Abstract
본 발명은 epcam 유전자 변이 제브라피쉬 및 이의 용도에 관한 것으로, epcam(epithelial cell adhesion molecule) 유전자 변이 제브라피쉬를 제작하는 방법, 담도 손상 진단 또는 모니터링하기 위한 정보 제공 방법 및 담도 손상 관련 질환의 치료제의 스크리닝 방법을 제공한다. 본 발명에 따르면, 본 발명의 epcam 변이 유전자 제브라피쉬 모델에서 간 손상 후 담도 재생시 담도가 비정상적으로 재생되는 것을 발견하였다. 따라서, 본 발명의 제브라피쉬 모델을 이용하는 경우, 부작용이 적으면서도 효능이 우수한 담도 손상 관련 질환 치료제를 효과적으로 스크리닝할 수 있으며, 담도 손상 관련 질환의 진단 또는 그 분자적 기전에 관한 연구과정 등에 널리 이용될 수 있다.
Description
본 발명은 epcam 유전자 변이 제브라피쉬 및 이의 용도에 관한 것이다.
간(liver)은 동물의 주요 장기로 대표적인 기능으로는 외부의 독소를 분해하는 해독 작용, 단백질 합성, 양분 저장, 쓸개즙, 요소 등을 생성한다. 간은 동물의 생존에 꼭 필요하며, 현재는 오랜 기간 간의 손실을 메울 수 있는 방법은 없다. 간은 모든 내장 기관 중 가장 크다. 간은 물질대사를 주로 담당한다. 또한, 글리코겐 저장, 적혈구 분해, 혈청 단백질 합성, 호르몬 생산, 해독작용 등 기타 여러 역할을 한다. 간은 배에서 복부-골반 부분의 횡격막 아래에 놓여 있다. 간은 지질의 유화로 소화를 돕는 알칼리성의 혼합물인 담즙을 생산한다. 간의 고도로 전문화된 조직들은 작고 복잡한 분자들의 합성과 분해와 같은 매우 다양한 대량의 생화학적 작용을 조절하며, 이들은 평상시의 주요 기능을 위해 필요하다.
담도(Bile ducts)는 간에서 담낭(gall bladder)을 거쳐 소장까지 담즙을 운반하는 작은 관(담관)으로 구성된다. 담즙(bile)은 녹황색의 농후한 점액 즙으로 간에서 생성되며 간에 뿌리같이 뻗어있는 미세관들로 이루어진 담도를 통해 담낭에 모여 저장되고 얼마 뒤 십이지장으로 분비된다. 담도는 간과 쓸개에서 즙을 받아 십이지장으로 보내는 관을 통틀어 이르는 말이다. 상황에 따라 간관을 통한 직접적인 분비도 가능하다. 담즙은 콜레스테롤과 지방, 지용성 비타민을 장에서 쉽게 흡수되도록 만들어 소화를 도와준다. 뿐만 아니라 담즙은 특정 폐기물(주로 빌리루빈과 초과 콜레스테롤)과 약물의 부산물을 체내에서 배출하는 데도 도움이 된다. 담즙의 주 성분은 담즙색소(bile pigment)와 담즙산염(bile acid)이다.
현재까지 간의 담즙의 흐름은 아래의 원인 때문에 차단될 수 있다고 보고 되고 있다. 담낭에서 담관으로 통과하는 담석, 담낭 수술 시 손상되는 담관, AIDS 관련 감염, 일차성 경화성 담관염과 같은 요인들로 인한 담관 협착, 췌장을 통과하는 담관을 협착시킬 수 있는 췌장 질환, 췌장, 담낭 또는 담관 종양, 아시아에서 기생충 감염 등으로 담도의 이상을 야기할 수 있다. 담관이 차단되면 담낭에 염증이 생겨 담낭염을 야기하며, 결석이 없는 담도성 통증(무결석 담도성 통증) 또한 발생할 수 있다. 담석은 초음파로 진단이 가능하나 담낭 수술, 간의 손상 등으로 손상된 담관을 진단하기 위한 방법은 개발되지 않았기 때문에 효과적이면서도 안전한 진단 방법 및 담관 손상 치료제를 개발하기 위한 치료 물질의 스크리닝 방법의 개발이 시급하다.
현재까지 많은 간의 담도 재생에 관여하는 물질이 발견되고 연구가 되어왔으나, 간의 손상에 반응하여 담도의 생리적 기능이 어떻게 재생이 일어나고 조절되는지에 대해서는 많이 알려져 있지 않다.
뿐만 아니라, 포유류 동물 모델에서는 담도 재생에 관한 연구가 상대적으로 많이 이루어진 반면, 제브라피쉬 동물모델에서는 거의 이루어지지 않았다.
또한, 유해한 자극에 의한 간의 손상과 담도의 재생에 대한 연구가 많이 이루어져 왔으나, 여전히 담도의 손상의 진단 및 담도 재생에 관여하는 신약 개발은 많이 이루어지지 않았고, 많은 비용과 시간이 걸리고 있기 때문에 최근 이를 줄이기 위한 시도가 지속되고 있으며, 제브라피쉬가 하나의 대안으로 주목받고 있다.
