Abstract
本发明提供了一种双特异性T细胞接合器分子。具体地,本发明提供了一种双特异性细胞接合器分子,其包括靶向uPAR的第一结合结构域、连接段和抗人CD3分子的第二结合结构域。第一结合结构域与uPAR阳性的靶细胞(肿瘤细胞、衰老细胞以及其他靶细胞)结合,第二结合结构域与T淋巴细胞结合,从而有效地将T细胞与靶细胞连接、使T细胞靶向并杀死靶细胞,具有在抗肿瘤、抗衰老等药物制备中应用的潜力。
Description
技术领域
本发明涉及生物医药领域。具体地说,本发明涉及双特异性T细胞接合器分子的制备及其应用。
背景技术
细胞毒性T淋巴细胞(CTL)在癌症治疗的免疫应答过程中起着关键作用。由于肿瘤细胞可通过自身分泌的相关细胞因子或与肿瘤微环境的相互作用营造出免疫抑制性环境,使肿瘤微环境中的CTL细胞功能失调。调节免疫细胞的一个策略就是利用双特异性细胞接合器使免疫细胞激活并杀死靶细胞,如肿瘤细胞。双特异性T细胞接合器(Bispecific T-cell Engager,BiTE),因其一端能特异性抗T细胞表达的抗原而得名,可引导T细胞杀死靶细胞,这个过程伴随着T细胞和靶肿瘤细胞之间形成瞬时的溶细胞突触形成,随后T细胞增殖和激活导致肿瘤细胞裂解。双特异性细胞接合器分子由两种结合不同靶蛋白的蛋白或多肽序列(抗体最为常见)连接而成。作为蛋白质大分子药物,双特异性T细胞接合器分子的大小至关重要,分子量越小,在生产经济性、分子稳定性、体内半衰期和组织渗透性等方面具有极大优势,分子量较小的纳米抗体在双特异性细胞接合器分子中的应用渐露头角。
尿激酶型纤溶酶原激活受体(uPAR)是一种GPI锚定的细胞膜受体,由三个同源结构域(DI,DII,DIII)组成,其主要功能是激活尿激酶(uPA)蛋白水解活性,调节细胞外基质(ECM)组分降解。uPA酶原和uPA都能结合uPAR,uPAR的一部分在与配体结合时被蛋白酶水解切割,产生可溶性uPAR(suPAR)。有大量文献证明了uPAR在大多数肿瘤进展中的重要性,在大多数的肿瘤细胞和肿瘤基质中过表达,包括乳腺癌,结直肠癌,前列腺癌,胰腺癌,卵巢癌,肺癌和胶质瘤等实体瘤,以及急性白血病和骨髓瘤等血液系统恶性肿瘤。然而正常组织仅在胸骨上皮中、单核细胞、巨噬细胞和嗜中性粒细胞的亚群检测到低uPAR表达。研究表明,(1)uPAR表达升高与不同类型癌症患者的不良结局(侵袭、转移和复发)有关;(2)uPAR与肿瘤代谢密切相关;(3)uPAR表达与非小细胞肺癌(NSCLC)和结直肠癌(CRC)患者的RAS基因突变相关;(4)uPAR是调节肿瘤细胞休眠和增殖之间转变的关键参与者;(5)uPAR与肿瘤细胞的多药耐药性(MDR)有关;(6)uPAR与肿瘤血管生成有关。
衰老(aging)是受遗传和环境因素共同影响的复杂过程,与永久性、渐进性的生理细胞衰退密切相关,是导致人口老龄化日渐严重的原因之一。衰老能够显著提高机体的患病率,即衰老相关疾病,如糖尿病、阿尔茨海默症、心血管疾病及神经退行性疾病等。个体衰老与衰老细胞既有联系,但又完全不同。衰老细胞是细胞处于稳定的细胞周期阻滞和分泌调节组织微环境的相关细胞因子。在生理上,衰老细胞是一种抑制肿瘤的机制,可以阻止癌前细胞的扩张,并在伤口愈合反应中发挥有益的作用。随着个体衰老,组织内会积累衰老细胞。衰老细胞在老年个体组织中的比例很少,但衰老细胞的积累会促进个体衰老及产生炎症环境、导致慢性组织损伤,如肝和肺纤维化、动脉粥样硬化、糖尿病和骨关节炎等疾病。在老年个体中消除衰老细胞可以改善这些病理症状,甚至促进长寿。已有研究证实,uPAR是细胞衰老的重要表面标志物,并且衰老细胞可分泌可溶性uPAR(suPAR)、作为衰老相关分泌表型(SASP)的一部分。
鉴于uPAR在肿瘤和衰老发生发展中的重要作用,已经有大量的研究探究uPAR在肿瘤和衰老,以及衰老相关疾病的诊断、预后方面的作用;以及将uPAR作为恶性肿瘤、衰老和衰老相关疾病的治疗靶标,探究靶向uPAR在相关治疗中的作用。
uPA蛋白含有N端的ATF结构域(Amino-Terminal Fragment)和C端的催化结构域,ATF结构域又可分为两个小的结构单元:较小的GFD(Growth Factor-like Domain)和较大的Kringle结构域(图1)。虽然GFD结构域是uPA结合uPAR所必需的,但已有研究多用ATF来研究uPA与uPAR的结合,以及用ATF结构域和EGF(epidermal growth factor)构建双特异性抗体靶向EGFR和uPAR、用ATF结构域构建靶向uPAR的CAR-T。ATF中的Kringle结构域可能与其他蛋白结合,从而影响双特异性抗体的靶点特异性,产生副作用。分子大小是蛋白质多肽药物在成本、稳定性等方面的重要考虑因素,更小的GFD结构域是否能用于构建双特异性T细胞接合器以引导T细胞攻击uPAR阳性的靶细胞并不清楚。
因此,本领域需要开发一种以uPAR为靶点双特异性T细胞接合器分子。
发明内容
本发明的目的就是提供一种以GFD(uPA的N端结构域)结合蛋白为靶点的双特异性细胞接合器分子。
在本发明的第一方面,提供了一种细胞接合器分子,其包括:
(a)第一结合结构域,所述第一结合结构域具有来源于uPA的GFD结构域的结构,并且所述第一结合结构域特异性结合GFD结合蛋白。
(b)第二结合结构域,所述第二结合结构域特异性结合CD3分子。
在另一优选例中,所述的细胞接合器分子为双特异性细胞接合器分子。
在另一优选例中,所述的细胞接合器分子特异性连接GFD结合蛋白和CD3分子。
在另一优选例中,所述的GFD结合蛋白指能够结合uPA的GFD结构域的蛋白,优选地为uPAR。
在另一优选例中,所述的第一结合结构域的氨基酸序列选自下组:
(i)如SEQ ID NO.2所示的氨基酸序列;和
(ii)在如SEQ ID NO.2所示序列的基础上,进行一个或多个氨基酸残基的替换、缺失、改变或插入,或在其N端或C端添加1至10个氨基酸残基,较佳地1至5个氨基酸残基,更佳地1至3个氨基酸残基,从而获得的氨基酸序列;并且所述获得的氨基酸序列与如SEQ IDNO.2所示序列具有≥85%(优选地≥90%,更优选地≥95%,例如≥96%、≥97%、≥98%或≥99%)的序列同一性;并且所获得的氨基酸序列与(i)所示的序列具有相同或相似的功能。
在另一优选例中,所述的第二结合结构域特异性结合人CD3。
在另一优选例中,所述的第二结合结构域具有选自下组的结构:单域抗体(sdAb)、单链抗体(scFv)、抗原结合片段(Fab片段)、配体,或其多聚体,或其组合。
在另一优选例中,所述的第二结合结构域具有来源于抗人CD3抗体的肽段。
在另一优选例中,所述的第二结合结构域包括VH段,所述VH段具有如下的互补决定区CDR:
SEQ ID NO.5所示的VH-CDR1,
SEQ ID NO.6所示的VH-CDR2,和
SEQ ID NO.7所示的VH-CDR3;和/或
所述的第二结合结构域包括VL段,所述VL段具有如下的互补决定区CDR:
SEQ ID NO.8所示的VL-CDR1,
SEQ ID NO.9所示的VL-CDR2,和
