본 출원은 2016년 12월 29일 출원된 미국가특허 출원 제62/440,075호의 이익을 주장하며, 그 전체 내용이 본원에서 참조로 통합된다.
본 발명은 당단백질을 변형시켜 당단백질이 하나 이상의 트리-만노실 코어를 포함하게 하는 방법에 관한 것이다. 본 발명은 또한 본 발명의 당단백질 및 관심 페이로드(payload)를 포함하는 당단백질-페이로드 컨주게이트(glycoprotein-payload conjugate)에 관한 것이다.

이들 임상 ADC에서, Kadcyla 및 Mylotarg는 라이신 잔기의 아민 기에 페이로드 또는 링커를 무작위로 컨주게이션시킴으로써 형성되지만, Adcetris®, 브렌툭시맙 베도틴(brentuximab vedotin)(cAC10-vcMMAE, SGN-35)은 쥐 항-CD30 항체 AC10의 가변성 중쇄 및 경쇄 영역의 융합에 의한 키메라 항-CD30 모노클로날 항체이다. 평균 4개(2-8개)의 MMAE 분자가 SGN-30 스캐폴드(SGN-30 scaffold)에 컨주게이션된다. MMAE의 컨주게이트된 지점은 사슬간 디설파이드 결합의 온화한 환원에 의해서 생성되는 시스테인 잔기의 무작위 -SH 기이다. 링커는 티올-반응성 말레이미도카프로일 스페이서(thiol-reactive maleimidocaproyl spacer), 디펩티드 발린-시트룰린 링커, 및 PABC 스페이서로 이루어진다(Francisco JA, Cerveny CG, Meyer DL, Mixan BJ, Klussman K, Chace DF, Rejniak SX et al. cAC10-vcMMAE. 제 1 세대 ADC의 이들 단점을 해소시키기 위해서, SMART-Tag, 비-천연 아미노산, 즉, 임의의 티로신, 치료 소르타제(therapeutic sortase), Thio-Bridge 등을 포함한 부위-특이적 ADC 플랫폼이 개발되었다. 본 발명자들이 예상한 바와 같이, 이들 기술은 모체 항체 내의 일부 특이적 부위 또는 도메인을 조작함으로써 균질의 ADC 생성물을 생성시킬 수 있다. 예를 들어, Thio-Bridge 기술은 링커와 페이로드를 항체의 부분적으로 환원된 디설파이드 결합에 연결시킨다. SMART-Tag는 박테리아 옥시다제의 기질 서열로서 항체의 인접 서열을 돌연변이시키는 기술이다. 포름알데하이드를 갖는 생성되는 생성물은 링커 및 페이로드의 연결 부위로서 사용된다. 예상된 바와 같이, 이들 제 2 세대 ADC 기술은 독특한 DAR 및 높은 균일성을 갖는 ADC 생성물을 생성시킬 수 있다. 그러나, 천연 항체의 돌연변이로 인해서, ADC 생성물은 PK 및 면역원성 문제를 가질 수 있다. 현재, N-글리코실화를 통한 항체에의 페이로드의 컨주게이션(conjugation)은 항체의 당조작(glycolengineering)의 성공적인 개발로 인해서 많은 주목을 끌었다.
모든 천연 IgGs 및 재조합 항체는 N-글리코실화 부위인 중쇄 CH2 불변영역의 각각 내의 위치 297(Asn297)에서 아미노산 아스파라긴을 갖는다. 포유동물 세포에서의 글리코실화 및 후 변형에 의해서, 두 개의 디-안테나-모양 모이어티(moiety)가 IgG 상의 N-글리코실화를 통해서 형성되며 디-안테나 모양 글리칸 모이어티의 각각은 근본적으로는 하기 화학식을 갖는 적어도 7개의 당 모이어티에 의해서 구성된다:

WO 2014/164534 A2, WO 2014/065661 A1, WO 2015/032899 A1, WO 2015/057064 A1, WO 2015/157446 A1, US 8716033 B2, US 7416858 B2, EP 2753752 B3 및 검토 논평(Bioconjug Chem.; 2015 Nov. 18; 26(11):2070-5)은 항체 약물 컨주게이션을 위한 많은 변형된 글리칸 모이어티를 개시하고 있다. 그러나, 항체-약물 컨주게이션에서의 약물 항체 비율(DAR)이 잘 조절될 수 없고, 페이로드 다양성이 이들 기술에서 수행될 수 없고, 그에 따라서, 약물 항체 비율을 조절하고 ADC의 페이로드 다양성을 증가시키기 위한 본 기술분야에서의 요구가 있다. 본 발명은 그러한 요구를 충족시키고 있으며, 다른 이익을 제공하고 있다.