이러한 제브라피쉬(zebrafish)는 열대성 어류의 한 종류로 실험 동물모델로 널리 이용되고 있다. 제브라피쉬는 척추동물로 쥐, 랫트와 같은 동물모델을 대체할 수 있음은 물론 이들 동물에 비해 배아가 투명하고 48시간의 짧은 발생기간을 가져 실시간 관찰이 가능하다. 특히, 기관 특이적으로 형광단백질과 같은 생물표지자를 발현시키면 생체에서 실시간으로 특정기관의 관찰이 가능하고 배아 단계에서는 세포 수준의 추적이 가능한 장점이 있다. 더불어, 체외수정을 하기 때문에 수정란의 난황으로 손쉽게 유전 물질을 주입하여 유전자를 조작하는 것이 용이하다.
즉, 대용량의 치료 물질을 스크리닝하기 위해서는 동물 사육 및 관리 비와 분석 비용이 많이 들고 시간이 많이 소요되는 동물들보다는 경제적, 시간적 효용성이 좋은 제브라피쉬가 훨씬 편리하다.
이에 따라 현재 제브라피쉬에서도 포유류와 같이 다양한 간 재생 모델이 확립되어 연구가 활발히 진행되고 있다. 제브라피쉬에서 간 재생에 관한 생리학은 잘 확립되어 있다. 특히, 간 재생시 담도의 재생 기작은 포유류와 거의 유사한 것으로 알려져 있다.
한편, epcam(Epithelial cell adhesion molecule) 유전자는 제브라피쉬 동물모델에서 간 발생에 관여하는것으로 알려져 있고(Lu et al., 2013, Dev. Cell.), 포유류의 간 재생시에 전구세포에서 발현되는 단백질로 알려져 있다(Okabe et al., 2009, Development). 그러나, 간의 손상 후 담도 재생에서 EPCAM의 기능은 알려져 있지 않다.
따라서, 생체 내 기능을 모사할 수 있으며 대용량 스케일의 스크리닝이 가능한 질환 동물 모델을 개발할 필요성이 있으며, 이러한 필요에 따라 제브라피쉬의 epcam 유전자 변이를 이용한 모델을 적용함으로써 효과적이면서도 안전한 진단 방법 및 담관 손상 치료제를 개발하기 위한 치료 물질의 스크리닝 방법을 개발할 수 있다.
그러나, 현재까지 간의 심각한 손상 후 재생 과정 중에 EPCAM 유전자를 대상으로 넉다운시킨 제브라피쉬 동물 모델에서 담관 형성의 영향에 대한 보고는 없다.
이러한 상황하에서, 본 발명자들은 담도 손상을 효과적이면서도 안전하게 진단하고 이를 치료할 수 있는 치료 물질을 스크리닝하는 방법을 개발하기 위하여 예의 노력하였다. 그 결과, 본 발명자들은 epcam 유전자가 담도 재생에 중요한 영향을 미치는 것을 발견하였다. 이를 기초로, 간 손상을 유발하는 특정 조건에 따라 담도 손상 증상을 모사하는 epcam 변이 유전자 제브라피쉬 돌연변이체 모델을 개발하였고, 상기 모델을 이용하여 담도 손상을 효과적이면서도 안전하게 진단하고, 담도 손상 관련 질환 타겟 치료 물질을 신속하고 효과적으로 스크리닝할 수 있음을 규명함으로써 본 발명을 완성하였다.
따라서, 본 발명의 일 목적은 epcam 유전자 변이 제브라피쉬 모델을 제작하는 방법 및 이에 따라 제작된 제브라피쉬 모델을 제공하는데 있다.
또한, 본 발명의 다른 목적은 상기 제브라피쉬 모델을 이용하여 담도 손상을 진단 또는 모니터링하기 위한 정보 제공 방법을 제공하는데 있다.
또한, 본 발명의 또 다른 목적은 상기 제브라피쉬 모델을 이용하여 담도 손상 관련 질환 치료제를 스크리닝하는 방법을 제공하는데 있다.
본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 설명 및 청구범위에 의해 보다 명확하게 된다.

이하, 본 발명에 대해 상세히 설명한다.
본 발명의 일 양태에 따르면, 본 발명은 다음 단계를 포함하는, epcam(epithelial cell adhesion molecule) 유전자 변이 제브라피쉬를 제작하는 방법 및 이에 따라 제작된 epcam 유전자 변이 제브라피쉬를 제공한다:
(a) 간-특이 형광 단백질 및 NTR(nitoreductase)을 발현하는 형질전환 제브라피쉬와 담도 세포(biliary epithelial cells)의 핵-특이 형광 단백질을 발현하는 형질전환 제브라피쉬를 교배하여, 형질전환된 제브라피쉬를 제작하는 단계;
(b) 서열번호 2로 표시되는 epcam 변이 유전자가 도입된 형질전환 제브라피쉬; 또는 서열번호 1로 표시되는 epcam 정상 유전자가 결실된 epcam 녹아웃 형질전환 제브라피쉬를 제작하는 단계;
(c) 상기 (a) 단계의 형질전환 제브라피쉬 및 (b) 단계의 형질전환 제브라피쉬를 교배하는 단계; 및
(d) 상기 (c) 단계에서 획득된 형질전환 제브라피쉬에 MTZ(metronidazole)를 처리하여 간 손상에 의한 담도 세포 손상을 유발시키는 단계.
