Abstract

Description
발명의 설명
본 발명은 미세입자의 유리한 구조 특성을 설명하는, 모두 규칙적이고 결정질인, 다음의 5가지 상이한 유형의 구리 화합물인 안틀러라이트(Antlerite) Cu3 +2(SO4)(OH)4, 브로칸타이트(Brochantite) Cu4 +2SO4(OH)6, 칼칸타이트(Chalcantite) Cu+2SO4·5H2O, 나트로칼사이트(Natrochalcite) NaCu2 +2(SO4)2OH·H2O, 구리 설페이트 히드록시드 Cu3(SO4)2(OH)2·4H2O/2CuSO4·Cu(OH)2를 이의 구조에 포함하는 항균 및/또는 살생물 활성을 갖는 미세구조 다중복합 구리 미세입자에 관한 것이다. 미세구조 다중복합 미세입자의 균질한 조성물은 5가지 상이한 구리 화합물 중 각각 하나의 차별적인 방출 속도 특성이 이들이 포함된 재료에 걸쳐 균일하게 할 수 있도록 하며, 각각의 구리 화합물은 미세입자 구조가 존재하는 모든 장소에서 동시에 이의 항균 및/또는 살생물 효과를 개별적으로 수행한다. 동일한 미세구조 미세입자에 5가지 구리 화합물이 포함되어 있기 때문에, 이러한 화합물이 균질하고 균일하게 분포되어, 더 적은 용량을 사용할 수 있으므로, 재료의 반투명 특성에 기여한다.

다제-내성 미생물의 출현은 현재 사용되고 있는 약제의 경우와 같이 내성을 획득하지 않고 세균 증식을 막을 수 있는 새로운 화합물에 대한 검색을 촉발시켰다(문헌[Betancourt et al. 2016]).
나노기술의 발달로 마이크로미터 및 나노미터 정도의 치수를 가질 수 있는 물질, 항균제로서 매우 유망한 특성을 나타내는 물질의 생성이 가능해졌다(문헌[Chatterjee et al. 2014]). 나노 입자 및 마이크로 입자는 독립적으로 또는 다른 분자의 운반체로 작용할 수 있으며, 표면적, 부피, 및 구조의 특성으로 인해 생물학적 반응을 유발할 수 있다(문헌[Chatterjee et al. 2014]).
구리는 나노 및 마이크로입자를 형성하는데 사용되는 화학 원소 중 하나이다. 구리 화합물 및 복합체는 액체, 고체, 및 인간 조직의 소독을 위한 제제 및 제품에 사용되어왔다(문헌[Delgado et al. 2011]).
종래 기술과 상이한 문헌은 항균 및/또는 살생물 특성을 가진 제품을 생산할 목적으로 구리 화합물 및 구리 나노-/마이크로 입자를 고체에 혼입하는 것을 기술하였다.
직물 유형의 재료 및 섬유의 경우, 문헌 미국 특허 US 8183167 (B1)호는 항균 활성을 나타내는 구리 및/또는 은 나노입자와 공유결합된 직물 기질을 개시한다.
문헌 미국 특허 US 5458906 A호는 항균 화합물, 그 중에서도 구리로 생분해성 기질을 처리하는 방법을 제시한다. 미국 특허 US 7754625B2호는 항균제와 결합된 하나 이상의 천연 또는 합성 섬유 또는 필라멘트를 포함하는 항균 직물을 기술하며, 여기서 상기 항균제는 적어도 하나의 항균 은 이온 공급원 및 적어도 하나의 구리 이온 공급원과 조합된 수용성 아연 염을 주로 포함한다.

문헌 CL201300332(국제공개 WO 2014117286 A1호)에서, Cu4SO4(OH)6에 해당하는 항균 화합물을 함유하는 상이한 비율의 식물성, 동물성, 또는 합성 기원 또는 이들의 혼합물의 함침성 매트릭스가 개시되어 있다.
