Abstract
본 발명은 티에노피리미딘 화합물, 또는 이의 약학적으로 허용가능한 염, 또는 이의 약학적으로 허용가능한 염의 수화물 및 유연물질의 생성을 저해하는 하나 이상의 저융점 안정화제를 포함하는 약학적 조성물, 및 이를 사용 및 포함하여 제조된 약학적 제형에 관한 것이다.
Description
본 발명은 암 세포 성장을 효과적으로 저해하는 아민 유도체 또는 이의 약학적으로 허용가능한 염 또는 이의 약학적으로 허용가능한 염의 수화물 및 저융점을 갖는 안정화제를 포함하는 약학적 조성물, 및 이를 이용하고 포함하여 제조된 약학적 제형에 관한 것이다.
세포는 다양한 신호 전달 경로와 관련되어 있다. 이와 같은 신호 전달 경로는 유기적으로 서로 연결되어 있고, 복잡한 기전을 형성하고 세포 증식, 성장, 사망 등을 조절한다 (William G. Kaelin et al., Nature Reviews Cancer, 2005, 5:689-698). 이에 따라, 세포 내 조절 기능이 유전적 또는 환경적 원인으로 인해 불균형하면, 신호 전달은 비정상적으로 증폭되거나 또는 파괴되고, 종양과 같은 질병을 일으킨다 (Douglas Hanahan et al., The Hallmarks of Cancer, 2000, 100:57-70).
단백질 티로신 키나제가 세포간 신호 전달에 있어 매우 중요한 역할을 하고 (Irena Melnikova, et al., Targeting protein kinases, 2004, 3:993-994), 이의 비정상적 발현 또는 변이가 암세포에서 흔히 관찰되는 것이 보고되어 있다. 단백질 티로신 키나제는 ATP로부터 단백질 기질 상의 티로신으로의 인산기 이동을 촉매하는 효소이다. 다수의 성장 인자 수용체 단백질이 이와 같은 티로신 키나제 활동에 의해 세포간 신호 전달을 수행한다. 성장 인자와 이의 수용체 사이의 상호작용이 세포 성장의 정상 조절에 필수적이다. 그러나, 수용체에 의한 신호 조절이 수용체 변이 또는 과발현으로 인해 기능부전이 이루어지면, 종양 세포가 발생하고 최종적으로 암을 유발한다.
다수의 성장 인자 및 이의 수용체가 단백질 티로신 키나제로서의 그 역할에 대해 연구되고 있다. 이들 중, 상피세포성장인자 (EGF) 및 이의 수용체 (EGFR) 티로신 키나제에 대한 연구가 활발히 진행되고 있다 (Nancy E. Hynes et al., Nature Reviews Cancer, 2005, 5:341-354). 수용체 부분 및 티로신 키나제 부분을 포함하는 EGFR 티로신 키나제는 세포막을 관통하여 위치하므로, 세포외 신호를 세포 내로 전달하는 역할을 한다.
의약의 활성성분의 순도는 안전하고 효과적인 약학적 제제를 제공하는데 있어 중요한 요소이다. 의약의 유연물질은 치료 시에 다양한 부작용을 유발할 수 있다. 문제가 되는 의약 유연물질은, 완전히 제거가 불가능한 유연물질 또는 활성성분 및/또는 최종 의약 제제/약학적 제형 제조 공정에서 생성되는 유연물질; 및 온도, 습도, 및 빛과 같이 다양한 환경적 요인에 의하여 최종 약학적 제형에서 생성되는 보관 시 분해 산물을 포함한다.
본 발명의 목적은 최종 약학적 제형 제조 후에 낮은 유연물질 수준을 갖는 약학적 조성물을 제공하는 것으로, 상기 약학적 조성물은 암세포의 성장을 저해하는 아민 유도체 또는 이의 약학적으로 허용가능한 염 또는 이의 약학적으로 허용가능한 염의 수화물을 포함한다.
본 발명의 또 다른 목적은 상기 약학적 조성물을 사용하고 포함하여 제조된 약학적 제형을 제공하는 것이다.
상기의 목적을 달성하기 위하여, 본 발명은 약학적 활성성분(API)으로서 화학식 1로 표시된 화합물 N-(3-(2-(4-(4-메틸피페라진-1-일)페닐아미노)티에노[3,2-d]피리미딘-4-일옥시)페닐)아크릴아미드, 또는 이의 약학적으로 허용가능한 염 또는 이의 약학적으로 허용가능한 염의 수화물; 및 하나 이상의 저융점 안정화제, 및 선택적으로 하나 이상의 약학적으로 허용가능한 부형제를 포함하는 약학적 조성물을 제공한다.