본 발명의 바람직한 구현예에 따르면, 상기 epcam(epithelial cell adhesion molecule) 유전자 변이 제브라피쉬는, MTZ 처리로 간 손상에 의한 담도 세포 손상이 유발되었을 때, 일정 시간 경과 후 간은 정상적으로 재생되는 반면, 담도는 비정상적으로 재생되는 것을 특징으로 한다.
본 명세서에서 사용되는 용어 “간 특이적”, “간-특이" 또는 "간세포 특이적"은 간 이외의 기관에서는 발현되지 않거나 낮은 정도로 발현되는 반면, 간에서는 매우 높은 정도로 발현되는 특징을 의미하며, 본 발명의 간 특이적 발현은 간 세포-특이적 발현을 유도하는 fabp10a 프로모터를 활용한 형광 단백질에 의한 것일 수 있으나, 이에 한정되는 것은 아니다.
본 명세서에서 사용되는 용어 "담도 특이적", "담도 세포-핵 특이" 또는 "담도 세포 특이적"은 담도 이외의 기관에서는 발현되지 않거나 낮은 정도로 발현되는 반면 담도에서는 매우 높은 정도로 발현되는 특징을 의미하며, 본 발명의 담도 특이적 발현은 간에서 담도 세포의 핵 특이적으로 발현되는 형광 단백질에 의한 것일 수 있으나, 이에 한정되는 것은 아니다.
본 명세서에서 사용되는 용어, "형질전환"은 플라스미드 형태의 DNA를 제브라피쉬 발생배에서 분열하는 세포에 물리화학적으로 직접 도입하여 외래 유전자를 발현시키는 것을 말하며, 본 발명에서 용어, "형질전환 제브라피쉬"는 목적 뉴클레오타이드 서열의 통합에 의하여 유전체(genome)가 변형된 제브라피쉬를 말한다.
상기 제작 방법에 있어서, (a) 단계의 형광 단백질은 생체 안에서 빛을 내는 단백질로, 적색(red) 형광단백질, 녹색(green) 형광단백질, 청색(blue) 형광단백질 등을 포함하며, 바람직하게는 서열번호 7 또는 8로 표시되는 염기서열을 포함하는 벡터의 도입으로 인해 발현되는 형광단백질일 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 일 실시예에서, 상기 (a) 단계의 간-특이 형광 단백질 및 NTR(nitoreductase)을 발현하는 형질전환 제브라피쉬는 서열번호 7로 표시되는 염기서열을 포함하는 벡터에 의해 형질전환된 것으로, 간세포에 특이적 발현을 유도하는 프로모터인 fabp10a 프로모터에 작동적으로 연결된(operatively linked) CFP-NTR이 발현될 때, 형광 단백질도 함께 발현될 뿐만 아니라, MTZ을 처리하면, 간 및 담도 손상이 유발된다. 또한, 상기 (a) 단계의 담도 세포(biliary epithelial cells)의 핵-특이 형광 단백질을 발현하는 형질전환 제브라피쉬는 서열번호 8로 표시되는 염기서열을 포함하는 벡터에 의해 형질전환된 것으로, 담도 세포 핵에 특이적 발현을 유도하는 프로모터인 Tp1(Notch-responsive elements) 프로모터에 작동적으로 연결된(operatively linked) H2B-mCherry이 발현될 때, 담도 세포의 위치 및 형태를 나타내는 형광 단백질도 함께 발현된다.
따라서, 이들을 교배하여 획득한 형질전환 제브라피쉬 Tg(fabp10a:CFP-NTR);Tg(Tp1:H2BmCherry)와 서열번호 2로 표시되는 epcam 변이 유전자가 도입된 형질전환 제브라피쉬를 다시 교배함으로써 최종적으로 획득한 형질전환 제브라피쉬 Tg(fabp10a:CFP-NTR);Tg(Tp1:H2BmCherry);epcam+/-를 이용하여, MTZ로 유발된 간담도 손상과 epcam 변이 유전자의 상관관계를 시각적으로 관찰함으로써, 담도 손상 질환 치료 물질 개발에 관한 연구에 유용하게 사용될 수 있다.
본 발명의 담도 세포-특이 손상이 유발되는 epcam 유전자 변이 제브라피쉬에서, 상기 변이는 epcam 유전자의 넉아웃(knockout) 또는 넉다운(knockdown); 또는 서열번호 2로 표시되는 epcam 변이 유전자 도입에 의해 이루어진다.
본 명세서에서 사용되는 용어, "변이(variant)"또는 "변이체"는 연속적 DNA 단편들 또는 서열이 유사한 DNA 단편들에서 특정 위치에 존재하는 DNA 서열의 뉴클레오타이드 다양성을 의미한다.
본 발명에서 epcam 유전자를 언급하면서 사용되는 용어, epcam 유전자 변이 또는 epcam 유전자 변이는 차이가 없으며, 본 명세서에서 혼용된다.
또한, 본 발명에서, 상기 epcam 유전자 변이는 참조 서열인 야생형 서열(서열번호 1)에 비해 염기서열이 변화된 것을 의미하고, 1개 이상의 염기가 치환, 결실, 삽입, 증폭 및 재배열된 것을 포함한다.
