이(H)를 디지털 캘리퍼스를 사용하여 3일마다 기록했고 종양 부피(mm3)는 하기 공식 (L x H x W)/2를 사용하여 계산되었다. 종양이 ~1,500 mm3인 마우스를 안락사시켰고 모든 동물 실험은 National Institutes of Health Guidelines에 요약된 일반 원칙과 절차에 따라 수행되었으며 모든 프로토콜은 전남대학교 동물 관리 및 사용위원회의 승인을 받았다(허가 번호: HCRL 16-001).
통계분석
모든 데이터는 평균±표준편차(SD)로 나타내었고 통계분석은 GraphPad Prism 5.0(GraphPad, USA)을 이용하였다. 생존분석은 Kaplan-Meier 방법과 log-rank test를 이용하였다. P-값 <0.05는 유의(*), <0.01은 매우 유의(**), <0.001은 매우 유의(***)로 평가되었다.
실시예 1: PASylated CRT 표적 L-ASNase의 설계 및 특성화
CRT-표적 모노바디(CRT3 및 CRT4) 및 대조군(#DGR)은 L-ASNase 및 PAS200 태그와 융합되도록 설계되었다(도 2a). 상기 재조합 단백질은 각각 CRT3LP, CRT4LP 및 #DGRLP로 명명하였다. 모든 단백질은 C-말단에 His6 태그를 가질 것으로 예상되었다. 컴퓨터 시뮬레이션을 통해 CRT3LP의 구조를 추정했다(도 2b). L-ASNase는 호모테트라머(homotetramer)이고 각 효소 서브유닛의 N-말단이 효소 복합체 외부에 노출되어 있기 때문에 4개의 CRT3 모노바디가 효소 복합체에 연결되었다. 각 효소 서브유닛의 C-말단에 있는 PAS200 태그도 외부로 노출되어 효소 복합체를 둘러쌌다. 시뮬레이션된 구조를 기반으로 CRT3 모노바디는 결합 특이성을 유지하고 재조합 단백질의 L-ASNase 구조에 영향을 미치지 않을 것으로 예상되었다. PAS200 태그는 효소 복합체를 프로테아제 공격으로부터 보호할 것으로 예상되었다. His6 태그도 노출되어 친화성 크로마토그래피에 의한 정제가 효율적일 것임을 시사한다.
재조합 단백질은 IPTG 유도 후 발현 벡터로 형질전환된 대장균에서 발현되었다(도 2c). 재조합 단백질은 IPTG의 존재하에서만 검출되었다. 그러나 예상 분자량(MW)은 약 64 kDa였지만 SDS-PAGE 젤 및 웨스턴 블롯에서 검출된 밴드의 MW는 100 kDa 이상으로 추정되었다. 불일치를 설명하기 위해 상기 재조합 단백질을 정제하고 질량 분석법으로 실제 MW를 측정했고(도 2d) 측정값은 예상 MW와 동일했다.
상기 결과는 PASylation이 표면 친수성의 변형을 통해 단백질 유체역학적 부피를 증가시켜 겔 전기영동에서 전기영동 이동도를 감소시킨다는 종래 연구보고와 유사하다(S. Aghaabdollahian et al., Scientific reports, 9(1) 1-14. 2019).또한, PASylated 단백질은 E. coli에서 PASylated가 아닌 단백질보다 3.8배 높게 발현되었다(도 3a 및 3b). 박테리아에서 재조합 단백질의 높은 발현은 종종 봉입체 (inclusion bodies) 형성으로 이어지기 때문에(A. Singh, et al., Frontiers in Microbiology. 11, 876. 2020), 상기 결과는 PASylation이 응집을 감소시키기 위해 단백질 용해도를 증가시켰고 궁극적으로 생산 수율을 증가시켰다는 것을 나타낸다.
다음으로 PASylated CRT-표적 L-ASNase를 특성화했다(도 4). SDS-PAGE에서, 상기 정제된 단백질은 MW가 100 kDa를 초과하는 도 2c에 표시된 박테리아 펠렛의 것과 유사한 이동 패턴을 보였다(도 4a, 왼쪽 패널). 상기 단백질은 웨스턴 블롯 분석에서 항-His 태그 및 항-L-ASNase 항체에 의해 검출되었다(도 4a, 상부 및 하부 패널). 재조합 단백질의 제타 전위는 수상(aqueous phase)에서 측정되었다(도 4b). 재조합 단백질의 등전점(pIs)이 5.45이므로 수용액에서 음전하를 띠는 것으로 예상하였다. 제타 전위(약 -12.5 mV)는 상기 예상과 일치했다.