본 발명은 화학식 1의 화합물, 또는 이의 약학적으로 허용가능한 염, 또는 이의 약학적으로 허용가능한 염의 수화물, 및 하나 이상의 저융점 안정화제를 포함하는, 암, 종양, 염증질환, 자가면역질환 또는 면역-매개 질환을 예방 및/또는 치료용 약학적 조성물을 제공한다.
상기의 목적을 달성하기 위하여, 본 발명은 또한 상기 약학적 조성물을 사용하고 포함하여 제조된 약학적 제형을 제공한다.
본 발명의 약학적 조성물은 제형 제조 공정 시 생성되는 약학적 활성 성분의 더 낮은 수준의 유연물질을 제공한다. 상기 약학적 조성물을 사용하고 포함하여 제조된 약학적 제형은 장기간 보관 시에도 품질을 유지할 수 있다.
도 1은 실시예 1의 정제 제조 시 생성된 유연물질 총량(중량% 기준)을 폴리에틸렌 글리콜을 저융점 안정화제로 포함하지 않은 비교예 1 내지 4의 정제와 비교한 안정성 테스트 결과를 나타낸다.

본 발명자들은 암 세포의 성장을 저해하는 아민 유도체 또는 이의 약학적으로 허용가능한 염 또는 이의 약학적으로 허용가능한 염의 수화물을 포함하는 향상된 안정성을 지닌 약학적 조성물을 제조하기 위해 연구해왔다. 본 발명자들은 약학적 조성물에서 저융점 안정화제를 사용하는 것이 특정 유연물질을 포함하는 약학적 투여 제형이 특정 유연물질을 더 낮은 수준으로 포함하도록 유도하는 것을 발견하였다.
본 발명의 일 양상는 약학적 활성 성분으로 화학식 1로 표시된 화합물 또는 이의 약학적으로 허용가능한 염 또는 이의 약학적으로 허용가능한 염의 수화물; 하나 이상의 저융점 안정화제 및, 선택적으로, 하나 이상의 약학적으로 허용가능한 부형제를 포함하는 약학적 조성물을 제공한다.
이하에서는 상기 양상의 본 발명의 약학적 조성물의 특징 및 성분의 종류를 더욱 상세히 설명한다.
(1) 약학적 활성성분
발명의 약학적 조성물에서, 하기 화학식 1의 화합물 또는 이의 약학적으로 허용가능한 염 또는 이의 약학적으로 허용가능한 염의 수화물은 약학적 활성성분(API)으로서 사용될 수 있다.

본 발명에 따른 상기 화학식 1의 화합물은 다양한 방법으로, 예를 들어 WO 2011/162515에 기재된 방법으로 제조될 수 있다.
본 발명에서, 약학적으로 허용가능한 염은 관련 분야에서 일반적으로 사용되는 것일 수 있다. 예를 들면, 염산, 황산, 이황산, 질산, 인산, 과염소산, 브롬산 등과 같은 무기산과의 염; 포름산, 초산(아세트산), 프로피온산, 옥살산, 숙신산, 벤조산, 구연산(시트르산), 말레인산, 말론산, 말산, 타르타르산, 글루콘산, 락트산, 겐티스산, 푸마르산, 락토비온산, 살리실산, 프탈산, 엠본산, 아스파르트산, 글루탐산, 캄실산, 베실산 또는 아세틸살리실릭산(아스피린)과 같은 유기산과의 염일 수 있다. 또한 상기 약학적으로 허용가능한 염은 칼슘, 나트륨, 마그네슘, 스트론튬, 칼륨 등의 알칼리 금속과의 반응에 의한 금속염 등의 형태일 수도 있다. 하지만 본 발명은 이에 한정되지 않는다.
본 발명의 일 실시예에서, 약학적 활성성분은 상기 화학식 1의 화합물의 염산염이다.
일 구체예에서, 상기 화학식 1의 화합물의 염산염은 무정형이다.
다른 일 구체예에서, 상기 화학식 1의 화합물의 염산염은 결정형이다.
본 발명의 특정 일 구체예에서, 상기 결정질 염산염은 무수화물이다.