SEQ ID NO.10所示的VL-CDR3;
并且,上述CDR序列中任意一种氨基酸序列还包括任选地经过添加、缺失、修饰和/或取代至少一个氨基酸的,并使得含有所述衍生CDR序列的重链和轻链所构成的衍生抗体能够保留CD3结合亲和力的衍生序列。
在另一优选例中,所述VH段具有SEQ ID NO.4所示的氨基酸序列,或具有与SEQ IDNO.4所示的氨基酸序列至少有80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%或99%的序列同源性或序列相同性的氨基酸序列;和/或
所述VL段具有SEQ ID NO.9所示的氨基酸序列,或具有与SEQ ID NO.9所示的氨基酸序列至少有80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%或99%的序列同源性或序列相同性的氨基酸序列。
在另一优选例中,所述的第二结构域为单链抗体。
在另一优选例中,所述的第一结构域和/或第二结构域是单价的、或多价的(如二价的)。

在另一优选例中,所述的额外靶抗原包括免疫细胞表面抗原,优选地包括:CD2、CD4、CD8、CD11、CD18、或其组合。

具体实施方式
本发明人经过广泛而深入的研究,经过大量的筛选,首次开发了一种基于uPA构建的双特异性细胞接合器分子的制备及其应用。实验结果表明,本发明的靶向uPAR受体的BiTE具有对靶细胞杀伤效果显著、特异性抗肿瘤细胞的效果。在此基础上完成了本发明。
本发明所提供的双特异性细胞接合器分子由三部分组成:靶向肿瘤细胞表面抗原受体的第一结合结构域(配体段)、连接段和抗人CD3分子的第二结合结构域(抗体段),通过配体段连接肿瘤细胞,同时抗CD3抗体段连接T淋巴细胞,从而有效将效应性免疫细胞趋化到肿瘤局部,使机体更有效地发挥抗肿瘤效应。

为了更容易理解本发明,以下具体定义了某些技术和科学术语。除非在本文中另有明确定义,本文使用的所有其它技术和科学术语都具有本发明所属领域的一般技术人员通常理解的含义。在描述本发明之前,应当理解本发明不限于所述的具体方法和实验条件,因为这类方法和条件可以变动。还应当理解本文所用的术语其目的仅在于描述具体实施方案,并且不意图是限制性的,本发明的范围将仅由所附的权利要求书限制。
除非另外定义,否则本文中所用的全部技术与科学术语均具有如本发明所属领域的普通技术人员通常理解的相同含义。如本文所用,在提到具体列举的数值中使用时,术语“约”意指该值可以从列举的值变动不多于1%。例如,如本文所用,表述“约100”包括99和101和之间的全部值(例如,99.1、99.2、99.3、99.4等)。
本发明所用氨基酸三字母代码和单字母代码如J.biol.chem,243,p3558(1968)中所述。
如本文所用,术语“治疗”指给予患者内用或外用治疗剂,包含本发明的靶向uPAR受体的BiTE、表达本发明BiTE的T细胞及其组合物,所述患者具有一种或多种疾病症状,而已知所述治疗剂对这些症状具有治疗作用。通常,以有效缓解一种或多种疾病症状的治疗剂的量(治疗有效量)给予患者。
如本文所用,术语“任选”或“任选地”意味着随后所描述的事件或情况可以发生但不是必须发生。例如,“任选包含1-3个抗体重链可变区”是指特定序列的抗体重链可变区可以有但不是必须有,可以是1个、2个或3个。
本发明所述的“序列同一性”表示当具有适当的替换、插入或缺失等突变的情况下最佳比对和比较时,两个核酸或两个氨基酸序列之间的同一性程度。本发明中所述的序列和其具有同一性的序列之间的序列同一性可以至少为85%、90%或95%,优选至少为95%。非限制性实施例包括85%,86%,87%,88%,89%,90%,91%,92%,93%,94%,95%,96%,97%,98%,99%,100%。
纤溶酶原激活系统
纤溶酶原激活剂(PA)系统是一种与各种生理和病理生理过程相关的细胞外蛋白水解酶系统。PA系统的主要生理功能是将无活性纤溶酶原转化为纤溶酶,可由两种类型的PA介导:组织型纤溶酶原激活剂(tPA)和尿激酶型纤溶酶原激活剂(uPA)。PA系统最初被认为在纤维形成的凝块溶解中发挥作用。然而,后来的研究表明,PA系统在其他生物过程中具有额外的功能,例如胚胎发生,血管生成,细胞迁移,伤口愈合,炎症反应以及凋亡细胞死亡。在癌症中,PA系统在肿瘤生长,血管生成,肿瘤细胞侵袭,迁移和转移中起主导作用。
uPAR与肿瘤:几乎90%的人类癌症相关死亡是由肿瘤细胞的转移扩散引起的。转移背后的主要事件之一是细胞外基质(ECM)的蛋白水解、降解,以促进肿瘤细胞侵袭,迁移和归巢到远处器官。尽管在此过程中涉及几个蛋白酶系统,但大量证据表明uPA-uPAR系统是介导癌症侵袭和转移期间蛋白水解的核心参与者。uPA-uPAR系统的功能不仅限于蛋白水解,其在从肿瘤发生到转移的癌症的多个环节发挥更广泛的作用。
尿激酶型纤溶酶原激活受体(uPAR)是一种GPI锚定的细胞膜受体,由三个同源结构域(DI,DII,DIII)组成。其主要功能是聚焦于细胞表面负责细胞外基质(ECM)组分降解的尿激酶(uPA)蛋白水解活性。有大量文献证明了uPAR在大多数肿瘤进展中的重要性,在绝大数的肿瘤细胞和肿瘤基质中过表达,包括乳腺癌,结直肠癌,前列腺癌,胰腺癌,卵巢癌,肺癌和胶质瘤等实体瘤,以及急性白血病和骨髓瘤等血液系统恶性肿瘤。然而正常组织仅在胸骨上皮中、单核细胞、巨噬细胞和嗜中性粒细胞的亚群检测到低uPAR表达。研究表明,(1)uPAR表达升高与不同类型癌症患者的不良结局(侵袭、转移和复发)有关;(2)uPAR与肿瘤代谢密切相关;(3)uPAR表达与非小细胞肺癌(NSCLC)和结直肠癌(CRC)患者的RAS突变相关;(4)uPAR是调节单细胞肿瘤休眠和增殖之间转变的关键参与者;(5)uPAR与肿瘤细胞的多药耐药性(MDR)有关;(6)uPAR与肿瘤血管生成有关。鉴于uPAR在肿瘤发生发展中的重要作用,已经大量的研究:(1)探究uPAR在肿瘤的诊断、预后方面的作用;(2)将uPAR作为恶性肿瘤的治疗靶标,探究靶向uPAR在肿瘤治疗中的作用。
uPAR与衰老及相关疾病:衰老(aging)是受遗传和环境因素共同影响的复杂过程,与永久性、渐进性的生理细胞衰退密切相关,是导致人口老龄化日渐严重的原因之一。衰老能够显著提高机体的患病率,即衰老相关疾病,如糖尿病、阿尔茨海默症、心血管疾病及神经退行性疾病等。
细胞衰老的特点是稳定的细胞周期阻滞和调节组织微环境的衰老相关分泌程序。在生理上,衰老是一种抑制肿瘤的机制,可以阻止癌前细胞的扩张,并在伤口愈合反应中发挥有益的作用。病理上,衰老细胞的异常积累会产生炎症环境,导致慢性组织损伤,并导致肝和肺纤维化、动脉粥样硬化、糖尿病和骨关节炎等疾病。从机体受损组织中消除衰老细胞可以改善这些病理症状,甚至促进长寿。
已有研究证实,uPAR是衰老细胞重要的表面标志物。uPAR的一部分在与配体结合时被蛋白酶水解切割,产生可溶性uPAR(suPAR)。衰老细胞分泌suPAR,作为衰老相关分泌表型(SASP)的一部分。
