[ Materials and Methods]

1. Extract of Nuri-japonica tree and its object Hexane Fraction  Produce

The extracts were collected from 70% (v / v) ethanol and filtered. The extracts were concentrated under reduced pressure to obtain the extracts of Nuri saplings. It was suspended and fractionated to obtain hexane, CH 2 Cl 2 , EtOAc and BuOH fractions.

2. Cell culture

HaCaT cells were cultured in Dulbecco's Modified Eagle Medium (DMEM, Gibco) supplemented with 10% fetal bovine serum (FBS, Gibco) and 1% penicillin-streptomycin And cultured in medium containing penicillin-streptomycin (PS, Gibco) at 37 ° C and 5% CO 2 .

Example  One. Cell survival rate  analysis

Example  2. MMP -1 expression level

Each sample was treated with HaCaT cells for 24 hours and irradiated with ultraviolet light (UV Crosslinker, Ultra Lum) at 20 mJ / cm 2. Then , using an MMP-1 ELISA kit (R & D Systems, Inc., Minneapolis, The expression level of MMP-1 was measured.

Specifically, HaCaT cells were plated on a 96-well plate, and then the hexane fraction, martynoside, and isomartynoside of the present invention were treated for each concentration. In addition, as a comparative example, the extracts of Nomad saponin, water fraction, ethyl acetate fraction, butanol fraction and verbascoside were treated by concentration. Then, ultraviolet (UV) irradiation was performed, and the supernatant of the culture was collected and centrifuged at 13,000 rpm for 5 minutes. The supernatant was then subjected to fluorescent assay using an ELISA kit to quantitatively analyze the expression level of MMP-1.

As a result, it was confirmed that the expression amount of MMP-1 remarkably increased in the group irradiated with ultraviolet light as compared with the negative control group not irradiated with ultraviolet light as disclosed in Figs. 3 and 4, and the sample of the present invention (martynoside ), Isomartynoside and hexane fractions of the extract of Quercus mongolica extract) significantly decreased the expression level of MMP-1 compared to the ultraviolet irradiation group.

In addition, it was confirmed that the amount of MMP-1 expression was remarkably decreased even when the sample of the present invention was compared with the control group (Nuri-Typha extract, water fraction, ethyl acetate fraction, butanol fraction and verbascoside).