아울러, CRT에 대한 재조합 단백질의 친화도를 ELISA 분석으로 평가하였다(도 4c). 해리상수(Kd) 값은 각각 CRT3LP에서 2.067 ± 0.173 nM, CRT4LP에서 2.297 ± 0.166 nM이었다. 상기 값은 전형적인 모노바디의 값과 비슷했고. 상기 단백질의 친화성은 CRT3 및 CRT4 모노바디보다 4배 더 높았다(Y. Zhang, et al., Cancers, 13(11), 2801. 2021). 예상대로 #DGRLP는 CRT에 대한 결합 친화성을 나타내지 않았다. PASylated CRT-표적 L-ASNase의 효소 활성을 측정했다(도 4d). 효소 활성은 L-ASNase와 유사했다(PASylated 단백질에서 ~6.5 IU/nmol 대 L-ASNase에서 ~7.2 IU/nmol). 상기 결과는 모노바디 및 PAS200 태그가 PASylated L-ASNase의 효소 활성을 거의 방해하지 않는다는 것을 나타낸다. 상기 효소의 열 안정성은 생체 내 적용에 중요한 매개변수이기 때문에 55℃에서 180분 동안 효소 활성을 측정했다(도 4e). PASylated 단백질의 상대적 활성은 시간이 지남에 따라 점차 감소했다. 그러나 상기 감소는 L-ASNase에서 관찰된 것보다 적었다. 180분 후, PASylated 단백질의 잔류 효소 활성은 초기 활성의 약 35.1%인 반면, 자유 L-ASNase의 잔류 효소 활성은 16.4%였다. 이는 모노바디와 PAS200 태그(A. Skerra, et al., Journal of Molecular Recognition, 13(4), 167-187. 2000)와의 융합에 의해 주어진 L-ASNase 구조의 제한 때문인 것으로 사료된다. 마지막으로 BALB/c 마우스에서 PASylated 단백질의 약동학을 평가했다(도 4f). PASylated 단백질 또는 L-ASNase(10 IU)를 꼬리 정맥을 통해 주입하고, 표시된 시간에 혈청 샘플을 수집했다. PASylated 재조합 단백질은 반감기에서 ~5배 향상을 나타내었다. #DGRLP, CRT3LP, CRT4LP의 반감기는 각각 9.20±1.22h, 9.34±0.91h, 9.02±0.28h인 반면, L-ASNase는 1.79±0.22h로 나타났다.
실시예 2: 종양 세포에 대한 세포 독성
CRT3LP 및 CRT4LP는 특히 ecto-CRT를 표적으로 하고 ICD-유도 약물로 치료된 종양 세포에 대한 세포 독성을 강화한다. 본 발명자들은 ICD-유도 항암제로 처리된 종양 세포에서 CRT3LP 및 CRT4LP의 ecto-CRT에 대한 특이적 결합을 유동 세포측정법으로 평가하였다(도 5). 그 결과, CRT3 및 CRT4 모노바디와 유사하게 상기 단백질은 DOX 및 MTX로 처리된 CT-26(도 5a 및 5c) 및 MC-38(도 5b 및 5d) 세포에 효율적으로 결합하였으나 GEM 또는 PBS 처리 시에는 그러지 않았다. 음성 대조군 #DGRLP는 DOX 및 MTX가 존재하는 경우에도 세포에 결합하지 않았다. 또한, CRT3LP 및 CRT4LP의 결합은 DOX 또는 MTX로 처리된 세포에서 항-CRT 항체의 전처리에 의해 유의하게 억제되었다. CRT3LP 및 CRT4LP가 ICD-유도 항암제에 의해 노출된 ecto-CRT에 특이적으로 결합함을 나타낸다. 