因此,本发明的靶向uPAR的双特异性细胞接合器分子对实体瘤、血液瘤、衰老、衰老相关疾病或其它uPAR表达异常相关性疾病的治疗均具有重要价值。
uPA
尿激酶型纤溶酶原激活剂(尿激酶,uPA)是参与将无活性纤溶酶原转化为活性纤溶酶的关键丝氨酸蛋白酶,其又在转移级联的一系列事件中起作用、。它于1947年由MacFarlane和Pilling首次在尿液中发现,其中报告了一种新型的具有纤维蛋白溶解活性的“未命名”蛋白质。五年后,Sobel及其同事将这种“未命名”的蛋白质命名为“尿激酶”。进一步的研究表明,尿激酶还存在于血浆,和ECM的许多组织中。
pro-uPA(411个氨基酸)是uPA的前体结构,pro-uPA由三个结构域组成:与表皮生长因子(EGF)同源的生长因子样结构域(GFD),kringle结构域(KD)和丝氨酸蛋白酶结构域。GFD(从1到49个氨基酸)和KD(50-131个氨基酸)位于N-末端,而催化丝氨酸蛋白酶结构域(159-411个氨基酸)位于“C-末端”。在N端和C端间,有一个连接子区域(132-158个氨基酸)。一旦pro-uPA需要分泌,则位于连接子区域的Lys158和IIe159之间的肽键就裂解,以产生二硫键连接的双链形式的uPA。uPA的双链形式在Lys135和Lys136之间的肽键上进行另一轮蛋白水解之后,可以进一步切割成两部分:(1)具有丝氨酸蛋白酶结构域的催化活性低分子量形式的uPA和(2)由GFD和KD组成的无活性氨基末端片段(ATF)。研究表明,GFD结构域是uPA与uPAR发挥特异性结合的关键区域,并且已经证实无论是ATF片段、双链形式的uPA,亦或是pro-uPA都能以几乎一致的亲和力与uPAR特异性结合。
uPA与uPAR特异性结合依赖于其GFD结构域,本发明以GFD结构域作为双特异性细胞接合器分子的第一分子结合结构域去识别靶点(uPAR)阳性的细胞,从而发挥效应。
抗体
本发明的细胞接合器分子的第二结合结构域特异性结合CD3。该第二结合结构域可以是抗CD3的抗体、或抗体的抗原结合片段。
如本文所用,术语“抗体”或“免疫球蛋白”是有相同结构特征的约150000道尔顿的异四聚糖蛋白,其由两个相同的轻链(L)和两个相同的重链(H)组成。每条轻链通过一个共价二硫键与重链相连,而不同免疫球蛋白同种型的重链间的二硫键数目不同。每条重链和轻链也有规则间隔的链内二硫键。每条重链的一端有可变区(VH),其后是多个恒定区。每条轻链的一端有可变区(VL),另一端有恒定区;轻链的恒定区与重链的第一个恒定区相对,轻链的可变区与重链的可变区相对。特殊的氨基酸残基在轻链和重链的可变区之间形成界面。
如本文所用,术语“可变”表示抗体中可变区的某些部分在序列上有所不同,它形成了各种特定抗体对其特定抗原的结合和特异性。然而,可变性并不均匀地分布在整个抗体可变区中。它集中于轻链和重链可变区中称为互补决定区(CDR)或超变区中的三个片段中。可变区中较保守的部分称为构架区(FR)。天然重链和轻链的可变区中各自包含四个FR区,它们大致上呈β-折叠构型,由形成连接环的三个CDR相连,在某些情况下可形成部分β折叠结构。每条链中的CDR通过FR区紧密地靠在一起并与另一链的CDR一起形成了抗体的抗原结合部位(参见Kabat等,NIH Publ.No.91-3242,卷I,647-669页(1991))。恒定区不直接参与抗体与抗原的结合,但是它们表现出不同的效应功能,例如参与抗体的依赖于抗体的细胞毒性。
术语“抗体片段”或“抗原结合片段”用于指抗体的一部分,例如F(ab')2,F(ab)2,Fab',Fab,Fv,单链Fvs(scFv),单链抗体,二硫键连接的Fvs(sdFv),包含VL或VH结构域的片段,Fab表达文库产生的片段以及抗个体遗传型(anti-Id)抗体。不论结构如何,抗体片段都与完整抗体所识别的相同抗原结合。术语“抗体片段”包括DART和双抗体。术语“抗体片段”还包括任何包含免疫球蛋白可变区的合成蛋白或基因工程改造蛋白,其通过与特定抗原结合形成复合物而像抗体一样起作用。“单链片段可变区”或“scFv”是指免疫球蛋白的重链(VH)和轻链(VL)的可变区的融合蛋白。在一些方面,所述区结构域与10至约25个氨基酸的短接头肽连接。接头可富含甘氨酸以具有柔韧性和丝氨酸或苏氨酸以具有溶解度,并且可以连接VH的N末端或VL的C末端,反之亦然。尽管去除了恒定区并引入了接头,但是这种蛋白仍保留了原始免疫球蛋白的特异性。关于IgG,标准免疫球蛋白分子包含两个相同的分子量约为23,000道尔顿的轻链多肽和两个相同的分子量为53,000-70,000的重链多肽。四个链通常以“Y”构型通过二硫键连接,其中轻链从“Y”的口连接(bracket)重链并延伸通过可变区。
如上所述,可变区允许抗体选择性地识别并特异性结合抗原上的表位。即,抗体的VL结构域和VH结构域或抗体的互补决定区(CDR)子集(subset)结合以形成限定三维抗原结合位点的可变区。这种四元抗体结构形成了各Y构型的各臂的末端处存在的抗原结合位点。更具体地说,该抗原结合位点由VH和VL链中的每一个上的三个CDR(即HCDR1,HCDR2,HCDR3,LCDR1,LCDR2和LCDR3)限定。在某些情况下,例如某些免疫球蛋白分子衍生自骆驼科动物物种或基于骆驼科动物免疫球蛋白进行工程改造。或者,免疫球蛋白分子可以由仅不具有轻链的重链或仅不具有重链的轻链组成。
如本文所用,抗体、抗体片段或抗体结构域还包括其“变体”,所述的“变体”是指如下,抗体、抗体片段或抗体结构域:(1)与原始抗体、抗体片段或抗体结构域具有至少80%,85%,90%,95%,96%,97%,98%或99%的序列同一性,和(2)特异性结合至与原始抗体,抗体片段或抗体结构域特异性结合的相同靶标。应当理解,在以“至少x%相同”或“至少x%同一性”的形式表示序列同一性的情况下,这样的实施方案包括等于或高于下限的任何和所有数值百分比。此外,应当理解,在本申请中存在氨基酸序列的情况下,应将其解释为另外公开或包含与该氨基酸序列具有至少80%,至少85%,至少90%,至少95%,至少96%,至少97%,至少98%或至少99%的同一性。
本发明细胞接合器分子中所包含的抗体可以可以是具有免疫活性的抗体片段,如Fab或(Fab')2片段;抗体重链;抗体轻链。本发明所用的抗体优选地为单链抗体形式,所述单链抗体(scFv)含有抗体重链可变区、轻链可变区,但没有恒定区,并具有全部抗原结合位点的最小抗体片段。一般的,Fv抗体还包含VH和VL结构域之间的多肽接头,且能够形成抗原结合所需的结构。
术语“表位”或“抗原决定簇”是指抗原上免疫球蛋白或抗体特异性结合的部位。表位通常以独特的空间构象包括至少3,4,5,6,7,8,9,10,11,12,13,14或15个连续或非连续的氨基酸。表位可以是抗原上不连续的,由本发明抗体或抗原结合片段识别的三维空间位点。
术语“特异性结合”、“选择性结合”、“选择性地结合”和“特异性地结合”是指抗体对预先确定的抗原上的表位的结合。通常,抗体以大约小于10-7M,例如大约小于10-8M、10-9M或l0-10M或更小的亲和力(KD)结合。