또한, 본 발명의 서열번호 2로 표시되는 epcam 유전자 변이의 뉴클레오티드 서열은 서열번호 3으로 표시되는 epcam 변이 단백질을 코딩한다.
본 발명에서 상기 epcam 변이 단백질을 코딩하는 것이면, 어떠한 뉴클레오티드 서열을 가질 수 있으나, 바람직하게는, 상기 뉴클레오티드는 서열번호 2의 뉴클레오티드 서열을 갖는 것이다.
본 명세서에서 사용되는 용어 “녹아웃(knockout)”은 정상 유전자의 결손을 유발하여 완전 불활성화를 유도하는 것을 의미하며, 용어 “녹다운(knockdown)”은 당업계에 공지된 다양한 방법을 통해 정상의 mRNA 발현량을 줄이도록 유도하는 것을 의미한다.
또한, 본 발명의 변이는 목적 효과로서 epcam 유전자의 기능을 억제 또는 감쇠시키는 효과를 발휘할 수 있는 한, 당업계에 공지된 다양한 임의의 방법을 이용하여 달성할 수 있다.
본 발명에서 “동물 모델(animal model)” 또는 “질환 모델(disease model)”은 사람의 질병과 유사한 특정 질환을 가지고 있어, 병인을 규명하고 병태를 확인할 수 있는 연구 대상이 될 수 있는 모델이 되는 동물을 의미한다. 동물 모델로서 본 발명에서 채택한 제브라피쉬는 인간에서와 같은 효과를 예측할 수 있으며, 쉽게 만들수 있고, 재현성이 높다는 장점이 있다.
또한, 본 발명의 다른 양태에 따르면, 본 발명은 다음 단계를 포함하는, 담도 손상을 진단 또는 모니터링하기 위한 정보 제공 방법을 제공한다:
(a) 대상 제브라피쉬에 MTZ(metronidazole)를 처리하여 간 손상을 유발하는 단계;
(b) 상기 (a) 단계의 제브라피쉬로부터 분리된 생물학적 시료에서 서열번호 2로 표시되는 epcam(epithelial cell adhesion molecule) 유전자 변이의 존재 여부를 확인하는 단계; 및
(c) 상기 (b) 단계의 생물학적 시료 내에서 서열번호 2로 표시되는 epcam(epithelial cell adhesion molecule) 유전자 변이가 존재하는 것으로 확인된 경우, 대상의 담도가 손상 또는 비정상적으로 재생된 상태인 것으로 진단하는 단계.
상기 epcam(epithelial cell adhesion molecule) 유전자 변이의 존재 여부는, 상기 목적 유전자 변이의 발현 여부를 확인할 수 있는 한, 당업계에 공지된 임의의 방법들을 이용하여 확인할 수 있다.
예컨대, 유전자 변이의 발현 여부를 RTPCR(Reverse Transcription Polymerase Chain Reaction), 노던 블롯(Northern blot), cDNA 마이크로어레이 혼성화 반응, 및 생체 내(in situ) 혼성화 반응 등의 방법을 통해 측정하는 것일 수 있으며, 단백질 변이의 활성 정도를 면역침강법(immunoprecipitation), 방사능면역분석법(RIA), 효소면역분석법(ELISA), 면역조직화학법(immunohistochemistry), 웨스턴 블롯(Western Blotting) 및 유세포 분석법(FACS) 등의 방법을 통해 측정하는 것일 수 있으나, 이에 한정되지 않는다.
또한, 본 발명의 또 다른 양태에 따르면, 본 발명은 다음 단계를 포함하는, 담도 손상 관련 질환의 치료제의 스크리닝 방법을 제공한다:
(a) 본 발명에 따른 epcam 유전자 변이 제브라피쉬에 담도 손상 관련 질환의 치료제 후보물질을 투여하는 단계;
(b) 단계 (a)의 담도 손상 관련 질환의 치료제 후보물질이 투여된 제브라피쉬의 담도 손상 수준을 측정하는 단계; 및
(c) 상기 후보물질을 투여하지 않은 대조군 제브라피쉬와 비교하여 담도 손상 수준을 유의하게 회복시킨 후보물질을 선별하는 단계.
상기 담도 손상 관련 질환은 담도 손상으로 인해 야기되는 모든 질환을 포함하며, 예컨대, 담도 폐색, 담낭 장애, 담석증, 췌염, 담도 운동실조증, 담관염, 담낭염, 담낭암, 담관암 및 원발성 담즙성 간경변으로 구성된 군으로부터 선택된다.
본 발명에서 분석하고자 하는 후보물질은 다양한 물질을 포함한다. 예를 들어, 상기 후보물질은 담도 손상 질환에 치료에 효과가 있는지 여부를 확인하기 위해 처리하는 물질로서, 화학물질, 단백질, 펩타이드, 항체, 핵산 및 천연 추출물을 포함하나, 이에 한정되는 것은 아니다.