상기 유세포 분석 결과는 동일한 세포에 대한 면역형광 염색 결과와 일치하였다(도 5e 및 5f). CRT3LP 및 CRT4LP는 DOX 또는 MTX로 처리된 CT-26 및 MC-38 세포에만 결합하고 GEM은 결합하지 않았다. 음성 대조군 #DGRLP는 DOX 및 MTX의 존재 하에서도 세포에 결합하지 않았다. L-ASNase는 고형 종양에서 유래한 세포에서도 세포 사멸을 유도했다(M. van Trimpont, et al., Cancers, 14(4), 902. 2022). 상기 세포사멸이 ICD인지 여부를 테스트하기 위해 CT-26 세포(200 IU/mL)를 L-ASNase로 24시간 동안 처리했다(도 6). 상기 효소로 처리된 세포는 생존력이 점진적으로 감소했다(도 6a). 그러나, 유의한 CRT 노출은 세포에서 검출되지 않았다(도 6b). 이는 상기 효소가 ICD 이외의 경로를 통해 세포사멸을 일으켰다는 것을 나타낸다. 다음으로, ICD-유도 약물로 전처리된 세포에서 CRT3LP 및 CRT4LP의 항암 활성을 측정하였다(도 6c 및 6d). CT-26 세포의 생존력은 저농도의 CRT3LP 또는 CRT4LP(1 IU/mL) 존재 시 거의 영향을 받지 않았다(도 6c). 항암제에 의한 전처리는 세포 생존율의 현저한 감소를 야기하였다(GEM에서 45.1%, DOX에서 57.9%, MTX에서 62.3%). L-ASNase 또는 #DGRLP의 존재 하에서 세포 생존율은 항암제로 전처리된 세포에서 거의 감소하지 않았다. 그러나 CRT3LP 및 CRT4LP는 DOX 및 MTX로 전처리된 세포의 생존율을 2배 더 높게 감소시켰다. 대조적으로, 세포 생존력의 이러한 감소는 GEM으로 전처리된 세포에서 CRT3LP 및 CRT4LP에 의해 관찰되지 않았다. 상기 결과는 MC-38 세포에서도 동일하였다(도 6d). 상기 결과는 CRT3LP 및 CRT4LP만이 ICD-유도 항암제로 전처리된 종양 세포에 대해 효율적인 세포독성을 나타냄을 시사하는 것이다.
실시예 3: 항암제로 처리한 종양-보유 마우스에서 CRT3LP 및 CRT4LP의 효과
3-1: DOX로 처리된 CT-26 종양 보유 마우스
본 발명자들은 생체 내에서 항암 효과를 나타내는 PASylated L-ASNase의 최적 용량을 평가하기 위해, CT-26 종양-보유 BALB/c 마우스(그룹당 n = 3)를 항암제와 CRT3LP로 처리했다(도 7). 처리 절차는 도 7a에 도시한 바와 같이 수행하였다. 먼저, 다양한 양의 CRT3LP(5회, 2일 간격)를 DOX(10 mg DOX/kg/복강내 주사, 3회, 2일 간격)로 처리한 마우스에 정맥내 투여하였다(도 7b). 그 결과, DOX로 처리된 그룹의 종양은 처리되지 않은 종양보다 더 느리게 성장했다. DOX 단독으로 처리한 마우스보다 DOX + CRT3LP로 처리한 마우스에서 종양 성장이 더 크게 억제되었다. 8 IU의 CRT3LP로 처리된 마우스에서 종양 성장 억제는 12 IU로 처리된 마우스와 유사했으며 4 IU로 처리된 마우스보다 더 우수했다. 특히, 20 IU의 CRT3LP 처리는 12일까지 일부 마우스의 급사를 유발했다(3마리 마우스 중 2마리). 상기 결과를 바탕으로 본 발명자들은 CRT3LP의 최적 치료 용량이 8 IU/injection/mouse라고 결론지었다(도 7c).