本发明所用的抗体不仅包括完整的抗体,还包括具有免疫活性的抗体的片段或抗体与其他序列形成的融合蛋白。因此,本发明所用的抗体还包括抗体的片段、衍生物和类似物。
在本发明中,抗体可以是单特异性、双特异性、三特异性、或者更多的多重特异性。
如本文所用,术语“重链可变区”与“VH”可互换使用。
如本文所用,术语“可变区”与“互补决定区(complementarity determiningregion,CDR)”可互换使用。
术语“CDR”是指抗体的可变结构域内主要促成抗原结合的6个高变区之一。所述6个CDR的最常用的定义之一由Kabat E.A等人,(1991)Sequences of proteins ofimmunological interest.NIH Publication91-3242)提供。
在本发明的一个优选例中,所述的双特异性细胞接合器分子为单链多肽,其包括抗CD3单链抗体段、连接肽和Pep42配体段,其中抗CD3单链抗体为本领域常规的单链抗体,其包括重链可变区、轻链可变区。
本发明中,所述动物优选为哺乳动物,如鼠。
同时,本领域技术人员应当理解,虽然本发明BiTE的第一结合结构域优选为uPAR的配体,但该第一结合结构域也可以选择为特异性抗uPAR的抗体,只要其能够获得本发明的细胞结合效果。
双特异性细胞接合器分子
如本文所用,术语“双特异性细胞接合器分子”、“双特异性细胞接合器”、“细胞接合器”、“双特异性抗体”、“BiTE”可互换使用,均指本发明第一方面提供能够同时结合uPAR和CD3的细胞接合器分子。
双特异性细胞接合器分子由两种结合不同靶蛋白的蛋白或多肽序列(抗体最为常见)连接而成。本发明的BiTE的功能是由uPA配体段和CD3抗体段特异性基因序列决定的。本发明的BiTE可以同时结合uPAR和CD3,通过配体段连接肿瘤细胞,同时抗CD3抗体段连接T淋巴细胞,从而有效将效应性免疫细胞与肿瘤细胞连接,更有效地发挥抗肿瘤效应。利用本发明的VL、VH段基因或互补决定区(CDR)基因,可在利用原核和真核细胞的任何表达系统中改造和生产不同形式的基因工程细胞接合器分子。
如本文所用,术语“双特异性”是指包含至少两个具有不同结合特异性的结合结构域的分子。每个结合结构域都能够与靶分子特异性结合。在一些实施方式中,双特异性细胞接合器是具有两个或更多个肽的聚合物分子。在一些实施方式中,结合结构域包含与靶蛋白特异性结合的单域抗体、抗体的抗原结合片段、单链可变片段,或可变区,或CDR、或其组合。在一些实施方式中,结合结构域包含与靶蛋白特异性结合的配体或其片段。在一些实施方式中,结合结构域包含上述结构的组合。
本发明的细胞接合器分子的至少两个靶向结构域可选地由连接肽相连。优选的连接肽序列如SEQ ID NO.16所示,但不限于此。
在本发明中,本发明的BiTE还包括其保守性变异体,指与本发明BiTE的氨基酸序列相比,有至多10个,较佳地至多8个,更佳地至多5个,最佳地至多3个氨基酸被性质相似或相近的氨基酸所替换而形成多肽。这些保守性变异多肽最好根据表1进行氨基酸替换而产生。

本发明上述内容中,所述添加、缺失、修饰和/或取代的氨基酸数量,优选为不超过初始氨基酸序列总氨基酸数量的40%,更优选为不超过35%,更优选为1-33%,更优选为5-30%,更优选为10-25%,更优选为15-20%。
本发明上述内容中,更优选地,所述添加、缺失、修饰和/或取代的氨基酸数量,可以是1-7个,更优选为1-5个,更优选为1-3个,更优选为1-2个。
重组蛋白
本发明还提供一种重组蛋白,其包括本发明的细胞接合器分子。
本发明的重组蛋白可以包括本发明细胞接合器分子的单体、二聚体、或多聚体;或是多特异性(如三特异性)的细胞接合器。
所述重组蛋白的制备方法为本领域常规的制备方法。所述制备方法较佳地为:从重组表达该蛋白质的表达转化体中分离获得或者通过人工合成蛋白质序列获得。所述的从重组表达该蛋白质的表达转化体中分离获得优选如下方法:将编码所述蛋白质并且带有点突变的核酸分子克隆到重组载体中,将所得重组载体转化到转化体中,得到重组表达转化体,通过培养所得重组表达转化体,即可分离纯化获得所述重组蛋白。
核酸
本发明还提供了编码上述细胞接合器分子的多核苷酸分子。本发明的多核苷酸可以是DNA形式或RNA形式。DNA形式包括cDNA、基因组DNA或人工合成的DNA。DNA可以是单链的或是双链的。DNA可以是编码链或非编码链。编码成熟多肽的编码区序列可以与本发明细胞接合器的编码区序列相同或者是简并的变异体。如本文所用,“简并的变异体”在本发明中是指编码具有与本发明的多肽相同的氨基酸序列,但其编码区序列有差别的核酸序列。
编码本发明的成熟多肽的多核苷酸包括:只编码成熟多肽的编码序列;成熟多肽的编码序列和各种附加编码序列;成熟多肽的编码序列(和任选的附加编码序列)以及非编码序列。
术语“编码多肽的多核苷酸”可以是包括编码此多肽的多核苷酸,也可以是还包括附加编码和/或非编码序列的多核苷酸。
本发明还涉及与上述的序列杂交且两个序列之间具有至少50%,较佳地至少70%,更佳地至少80%相同性的多核苷酸。本发明特别涉及在严格条件下与本发明所述多核苷酸可杂交的多核苷酸。在本发明中,“严格条件”是指:(1)在较低离子强度和较高温度下的杂交和洗脱,如0.2×SSC,0.1%SDS,60℃;或(2)杂交时加有变性剂,如50%(v/v)甲酰胺,0.1%小牛血清/0.1%Ficoll,42℃等;或(3)仅在两条序列之间的相同性至少在90%以上,更好是95%以上时才发生杂交。并且,可杂交的多核苷酸编码的多肽与SEQ ID NO.4、SEQ ID NO.9所示的成熟多肽有相同的生物学功能和活性。
本发明的细胞接合器分子的核苷酸全长序列或其片段通常可以用PCR扩增法、重组法或人工合成的方法获得。一种可行的方法是用人工合成的方法来合成有关序列,尤其是片段长度较短时。通常,通过先合成多个小片段,然后再进行连接可获得序列很长的片段。此外,还可将其编码序列和表达标签(如6His)融合在一起,形成融合蛋白。
载体和宿主细胞
本发明还提供一种包含所述核酸的重组表达载体。
其中所述重组表达载体可通过本领域常规方法获得,即:将本发明所述的核酸分子连接于各种表达载体上构建而成。所述的表达载体为本领域常规的各种载体,只要其能够容载前述核酸分子即可。所述载体较佳地包括:各种质粒、粘粒、噬菌体或病毒载体等。
本发明还提供一种包含上述重组表达载体的重组表达转化体。
其中,所述重组表达转化体的制备方法为本领域常规的制备方法,较佳地为:将上述重组表达载体转化至宿主细胞中制得。所述的宿主细胞为本领域常规的各种宿主细胞,只要能满足使上述重组表达载体稳定地自行复制,且所携带所述的核酸可被有效表达即可。较佳地,所述宿主细胞为E.coli TG1或E.coli BL21细胞,或者HEK-293T或CHO细胞。将前述重组表达质粒转化至宿主细胞中,即可得本发明优选的重组表达转化体。其中所述转化方法为本领域常规转化方法,较佳地为化学转化法,热激法或电转法。