상기 화학식 1의 화합물의 염의 결정형
상기 화학식 1의 화합물의 염은 결정형, 무정형, 또는 이의 혼합물로, 바람직하게는 결정형으로 제조될 수 있다. 상기 화학식 1의 화합물의 염산염의 결정형, 특히 결정형 이염산염 일수화물은 우수한 안정성을 가지며 그러므로 제제화를 용이하게 하는 물리화학적 특성을 가졌다는 점에서 바람직하다.
본 발명의 약학적 조성물의 일 구체예에서, 이염산염 수화물의 결정형, 바람직하게는 이염산염 일수화물의 결정형이 약학적 활성성분(API)이다. 가장 바람직하게는, 상기 이염산염 일수화물은 1X로 지정된 결정형이다(PCT/KR2016/015535 참조). 상기 결정형 1X는 높은 수용성, 탁월한 비흡습성/비가습성 및 안정성을 갖는다.
상기 바람직한 결정형 1X는 Cu-Kα 광원으로 조사하였을 때 5.6°±0.2° 및 27.3°±0.2°의 회절각 2θ에서의 피크를 포함하는 X-선 분말 회절(XRPD) 패턴을 나타낸다(XRPD1-1).
더욱 구체적으로, 상기 결정형 1X는 Cu-Kα 광원으로 조사하였을 때 5.6°±0.2°, 21.1°±0.2°, 및 27.3°±0.2°의 회절각 2θ에서의 피크를 포함하는 XRPD 패턴을 나타낸다(XRPD1-2).
더욱 구체적으로, 상기 결정형 1X는 Cu-Kα 광원으로 조사하였을 때 5.6°±0.2°, 11.1°±0.2°, 및 27.3°±0.2°의 회절각 2θ에서의 피크를 포함하는 XRPD 패턴을 나타낸다(XRPD1-3).

더욱 구체적으로, 상기 결정형 1X는 Cu-Kα 광원으로 조사하였을 때 5.6°±0.2°, 10.7°±0.2°, 11.1°±0.2°, 14.0°±0.2°, 20.8°±0.2°, 21.1°±0.2°, 22.5°±0.2°, 및 27.3°±0.2°의 회절각 2θ에서의 피크를 포함하는 XRPD 패턴을 나타낸다(XRPD1-7).
상기 피크들은 약 10% 이상의 상대 강도 (I/Io)를 갖는 것일 수 있다.
더욱 구체적으로, 상기 결정형 1X는 Cu-Kα 광원으로 조사하였을 때 표 A에 포함된 것과 같은 회절 각에서의 피크를 갖는 XRPD 패턴을 나타낸다.

더욱 구체적으로, 상기 결정형 1X는 그13C CP/MAS TOSS ssNMR 스펙트럼이44.6 ± 0.2 ppm, 45.4 ± 0.2 ppm, 50.8 ± 0.2 ppm, 및 56.6 ± 0.2 ppm의 13C 화학적 이동에서의 피크를 포함할 수 있다(ssNMR1-2).
상기 결정형 1X는 그13C CP/MAS TOSS ssNMR 스펙트럼이 149.6 ± 0.2 ppm, 152.6 ± 0.2 ppm, 및 164.3 ± 0.2 ppm의 13C 화학적 이동에서의 피크를 포함할 수 있다(ssNMR1-3).

(b) 44.6 ± 0.2 ppm 및 56.6 ± 0.2 ppm의 13C 화학적 이동에서의 피크를 포함하는 13C CP/MAS TOSS ssNMR.
상기 결정형 1X는 하기 특징을 가질 수 있다:
(a) Cu-Kα 광원으로 조사하였을 때 5.6°± 0.2° 및 27.3°± 0.2°의 회절각 2θ에서의 피크를 포함하는 XRPD 패턴; 및
(b) 149.6 ± 0.2 ppm, 152.6 ± 0.2 ppm, 및 164.3 ± 0.2 ppm의 13C 화학적 이동에서의 피크를 포함하는 13C CP/MAS TOSS ssNMR.
상기 결정형(예. 1)은 또한 상기 기재된 XRPD 피크(XRPD1-1 내지 XRPD1-7) 및 13C 화학적 이동(ssNMR1-1 내지 ssNMR1-5)의 임의의 다른 조합이 특징일 수 있다.
샘플의 XRPD 분석은 3° 2θ 내지 40°2θ의 범위에서 D8 Advance (Bruker ASX, 독일) 분석기를 이용하여 수행하였다. 샘플의 양이 100 mg 미만인 경우, 약 5 mg 내지 10 mg의 샘플을 샘플 홀더에 고정된 유리 슬라이드 상에 부드럽게 압착시켰다. 샘플의 양이 100 mg 초과인 경우, 약 100 mg의 샘플을 샘플 표면이 평활하도록 플라스틱 샘플 홀더에 부드럽게 압착시키고, 샘플 홀더 수준 바로 위에 배치하였다.
측정은 하기와 같이 수행하였다:

고체상 핵자기 공명 분광법(Solid State Nuclear Magnetic Resonance Spectroscopy: ssNMR)은 고체 상에서 예를 들어 하기와 같이 수행하였다.
100 mg 양의 샘플을 칭량하여 4 mm 샘플 튜브에 부가하였다. 13C NMR 스펙트럼(13C CP/MAS TOSS ssNMR)을 하기의 조건에서 4 mm 프로브 타입(probe type) CP/MAS BB-1H를 갖는 Bruker Avance II 500 MHz Solid NMR 시스템 (Bruker, 독일) 분석기를 이용하여 실온에서 기록하였다:
주파수: 125.76 MHz,
스펙트럼 폭: 20 kHz,
매직 각(magic angle)에서의 샘플의 회전 속도: 5 kHz,

상기 결정형 1X는 칼 피셔(Karl Fischer) 적정기(795KFT Titrino (Metrohm, 스위스)를 사용하여 측정시 약 3.1 %의 함수량을 가질 수 있고(일수화물의 이론 함수량 수치는 3.11 %), 시차주사 열량측정법(STA-1000, Scinco, 한국)으로 측정 시 약 202 ℃ 내지 225 ℃의 용융점을 가질 수 있다.
일 구체예에서, 본 발명에 따른 약학적 조성물은 상기 화학식 1의 화합물의 염산염을 약학적 활성성분으로 포함하고, 그 약학적 활성 성분의 50 중량% 이상은 상기에 기재된 바와 같은 결정형 1X 형태로 존재한다. 바람직하게는 상기 약학적 조성물에서 약학적 활성성분의 80 중량% 이상, 더욱 바람직하게는 90 중량% 이상, 가장 바람직하게는 95 중량% 이상의 약학적 활성 성분은 상기 정의된 바와 같은 결정형 1X 형태로 존재한다.
상기 약학적 활성성분은 약리 활성을 나타내기에 일반적으로 효과적인 중량으로 함유되고, 상기 화학식 1의 화합물의 형태로서 본 발명의 단위 약학적 제형 당 1 mg 내지 1,000 mg, 예를 들어, 50 mg 또는 75 mg 또는 150 mg 또는 200 mg 또는 400 mg 또는 600 mg 또는 800 mg으로 함유되거나 또는 이의 상응하는 중량의 약학적으로 허용가능한 염 또는 약학적으로 허용가능한 염의 수화물로 포함될 수 있다. 일 구체예에서 단위 약학적 제형 당 이염산염 일수화물의 함량은 200 mg의 유리 염기(화학식 1)에 상응하는 양이다. 다른 구현예에 있어서 단위 약학적 제형 당 이염산염 일수화물의 함량은 400 mg의 유리 염기(화학식 1)에 상응하는 양이다. 약학적 조성물에서 약학적 활성성분의 양은 (중량% 기준) 약 1 % 내지 65 %, 바람직하게는 약 55 % 내지 65 % 범위이다.
(2) 저융점 안정화제
본 발명의 약학적 조성물은 특정 특성을 갖는 저융점 안정화제를 하나 이상 포함한다.
본 발명에 따른 약학적 조성물에 사용하기에 적합한 저융점 안정화제는 제형 제조 공정 시 특정 유연물질 생성을 저해하고 80 ℃ 이하의 용융점을 갖는다. 예를 들어, 상기 저융점 안정화제는 폴리에틸렌 글리콜 (예를 들어, PEG 1000, 1500, 1540, 2000, 3000, 4000, 6000, 8000, 20000 및 35000), 글리세릴 베헤네이트, 글리세릴 모노스테아레이트, 소르비탄 모노팔미테이트 또는 소르비탄 모노스테아레이트와 같은 소르비탄 지방산 에스테르, 폴리옥실 150 디스테아레이트와 같은 폴리옥시에틸렌-폴리옥시프로필렌 블록 공중합체 및 폴록사머 (예를 들어 188 및 407), 에틸렌 글리콜 스테아레이트, 라우르 산과 같은 융점이 80 ℃ 이하인 지방산, 팔미트산 또는 스테아르산, 및 이들의 임의의 혼합물로 구성된 군으로부터 선택될 수 있다.