3-2: CT-26 종양 모델에서 PASylated CRT- 표적 L-ASNase의 병용효과
다음으로, 본 발명자들은 CT-26 종양-보유 마우스에서 DOX와 조합된 PASylated CRT 표적 L-ASNase(8 IU/마우스)의 최적 치료 용량의 치료 효능을 조사했다(도 8). 마우스에 대한 처리는 설명된 일정대로 수행되었다(도 8a). 그 결과, DOX + L-ASNase 및 DOX + #DGRLP는 무처리보다 종양 성장을 더 잘 억제했지만, 억제 수준은 DOX 단독(DOX + PBS)으로 처리한 후 관찰된 것과 유사했다. L-ASNase 및 #DGRLP의 효소 활성이 DOX에 의한 항암 효과를 증가시키지 않음을 나타낸다. DOX + CRT3LP 및 DOX + CRT4LP는 DOX + L-ASNase 및 DOX + #DGRLP보다 더 높은 수준으로 종양 성장을 억제했다(도 8b, 8c 및 도 9). DOX + CRT3LP 및 DOX + CRT4LP 그룹의 마우스 생존 시간은 DOX + L-ASNase 및 DOX + #DGRLP 그룹보다 1주 이상 연장되었지만 종양이 완전히 제거되지는 않았다(도 8d). 상기 결과는 12일째 측정된 종양 중량에서도 확인되었다(도 8e 및 8f). 평균 종양 무게는 DOX 단독 그룹(DOX + PBS에서 304.8 mg, DOX + L-ASNase에서 263.3 mg 및 DOX + L-ASNase에서 278 mg)보다 DOX + L-ASNase 및 DOX + #DGRLP 그룹에서 약간 감소했다. 체중은 DOX + CRT3LP 및 DOX + CRT4LP 그룹에서 가장 적었다(DOX + CRT3LP에서 136.2 mg 및 DOX + CRT4LP 그룹에서 143.3 mg). 모든 결과는 종양 성장 속도 및 생존율 측정 결과와 일치했다. DOX-처리된 CT-26 종양에 대한 CRT3LP 및 CRT4LP의 표적화를 6일째에 공초점 현미경으로 관찰하였다(도 8g 및 8h). 항-His 태그 항체로 염색된 종양 조직에서, DOX + CRT3LP 및 DOX + CRT4LP 그룹에서는 형광 신호가 검출되었지만 무처리(PBS) 그룹 및 DOX + #DGRLP 그룹에서는 형광 신호가 검출되지 않았다. DOX + #DGRLP 및 DOX + PBS의 형광 강도는 배경 수준인 반면, DOX + CRT3LP 및 DOX + CRT4LP의 형광 강도는 현저히 증가했다(DOX+CRT3LP에서 7.1배 더 높음, DOX+CRT4LP에서 6.7배 더 높음). 상기 결과는 CRT3LP 및 CRT4LP가 생체 내에서 DOX로 처리된 종양 세포에서 ecto-CRT를 특이적으로 표적으로 한다는 것을 시사한다.
마지막으로, 혈청 샘플의 생화학적 분석을 12일째에 수행하였다(도 8i 내지 8k). 간 기능의 지표인 Ala aminotransferase(ALT) 및 Asp transaminase(AST) 수치는 고용량의 DOX 처리로 인해 증가했다(Y. Sadzuka, et al., nternational journal of pharmaceutics, 432(1-2), 42-49. 2012). 상기 수준은 본 발명에서 무-처리 그룹과 비교하여 모든 처리된 그룹에서 거의 증가하지 않았다(도 8i 및 8j). 이러한 데이터는 본 발명의 DOX 및 PASylated CRT-표적 L-ASNase의 용량이 임상 범위 내에 있음을 나타낸다. 나아가, CRT3LP 및 CRT4LP에 대한 1차 항체 반응을 평가하기 위해 혈청에서 L-ASNase에 대한 IgM 수준을 측정했다(도 8k). 모든 그룹에서 수준의 유의한 변화가 감지되지 않았다. 이는 PASylated CRT-표적 L-ASNase가 DOX와 결합된 처리의 초기 시점에서 항체 반응을 유도하지 않았음을 나타낸다. MTX와 결합된 PASylated CRT-표적 L-ASNase(8 IU/마우스)의 치료 효능을 CT-26 종양 보유-마우스에서 측정했다. 처리는 도 6a에 기재된 동일한 일정으로 MTX(2 mg/kg/injection, 3회 및 2일 간격)로 수행하였다. 그 결과, MTX-처리된 마우스에서 CRT3LP 및 CRT4LP의 효과는 DOX-처리된 마우스에서와 유사하였다(도 8l 내지 8n 및 도 10). MTX + L-ASNase 및 MTX + #DGRLP 마우스와 비교하여 MTX + CRT3LP 및 MTX + CRT4LP 마우스에서 현저한 종양 억제 효과 및 연장된 생존율이 관찰되었다.