도 1은 트리-만노실 항체 약물 컨주게이트를 제조하기 위한 원-스텝(one-step) 및 순차적 전략을 나타낸다.

실시예 1. β1,4-갈락토시다제 및 뉴라미니다제를 사용함에 의한 헤르셉틴 항체의 제조
헤르셉틴 항체(Roche Inc)로부터 N-글리칸의 갈락토오스 및 시알산 모이어티를 제거하기 위해서, 10 mg의 헤르셉틴 항체를 37℃에서 24시간 동안 1X 글리코버퍼 1(GlycoBuffer 1)(NEB, 전체 부피 1 ml) 중의 20 ㎕의 β1,4-갈락토시다제(NEB, P0745L, 8 단위/㎕) 및 5 ㎕의 α2-3,6,8 뉴라미니다제(NEB, P0720L, 50 단위/㎕)로 처리하였다. 10 ㎕의 β1,4-갈락토시다제(NEB, P0745L, 8 단위/㎕)를 반응물에 추가로 첨가하고, 반응을 37℃에서 추가로 24시간 동안 수행시켜 G0F/G0 항체 샘플을 얻었다. 항체 샘플을 rProtein A Sepharose Fast Flow(GE Healthcare, 17-1279-02)를 사용하여 정제하였다. 정제 후에, 항체 샘플을 감소된 질량 크로마토그래피 분석에 가하였다. 도 2에 도시된 결과는 샘플 중의 주된 양의 항체가 GOF(50,600 Da의 분자량을 갖는 중쇄를 가짐)이고, 단지 소량이 GO(푸코오스 당 없이; 50,451 Da의 분자량을 갖는 중쇄를 가짐)임을 나타내고 있다.

트리-만노실 헤르셉틴-4(GlcNAc-트리아졸-DBCO-(PEG)4-DM1) ADC를 상기 기재된 공정에 의해서 구성시켰다. Her2/Neu 분자에 대항하는 Kadcyla를 (Roche Inc)로부터 구매하였다. OD 판독 및 항체의 농도를 사용하여 Prism 소프트웨어를 사용한 다중 산란 플롯(multiple scatter plot)을 작성하였다. 도 14에 나타낸 결과는 트리-만노실 헤르셉틴-4(GlcNAc-트리아졸-DBCO-(PEG)4-DM1) ADC의 곡선이 양성 대조군 Kadcyla 및 트리-만노실 헤르셉틴-4GlcNAz의 것들과 거의 동일하였음을 나타낸다. 음성 대조군 항-메소텔린(negative control anti-mesothelien)은 Her2/Neu 분자에 대한 결합 친화성을 나타내지 않는다. 이러한 결과는 Her2/Neu 분자에 대한 트리-만노실 헤르셉틴-4(GlcNAc-트리아졸-DBCO-(PEG)4-DM1) ADC의 결합 친화성이 형성된 변형에 영향을 받지 않음을 암시한다.