상기 저융점 안정화제는 약학적 활성성분 1 중량부를 기준으로 약 0.15 내지0.6 중량부 수준일 수 있다. 상기 범위에서 사용되는 상기 저융점 안정화제는제형 제조 시, 예를 들면 정제 제조 시, 가공성 및 약물 방출 특성의 타협없이 유연물질 생성의 감소를 제공한다.
본 발명의 약학적 조성물의 일 구체예에서, 상기 저융점 안정화제는 폴리에틸렌 글리콜이다. 일 구체예에서, 분자량 1,000 내지 35,000의 폴리에틸렌 글리콜이 사용된다. 다른 일 구체예에서, 분자량 1,000 내지 8,000의 폴리에틸렌 글리콜이 사용된다. 예를 들어, 2,000 내지 8,000의 분자량 범위가 고형 제제에서 바람직하게 사용될 수 있다. 용융점과 같은 특성은 등급에 따라 다양할 수 있다. 예를 들어, 마크로골 8000(Ph. Eur.)으로도 불리는 PEG 8000이 가장 바람직하며 55 ℃ 내지 63 ℃의 저융점을 갖는다
다른 구체예에 있어서, 상기 저융점 안정화제는 API 1 중량부 기준으로, 약 0.15 중량부 내지 0.6 중량부, 더 구체적으로 약 0.2 중량부 내지 0.5 중량부, 더욱 구체적으로 약 0.2 중량부 내지 0.3 중량부, 및 더더욱 구체적으로 약 0.21 중량부 내지 0.26 중량부 함량의 폴리에틸렌 글리콜이다. 바람직하게는, 사용되는 상기 폴리에틸렌 글리콜은 PEG 8000이다.
본 발명의 작동 원리를 특정 이론에 의하여 한정하려는 것은 아니나, 단지 본 발명을 더 잘 이해할 수 있기 위한 측면에서, 저융점 안정화제는 마찰에 대한 보호를 제공하는 것으로 여겨진다:
건조 과립화 및 압축 시, 압력이 약학적 조성물에 가해진다. 분말 혼합물 내 입자는 이동하고, 혼합물은 더 조밀해 진다. 상기 입자 움직임은 입자들 간의 마찰을 유발하고 이는 코어 정제 제제 내 특정 유연물질의 증가의 원인일 수 있다. 폴리에틸렌글리콜 8000(PEG 8000)과 같은 저융점 안정화제는 건조 과립화 및 압축 시 기계적 응력(mechanical stress)으로부터 약학적 활성 성분을 보호하기 때문에 안정화제로서 작용할 수 있다.
상기 저융점 안정화제를 약 0.15 중량부 미만의 함량으로 사용할 경우, 유연물질 생성을 저해하지 못할 수 있다. 상기 저융점 안정화제를 약 0.6 중량부 초과의 함량으로 사용할 경우, 약물 방출에 문제가 생길 수 있다.
(3) 약학적으로 허용가능한 부형제
본 발명에 따른 약학적 조성물은 하나 이상의 약학적으로 허용가능한 부형제를 포함할 수 있다. 상기 약학적으로 허용가능한 부형제는 해당 분야에서 일반적으로 사용되는 것일 수 있다. 예를 들어, 상기 약학적으로 허용가능한 부형제는 희석제, 결합제, 붕해제, 활택제, 착색제, 유동화제, 흡착제, 및 이의 임의의 혼합물, 그리고 약학 분야에서 일반적으로 사용되는 임의의 부형제로 구성되는 군으로부터 선택될 수 있다.
희석제(충진제라고도 함)는 고형 약학적 조성물의 크기를 증가 시키서, 환자 및 보호자가 다루기에 더 용이한 약학적 조성물을 함유하는 약학적 제형을 제조할 수 있다. 약학적 조성물에서 사용되는 희석제는 고형 약학적 조성물에서 일반적으로 사용되는 희석제를 포함한다. 본 발명의 일 구체예에서, 희석제는: 고체 유기물로서,