문헌 CL201500921은 구리 화합물, 혼합된 구리 미세입자 또는 나노입자에 해당하는 항균제를 포함하는 셀룰로스-기재 물질이 개시되어 있다. 구리 미세입자 또는 나노입자를 첨가하는 단계를 포함하는 셀룰로스-기재 물질의 제조 방법이 포함된다.
한편, 문헌 CL 201503652는 표면-보호 항균 활성을 나타내는 반투명 접착 필름, 구리, 키토산, 젤라틴, 및 글루타르알데히드의 나노입자를 포함하는 접착 필름을 제시한다.
식품 포장용 식품 산업을 위해 특별히 다양한 재료가 개발되었다. 문헌 국제공개 WO 2014001541 A1호, 국제공개 WO 2012127326 A1호, 및 국제공개 WO 2000053413 A1호는 항균 및/또는 살생물 특성을 가진 식품 포장에 사용하기 위한 구리 입자를 함유하는 중합체 물질의 생산을 기술한다. 이러한 문헌에서 고려되는 물질은 구리 화합물 또는 구리 입자를 포함하며, 그 중에는 구리 산화물 1차(copper oxide primary)가 있다.
항균 특성을 부여하기 위해 매트릭스 및 중합체 물질에서 상이한 유형의 담체 또는 이들의 혼합물에 지지된 구리 화합물 또는 구리 미세입자의 첨가에 대한 이전의 몇 가지 선행문헌이 있지만, 구리의 산화 상태에 따라 제안이 다양하다. 구리의 산화 상태는 구리 종류의 용해도, 및 결과적으로 중합체 물질로부터의 이들의 방출 거동에 영향을 미치며; 더욱이, 사용되는 상이한 담체는 필수 입자 부피를 크게 만드는 경향이 있다.
포장 또는 용기용 항균 및/또는 살생물 물질과 관련하여, 식품을 악화시키고 포장 시 존재하는(RAM 박테리아) 미생물의 표면 성장을 억제하기 위해 용기에서 신속하고 즉각적인 방출을 생성해야 한다. 특히, 각 작업, 온도 변화, 및 보관의 특정 처리로 인한 발생을 억제하기 위해 중기-방출이 또한 필요하다. 따라서, 실제로는 시간이 지남에 따라 즉각적이고, 지속적이고, 잔류 방출을 가능케 하고, 항균 및/또는 살생물 효과를 가질 수 있게 하는 구리 농도를 유지하지만, 화합물과 관련된 독성이나 접촉하는 식품으로의 과도한 이동을 방지하는 구리 농도를 유지하는 용해도(Kps)를 가진 구리 미세입자가 재료에 포함되어야 한다. 즉, 더 낮은 구리 농도에서 개선된 항균 및/또는 살생물 효과를 가진 재료가 필요하다.
본 발명의 미세입자의 특정 조성물은 식품 포장 재료의 경우 중요한 특성인 반투명 재료를 생산할 수 있도록 하며, 이는 입자가 이를 지지할 담체가 전혀 없고 부피가 커지게 하여 재료에 반투명성을 부여하기 때문이다.

본 발명은 미세입자의 유리한 구조 특성을 부여하는, 모두 규칙적이고 결정질 형상인, 다음의 5가지 상이한 유형의 구리 화합물인 안틀러라이트 Cu3 +2(SO4)(OH)4, 브로칸타이트 Cu4 +2SO4(OH)6, 칼칸타이트 Cu+2SO4·5H2O, 나트로칼사이트 NaCu2 +2(SO4)2OH·H2O 및 수화된 구리 설페이트 히드록시드 Cu3(SO4)2(OH)2·4H2O/2CuSO4·Cu(OH)2·4H2O를 이의 구조에 포함하는 미세구조 다중복합 구리 미세입자에 관한 것이다. 본 발명의 범위의 부분인 미세입자는 5 μm 내지 50 μm 사이, 특히 10 μm 내지 40 μm 사이, 보다 특히 10 μm 내지 15 μm 사이의 정의된 크기를 갖는다.
