Abstract
Description
The present invention relates to a gold-naringin nanocomposite cluster for mercury sensing and fluorescence image sensor and a method for preparing the same, and more particularly, to a multicolor image, photostability and non-toxicity in vivo with high selectivity and sensitivity to mercury. It relates to a gold-naringin nanocomposite cluster having properties at the same time and a method for manufacturing the same.
Mercury may exist in various forms, such as inorganic compounds, ions in solution, and solids, and among them, the ionic form in aqueous solution has the most stable structure and is known as a material that causes fatal environmental pollution and human damage. Therefore, the development of a high-sensitivity detection sensor with high selectivity under various conditions is required.
Chem. As an example of a high-sensitivity sensor for mercury detection, research on the development of an optical sensor, particularly a fluorescent sensor (Anal. Chem. 2008, 80, 3716-3721), has been recently reported. In particular, as disclosed in Korean Patent No. 10-0862606, a high-sensitivity fluorescence-sensitive chemical sensor that is selective and highly sensitive by introducing a fluorescence-sensitive moiety to a compound capable of selectively binding to mercury ions is disclosed. However, there is a limitation in lowering the detection limit, and since a fluorescent material, which is a labeling material, must be introduced in the signal conversion step, there is a problem in that the direct detection of label-free and photo-bleaching phenomenon cannot be overcome.
Chem. Int. To solve this problem, the development of a biosensor using a local surface plasmon phenomenon using gold nanoparticles (Angew. Chem. Int. Ed. 2007, 46, 4093-4096; Gold Blutein 2008, 41/1, 37-41) reported in this academic circle.
In this study, we succeeded in developing a selective sensor through color change using aggregation of gold nanoparticles and biomaterials that can selectively bind mercury ions. There was a problem in that the stability of the sensor was lowered in terms of use.
A biosensor using such gold nanoparticles is difficult to inject into a living body due to the toxicity of the nanoparticles, so it is difficult to apply it as an in vivo sensor or an image sensor.

In another aspect, the present invention provides
금 전구체 수용액에 나린진, 글루타티온 및 혈청 알부민을 혼합하는 단계 ;mixing naringin, glutathione and serum albumin in an aqueous gold precursor solution;
It relates to a method for manufacturing a nanocomposite cluster for sensing mercury and a fluorescence image sensor, comprising the step of reacting after adjusting the pH of the aqueous solution to 8.5-10.
In another aspect, the present invention provides
상기 수은 감지 및 형광 이미지 센서용 나노복합 클러스터를 수은 함유 용액에 넣어 혼합하는 단계 ; mixing the mercury-sensing and fluorescent image sensor nanocomposite clusters into a mercury-containing solution;

It relates to a method for measuring mercury using a nanocomposite cluster for sensing mercury, comprising the step of calculating a mercury concentration from a detected extinction degree.
The nanocomposite cluster for mercury sensing and fluorescence image sensor of the present invention includes a gold cluster exhibiting green fluorescence properties, glutathione bound thereto, naringin surrounding the gold cluster, and the serum albumin.
Since the nanocomposite cluster of the present invention has a thiol group derived from glutathione that selectively binds to mercury ions (Hg 2+ ), the decrease in fluorescence intensity (quenching) of the gold cluster is very large compared to other metal ions, so the detection sensitivity and Selectivity is very good.
The nanocomposite cluster of the present invention has a size of 2 to 6 nm, and surrounds the gold cluster having toxicity with naringin, a biomolecule, and the serum albumin, so that the toxicity of gold can be blocked, so that mercury ions or cell images in living cells can be taken. .

In addition, the present invention may include injecting the nanocomposite cluster into living cells, measuring the extinction degree of fluorescence by irradiating the living cells with light, and calculating the mercury concentration from the detected extinction degree.
The method includes the steps of measuring fluorescence intensity by irradiating light to the solution or living cells, calculating the extinction degree of the nanocomposite cluster using the fluorescence intensity, and the extinction degree in a graph of extinction degree versus previously measured mercury concentration input to determine the mercury concentration.
The method may irradiate the solution with light having an excitation wavelength, for example, a wavelength of 394 nm.
The nanocomposite cluster exhibits the highest fluorescence intensity at pH 7.2.
When the nanocomposite clusters are mixed with a mercury-containing solution, the thiol groups (derived from glutaone) of the nanocomposite clusters bind to mercury ions.
The extinction degree may be calculated as F 0 -F/F 0 .