실시예 15. 트리-만노실 헤르셉틴-4(GlcNAc-트리아졸-DBCO-(PEG)
4
-DM1) ADC의 세포 독성 효과
Her2/Neu 고-발현 세포주 SK-BR-3, Her2/Neu 중간-발현 세포주 HCC-1954 및 Her2/Neu 저-발현 세포주 MDA-MB-231를 106 세포/ml로 희석시켰다. 96-웰 플레이트의 웰에 100 μL의 희석된 세포 배양물을 첨가한 후에, 세포를 37℃에서 24 시간 동안 인큐베이션하였다. 80 ㎕의 완전 배지를 각각의 웰에 첨가하고, 이어서, 상이한 투입량으로 20 ㎕의 트리-만노실 헤르셉틴-4(GlcNAc-트리아졸-DBCO-(PEG)4-DM1) ADC를 상이한 웰에 첨가하였다. 플레이트를 37℃에서 48 시간 동안 인큐베이션시킨 후에, 100 ㎕의 CellTiter-Glo® Reagent을 각각의 웰에 첨가하였다. 실온에서 10분 동안 추가의 인큐베이션 후에, 웰의 발광(광)을 광도계에 의해서 측정하였다. Her2/Neu 고-발현 세포주 SK-BR-3, 및 Her2/Neu 중간-발현 세포주 HCC-1954에 대한 트리-만노실 헤르셉틴-4(GlcNAc-트리아졸-DBCO-(PEG)4-DM1) ADC의 IC50 값은 각각 4.7 nM 및 14 nM이었다. 실시예 16. F293 세포에 의한 트리-만노실 코어 트라스투주맙 항체 및 트리-만노실 코어 항-TMC33 항체의 생성
β-N-아세틸글루코사미니다제 S를 엔코딩하는 cDNA를 함유하는 플라스미드 pTCAE8.3-exo-Gal을 구성시키고, 두 개의 F293 세포주 내로 공동-형질감염(co-transfection)시켜 각각 트리-만노실 트라스투주맙(항-Her2 항체) 및 트리-만노실 항-TMCC3(막관통 및 이중 코일 도메인 패밀리 3(transmembrane and coiled-coil domain family 3)) 항체를 발현시켰다. 인큐베이션 후에, 두 개의 세포 배양물의 상등액을 수거하고, 그에 함유된 항체를 각각 rProtein A Sepharose Fast Flow(GE Healthcare, 17-1279-02)를 사용하여 정제하였다. 정제 후에, 정제된 항체 샘플을 감소된 질량 크로마토그래피 분석에 가하였다. 도 15a에 나타낸 결과는, 단지 트라스투주맙 유전자로 형질감염된 F293 세포로부터 단리된 항체와 비교할 때, β-N-아세틸글루코사미니다제 S 및 트라스투주맙 유전자로 또한 형질감염된 동일한 세포로부터 얻은 트라스투주맙 항체의 중쇄가 50,195 Da의 분자량을 갖는 피크를 나타내고, 이는 생성되는 트라스투주맙 항체가 트리만노실 코어 항체이었음을 나타낸다는 것을 밝히고 있다. 유사한 결과가 항-TMCC3 항체 유전자로 형질감염된 동일한 세포에서 나타났다(도 15b). 결과는, 트리-만노실 항체가 상업적 세포주로부터 대량생산 가능하고 산업적 CMC 증폭에 적용될 수 있음을 암시한다.