在本发明的优选实施方式中,可选用的载体包括:pCDH、pTOMO、pGEM、pELNS、pMSGV,或其组合。
在本发明的优选实施方式中,可选用的宿主细胞包括:T细胞、NK细胞、或其组合。
细胞接合器分子的制备
本发明细胞接合器分子或其片段的DNA分子的序列制备方法优选为将配体段和抗体段的编码序列融合在一起,形成单链多肽。此外,可以用常规技术,比如利用PCR扩增或基因组文库筛选等方法获得。
一旦获得了有关的序列,就可以用重组法来大批量地获得有关序列。这通常是将其克隆入载体,再转入细胞,然后通过常规方法从增殖后的宿主细胞中分离得到有关序列。
此外,还可用人工合成的方法来合成有关序列,尤其是片段长度较短时。通常,通过先合成多个小片段,然后再进行连接可获得序列很长的片段。
目前,已经可以完全通过化学合成来得到编码所述的本发明的细胞接合器(或其片段,或其衍生物)的DNA序列。然后可将该DNA序列引入本领域中已知的各种现有的DNA分子(或如载体)和细胞中。此外,还可通过化学合成将突变引入本发明蛋白序列中。
本发明还涉及包含上述的适当DNA序列以及适当启动子或者控制序列的载体。这些载体可以用于转化适当的宿主细胞,以使其能够表达蛋白质。
宿主细胞可以是原核细胞,如细菌细胞;或是低等真核细胞,如酵母细胞;或是高等真核细胞,如哺乳动物细胞。优选的细胞包括(但并不限于):T细胞。
通常,在适合本发明细胞接合器分子表达的条件下,培养转化所得的宿主细胞。然后用常规的免疫球蛋白纯化步骤,如蛋白A-Sepharose、羟基磷灰石层析、凝胶电泳、透析、离子交换层析、疏水层析、分子筛层析或亲和层析等本领域技术人员熟知的常规分离纯化手段纯化得到本发明的细胞接合器分子。
所得细胞接合器可用常规手段来鉴定。比如,其结合特异性可用免疫沉淀或体外结合试验(如放射性免疫测定(RIA)或酶联免疫吸附测定(ELISA))来测定。其结合亲和力例如可用Munson等,Anal.Biochem.,107:220(1980)的Scatchard分析来测定。
本发明的细胞接合器可在细胞内、或在细胞膜上表达、或分泌到细胞外。如果需要,可利用其物理的、化学的和其它特性通过各种分离方法分离和纯化重组的蛋白。这些方法是本领域技术人员所熟知的。这些方法的例子包括但并不限于:常规的复性处理、用蛋白沉淀剂处理(盐析方法)、离心、渗透破菌、超声处理、超离心、分子筛层析(凝胶过滤)、吸附层析、离子交换层析、高效液相层析(HPLC)和其它各种液相层析技术及这些方法的结合。
本领域常规的双特异性细胞接合器分子制备方法如以下两种;
1.首先利用PCR制备双特异性抗体基因,再将此基因克隆到表达载体pRB199转化到大肠杆菌菌株BL21(λDE3),制备包涵体(Inclusion bodies)。随后,在包涵体中加入6Mguanidine-HCl和DET(dithioerythritol)进行变性,然后使用到复性缓冲液进行100稀释,在4℃快速混合,随后在4℃孵育72h,使蛋白重新折叠。复性后,以1:10的比例加入0.1MTris和0.5M NaCl进行透析,重复三次后进行过滤(0.2μm),随后进行金属离子亲和层析。随后,采用快速蛋白液相色谱仪(BioLogic DuoFlow 10System;Bio-Rad)进行纯化,采用组氨酸标签融合蛋白质纯化柱分离。蛋白质是由咪唑逐步梯度洗脱,流速1毫升/分钟。将该产物进行过柱(Sartorius Stedim Biotech)处理,去掉分子量大于10000的蛋白,用PBS透析,过滤除菌。测定浓度后使用SDS/PAGE进行银染鉴定(参见PNAS,2013,110(1):270-275);
2.将含有双特异性抗体的慢病毒感染CHO细胞,待感染后培养72h,可以观察到CHO细胞有荧光表达。将感染成功的细胞株扩大培养。稳定表达双特异性抗体的CHO细胞能持续分泌表达。收集细胞上清进行蛋白纯化与浓缩。随后,采用快速蛋白液相色谱仪(BioLogicDuoFlow 10System;Bio-Rad)进行纯化,采用组氨酸标签融合蛋白质纯化柱分离。用样本的五倍体积的平衡缓冲液过镍柱,流速0.5-1ml/min。平衡后,将样品流过镍柱,流速0.5ml/min。用五倍体积的平衡缓冲液清洗镍柱,洗去背景蛋白,直到280nm液吸光度的洗出液为0。目的蛋白由咪唑洗脱,流速0.5ml/min。后用超滤管浓缩蛋白并置换盐溶液。测定浓度后使用western blot进行鉴定(参见Oncoimmunology,2015,4(4):e989776。)。
本领域技术人员能够对上述方法进行常规选择或等价改动,以制备或生产本发明的双特异性细胞接合器分子。
抗体-药物偶联物(ADC)
本发明所用的术语“抗体-药物偶联物(antibody-drug conjugate,ADC)”是指本发明细胞接合器分子与效应分子形成的偶联物。
典型地,所述抗体-药物偶联物包括所述细胞接合器分子、以及效应分子,所述细胞接合器分子与所述效应分子偶联,并优选为化学偶联。其中,所述效应分子优选为具有治疗活性的药物。此外,所述效应分子可以是毒蛋白、化疗药物、小分子药物或放射性核素中的一种或多种。
本发明细胞接合器分子与所述效应分子之间可以是通过偶联剂进行偶联。所述偶联剂的例子可以是非选择性偶联剂、利用羧基的偶联剂、肽链、利用二硫键的偶联剂中的任意一种或几种。所述非选择性偶联剂是指使效应分子和细胞接合器分子形成共价键连接的化合物,如戊二醛等。所述利用羧基的偶联剂可以是顺乌头酸酐类偶联剂(如顺乌头酸酐)、酰基腙类偶联剂(偶联位点为酰基腙)中的任意一种或几种。
细胞接合器分子上某些残基(如Cys或Lys等)用于与多种功能基团相连,其中包括成像试剂(例如发色基团和荧光基团),诊断试剂(例如MRI对比剂和放射性同位素),稳定剂(例如乙二醇聚合物)和治疗剂。细胞接合器分子可以被偶联到功能剂以形成细胞接合器分子-功能剂的偶联物。功能剂(例如药物,检测试剂,稳定剂)被偶联(共价连接)至细胞接合器分子上。功能剂可以直接地、或者是通过接头间接地连接于细胞接合器分子。
细胞接合器分子可以偶联药物从而形成抗体药物偶联物(ADCs)。典型地,ADC包含位于药物和细胞接合器分子之间的接头。接头可以是可降解的或者是不可降解的接头。可降解的接头典型地在细胞内环境下容易降解,例如在目标位点处接头发生降解,从而使药物从细胞接合器分子上释放出来。合适的可降解的接头包括,例如酶降解的接头,其中包括可以被细胞内蛋白酶(例如溶酶体蛋白酶或者内体蛋白酶)降解的含有肽基的接头,或者糖接头例如,可以被葡糖苷酸酶降解的含葡糖苷酸的接头。肽基接头可以包括,例如二肽,例如缬氨酸-瓜氨酸,苯丙氨酸-赖氨酸或者缬氨酸-丙氨酸。其它合适的可降解的接头包括,例如,pH敏感接头(例如pH小于5.5时水解的接头,例如腙接头)和在还原条件下会降解的接头(例如二硫键接头)。不可降解的接头典型地在细胞接合器分子被蛋白酶水解的条件下释放药物。
药物可以是任何细胞毒性,抑制细胞生长或者免疫抑制的药物。在实施方式中,接头连接细胞接合器分子和药物,而药物具有可以和接头成键的功能性基团。例如,药物可以具有可以和连接物成键的氨基,羧基,巯基,羟基,或者酮基。在药物直接连接到接头的情况下,药物在连接到细胞接合器分子之前,具有反应的活性基团。
有用的药物类别包括,例如,抗微管蛋白药物、DNA小沟结合试剂、DNA复制抑制剂、烷化试剂、抗生素、叶酸拮抗物、抗代谢药物、化疗增敏剂、拓扑异构酶抑制剂、长春花生物碱等。特别有用的细胞毒性药物类的例子包括,例如,DNA小沟结合试剂、DNA烷基化试剂、和微管蛋白抑制剂、典型的细胞毒性药物包括、例如奥瑞他汀(auristatins)、喜树碱(camptothecins)、多卡霉素/倍癌霉素(duocarmycins)、依托泊甙(etoposides)、美登木素(maytansines)和美登素类化合物(maytansinoids)(例如DM1和DM4)、紫杉烷(taxanes)、苯二氮卓类(benzodiazepines)或者含有苯二氮卓的药物(benzodiazepine containingdrugs)(例如吡咯并[1,4]苯二氮卓类(PBDs),吲哚啉苯并二氮卓类(indolinobenzodiazepines)和噁唑烷并苯并二氮卓类(oxazolidinobenzodiazepines))和长春花生物碱(vinca alkaloids)。
在本发明中,药物-接头可以用于在一个简单步骤中形成ADC。在其它实施方式中,双功能连接物化合物可以用于在两步或多步方法中形成ADC。例如,半胱氨酸残基在第一步骤中与接头的反应活性部分反应,并且在随后的步骤中,接头上的功能性基团与药物反应,从而形成ADC。
通常,选择接头上功能性基团,以利于特异性地与药物部分上的合适的反应活性基团进行反应。作为非限制性的例子,基于叠氮化合物的部分可以用于特异性地与药物部分上的反应性炔基基团反应。药物通过叠氮和炔基之间的1,3-偶极环加成,从而共价结合于接头。其它的有用的功能性基团包括,例如酮类和醛类(适合与酰肼类和烷氧基胺反应),膦(适合与叠氮反应);异氰酸酯和异硫氰酸酯(适合与胺类和醇类反应);和活化的酯类,例如N-羟基琥珀酰亚胺酯(适合与胺类和醇类反应)。这些和其它的连接策略,例如在《生物偶联技术》,第二版(Elsevier)中所描述的,是本领域技术人员所熟知的。本领域技术人员能够理解,对于药物部分和接头的选择性反应,当选择了一个互补对的反应活性功能基团时,该互补对的每一个成员既可以用于接头,也可以用于药物。
应用
本发明还提供了本发明双特异性细胞接合器分子、抗体偶联物ADC、重组蛋白、嵌合抗原受体(CAR)构建物和/或免疫细胞的用途,例如用于制备诊断制剂或制备药物。
较佳地,所述的药物是用于预防和/或治疗与uPAR表达或功能异常相关的疾病的药物。
本发明中,所述与uPAR表达或功能异常相关的疾病是本领域常规的与uPAR表达或功能异常相关的疾病,包括:肿瘤、衰老、衰老相关疾病,以及他们的组合。
本发明中,所述癌症为本领域常规的癌症,包括血液肿瘤和实体瘤。
检测用途和试剂盒
本发明的双特异性细胞接合器分子或其ADC可用于检测应用,例如用于检测样本,从而提供诊断信息。
本发明中,所采用的样本(样品)包括细胞、组织样本和活检标本。本发明使用的术语“活检”应包括本领域技术人员已知的所有种类的活检。因此本发明中使用的活检可以包括例如肿瘤的切除样本、通过内窥镜方法或器官的穿刺或针刺活检制备的组织样本。
本发明中使用的样本包括固定的或保存的细胞或组织样本。
本发明还提供了一种指含有本发明的细胞接合器(或其片段)的试剂盒,在本发明的一个优选例中,所述的试剂盒还包括容器、使用说明书、缓冲剂等。在优选例中,本发明的细胞接合器可以固定于检测板。
药物组合物
本发明还提供了一种组合物。在优选例中,所述的组合物是药物组合物,它含有上述的细胞接合器或其活性片段或其融合蛋白或其ADC或相应的免疫细胞,以及药学上可接受的载体。通常,可将这些物质配制于无毒的、惰性的和药学上可接受的水性载体介质中,其中pH通常约为5-8,较佳地pH约为6-8,尽管pH值可随被配制物质的性质以及待治疗的病症而有所变化。
配制好的药物组合物可以通过常规途径进行给药,其中包括(但并不限于):瘤内、腹膜内、静脉内、或局部给药。典型地,本发明所述的药物组合物的给药途径较佳地为注射给药或口服给药。所述注射给药较佳地包括静脉注射、动脉注射、肌肉注射、腹腔注射、皮内注射或皮下注射等途径。所述的药物组合物为本领域常规的各种剂型,较佳地为固体、半固体或液体的形式,可以为水溶液、非水溶液或混悬液,更佳地为片剂、胶囊、颗粒剂、注射剂或输注剂等。
本发明所述细胞接合器也可以是由核苷酸序列在细胞内表达用于的细胞治疗。
本发明所述的药物组合物是用于预防和/或治疗与uPAR和/或CD3表达或功能异常相关的疾病的药物组合物。
本发明的药物组合物含有安全有效量(如0.001-99wt%,较佳地0.01-90wt%,更佳地0.1-80wt%)的本发明上述的细胞接合器(或其偶联物)或者安全有效量(1×103-1×108个细胞/ml,更优地1×104-1×107个细胞/ml)的工程化免疫细胞以及药学上可接受的载体或赋形剂。这类载体包括(但并不限于):盐水、缓冲液、葡萄糖、水、甘油、乙醇、及其组合。药物制剂应与给药方式相匹配。本发明的药物组合物可以被制成针剂形式,例如用生理盐水或含有葡萄糖和其他辅剂的水溶液通过常规方法进行制备。药物组合物如针剂、溶液宜在无菌条件下制造。活性成分的给药量是治疗有效量,例如每天约1微克/千克体重-约5毫克/千克体重。此外,本发明的多肽还可与其他治疗剂一起使用。
在本发明的一个优选例中,本发明的多肽可以与其他治疗和/或预防癌症和/或癌症转移的治疗剂联用。
本发明中,较佳地,本发明所述的药物组合物还包括一种或多种药用载体。所述的药用载体为本领域常规药用载体,所述的药用载体可以为任意合适的生理学或药学上可接受的药物辅料。所述的药物辅料为本领域常规的药物辅料,较佳的包括药学上可接受的赋形剂、填充剂或稀释剂等。更佳地,所述的药物组合物包括0.01~99.99%的上述蛋白质和0.01~99.99%的药用载体,所述百分比为占所述药物组合物的质量百分比。
本发明中,较佳地,所述的药物组合物的施用量为有效量,所述有效量为能够缓解或延迟疾病、退化性或损伤性病症进展的量。所述有效量可以以个体基础来测定,并将部分基于待治疗症状和所寻求结果的考虑。本领域技术人员可以通过使用个体基础等上述因素和使用不超过常规的实验来确定有效量。
使用偶联物时,是将安全有效量的偶联物施用于哺乳动物,其中该安全有效量通常至少约10微克/千克体重,而且在大多数情况下不超过约50毫克/千克体重,较佳地该剂量是约10微克/千克体重-约20毫克/千克体重。当然,具体剂量还应考虑给药途径、病人健康状况等因素,这些都是熟练医师技能范围之内的。
检测样品中uPAR和/或CD3的方法、组合物
本发明还提供一种检测样品中uPAR和CD3(例如检测过表达uPAR和CD3的方法,包括如下的步骤:上述的细胞接合器与待检样品在体外接触,检测上述的细胞接合器与所述待检样品是否结合形成抗原-抗体复合物和/或受体-配体复合物即可。