더 나아가, 36일 동안 병용 요법 후 마우스의 체중 변화를 측정했다(도 8o). 체중은 모든 그룹에서 처리 후 약간 감소했으며 점차 회복되었다. 항암제 처리로 체중이 감소했다는 보고가 있다(K. Takayoshi, et al., Nutrition and cancer 69(3), 408-415. 2017). 따라서 병용 요법에서의 체중 감소는 아마도 항암제에 의해 야기되었을 것으로 PASylated CRT-표적 L-ASNase에 의해 야기된 것은 아니다. CRT3LP 및 CRT4LP의 치료 효과는 ICD를 유발하지 않는 항암제인 GEM으로 처리한 마우스에서도 측정되었다. CRT3LP 및 CRT4LP는 상기 약물로 처리된 마우스에서 종양 억제 효과를 향상시키지 않았다(도 8p). 이런 결과는 CRT3LP와 CRT4LP가 ICD 유도 항암제를 처리한 CT-26 종양-보유 마우스에서 부가적인 항암 효과를 나타냄을 시사하는 것이다.
3-3: MC-38 종양 모델에서 DOX 및 PASylated CRT-표적 L-ASNase와의 병용효과
마지막으로, 우리는 DOX로 처리된 MC-38 종양 보유 C57BL/6 마우스에서 CRT3LP 및 CRT4LP의 치료 효능을 평가했습니다(도 11). 모든 치료는 CT-26 종양-보유 마우스에서 사용된 것과 동일한 절차로 수행되었다. 다른 그룹에 비해 DOX + CRT3LP 및 DOX + CRT4LP 그룹에서 종양 성장 억제 및 증가된 생존율이 관찰되었다. 체중 변화의 결과는 CT-26 종양-보유 마우스에서 관찰된 것과 유사했다. 결론적으로 상기 결과는 PASylated CRT-표적 L-ASNase, CRT3LP 및 CRT4LP가 고형 종양에 대한 ICD-유도 항암제의 효능을 향상시켰다는 것을 시사하는 것이다.
실시예 4: 방사선 조사 및 CRT-표적 L-ASNase와의 병용효과
본 발명자들은 CT-26 및 MC-38 종양 세포에 방사선을 조사 후 본 발명의 융합단백질의 처리에 따른 병용효과를 조사하였다. 먼저, 방사선 조사 유도 후 세포 독성 및 ecto-CRT 전좌의 측정을 위해 CT-26 및 MC-38 세포(5x105)에 방사선을 조사 후(10 Gy, 3.3 Gy/min) 생존력 및 CRT 노출 여부를 분석한 결과 조사 처리 후 지정된 시간에 따라 세포 독성을 나타내었고 외부 CRT가 검출됨을 확인하였다(도 12). 또한, 상기 종양세포에 ICD 유도 방사선 조사 후 CRT 표적 L-ASNase의 결합을 조사하기 위해 48시간 동안 10 Gy 조사로 처리된 세포를 CRT-표적 L-ASNase로 염색하였고 항-His 항체 및 FITC-컨쥬게이션된 이차 항체로 염색한 후 형광현미경으로 관찰한 결과, 방사성 조사 후 CRT-표적 L-ASNase의 결합이 증가하는 것으로 나타났다(도 13). 아울러, 방사선 조사 후 본 발명의 융합단백질의 항암효능을 확인하기 위하여 CT-26 및 MC-38 세포를 24시간 동안 방사선 조사로 처리하였고 CRT 표적 L-ASNase(1 IU/mL)와 함께 24시간 동안 함께 배양한 다음 상기 세포를 현미경으로 관찰하고 상대적인 세포 생존력을 분석한 결과 본 발명의 CRT-표적 L-ASNase는 종양 세포에서 방사선 유발 세포 독성을 향상시키는 것으로 나타났다(도 14).
본 발명은 상술한 실시예를 참고로 설명되었으나 이는 예시적인 것에 불과하며, 당해 기술분야에서 통상의 지식을 가진 자라면 이로부터 다양한 변형 및 균등한 다른 실시예가 가능하다는 점을 이해할 것이다. 따라서 본 발명의 진정한 기술적 보호 범위는 첨부된 특허청구범위의 기술적 사상에 의하여 정해져야 할 것이다.
Claims (20)
- 인간 피브로넥틴 도메인 Ⅲ(Fn3)의 BC 루프 및 FG 루프 중 적어도 어느 하나 이상에 칼레티큘린(calreticulin) 특이적 결합 펩타이드가 삽입되어 상기 칼레티큘린에 특이적으로 결합하는 재조합 모노바디의 C-말단에 L-아스파라기나제에 연결된, 융합단백질.
- 제1항에 있어서,
상기 재조합 모노바디는 종양 항원 특이적 결합 펩타이드가 상기 BC 루프 및 FG 루프 모두에 삽입된, 융합단백질. - 제1항에 있어서,
상기 재조합 모노바디는 서열번호 9 또는 10으로 기재되는 아미노산 서열로 구성되는, 융합단백질. - 제1항에 있어서,
상기 재조합 모노바디는 인간 피브로넥틴 도메인 Ⅲ의 BC 루프에 서열번호 15로 기재되는 칼레티큘린 결합 펩타이드가 삽입되고 FG 루프에 서열번호 16으로 기재되는 칼레티큘린 결합 펩타이드가 삽입되거나, 상기 BC 루프에 서열번호 16으로 기재되는 칼레티큘린 결합 펩타이드가 삽입되고 상기 FG 루프에 서열번호 15로 기재되는 칼레티큘린 결합 펩타이드가 삽입된 것인, 융합단백질. - 제1항에 있어서
상기 융합단백질은 PAS(Pro-Ala-Ser) 반복체를 추가적으로 포함하는, 융합단백질. - 제5항에 있어서,
상기 PAS 반복체는 20 내지 500 단위체로 구성되는, 융합단백질. - 제5항에 있어서,
상기 PAS 반복체는 상기 융합단백질의 N-말단 또는 C-말단에 연결된, 융합단백질. - 제1항에 있어서,
서열번호 23 또는 24로 기재되는 아미노산 서열로 구성되는, 융합단백질. - 제1항 내지 제8항 중 어느 한 항의 융합단백질을 유효성분으로 포함하는, 고형암 치료용 약학적 조성물.

상기 항암 화합물은 면역원성 세포사멸 유도제(immunogenic cell death agent), 면역 검문소 억제제(immune checkpoint inhibitor), 유사분열 억제제, 대사길항제(antimetabolite), 호르몬제, 알킬화제(alkylating agent), 및 토포이소머라제 억제제(topoisomerase inhibitors)로 구성되는 군으로부터 선택되는, 약학적 조성물. - 제12항에 있어서,
상기 면역원성 세포사멸 유도제는 안트라사이클린계열 항암제, 탁산계열 항암제, 항-EGFR 항체, BK 채널작용제, 보르테조밉(Bortezomib), 강심성 배당체(cardiac glycoside), 사이클로포스마이드 계열 항암제, GADD34/PP1 저해제, LV-tSMAC, Measles 바이러스, 또는 옥살리플라틴인, 약학적 조성물. - 제13항에 있어서,
상기 안트라사이클린 계열 항암제는 다우노루비신(daunorubicin), 독소루비신(doxorubicin), 에피루비신(epirubicin), 이다루비신(idarubicin), 픽산트론(pixantrone), 사바루비신(sabarubicin), 또는 발루비신(valrubicin)인, 약학적 조성물. - 제11항에 있어서,

상기 고형암은 폐암, 위암, 간암, 골암, 췌장암, 담낭암, 담관암, 피부암, 두경부암, 피부 흑색종, 자궁암, 난소암, 직장암, 대장암, 결장암, 유방암, 자궁 육종, 나팔관 암종, 자궁내막 암종, 자궁경부 암종, 질 암종, 외음부 암종, 식도암, 후두암, 소장암 및 갑상선암으로 구성되는 군으로부터 선택되는, 약학적 조성물. - 치료적으로 유효한 양의 제1항 내지 제8항 중 어느 한 항의 융합단백질 또는 제9항 내지 제18항 중 어느 한 항의 약학적 조성물을 고형암에 걸린 개체에 투여하는 단계를 포함하는, 상기 개체의 고형암 치료방법.
- 방사선 조사와 동시에 또는 방사선 조사 전후 치료적으로 유효한 양의 제1항 내지 제8항 중 어느 한 항의 융합단백질 또는 제9항 내지 제18항 중 어느 한 항의 약학적 조성물을 고형암에 걸린 개체에 투여하는 단계를 포함하는, 상기 개체의 고형암 치료방법.