붕해제는 환자의 체내에서 고형 약학적 조성물의 용해도를 높인다. 상기 약학적 조성물에서 사용되는 붕해제는 고형 약학적 조성물에서 일반적으로 사용되는 붕해제를 포함한다. 본 발명의 일 구체예에서, 붕해제는 전분 글리콜산 나트륨, 크로스포비돈, 크로스카멜로스, 카르복시메틸셀룰로오스 나트륨 및 건조 옥수수 전분, 및 이의 임의의 혼합물로 구성된 군으로부터 선택되지만, 이에 한정되는 것은 아니다. 바람직한 붕해제는 크로스포비돈이다.
상기 붕해제는 약학적 조성물의 총 중량 기준으로 약 1 내지 30 중량%, 구체적으로 약 2 내지 6 중량%, 바람직하게는 약 3 중량%으로 함유될 수 있다.
활택제는 공정 시에 장비에의 점착을 방지하여 공정을 용이하게 하기 위해 약학적 조성물에 부가한다. 약학적 조성물에서 사용되는 활택제는 고형 약학적 조성물에서 일반적으로 사용되는 활택제를 포함한다. 본 발명의 일 실시예에서, 활택제는 스테아르산, 마그네슘 스테아레이트, 칼슘 스테아레이트, 스테아릴푸마르산나트륨, 글리세롤 트리베헤네이트, 폴리에틸렌 글리콜, 및 이의 임의의 혼합물로 구성된 군으로부터 선택되지만, 이에 한정되는 것은 아니다. 바람직한 활택제는 마그네슘 스테아레이트이다.
상기 활택제는 약학적 조성물의 총 중량 기준으로 약 0.5 내지 5 중량%, 구체적으로 0.5 내지 2 중량%, 바람직하게는 약 1 중량%으로 함유될 수 있다.

본 발명에서 사용된 예시적인 코팅제는 즉시 방출 필름 형성제로서 히드록시 프로필 셀룰로오스, 히드록시 프로필메틸 셀룰로오스, 폴리비닐 알코올 및 폴리비닐 알코올-폴리에틸렌 글리콜 그라프트 폴리머(Kollicoat IR, BASF); 장용성 코팅제로서 (메타)아크릴산 코폴리머 (EUDRAGIT, Evonik Industries), 프탈산 히드록시 프로필메틸 셀룰로오스 및 프탈산 셀룰로오스 아세테이트; 및 서방형 코팅제로서 셀룰로오스 아세테이트, 에틸 셀룰로오스 및 폴리비닐 아세테이트를 포함할 수 있다.

실시예 1 내지 3: 폴리에틸렌 글리콜을 함유하는 정제의 제조
약학적 활성성분(API)으로서, 화학식 1의 화합물의 이염산염 일수화물 염 분말(한미정밀화학(주))을 사용하였다. 상기 API, 만니톨(ROQUETTE), 미세결정질 셀룰로오스(MINGTAI), 히드록시프로필 셀룰로오스(L-형, NISSO), 및 폴리에틸렌 글리콜 8000(Ph. Eur, 일본: 마크로골 6000, SANYO)를 하기 표 1에 기재된 약학적 조성에 따라 혼합하였고, 그 결과 혼합물을 롤 컴팩터 (TFC-LAB, FREUND)를 사용하여 건조 과립화하였다.
크로스포비돈(BASF)을 상기 제조된 과립에 첨가 및 혼합하고, 마그네슘 스테아레이트(PETER GREVEN)를 이에 첨가하여 최종 혼합물을 획득하였다. 타정기(GRC-18, 세종파마텍(주))를 사용하여 상기 얻어진 최종 혼합물로부터 경도 14 kp의 정제를 제조하였다. 상기 제조된 정제를 자동 코팅기(SFC-30, 세종파마텍(주))를 사용하여 정제의 총 중량 기준으로 2 중량%의 Opadry Y-1-7000(COLORCON) 코팅제로 코팅하였다.

실험예 1: 유연물질 함량 측정
상기 실시예 및 비교예에서 제조된 제제의 제조를 평가하기 위해, 유연물질 총량을 측정하였다. 유연물질의 측정 및 정량은 하기와 같이 수행하였다.
<테스트 방법>
사용된 기기: 고성능 액체 크로마토그래피(HPLC, Hitachi 2000 series, 일본)
검출기: UV 흡광계 (측정 파장: 254 nm)
칼럼: Inertsil ODS-2 (4.6x150 mm, 5 ㎛) 또는 이와 동등한 칼럼
유속: 1.0 mL/분
칼럼 온도: 30 ℃

<계산 방법>
각 유연물질의 함량은 254 nm에서의 자외선 조사 조건에서 고성능 액체 크로마토그래피(HPLC)의 크로마토그램에서 해당하는 피크 하 면적을 계산하여 결정하였다. 모든 유연물질 종의 모든 유연물질 함량을 산출하기 위해 해당하는 유연물질 및 API 사이의 피크 면적의 비율을 계산하였다.
하기 표 8 내지 13 및 도 1 내지 6에 나타낸 바와 같이, 상기 저융점 안정화제를 포함하는 본 발명의 약학적 조성물은 정제 제조 시 모든 유연물질을 유의적으로 억제할 수 있다. 이와 같은 제조 안정성의 향상은 상기 테스트 방법에 따른HPLC실행에서 1.4 또는 1.8의 상대적 체류 시간(relative retention time: RRT)을 가진 것들의 경우와 같이 특정 유연물질에 더욱 효과적이었다(데이터는 미표시).

표 8, 및 도 1에 나타난 바와 같이, 저융점 안정화제가 포함되지 않은 비교예 1 내지 4의 제제에서, 제조 직후 초기 유연물질 총량은 폴리에틸렌 글리콜을 저융점 안정화제로 함유하는 실시예 1의 제제와 비교하여 더 높았다. 다시 말해, 정제화 후 테스트된 비교예 정제에서 더 많은 유연물질이 관찰되었고, 정제의 경도는 증가하였다.
제조 시의 안정화 효과를 더 상세히 살펴보기 위해, 하기 정제화된 제제(실시예 1 및 비교예 1, 모두 경도 14) 및 정제화되지 않은 제제(실시예 5 및 비교예 5)를 테스트 하였다.

폴리에틸렌 글리콜은 안정화제로서 기능하고 저융점을 갖는 것이 특징이다. 그러므로, 정제가 폴리에틸렌 글리콜과 유사한 특성을 갖는 저융점 안정화제를 채용하여 제조되는 경우, 표 12 및 도 5에 나타난 바와 같이, 상기 안정화제는 상기 정제가 타정기에서 압력이 높아지는 경우에 노출되어도 마찰을 경감시킴으로 API로부터의 특정 유연물질의 생성을 효과적으로 저해할 수 있다.

반면에, 정제가 80 ℃ 초과의 용융점을 갖는 안정화제를 사용하여 제조된 경우, 결과물 제제는 표 13, 및 도 6에 나타난 바와 같이 유연물질 프로파일이 안정화제가 결여된 비교예 1과 유사하다. 약학적 활성성분으로서 N-(3-(2-(4-(4-메틸피페라진-1-일)페닐아미노)티에노[3,2-d]피리미딘-4-일옥시)페닐)아크릴아미드 하이드로클로라이드에 저융점 안정화제의 첨가는 정제의 제조 후 낮은 수준의 유연물질을 보장하는데 있어 중요하다고 결론내릴 수 있다.

- 청구항 2에 있어서, 상기 약학적 활성성분은 화학식 1의 화합물의 염산염 결정형인 것인 약학적 조성물.
- 청구항 3에 있어서, 상기 약학적 활성성분은 화학식 1의 화합물의 염산염 결정형의 수화물인 것인 약학적 조성물.
- 청구항 3에 있어서, 상기 약학적 활성성분은 화학식 1의 화합물의 이염산염 결정형인 것인 약학적 조성물.
- 청구항 5에 있어서, 상기 약학적 활성성분은 화학식 1의 화합물의 이염산염 결정형의 수화물인, 바람직하게는 일수화물인 것인 약학적 조성물.

- 청구항 1 내지 17 중 어느 한 항에 있어서, 상기 약학적 조성물은 하나 이상의 희석제를 추가적으로 포함하는 것인 약학적 조성물.
- 청구항 18에 있어서, 상기 하나 이상의 희석제는 만니톨, 미세결정질 셀룰로오스, 및 이들의 임의의 혼합물로부터 선택되는 것인 약학적 조성물.