본 발명의 스크리닝 방법에 의해 분석되는 후보물질은 단일 화합물 또는 화합물들의 혼합물(예컨대, 천연 추출물 또는 세포 또는 조직 배양물)일 수 있다. 후보물질은 합성 또는 천연 화합물의 라이브러리로부터 얻을 수 있다. 이러한 화합물의 라이브러리를 얻는 방법은 당업계에 공지되어 있다. 합성 화합물 라이브러리는 Maybridge Chemical Co.(영국), Comgenex(미국), Brandon Associates(미국), Microsource(미국) 및 Sigma-Aldrich(미국)에서 상업적으로 구입 가능하며, 천연 화합물의 라이브러리는 Pan Laboratories(미국) 및 MycoSearch(미국)에서 상업적으로 구입 가능하다. 시료는 당업계에 공지된 다양한 조합 라이브러리 방법에 의해 얻을 수 있으며, 예를 들어, 생물학적 라이브러리, 공간 어드레서블 패러럴 고상 또는 액상 라이브러리(spatially addressable parallel solid phase or solution phase libraries), 디컨볼루션이 요구되는 합성 라이브러리 방법, "1-비드 1-화합물" 라이브러리 방법, 그리고 친화성 크로마토그래피 선별을 이용하는 합성 라이브러리 방법에 의해 얻을 수 있다. 분자 라이브러리의 합성 방법은, DeWitt et al., Proc. Natl. Acad. Sci. U.S.A. 90, 6909, 1993; Erb et al. Proc. Natl. Acad. Sci. U.S.A. 91, 11422, 1994; Zuckermann et al., J. Med. Chem. 37, 2678, 1994; Cho et al., Science261, 1303, 1993; Carell et al., Angew. Chem. Int. Ed. Engl. 33, 2059, 1994; Carell et al., Angew. Chem. Int. Ed. Engl. 33, 2061; Gallop et al., J. Med. Chem. 37, 1233, 1994 등에 개시되어 있다.
상기 스크리닝 방법에 있어서, 상기 (c) 단계의 선별 단계는 상기 (b) 단계 의 측정 결과를 정량적 또는 정성적으로 분석하여 후보물질을 처리한 제브라피쉬 모델과 후보물질을 처리하지 않은 대조군의 담도 세포의 손상 정도 비교를 통해 대조군에 비해 손상 회복 정도가 증가한 후보물질을 선별하는 것일 수 있다.
또한, 상기 담도 세포의 손상 정도는, 상기 담도 세포의 형광 발현을 통해 담도 세포의 손상을 확인할 수 있는 한, 당업계에 공지된 임의의 방법들을 이용하여 확인할 수 있다.
따라서, 본 발명의 epcam 유전자 변이 제브라피쉬는 담도 손상 질환의 치료, 예방, 진단 또는 그 분자적 기전에 관한 연구과정 등에 널리 이용될 수 있다.
본 발명에 따르면, 본 발명의 epcam 변이 유전자를 갖는 제브라피쉬 모델에서 간 손상 후 담도 재생시 담도가 비정상적으로 재생되는 것을 발견하였다. 따라서, 본 발명의 제브라피쉬 모델을 이용하는 경우, 부작용이 적으면서도 효능이 우수한 담도 손상 관련 질환 치료제를 효과적으로 스크리닝할 수 있으며, 담도 손상 관련 질환의 진단 또는 그 분자적 기전에 관한 연구과정 등에 널리 이용될 수 있다.
도 1은 제브라피쉬의 epcam 유전자가 정상 혹은 비정상 변이를 생성하는 개체를 판별한 결과이다.
도 2는 제브라피쉬의 epcam 유전자가 정상 혹은 비정상 변이를 생성하는 개체에서 간의 발생이 정상적으로 이루어짐을 확인한 결과, 제브라피쉬의 epcam 유전자가 비정상 변이를 생성하는 개체에서 담도의 형성 및 담즙 분비에 아무런 이상을 야기하지 않음을 확인한 결과이다.
도 3은 제브라피쉬의 epcam 유전자가 정상 혹은 비정상 변이를 생성하는 개체에서 간 세포에 손상을 야기하였을 때 epcam 유전자가 정상인 개체에서는 간 세포 및 담도 형성이 정상적으로 이루어지는 것을 확인하였으나 epcam 유전자가 비정상 변이를 생성하는 개체에서는 간 세포의 재생은 정상적으로 이루어지나 담도가 비정상적으로 재생됨을 확인한 결과이다.
도 2는 제브라피쉬의 epcam 유전자가 정상 혹은 비정상 변이를 생성하는 개체에서 간의 발생이 정상적으로 이루어짐을 확인한 결과, 제브라피쉬의 epcam 유전자가 비정상 변이를 생성하는 개체에서 담도의 형성 및 담즙 분비에 아무런 이상을 야기하지 않음을 확인한 결과이다.
도 3은 제브라피쉬의 epcam 유전자가 정상 혹은 비정상 변이를 생성하는 개체에서 간 세포에 손상을 야기하였을 때 epcam 유전자가 정상인 개체에서는 간 세포 및 담도 형성이 정상적으로 이루어지는 것을 확인하였으나 epcam 유전자가 비정상 변이를 생성하는 개체에서는 간 세포의 재생은 정상적으로 이루어지나 담도가 비정상적으로 재생됨을 확인한 결과이다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
실시예 1. 제브라피쉬의 사육 및 관리
본 발명자들은 담도 손상 및 재생에 미치는 epcam(epithelial cell adhesion molecule) 유전자의 기능 연구를 위하여 야생형(wild type) 제브라피쉬, 형질전환 제브라피쉬 및 epcam 유전자가 비정상인 개체를 사용하였다.