실시예 17. MGAT-1 및 MGAT-2에 의한 트리-만노실 헤르셉틴-4(GlcNAc-트리아졸-DBCO-(PEG)
4
-DM1) ADC를 생성시키기 위한 세포-발현된 트리-만노실 코어 트라스투주맙 항체에의 UDP-GlcNAz의 컨주게이션
실시예 7에 기재된 공정에 따라서, 800 ㎕ 1X 완충액 SP(25 mM MES, 10 mM MnCl2, pH 6.5) 중의 실시예 16으로부터 얻은 2mg의 트리-만노실 코어 트라스투주맙 항체(포유동물 세포에 의해서 생성됨) 및 UDP-GlcNAz(1 mM)를 토끼 MGAT-1(25 μg) 및 래트 MGAT-2(10 μg)의 존재 하에 37 ℃에서 16 시간 동안 인큐베이션하였다. 인큐베이션 후에, 반응 생성물을 감소된 질량 크로마토그래피 분석 및 온전한 질량 크로마토그래피 분석에 가하였다. 도 16a에 나타낸 감소된 질량 크로마토그래피 결과로서, 50,194 Da의 분자량의 중쇄를 갖는 트리-만노실 코어 트라스투주맙 항체에 비해서, 중쇄가 약 244 Da x 2 = 488의 분자량으로 GlcNAz 분자를 두 개 더 함유하고, 각각의 중쇄가 약 50,680 Da의 분자량을 갖는 트리-만노실 트라스투주맙-4GlcNAz 항체 생성물을 얻었다. 이러한 결과는, MGAT-1 및 MGAT-2를 통해서, GlcNAz가 포유동물 세포에 의해서 생성된 트리-만노실 코어 트라스투주맙 항체의 각각의 중쇄의 α-3 만노오스 및 α-6 만노오스에 컨주게이션될 수 있음을 암시한다.
실시예 9에 기재된 공정에 따르면, DBCO-(PEG)4-DM1을 상기 얻은 트리-만노실 트라스투주맙-4GlcNAz 항체를 함유하는 트리스 완충액(pH 7.0)에 서서히 첨가하여 25℃에서 16 시간 동안 클릭 화학 반응을 수행하였다. 반응 및 정제 후에, 생성물을 감소된 질량 크로마토그래피 분석 및 온전한 질량 크로마토그래피 분석에 가하였다. 도 16b에서의 감소된 질량 크로마토그래피 분석 결과는 생성물의 중쇄가 53,509 Da의 분자량을 가지며, 이는 두 개의 DBCO-(PEG)4-DM1 분자(1,413 Da x 2 = 2,826의 분자량)가 트리-만노실 트라스투주맙-4GlcNAz 항체의 중쇄에 컨주게이션되었음을 나타내고 있음을 밝히고 있다. 이러한 결과는 도 16c에서의 온전한 질량 크로마토그래피 분석 결과로부터 추가로 확인된다. 이러한 결과는, 4개의 DBCO-(PEG)4-DM1 분자(1,413 Da x 4 = 5,652 Da의 분자량)을 함유하며 약 153,868 Da의 분자량을 갖는 트리-만노실-4(GlcNAc-트리아졸-DBCO-(PEG)4-DM1) ADC 생성물이 얻어졌음을 밝히고 있다.

Claims (32)
- 화학식(1)의 구조를 포함하는 당단백질-페이로드 컨주게이트(glycoprotein-payload conjugate)를 제조하는 방법으로서,
(i) 화학식(2)를 갖는 글리칸을 포함하는 당단백질을 β-N-아세틸글루코사미니다제와 반응시켜 화학식(3)의 트리-만노실 코어를 포함하는 변형된 당단백질을 생성시키는 단계;
(ii) 만노실 (α-1,3-)-당단백질 β-1,2-N-아세틸글루코사미닐트랜스페라제 및 만노실 (α-1,6-)-당단백질 β-1,2-N-아세틸글루코사미닐트랜스페라제의 존재 하에 화학식(3)의 트리-만노실 코어를 포함하는 변형된 당단백질을 R이 아지도, 케톤 기 또는 알데하이드인 UDP-GlcNAc-(CH2)0-8-R과 반응시켜 두 개의 GlcNAc-(CH2)0-8-R 당이 Man2 및 Man3의 각각의 β-1,2 위치에 각각 결합되게 하여, 화학식(4)의 글리칸 모이어티가 형성되게 하는 단계; 및
(iii) 페이로드가 동일하거나 상이한 두 개의 컨주게이터-링커-페이로드(conjugator-linker-payload)를 화학식(4)의 글리칸 모이어티와 반응시켜 화학식(1)의 구조를 포함하는 당단백질-페이로드 컨주게이트를 생성시키는 단계를 포함하는 방법:
