실시예
대식세포는 선천성 및 적응성 면역 모두의 중요한 조절 인자이다. 대식세포 가소성이 세포 고유 인자에 의해 어떻게 조절되는 지는 불완전하게 이해되고 있다. 하기에 기술된 데이터는 인간 호메오박스 전사인자, Hom-1이 M1 표현형으로 대식세포의 분극(polarization)을 유도하는 중추적인 역할을 한다는 것을 입증한다. 가소성은 대식세포의 특징이다. 환경적인 단서에 반응하여, 대식세포는 고전적인 전-염증성 M1 표현형으로부터 구별된 특징을 갖는 다양한 M2 표현형의 범위로, 표현형의 스펙트럼을 나타낸다. 코르티코스테로이드는 염증성 질환을 관리하기 위해 광범위하게 사용되어왔고 대식세포의 M2 표현형을 유도하는 것으로 나타났다. 점막 궤양을 유발하는, 상피 세포의 세포사멸(apoptosis)은, IBD의 특징이다. 조직 염증은 점막 상피 세포의 세포 사멸의 주된 원인으로 생각되었다. 일괄 조직 배양을 사용하여, 정상 대조군 조직 내 상피 세포의 세포사멸 속도와 비교하여 염증성 점막으로부터 분리된 조직 내 점막 상피 세포의 세포 사멸의 속도가 더 빠르다는 것을 발견하였다. Hom-1 MO가 일괄 조직 배양에 첨가되었을 때, 대조군 MO가 아닌 Hom-1 MO가 조직 배양 내 상피 세포의 세포 사멸에 강한 억제 효과를 나타냄을 발견하였다. Hom-1 MO가 조직 염증을 완화시킬 수 있다는 발견과 함께, 상기 데이터는 Hom-1 MO가 염증을 조절하는 작용제로서 기능할 수 있음을 시사하였다.
단핵구의 분리 및 배양
보스톤 아동 병원의 건강한 성인 기증자로부터 말초 혈액 단핵구 세포 (PBMC)는 피콜(Ficoll) 밀도 구배 원심분리로 분리되었다. 인간 재료의 실험은 브리검 여성 병원의 기관 검토 위원회에 의해 승인을 받은 지침에 따라 수행되었다. CD14+ 단핵구는 항-CD14-코팅된 마이크로비드(Miltenyi Biotec)를 사용하여 PBMC로부터 정제되었다. 신선하게 분리된 CD14+ 단핵구의 순도는 유세포 분석에 의해 분석하였을 때 95% 이상이었다. 단핵구는 12-웰 플레이트에서 10% 소태아혈청(FBS)를 함유한 RPMI 1640 배지에 1 x 106 세포/ml로 배양되었다. RNA 간섭

FACS 분석
단핵구/대식세포의 표현형 분석은 형광 염료 접합된 항체로 세포의 면역 표지 후 유세포 분석을 사용하여 수행되었다. 하기의 항체는 사용되었다: PE-접합된 항-CD71, CD11b, CD11c, CD16, CD64, CD80, CD86, HLA-DR, CD14, TLR4, IL1-β 및 TNF-α, 및 FITC-접합된 항-CD40, CD36 (eBioscience); FITC-접합된 항-만노스 수용체(MR), 및 비접합된 마우스 항-MCSFR (R&D 시스템). 동위원소(isotope) 대조군 표 작성은 병행하여 수행되었다. 항체는 공급체에 의해 권장되는 대로 희석되었다. 마우스 IgG 항체에 대한 PE-접합된 토끼가 2차 M-CSFR 염색에 사용되었다. 표지된 세포는 셀퀘스트 소프트웨어(CellQuest software)를 사용하여 FACScan 유동 세포 계측기(BD Bioscience)로 분석되었다. 결과는 동위원소 대조군 항체로 수득된 MFI를 뺀 후 양성 세포의 백분율 및/또는 평균 형광 감도(MFI) 값으로 표현된다.