성체(adult) 및 배아(embryo) 제브라피쉬는 섭씨 28.5℃ 온도에서 14시간: 10시간의 명암 일주기로 사육하였다. 제브라피쉬 배아와 치어(larva)의 단계는 수정 후 시간(hours post-fertilization, hpf) 혹은 수정 후 며칠 (days post-fertilization, dpf) 형태학적 특징에 따라 정의하여 사용하였다. 수정 후 24시간 배아에 0.003% PTU(1-phenyl-2-thiourea) 색소 형성을 막기 위하여 배아 배지 (embryo media)를 처리하였다.
실시예 2. 담도 세포-특이 손상이 유발되는 epcam 유전자 변이 제브라피쉬의 제작
본 발명자들은 MTZ 처리로 인해 간세포 손상을 유발했을 때, 간 세포는 정상적으로 재생되나, 담도 세포는 정상적으로 재생되지 않는, 담도 세포-특이 손상이 유발되는 epcam 유전자 변이 제브라피쉬 동물 모델을 최초로 구축하였다.
즉, 당업계에서 공지된 바와 같이, 담도 손상을 유발하기 위해서는 간을 손상시켜야 하므로, 간 특이적 손상을 유발할 수 있는 형질 전환체를 플랫폼으로 이용하여, epcam 유전자 변이(variant) 형질을 가지는 제브라피쉬 형질전환 주를 제작하였다.
간략하게는, (i) 간 및 담도 손상을 위한 NTR(nitoreductase)/MTZ(metronidazol) 시스템을 기반으로 하는 형질전환체의 제작;
(ii) epcam 유전자 변이 서열(서열번호 2)을 포함하는 벡터를 도입시킨 형질전환체의 제작; 및
(iii) 최종적으로 이들을 교배함으로써,
간 특이적 손상을 유발할 수 있는 형질 전환체를 플랫폼으로 이용하여, epcam 유전자 변이(variant) 형질을 가지도록 형질전환된 제브라피쉬 형질전환체를 획득하였다.
보다 상세하게는, (i)의 간 및 담도 손상을 위한 NTR(nitoreductase)/MTZ(metronidazol) 시스템을 기반으로 하는 형질전환체는, 간-특이 형광 단백질(CFP) 및 NTR(nitoreductase)을 발현하는 형질전환 제브라피쉬 Tg(fabp10a:CFP-NTR);와 간에서 담도 세포의 핵 특이적으로 형광단백질(mCherry)을 발현하는 형질전환 제브라피쉬 Tg(Tp1:H2BmCherry)를 교배하여, 제작된 형질전환체 Tg(fabp10a:CFP-NTR);Tg(Tp1:H2BmCherry)이다.
상기 '간-특이 형광 단백질 및 NTR(nitoreductase)을 발현하는 형질전환 제브라피쉬'의 제작에 이용된 fabp10a 프로모터(Her, et al., 2003)는 제브라피쉬에서 간세포에 특이적으로 발현을 유도하는 프로모터이므로, 이 유전자를 프로모터로 이용하는 NTR(nitoreductase)/MTZ(metronidazol) 시스템(Curado et al., 2008)은 박테리아에서 분리된 NTR 유전자에 의해 발현된 NTR 단백질을 이용하여 MTZ를 첨가하면, NTR 단백질이 MTZ와 반응하여 세포독성이 있는 물질을 생성함으로써 NTR 단백질이 발현되는 간세포 특이적으로 세포를 사멸시켜 손상을 유발시키는 방법을 기반으로 한다.
이에, 본 실시예에서는 간 세포특이적으로 발현하는 fabp10a 프로모터를 이용하고 발현을 확인할 수 있는 형광단백질 Cyan Fluorescent Protein (CFP) 및 NTR 유전자를 재조합하여 제작한 fabp10a:CFP-NTR 재조합 벡터(서열번호 7)를 제브라피쉬에 도입시켜서, 간-특이 형광 단백질(CFP) 및 NTR(nitoreductase)을 발현하도록 형질전환된 제브라피쉬 Tg(fabp10a:CFP-NTR)를 이용하였다.
또한, 본 실시예에서는 담도 세포 핵-특이적으로 발현하는 Tp1(Notch-responsive elements) 프로모터를 이용하고 발현을 확인할 수 있는 형광단백질 mCherry를 재조합하여 제작한 Tp1:H2BmCherry 재조합 벡터(서열번호 8)를 제브라피쉬에 형질전환시켜서, 담도 세포 핵-특이 형광 단백질(mCherry)을 발현하도록 형질전환된 제브라피쉬 Tg(Tp1:H2BmCherry)를 이용하였다.
상기 Tg(fabp10a:CFP-NTR)와 Tg(Tp1:H2BmCherry)를 교배하여, 형질전환체 Tg(fabp10a:CFP-NTR);Tg(Tp1:H2BmCherry)을 제작하였다.
또한, (ii) 성체 제브라피쉬 암수를 교배하여 배아를 얻고, 상기 배아의 1 세포(cell) 시기에 epcam 유전자 변이 서열(서열번호 2)을 포함하는 pCS2+:nLacz-GTvirus 벡터를 도입시킴으로써, epcam 유전자 exon 2 위치에 변이 서열을 발현하는 epcam+/-, 예컨대, Tg(nLacz-GTvirus) 형질전환 제브라피쉬를 제작하였다.
최종적으로, (iii) 상기 제브라피쉬 형질전환 주들을 교배함으로써, MTZ 처리로 인해 간세포 손상을 유발했을 때, 간 세포는 정상적으로 재생되나, 담도 세포는 정상적으로 재생되지 않는, 담도 세포-특이 손상이 유발되는 epcam 유전자 변이 제브라피쉬 형질전환 주를 구축하였다.
실시예 2. 역전사 중합효소 연쇄 반응 및 제브라피쉬 epcam 유전자의 유전자형(genotyping)
본 발명자들은 제브라피쉬 epcam 유전자가 비정상 변이(variant)를 생성하는 개체, 즉, 제브라피쉬 epcam 변이(variant) 유전자가 존재하는 개체를 동정하기 위하여, 제브라피쉬 배아, 치어, 성체에서 각각 gDNA를 50mM NaOH로 추출하였다. 추출된 gDNA는 역전사 중합효소 연쇄반응 (DNA polymerase, invitrogen)을 사용하여 단편을 증폭하였다.
역전사 중합효소 연쇄반응을 위해 사용된 epcam 유전자 프라이머는 다음과 같다:
5'-GACCTCTGTGTTAATGATTAAAGTGCTC(서열번호 4);
5'-CACTGAACCACACCCTAGATCTCAT(서열번호 5); 및
5'-GCTAGCTTGCCAAACCTACAGGT(서열번호 6).
상기 디자인된 프라이머는 Ensemble로부터 얻은 염기서열을 이용하여 제조되었다(epcam: ENSDART00000059321.5).
PCR 프로그램은 Step1 = 98℃, 2min, Step2 = 98℃, 30sec, Step3 = 58℃, 1min 30sec, Step4 = 72℃, 1min, Step5 = goto Step2 (32 additional cycles)으로 증폭하여 증폭 산물을 확인하였다.
그 결과, 도 1에 나타낸 바와 같이, 증폭된 단편은 야생형(WT)의 경우 300bp; 비정상 개체의 경우 200bp 크기의 단편으로 확인되었고, 모두 증폭되면 Het으로 표기하였다.
실시예 3. 간의 담도 분비 표현형 분석
본 발명자들은 상기 실시예 2에서 확인한 제브라피쉬의 epcam 유전자가 비정상 변이(variant)를 생성하는 개체에서 간이 형성될 때 간의 담도 분비 표현형을 분석하기 위해, PED6 어세이를 실시하였다.
PED6(invitrogene)는 간에서 담즙의 분비를 확인하는 시약으로, 이를 epcam이 정상인 야생형 제브라피쉬 개체 및 epcam 유전자가 비정상 변이(variant)를 생성하는 개체에 각각 처리하였다. PED6의 농도는 제브라피쉬 개체 배양액에 10ug/ml의 농도로 2시간 동안 처리한 후 담낭에서 담즙이 분비되는 지를 형광 현미경을 통해 관찰하였다.
그 결과, 도 2에 나타낸 바와 같이, epcam 유전자가 비정상 변이를 생성하는 개체는 정상적으로 간으로부터 담즙이 분비되는 것을 확인하였다.
실시예 4. 담도 형성 표현형 분석
본 발명자들은 상기 실시예 2에서 확인한 제브라피쉬의 epcam 유전자가 비정상 변이를 생성하는 개체에서 담도의 형성이 정상적으로 이루어지는 지를 확인하기 위해, 온조직 조직 염색법(Wholemount Immunostaining)을 실시하였다.
이 때, Tg(fabp10a:CFP-NTR)는 간 세포를 표지하기 위해 사용하였고, Tg(Tp1:H2BmCherry)는 담도 세포의 핵을 표지하기 위해 사용되었으며 Alcam 항체는 담도의 형성을 표지하기 위해 사용하였다.
epcam 유전자가 정상인 개체 및 비정상 변이를 생성하는 개체에서 수정란을 받아 제브라피쉬 배양액에 5일동안 키우고 제브라피쉬 배아 및 치어를 마취제(ethyl 3-aminobenzoate methanesulfonate salt, 200 mg/L, MS222, Sigma)를 이용해 마취시킨 후, 4% 파라 포름알데하이드 (paraformaldehyde)/PBS (1X phosphate buffered saline)에 넣어 4℃에서 12~18시간 이상 고정시켰다. 1 시간 이상 반응시킨 후 1차 항체를 블록킹 용액(blocking solution)에 희석하여 이를 4℃에서 12~18시간 이상 반응시켰다. 이후 PBStx (1X PBS + 0.2% Triton-X)로 1-2시간 동안 씻어준 후 블록킹 용액에 1시간 반응시켰다. 그 후 PBStx (1X PBS + 0.2% Triton-X)로 1-2시간 동안 씻어준 후 2차 항체를 블록킹 용액에 희석해 4℃에서 12~18시간 이상 반응시켰다. PBStx로 10분간 6번 씻어준 후 현미경 촬영용액 (vectorshield;vectorlab)에 넣어 현미경으로 관찰하였다.