전술된 미세입자는 다음의 단계를 포함하는 방법에 의해 생성된다:

본 발명이 해결하고자 하는 기술적 문제는 규칙적인, 결정질이고 미세구조 조성물이 항균 및/또는 살생물 특성을 제공하는 구리 이온의 제어된 방출을 가능케 하는 5가지 유형의 구리를 포함하는 새로운 유형의 미세입자를 제공하는 것이다. 미세입자에 포함된 5가지 상이한 구리 화합물의 차등 방출 동력학은 접촉에 대한 1차 항균 및/또는 살생물 효과 및 2차 및 그 안에 존재하는 다른 종류의 구리의 후속 방출 덕분에 시간에 지남에 따라 지속되는 항균 및/또는 살생물 효과를 가능하게 한다. 반면에, 이러한 특성을 가진 구리 미세입자는 농축된 중합체 조성물 또는 마스터배치의 일부를 형성할 수 있으며, 이는 압출 공정 동안 미세입자를 포함하고 전술된 기술적 특징 및 이점을 가진 항균 및/또는 살생물 활성을 가진 필름, 시트, 또는 구조의 생산 목적으로 경직성 몰드, 섬유, 필라멘트, 및 시트를 형성하는데 사용되는 용융 중합체에 통합될 수 있다.
이러한 구조화된 미세입자는 또한 직접적으로 또는 혼합물로 사용되어 이것이 배치된 장소에서 항균 및/또는 살생물 효과를 생성할 수 있다.

본 발명은 또한 접촉 시 지속 효과로서의 항균 및/또는 살생물 활성을 갖는 재료의 제조에 유용한 구리 미세입자 및 농축된 중합체 조성물(마스터배치)의 용도를 제공한다. 미세입자에 포함된 구성요소의 미세구조 구성은 재료가 박테리아 및/또는 병원체와 접촉할 때, 구리 화합물과 동시에 직접 접촉하여 접촉 시 즉각적인 항균 및/또는 살생물 효과를 발휘하도록 한다. 상이한 용해도 및/또는 해리를 갖는 5가지 상이한 구리 화합물의 존재는 또한 개시된 미세입자의 항균 및/또는 살생물 활성의 지속 효과가 존재할 수 있게 한다.
본 발명에서 재료가 "살생물 활성"을 갖는다는 언급은 미세입자가 박테리아 및 진균의 발생 및 성장을 억제할 수 있음을 의미한다.
재료가 "항균 활성"을 갖는다는 언급은 미세입자가 박테리아의 증식을 억제하거나 방해한다는 것을 의미한다.
본 발명에서, 용어 "미세구조화"는 재료-이 경우 미세입자-가 이를 형성하는 일련의 상 또는 구성요소로 구성됨을 의미한다. 재료 과학 분야에서, 재료의 미세구조가 특성을 결정한다고 한다.
본 발명에서, 용어 "다중복합"은 미세입자가 특정 군의 상이한 화합물을 동시에 포함함을 의미한다.
용어 "마스터배치"는 생산될 재료에 혼입될 요소의 예비 혼합물을 포함하는 농축된 중합체 조성물을 지칭한다.
본 발명의 범위에 속하는 재료는 특히 중합체, 섬유, 직물, 유리 유형, 수지 등을 포함할 수 있지만 이에 제한되지 않는다. 중합체는 특히 폴리프로필렌(PP), 폴리에틸렌(PE), 폴리에틸렌 테레프탈레이트(PET), 에틸렌-비닐 아세테이트(EVA 고무)를 포함하지만 이에 제한되지 않는다.
본 발명의 일 실시형태에서, 미세입자는 항균 작용을 갖는 다른 금속 및 비금속 화합물을 추가로 포함할 수 있다. 이러한 화합물은 특히 아연 화합물, 납 화합물, 카드뮴 화합물, 은 화합물을 포함하지만 이에 제한되지 않는다.
이하에 제시된 예시적인 응용은 본 발명의 범위를 제한하지 않고 본 발명의 일 실시형태를 예시한다.
예시적인 응용
실시예 1: 구리 미세입자의 생성
100 g/l의 5수화된 구리 히드록시드를 함유하는 구리 설페이트 용액을 알칼리화하여 미세입자를 구성하는 구리 히드록시드를 제조하며, 이는 완전히 용해된 용액을 생성해야 한다. 그런 다음 소듐 히드록시드 또는 다른 알칼리를 OH 염기로 물에서 10%로 희석해야 한다; 상기 용액을 pH 4 내지 6 사이에 도달할 때까지 제조된 구리 설페이트 용액을 천천히 교반하면서 첨가하고, 다음 반응이 생성된다:
반응은 겔 상태의 용액 중의 구리 히드록시드의 침전을 생성하고, 24시간 후에 상등액을 제거한다.
구리 히드록시드를 함유하는 생성된 침전물을 1:5의 비율로 구리 설페이트 용액 중에 유화한다. 구리 설페이트 및 구리 히드록시드와 같은 겔 형태의 다른 것의 용해된 이온 상을 함유하는 이러한 현탁액을 220℃ 내지 280℃ 사이의 유입구 온도 및 80℃ 내지 100℃ 사이의 배출구 온도의 분무 건조기를 사용하여 건조 공정에 적용한다.
실시예 2: 구리 미세입자의 특성분석
실시예 1에 기술된 프로토콜에 의해 생성된 구리 미세입자의 구조, 크기 및 분포를 특성분석하기 위해, 주사 전자 현미경(SEM) 분석을 수행하였다.
SEM 분석은 Penta FET Precision 검출기인 Oxford Instruments X-act와 함께 EDS 모드를 사용하여 주사 전자 현미경(SEM), Zeiss 모델 EVO MA 10을 사용하여 수행하였다. 이러한 분석은 (고체) 구리 미세입자 분말의 대표 표본을 스캔하고 미세입자의 원소 조성을 결정하기 위해 대표 영역을 선택하는 것으로 구성하였다.
상이한 배율의 SEM 이미지에서, 입자가 규칙적인 구형과 이질적인 크기 분포를 갖는 것이 관찰되었다(도 1a 내지 도 1d). 현미경에 장착된 EDS 검출기를 이용하여, 미세입자의 원소 조성을 결정하고, 구리, 황, 및 산소 원소로 구성되어 있음을 확인하였다(도 2a 내지 도 2d).
입자의 크기와 관련하여, 두 개의 독립적인 표본으로부터 취한 이미지의 4개의 독립적인 대표 영역을 평가하였다. 입자가 7 마이크로미터 내지 22 마이크로미터 사이의 마이크로미터 크기를 갖는 것으로 결정하였다(표 1 참조).
미세입자의 특정 화학 조성을 결정하고 함유된 구리 유형의 산화 상태를 결정하기 위해 X-선 회절 분석을 수행하였다. 상기 분석을 기반으로, X-선 회절 패턴은 다양한 화합물의 존재에 의해 결정하였다(도 3).
미세입자에 존재하는 화합물을 정의하기 위해, 결정질 분말 PDF-2 데이터베이스를 참조하고 수득된 패턴과 비교하였다. 본 발명의 미세입자는 안틀러라이트, 브로칸타이트, 칼칸타이트, 나트로칼사이트, 및 수화된 구리 설페이트 히드록시드(표 2)의 5 가지 유형의 구리 화합물로 구성되어 있는 것으로 확인되었다.
실시예 3: 마스터배치 제조. 다층 중합체 시트에 구리 미세입자 첨가.
실시예 2에 기술된 미세입자는 다층 섬유 및 시트를 형성하기 위한 상이한 중합체 재료 및 수지에 포함될 수 있다. 본 실시예에서, 농축된 중합체 조성물(마스터배치)을 제조하는데 따르는 프로토콜 및 수지 및 중합체에 이들을 포함시키기 위한 조건이 기술된다.
a) 농축된 중합체 조성물(마스터배치)의 제조
마스터배치를 제조하기 위해, 중합체 재료 및 미세입자의 예비 혼합물을 제조한다. 이를 위해, 수지 또는 중합체 재료를 분쇄기에서 분쇄한 다음 미세입자와 냉간 혼합한다.
예비 혼합물은 수지 또는 중합체의 종류인 폴리프로필렌(PP), 폴리에틸렌(PE), PET, EVA, 고무에 따라 120℃ 내지 250℃ 사이의 온도에서 용융되고, 펠릿 압출기를 통과하여 마스터배치라 지칭되는 펠릿이 생성된다.
b) 용융된 중합체를 압출하여 다층 재료를 형성하는 공정에서 매스터배치가 포함된다.
마스터배치는 섬유, 필라멘트 및 시트와 같은 경질 또는 가요성 중합체를 형성하는데 사용되는 용융 중합체의 압출 공정에 추가될 수 있다. 상기 절차에 의해 생산된 시트는 각 시트의 두께가 5 μm 내지 120 μm 사이, 보다 구체적으로 5 μm 내지 30 μm 사이인 다층 구조에 해당할 수 있으며, 즉, PE, PA, PP 및/또는 PET와 같은 이러한 중합체 수지 층을 2, 3, 심지어 5개까지 함유할 수 있는 최종 재료를 형성할 수 있다.
실시예 4: 미세구조 구리 미세입자를 포함하는 다층 중합체 재료의 항균 및/또는 살생물 효과.
미세구조 구리 미세입자를 포함하는 다층 재료의 항균 및/또는 살생물 효과를 평가하기 위해, 서로 다른 중합체로 구성된 다층 재료의 표면에 이전에 성장한 박테리아 배양물을 첨가하는 시험을 수행하였다. 대장균(Escherichia coli )및 황색포도상구균(Staphylococcus aureus)에 대한 항균 및/또는 살생물 효과를 평가하였다.
다층 재료를 박테리아의 성장에 필요한 온도에서 24시간 동안 배양하였다. 배양 기간이 지나면, 재료에 대한 콜로니-형성 단위의 계수를 수행하고, 이를 구리 미세입자를 함유하지 않는 재료와 비교하였다. 마스터배치에 추가된 중합체의 유형에 따라 두 가지 유형의 시트를 시험하였다: 1) 폴리프로필렌 및 구리 미세입자로 제조된 시트(마스터배치에 2% w/w 첨가); 및 2) 마스터배치에 첨가된 미세구조 구리 미세입자와 함께 에틸렌-비닐 아세테이트(EVA 고무)로 제조된 시트.
결과는 폴리프로필렌과 미세구조 구리 미세입자(마스터배치에 첨가된 2% w/w)로 구성된 시트에 대한 콜로니-형성 단위(CFU) 계수를 마스터배치의 성분을 포함하지 않는 대조군 시트와 비교했을 때, 구리 미세입자를 포함하는 시트는 CFU의 양이 99% 감소했으며 박테리아 수가 3 배 더 감소했음을 나타낸다(표 3). 에틸렌-비닐 아세테이트(EVA 고무)와 구리 미세입자로 구성된 시트의 경우, 대조군 재료와 비교하여 마스터배치를 구성하는 시트에서 시험한 박테리아의 CFU 수가 감소했음을 반영하는 동등한 현상이 관찰되었다(표 4).
Claims (10)
- 항균 및/또는 살생물 활성을 갖는 구리 미세입자에 있어서, 각각의 미세입자가 5가지 상이한 구리 화합물인 안틀러라이트(Antlerite) Cu3 +2(SO4)(OH)4, 브로칸타이트(Brochantite) Cu4 +2SO4(OH)6, 칼칸타이트(Chalcantite) Cu+2SO4·5H2O, 나트로칼사이트(Natrochalcite) NaCu2 +2(SO4)2OH·H2O 및 수화된 구리 설페이트 히드록시드 Cu3(SO4)2(OH)2·4H2O/2CuSO4·Cu(OH)2를 포함하는 규칙적인, 결정질이고 미세구조 조성물을 갖는 것을 특징으로 하며,
상기 미세입자는 5 μm 내지 50 μm 사이의 크기를 갖는, 항균 및/또는 살생물 활성을 갖는 구리 미세입자. - 제1항에 있어서, 미세입자가 5 μm 내지 50 μm 사이, 특히 10 μm 내지 40 μm 사이의 크기를 갖는, 항균 및/또는 살생물 활성을 갖는 구리 미세입자.
- 제1항에 있어서, 미세입자가 10 μm 내지 15 μm 사이의 크기를 갖는, 항균 및/또는 살생물 활성을 갖는 구리 미세입자.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 다른 금속 및 비금속 항균 화합물 예컨대 특히 아연 화합물, 납 화합물, 카드뮴 화합물, 은 화합물을 추가로 포함할 수 있는 것을 특징으로 하는, 항균 및/또는 살생물 활성을 갖는 구리 미세입자.
- 제1항의 항균 및/또는 살생물 활성을 갖는 구리 미세입자의 제조 공정에 있어서,
a) 1:10 w/w의 비율로 증류수에 용해된 구리 설페이트 용액을 제조하는 단계;
b) 용액의 pH가 4 내지 6 사이로 될 때까지 단계 a)에서 수득된 용액에 10% w/v 소듐 히드록시드 용액 및 증류수를 첨가하는 단계;
c) 24시간 후에, 상등액 및 구리 히드록시드 겔인 침전물을 분리하는 단계;
d) 단계 c)에서 수득된 겔 상태의 침전물을 1:5 w/v의 비율로 제조된 증류수 중의 구리 설페이트 용액에 유화시키는 단계;
e) 단계 d)에서 수득된 에멀젼을 바람직하게는 220℃ 내지 280℃ 사이의 유입구 온도 및 바람직하게는 80℃ 내지 100℃ 사이의 배출구 온도에서 바람직하게는 분무 건조기에 의해 건조 공정에 적용하는 단계를 포함하는 것을 특징으로 하는, 항균 및/또는 살생물 활성을 갖는 구리 미세입자의 제조 공정. - 제1항 내지 제3항 중 어느 한 항의 구리 미세입자 및 적어도 하나의 중합체 또는 수지를 포함하는 것을 특징으로 하는, 섬유, 필라멘트, 및 시트와 같은 경직성 또는 가요성 제품 형성을 위한 용융된 중합체에 압출 공정 동안 통합되는 항균 및/또는 살생물 활성을 가진 농축된 중합체 조성물(마스터배치).
- 제4항에 있어서, 중합체 또는 수지가 특히 폴리프로필렌(PP), 폴리에틸렌(PE), 폴리에틸렌 테레프탈레이트(PET), 에틸렌-비닐 아세테이트(EVA 고무), 폴리스티렌(PS), 스티렌-부타디엔 고무(SBR), 폴리비닐 클로라이드(PVC), 폴리테트라플루오로에틸렌(PTFE), 메타크릴레이트(PMMA), 폴리에스터, 지방족 폴리아미드, 폴리테레프탈아미드, 아르아미드(방향족 폴리아미드), 경질 및 가요성 폴리우레탄, 실리콘 등을 포함하지만 이에 제한되지 않는 것을 특징으로 하는, 농축된 중합체 조성물(마스터배치).
- 제1항 내지 제4항 중 어느 한 항의 항균 및/또는 살생물 활성을 갖는 구리 미세입자의 용도에 있어서, 접촉 시 항균 및/또는 살생물 활성을 갖고 지속 효과를 갖는 재료의 제조에 유용한 것을 특징으로 하는, 용도