所述的过表达的含义为本领域常规,指uPAR和CD3在待检样品中的RNA或蛋白质的过表达(由于转录增加、转录后加工、翻译、翻译后加工以及蛋白质降解改变),以及由于蛋白质运送模式改变(核定位增加)而导致的局部过表达和功能活性提高(如在底物的酶水解作用增加的情况下)。
本发明中,上述是否结合形成复合物的检测方式是本领域常规的检测方式,较佳地为流式细胞实验(FACS)检测。
本发明提供一种检测样品中uPAR和CD3的组合物,其包括上述的细胞接合器、重组蛋白、抗体偶联物、免疫细胞、或其组合作为活性成分。较佳地,其还包括上述的抗体的功能片段组成的化合物作为活性成分。
本发明的主要优点
本发明构建的双特异性细胞接合器分子同时靶向uPAR肿瘤抗原和T细胞,通过直接输注入体内,或利用体内细胞(如NK细胞、T细胞、CAR-T细胞等)携带并在体内持续表达该蛋白,从而使该双特异性细胞接合器分子在体内发挥杀伤作用的同时,伴随足量的T效应细胞,更优化了效应发挥的效率。其主要优点包括:
1)高靶向性:制备携带针对uPAR阳性肿瘤的双特异性细胞接合器分子,能够有效的结合肿瘤靶细胞的受体位点。
2)分子量小:本发明的优选双特异性细胞接合器一端为scFv结构,一端为仅有48个氨基酸的结构,在血液中不仅有利于结合肿瘤细胞与T细胞表面抗原,而且总蛋白分子量小,易生产。

在本实施例中,构建了双特异性细胞接合器分子的序列。
本实施例中使用的结合uPAR的GFD配体序列来源于uPA分子(参见Kumar AA,etal.Biomolecules.2022;12(2):152.)。
本实施例中使用的结合uPAR的SMB序列来自于VTN配体(参见Smith HW,MarshallCJ.Nat Rev Mol Cell Biol.2010Jan;11(1):23-36.;Deng G,et al.J CellBiol.1996Sep;134(6):1563-71;Huai Q,et al.Nat Struct Mol Biol.2008Apr;15(4):422-3.)。

本实施例中使用的抗CD3的抗体序列来源于抗体克隆OKT3的序列(参见ArakawaF,Kuroki M,Kuwahara M,Senba T,Ozaki H,Matsuoka Y,Misumi Y,Kanda H,WatanabeT.Cloning and sequencing of the VH and V kappa genes of an anti-CD3monoclonalantibody,and construction of a mouse/human chimeric antibody.JBiochem.1996Sep;120(3):657-62.doi:10.1093/oxfordjournals.jbchem.a021462.PMID:8902633),OKT3的VH和VL链如图2所示。本发明中所使用的抗CD3的抗体序列VH链序列来源参见GenBank BAA11539.1,VL链序列来源参见GenBank AAC28463.1。
抗CD3抗体VH和VL之间由linker2(SEQ ID NO.13)连接,形成OKT3的VH-linker2-VL单链结构,该单链结构在下文中统称OKT3,其氨基酸序列及核苷酸序列如表2所示。
使用linker1(SEQ ID NO.16)连接GFD和OKT3,构建本发明的细胞接合器分子(GFD-linker1-OKT3),其结构如图3(a)所示,其全长氨基酸序列及核苷酸序列如表2所示。
实施例2质粒构建
质粒构建采用慢病毒载体pCDH-CMV-MCS-EF1-Puro(参见Myeloid Leukemia.MolTher,2016.24(9):p.1615-26.),其图谱如图3(b)所示。目的基因采用直接合成方法合成,并在其3’端和5’端分别加上EcoR I和Swa I酶切位点和保护碱基,用EcoR I和Swa I酶切后与同样经EcoR I和Swa I酶切的载体连接。经过测序结果比对正确后,转化感受态大肠杆菌(Stbl3)。所有质粒均用QIAGEN公司的无内毒素大抽试剂盒抽提,并用Hind III酶切鉴定,鉴定结果如图3(c)所示。
实施例3病毒包装
在15cm培养皿中培养HEK-293T细胞用于病毒包装。待HEK-293T细胞汇合度在80%-90%左右进行转染,准备2ml OPTIMEM溶解的质粒混合物(核心质粒20ug、pCMVΔR8.910ug、PMD2.G 4ug);在另一离心管中2ml OPTIMEM以及68ul的lipo 8000。室温静置5min后,将质粒复合物加入脂质体复合物中,室温静置20min。将上述混合物滴加入HEK-293T细胞中,37℃孵育6小时后去除培养基。重新加入预热的完全培养基。转染24h后镜下观察转染情况,结果见图4;收集48小时和72小时病毒上清后,于4℃3000rpm离心20分钟。用0.45um滤膜过滤后,于25000rpm 4℃离心2.5小时进行病毒浓缩。浓缩的病毒用30ul病毒溶解液过夜溶解后,病毒滴度用QPCR检测。结果显示,病毒滴度达到要求。
实施例4分泌双特异性细胞接合器分子的T细胞制备
用Ficool分离液从人外周血中分离单核细胞,由RosetteSep Human T CellEnrichment Cocktail(Stemcell technologies)获得纯化的CD3+T细胞。T细胞用CD3/CD28磁珠进行活化(Life technology),再加入200U/ml的IL2(PeproTech),刺激培养48小时后进行病毒感染。慢病毒在lentiboost存在时按照MOI=100感染T细胞制备分泌双特异性单链细胞接合器分子的T细胞。感染24小时后更换培养基。慢病毒感染48小时后观察感染T细胞的能力,同时测定T细胞增殖和活力。
结果:慢病毒感染的48小时后,与对照组相比,T细胞增殖和活力(图6)未见明显改变。该结果说明感染表达本发明细胞接合器分子的病毒后对T细胞活性无明显影响。
实施例5携带luciferase的靶细胞构建
pTomo-CMV-Luciferase-IRES-Puro慢病毒包装步骤与实施例1.2中相同。病毒感染(MOI=100)PANC1、BXPC3、ASPC1、MCF7、MDA-MB-468、MDA-MB-231细胞后用Puromycin(1ug/ml)筛选2周,成功获得PANC1、BXPC3、ASPC1、MCF7、MDA-MB-468、MDA-MB-231-luciferase稳转细胞。
实施例6靶向uPAR双特异性细胞接合器分子筛选—杀伤试验
将上述慢病毒感染48小时后的各组T细胞分别与PANC1、ASPC1、BXPC3、MCF7、MDA-MB-468、MDA-MB-231肿瘤细胞按效靶比10:1(T细胞为效应细胞,浓度为2*105/mL,每孔100uL;靶细胞,浓度2*104/mL,每孔100uL,每孔100uL)共孵育48小时(细胞培养液:advance1640培养基(Gibco)+10%胎牛血清(Gibco)+1%青霉素、链霉素(Gibco))。
细胞杀伤效果使用promega荧光检测试剂盒检测,首先细胞用30ul 1*PLB裂解液处理细胞30分钟,每孔加入30ul底物后立即用BioTek酶标仪检测。细胞毒性杀伤细胞%=(1-含效应细胞时靶细胞荧光值/无效应细胞时靶细胞荧光值)×100%。
结果:如图7所示,与对照组(NTD、CD19-BiTE)相比,分泌SMB-BiTE和Clone 20-BiTE的T细胞杀伤肿瘤细胞的能力无明显差异,但分泌GFD-BiTE双特异性单链抗体特异性杀伤PANC1、ASPC1、BXPC3、MCF7、MDA-MB-468、MDA-MB-231细胞的能力显著上调。本实施例的结果证明,并非所有能够结合uPAR的序列构建的细胞接合器分子都能够达到杀伤肿瘤细胞的效果,本发明的GFD-BiTE具有出乎意料的优异抗肿瘤效果。
实施例7靶向uPAR双特异性细胞接合器分子TNF-α释放试验
在本实施例中,检测能分泌双特异性细胞接合器分子的T细胞与靶细胞共孵育情况下的细胞因子的释放情况。采用细胞杀伤实验中共孵育的细胞上清进行检测。
方法如下:取实施例6中共孵育的细胞上清,按照人TNF-alpha双抗夹心法ELISA检测试剂盒(proteintech)检测TNF-α。
用Sample Diluent PT6溶解标准品,并进行梯度稀释成500pg/ml、250pg/ml、125pg/ml、62.5pg/ml、31.2pg/ml、15.6pg/ml、0pg/ml的标准品。
每孔中加入100ul检测样本、标准品或Sample Diluent PT6(作空白孔),混匀后盖上封口膜37℃孵育120分钟。
然后依次按照以下步骤进行操作:
用1*Wash Buffer洗孔4次,每次停留1分钟;
每孔加入100ul 1×检测抗体,,盖上封口膜,37℃孵育1h;
重复步骤(1)
每孔加入100ul 1×HRP标记二抗,37℃孵育40分钟。
重复步骤(1)
每孔加TMB显色液100ul,37℃避光孵育15min。
每孔加入100ul Stop solution后混匀。
450nm处检测吸光值。
TNF-α分泌情况如图8所示,结果进一步证实了并非所有能够结合uPAR的序列构建的细胞接合器分子都能够识别靶蛋白,活化T细胞,释放大量的细胞因子以发挥杀伤效应,本发明的GFD-BiTE具有出乎意料的优异抗肿瘤效果。
实施例8双特异性细胞接合器分子GFD-BiTE蛋白的杀伤肿瘤细胞能力检测
用上述慢病毒感染CHO细胞后,收集细胞上清并浓缩纯化目的蛋白。分别与MDA-MB-468和MDA-MB-231、BXPC3肿瘤细胞按效靶比10:1共孵育(T细胞为效应细胞,浓度为1*104/mL,每孔100uL;肿瘤细胞为靶细胞,浓度1*103/mL,每孔100uL),并加入相应浓度的蛋白,共孵育24小时(细胞培养液:advance 1640培养基(Gibco)+10%胎牛血清(Gibco)+1%青霉素、链霉素(Gibco))。
细胞杀伤效果使用promega荧光检测试剂盒检测,首先细胞用30ul 1*PLB裂解液处理细胞30分钟,每孔加入30ul底物后立即用BioTek酶标仪检测。细胞毒性杀伤细胞%=(1-含效应细胞时靶细胞荧光值/无效应细胞时靶细胞荧光值)×100%。
结果:如图9所示,与对照CD19-BiTE相比,GFD-BiTE双特异性单链抗体对肿瘤细胞具有很好的杀伤作用,IC50均低于0.21ng/ml。
实施例9双特异性细胞接合器分子GFD-BiTE对衰老细胞的细胞毒性试验
HEL1-P16是衰老细胞系,稳转了绿色荧光蛋白(EGFP),将HEL1-P16细胞与本发明的GFD-BiTE蛋白及对照(NTD(仅有T细胞)与CD19-BiTE蛋白)以2:1的效靶比共培养20h后,镜下观察杀伤效果,并计数细胞个数以统计;同时收集细胞上清检测细胞因子IFN-γ的释放。结果如图10所示,与对照组相比,GFD-BiTE组对HEL1-P16细胞展现出了明显的杀伤效果(图10A,B)和更高的细胞因子IFN-γ释放(图10C)。

Claims (10)
1.一种细胞接合器分子,其特征在于,所述细胞接合器分子包括:
(a)第一结合结构域,所述第一结合结构域具有来源于uPA的GFD结构域的结构,并且所述第一结合结构域特异性结合GFD结合蛋白。
(b)第二结合结构域,所述第二结合结构域特异性结合CD3分子。
2.如权利要求1所述的细胞接合器分子,其特征在于,所述的第一结合结构域的氨基酸序列如SEQ ID NO.2所示。
3.如权利要求1所述的细胞接合器分子,其特征在于,所述的第二结合结构域包括VH段,所述VH段具有如下的互补决定区CDR:
SEQ ID NO.5所示的VH-CDR1,
SEQ ID NO.6所示的VH-CDR2,和
SEQ ID NO.7所示的VH-CDR3;和/或
所述的第二结合结构域包括VL段,所述VL段具有如下的互补决定区CDR:
SEQ ID NO.8所示的VL-CDR1,
SEQ ID NO.9所示的VL-CDR2,和
SEQ ID NO.10所示的VL-CDR3;
并且,上述CDR序列中任意一种氨基酸序列还包括任选地经过添加、缺失、修饰和/或取代至少一个氨基酸的,并使得含有所述衍生CDR序列的重链和轻链所构成的衍生抗体能够保留CD3结合亲和力的衍生序列。
4.如权利要求1所述的细胞接合器分子,其特征在于,所述细胞接合器分子从N端到C端具有选自下式的结构:
S-D1-L1-D2-T (I);和
S-D2-L1-D1-T (II),
式中,
各“-”独立地为连接肽或肽键;
S是无或信号肽序列;
D1是第一结合结构域;
L1是无或连接肽;
D2是第二结合结构域;
T是无或标记蛋白。
5.如权利要求1所述的细胞接合器分子,其特征在于,所述的细胞接合器分子氨基酸序列选自:
(i)如SEQ ID NO.20所示的氨基酸序列;
(ii)在如SEQ ID NO.20所示序列的基础上,进行一个或多个氨基酸残基的替换、缺失、改变或插入,或在其N端或C端添加1至30个氨基酸残基,较佳地1至10个氨基酸残基,更佳地1至5个氨基酸残基,从而获得的氨基酸序列;并且所述获得的氨基酸序列与如SEQ IDNO.14所示序列具有≥85%(优选地≥90%,更优选地≥95%,例如≥96%、≥97%、≥98%或≥99%)的序列同一性;并且所获得的氨基酸序列与(i)所示的序列具有相同或相似的功能。
6.一种重组蛋白,其特征在于,所述的重组蛋白包括如权利要求1所述的细胞接合器分子。
7.一种多核苷酸,其特征在于,所述多核苷酸编码选自下组的多肽:
(1)如权利要求1所述的双特异性细胞接合器分子;和
(2)如权利要求6所述的重组蛋白。
8.一种载体,其特征在于,所述载体含有如权利要求7所述的多核苷酸。
9.一种工程化的宿主细胞,其特征在于,所述宿主细胞含有如权利要求8所述的载体或基因组中整合有如权利要求7所述的多核苷酸。
10.一种偶联物,其特征在于,所述偶联物含有:
(a)如权利要求1所述的细胞接合器分子;和
(b)与所述细胞接合器分子偶联的偶联部分,所述偶联部分选自下组:可检测标记物、药物、或其组合。