상기 실험에 사용한 1 차 및 2차 항체는 다음과 같다:
1차 항체; 마우스 항-Alcam 항체(mouse anti-Alcam, 1:20, ZIRC) 및
2차 항체; 488-, 568-, 647- 형광 시료가 부착된 2차 항체 (Alexa 488-, 568-, 647-conjugated secondary antibodies, 1:1000, Invitrogen, Cat. No. A-11001, A-11004, A11008, A-11011 and Abcam, Cat. No. ab96947).
그 결과, 도 2에 나타낸 바와 같이, epcam 유전자가 비정상 변이를 생성하는 개체에서 간이 정상적으로 발생하고 간의 담도에도 이상이 없다는 것을 확인하였다.
실시예 5. 형질전환개체를 활용한 간세포 손상 (MTZ/NTR Assay)
상기 제브라피쉬의 epcam 유전자가 정상 혹은 비정상 변이를 생성하는 개체에 간세포의 손상을 유도하기 위해 형질전환개체를 이용하였다.
Tg(fabp10a:CFP-NTR)는 간 세포 특이적으로 NTR(nitoreductase)를 발현하는 형질전환체로서, 이 형질전환체에 MTZ(metronidazole)를 처리하면 간 세포-특이 사멸을 유도한다(Choi et al., 2014, Gastroenterology).
또한, Tg(Tp1:H2BmCherry)는 간에서 담도 세포(biliary epithelial cells)의 핵만을 특이적으로 mCherry가 발현되도록 만든 형질전환체이다(Choi et al., 2014, Gastroenterology).
수정일로부터 5일이 지난 epcam 유전자가 정상인 야생형 개체 및 비정상 변이를 생성하는 제브라피쉬 개체들에, MTZ를 28.5℃에서 24시간동안 동안 처리하여 간 세포의 손상을 유도한 후 48 시간 동안 간의 재생이 이루어지도록 하였다. 48시간 동안 간의 재생이 이루어진 epcam 유전자가 정상인 야생형 개체 및 비정상 변이를 생성하는 개체로부터 수정란을 받아 제브라피쉬 배양액에 5일동안 키우고 제브라피쉬 배아 및 치어를 마취제 (ethyl 3-aminobenzoate methanesulfonate salt, 200 mg/L (MS222, Sigma))를 이용해 마취시킨 후, 4% 파라 포름알데하이드 (paraformaldehyde)/PBS (1X phosphate buffered saline)에 넣어 4℃에서 12~18 시간 이상 고정시켰다. 1 시간 이상 반응시킨 후 1차 항체를 블록킹 용액에 희석하여 이를 4℃에서 12~18시간 이상 반응시켰다. 이후 PBStx (1X PBS + 0.2% Triton-X)로 1-2시간 동안 씻어준 후 블록킹 용액(blocking solution)에 1시간 반응시켰다. 그 후 PBStx (1X PBS + 0.2% Triton-X)로 1-2 시간 동안 씻어준 후 2차 항체를 블록킹 용액에 희석해 4℃에서 12~18 시간 이상 반응시켰다. PBStw로 10분간 6번 씻어준 후 현미경 촬영용액(vectorshield; vectorlab)에 넣어 현미경으로 관찰하였다.
상기 실험에 사용 한 1차 및 2차 항체는 다음과 같다:
1차 항체로 마우스 항-CK18 항체(mouse anti-CK18, 1:20, RDI Division of Fitzgerald Industries); 및
2차 항체로 488-, 568-, 647- 형광 시료가 부착된 2차 항체 (Alexa 488-, 568-, 647-conjugated secondary antibodies, 1:1000, Invitrogen, Cat. No. A-11001, A-11004, A11008, A-11011 and Abcam, Cat. No. ab96947).
그 결과, 도 3에 나타낸 바와 같이, epcam 유전자가 비정상 변이를 생성하는 개체에서 간의 담도 형성의 이상을 야기하는 것이 확인되었다.
또한, 더욱 정확한 담도 형성을 확인하기 위해, 100ug/ml BODIPY FL C5(invitrogene) 시약을 제브라피쉬 치어 배양액에 넣고 어두운 조건에서 28.5℃에서 2시간 처리하고 커버 슬라이드로 슬라이드 글라스를 덮어준 후 공초점 현미경으로 관찰했을 때, 제브라피쉬의 epcam 유전자가 정상 혹은 비정상 변이를 생성하는 개체를 확인한 결과 담도가 비정상적으로 재생됨을 확인하였다.
결론적으로, 본 발명에서는 위와 같은 결과를 바탕으로 epcam 변이 유전자를 도입한 제브라피쉬 모델이 담도 손상 관련 질환 모델 동물로서의 활용이 가능함을 밝혔으며, 따라서, 본 발명의 제브라피쉬 모델은 관련 치료물질의 대량 스크리닝 기술에 유용하게 사용될 수 있다.

(b) 서열번호 2로 표시되는 epcam 변이 유전자가 도입된 형질전환 제브라피쉬; 또는 서열번호 1로 표시되는 epcam 정상 유전자가 결실된 epcam 녹아웃 형질전환 제브라피쉬를 제작하는 단계;