RT-PCR
트리졸(TRIzol) 시약에 의해 총 RNA는 분리되었고, 제조사의 프로토콜에 따르는 수퍼스크립트 III 제 1 가닥 합성 시스템(SuperScript III First-Strand Synthesis System)(Invitrogen)으로 제 1 가닥 cDNA 합성을 위해 사용되었다. 종래의 PCR로 Hom-1 cDNA를 증폭시키기 위하여, AccuPrimeTM Taq DNA 중합효소 시스템 (Invitrogen)은 제조사의 지시에 따라 사용되었다. 사이토카인 측정
대장균(E.coli) LPS (Sigma) 및 IFN-γ (PeproTech) 처리된 대식세포 또는 LPS 처리된 U937 세포의 상층액 내 IL-1β 및 TNF-α 및 IL12p70의 수준을 eBiosciences 로부터 수득된 ELISA 키트(kit)를 사용하여 정량화하였다. 분석은 제조사의 지시에 따라 수행되었다.
활성 산소 종(ROS) 및 산화 질소(NO)의 검출
활성화된 대식세포 내 ROS 수준은 결과가 형광 현미경 및 유동 세포 계측법에 의해 분석되는 것을 제외하고는 기본적으로 제조사의 지침에 따라 이미지-iT®LIVE 녹색 반응성 산소 종 검출 키트(Green Reactive Oxygen Species Detection Kit)(Invitrogen)로 검출되었다. NO 수준은 제조사가 제공한 프로토콜에 따라 아질산염 측정을 위한 Griess 시약 키트(Griess Reagent Kit for Nitrite Determination) (Invitrogen)에 의해 측정되었다.

통계 분석
데이터는 한 쌍의 스튜던트 t 테스트(Student's t test)(양측) 및 윌콕슨 순위합 테스트(Wilcoxon rank-sum test)를 사용하여 분석되었다. p 값이 <0.05의 차이로 통계적으로 유의한 것으로 고려되었다.

Claims (24)
- Hom-1의 발현을 억제하기 위한 핵산 분자를 이를 필요로 하는 대상체에게 투여하는 것을 포함하는, 대상체 내 염증성 질환을 치료하는 방법.
- 제1항에 있어서, 상기 핵산 분자는 RNAi 제제(agent) 또는 안티센스 올리고뉴클레오티드(antisense oligonucelotide)인 방법.
- 제2항에 있어서, 상기 핵산 분자는 모르폴리노(morpholino) 올리고뉴클레오티드인 방법.
- 제3항에 있어서, 상기 모르폴리노 올리고뉴클레오티드는 서열번호: 3, 4, 5 또는 6의 서열을 갖는 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 핵산 분자는 국소(topically), 경구(orally), 직장(rectally), 비강(nasally), 정맥 내(intravenously), 관절 내(intraarticularly), 결막 내(conjunctivally), 두개 내(intracranially), 복강 내(intraperitoneally), 흉강 내(intrapleurally), 근육 내(intramuscularly), 척수강 내(intrathecally) 또는 피하(subcutaneously)로 투여되는 방법.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 상기 핵산 분자는 노출되어 투여되는 방법.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 대상체로부터 수득된 염증성 조직 샘플은 대조군 수준과 비교하여 더 높은 수준의 Hom-1을 발현하는 방법.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 투여 단계에 앞서, 대조군 수준과 비교하여 상기 대상체로부터 수득된 염증성 조직 샘플 내 더 높은 발현 수준의 Hom-1을 검출하는 단계를, 추가로 포함하는 방법.
- 제8항에 있어서, 상기 투여 단계 후, 상기 대상체로부터 수득된 염증성 조직 샘플 내 Hom-1 또는 염증성 사이토카인의 발현 수준을 검출하는 단계를 추가로 포함하는 방법.
- 제1항 내지 제9항 중 어느 한 항에 있어서, 상기 대조군 수준은 비-염증성 조직 샘플 내 Hom-1의 발현 수준에 상응하는 방법.
- Hom-1의 발현을 억제하기 위한 핵산 분자 및 약제학적으로 허용 가능한 담체를 포함하는, 염증성 질환 치료용 조성물.
- 제11항에 있어서, 상기 핵산 분자는 서열번호: 3, 4, 5 또는 6의 서열을 갖는 모르폴리노 올리고뉴클레오티드인 조성물.
- 제11항 또는 제12항에 있어서, 상기 조성물은 국소, 경구, 직장, 비강, 정맥 내, 관절 내, 결막 내, 두개 내, 복강 내, 흉강 내, 근육 내, 척수강 내 또는 피하의 투여 경로를 위해 제형화되는 조성물.
- 염증성 질환을 위한 치료제를 동정하는 방법으로서,
염증성 조직 샘플을 시험용 치료제와 접촉시키는 단계; 및
상기 조직 샘플 내 Hom-1의 발현 수준을 검출하는 단계를 포함하고,
상기 발현 수준이 대조군 수준보다 낮거나 또는 이와 동일한 경우, 상기 시험용 치료제는 염증성 질환을 위한 후보군 치료제인 방법.
- 염증성 질환을 위한 치료제를 이를 필요로 하는 대상체에서 선택하는 방법으로서,
상기 대상체로부터 수득된 염증성 조직 샘플을 치료제와 접촉시키는 단계;
대조군 수준과 비교하여 상기 조직 샘플 내 Hom-1보다 낮거나 또는 이와 동일한 발현 수준을 검출하는 단계; 및
상기 대상체에게 상기 치료제를 투여하는 단계를 포함하는 방법.
- 염증성 질환용 치료제를 이를 필요로 하는 대상체에서 효능을 모니터링하는 방법으로서,
상기 대상체가 상기 치료제로 투여된 후에 상기 대상체로부터 수득된 염증성 조직 샘플 내 Hom-1의 발현 수준을 검출하는 단계;
상기 검출된 수준을 대조군 수준과 비교하는 단계; 및
상기 비교에 기초하여 치료 결정을 하고, 상기, 검출된 수준이 상기 대조군 수준보다 높은 경우, 상기 대상체에게 상기 치료제 또는 다른 치료제를 지속적으로 투여하는 단계를 포함하는 방법.
- 제15항 또는 제16항에 있어서, 상기 치료제가 스테로이드, 비-스테로이드성 항-염증제, 면역-억제제인 방법.
- 제15항 내지 제17항 중 어느 한 항에 있어서, 상기 치료제는 Hom-1 억제제인 방법.
- 제15항 내지 제18항 중 어느 한 항에 있어서, 상기 치료제는 단백질, 펩타이드, 펩타이드유사체, 펩토이드, 세포, 항체 또는 이의 단편, 소분자 화합물, 핵산 분자, 또는 식물 추출물인 방법.
- 제15항 내지 제19항 중 어느 한 항에 있어서, 상기 대조군 수준은 치료제와 접촉하기 전 비-염증성 조직 샘플 또는 염증성 조직 샘플 내 Hom-1의 발현 수준에 상응하는 방법.
- 염증성 질환의 치료를 필요로 하는 대상체에서 염증성 질환을 치료하는 방법으로서,
Hom-1 억제제로 치료되거나 또는 Hom-1 억제제를 발현하기 위한 발현 작제물(construct)을 함유하는 변형된 대식세포, 단핵구, 또는 수지상 세포를 제공하는 단계로서, 상기 변형된 대식세포, 단핵구, 또는 수지상 세포는 대조군 수준과 비교하여 더 낮은 수준의 Hom-1을 발현하는 단계; 및
유효량의 상기 변형된 대식세포, 단핵구, 또는 수지상 세포를 상기 대상체에게 투여하는 단계를 포함하는 방법.
- 제21항에 있어서, 상기 Hom-1 억제제는 Hom-1의 발현을 억제하는 방법.
