NaOH 추출물을 제외한 3종류 추출물(50℃ 열수 추출물, 100℃ 열수 추출물 및 HCl 추출물)은 50%(v/v) 이상에서 세포독성이 나타났다(도 2a-2d). 100%(v/v) 추출물을 처리한 경우, 모든 실험군에서 세포독성이 나타났다. 배스 추출물을 일반적인 사료로 사용할 경우, 세포에 20%(v/v) 이상으로 처리될 가능성은 없으므로 세포 안전성은 매우 높은 것으로 사료된다. 50℃ 열수 추출물과 NaOH 추출물에서 25%(v/v) 처리군까지 강한 세포 증식 효과를 보여주었다.Except for the NaOH extract, the three extracts (50°C hot water extract, 100°C hot water extract and HCl extract) showed cytotoxicity at 50% (v/v) or more ( FIGS. 2a-2d ). 2-1. 활성산소 제거 활성2-1. Active oxygen removal activity
에탄올에 0.25 mM DPPH를 녹여 최종 0.0019 g/20 mL의 농도를 만들고, 0.22 μm 필터 주사기로 필터링하였다. 4종류 추출물을 3차 증류수로 희석하여 농도별(0, 1, 10, 50, 100%)로 준비하였다. 음성대조군으로 증류수, 양성대조군으로 아스코르브산을 사용하였다. 튜브에 DPPH 시약을 350 μl씩 분주하고 대조군과 추출물 샘플을 동일한 양으로 분주하였다. 샘플의 보유 색을 제거해주기 위해 증류수를 350 μl씩 분주한 다음, 추출물 샘플을 동일한 양으로 분주하였다. 볼텍싱 후, 상온에서 20 분간 반응시키고 96웰 플레이트에 각 200 μl씩 3반복으로 분주하였다. ELISA reader기를 사용해 흡광도 517 nm에서 흡광도를 측정하였다. 0.25 mM DPPH was dissolved in ethanol to make a final concentration of 0.0019 g/20 mL, and filtered with a 0.22 μm filter syringe. Four types of extracts were diluted with tertiary distilled water and prepared by concentration (0, 1, 10, 50, 100%). Distilled water was used as a negative control and ascorbic acid was used as a positive control. 350 μl of DPPH reagent was dispensed into the tube, and the same amount of control and extract samples were dispensed. In order to remove the retained color of the sample, 350 μl of distilled water was dispensed, and then the extract sample was dispensed in the same amount. After vortexing, the reaction was performed at room temperature for 20 minutes, and 200 μl each was dispensed in 3 repetitions in a 96-well plate. Absorbance was measured at 517 nm using an ELISA reader.

7 mM ABTS와 2.4 mM potassium persulfate를 3차 증류수에 녹인 후 혼합하고 빛을 차단한 상태에서 상온에 12-16시간 동안 반응시켰다. ELISA reader기에서 흡광도 414 nm로 측정하여 1.5 ± 0.03의 수치가 될 때까지 3차 증류수로 희석하였다. 추출물 샘플 농도별로 3차 증류수로 희석하여 준비하였다. 7 mM ABTS and 2.4 mM potassium persulfate were dissolved in tertiary distilled water, mixed, and reacted at room temperature for 12-16 hours while blocking light. The absorbance was measured at 414 nm in an ELISA reader and diluted with tertiary distilled water until a value of 1.5 ± 0.03. The extract was prepared by diluting with tertiary distilled water for each sample concentration.
4종류 추출물을 3차 증류수로 희석하여 농도별(0, 1, 10, 50, 100%)로 준비하였다. 음성대조군으로 증류수, 양성대조군으로 아스코르브산을 사용하였다. 튜브에 DPPH 시약을 600 μl씩 분주하고 추출물 샘플을 200 μl 분주하였다. 샘플의 보유 색을 제거해주기 위해 증류수를 600 μl씩 분주한 다음, 추출물 샘플을 200 μl 분주하였다. 볼텍싱 후, 상온에서 10 분간 반응시키고 96웰 플레이트에 각 200 μl씩 3반복으로 분주하였다. DPPH 분석과 다르게 모든 샘플에서 농도가 증가할수록 항산화 효과가 증가하는 확인하였다(도 3). 4종류 추출물은 50% 농도까지 아스코르브산의 활성보다 높게 나타났으며, 최고농도 100%에서는 아스코르브산의 활성과 비슷한 효과를 나타냈다. 결론적으로 배스 추출물을 우수한 항산화 효과를 나타내기 때문에 면역력, 노화방지 등에 탁월한 효과를 줄 수 있을 것으로 사료된다. THP-1 세포를 24웰 플레이트에 시딩하고 10% 또는 20% 농도로 배지에 희석 추출물 샘플 및 LPS(1 μg/ml)를 처리하고 24시간 동안 배양하였다. 상층액을 모은 후 Griess reagent와 1:1로 혼합하여 상온에서 15분 동안 배양하고, 540 nm에서 흡광도를 측정하였다. THP-1 cells were seeded in a 24-well plate, treated with a diluted extract sample and LPS (1 μg/ml) in the medium at a concentration of 10% or 20%, and cultured for 24 hours. Human 유래 Monocyte 세포주인 THP-1 세포를 사용하여 면역 활성화에 의해 생성, 분비되는 Nitric Oxide(NO) 양을 정량하는 방법으로 배스를 포함하는 사료의 면역활성화 효과를 확인하였다. 배스에 대한 4종류 추출물을 세포 독성이 없는 농도로 희석하고, LPS(1 μg/ml)과 함께 24시간 배양하였다. 추출방법과 무관하게 모두 대조군 대비 NO값이 유의 있게 증가하는 것으로 확인되었다. 따라서, 배스 추출물은 면역활성화를 유도하는 것으로 판단된다(도 4). A cosmetic composition for antioxidants comprising a bath extract.
The method of claim 1, wherein the extract is water, HCl solution, NaOH aqueous solution, a linear or branched alcohol having 1 to 4 carbon atoms, acetone, ethyl acetate, butyl acetate, 1,3-butylene glycol, hexane and diethyl ether. An extract obtained by extraction with one or more solvents selected from the group consisting of, an antioxidant cosmetic composition.
