Description

Technical Field [0001] The present invention relates to a cosmetic composition for improving acne, which comprises a purified fraction of red ginseng ethanol extract,

The present invention relates to a cosmetic composition for improving acne comprising a purified fraction of red ginseng ethanol extract.

Acne is an inflammatory disease of the hair follicle that shows various skin lesions including facial, thoracic, back and arm open or closed skin, papules, cysts and nodules. Often acne causes a small size rash. These rashes are considered to be a characteristic symptom of acne.

The cause of acne has not yet been clarified, but it is presumed that various causes are involved. However, there are two factors that are caused by the hyperactivity of gonadal function, which is basically caused by hormones and genetic background, and by the action of acne bacteria that break down sebaceous glands and produce free fatty acids by breeding in fur branch lines. Particularly, sebaceous stasis in the hair follicles blocks the air circulation by blocking the hair follicles, and thus the environment in which the anaerobic bacteria residing in the hair follicles can grow well inside the hair follicle. The cotton fabric thus produced is used to create a living environment of microorganisms, such as P. acnes , Staphylococcus epidermidis , and Pityrosporum ovale . These are lipophilic microorganisms. Staphylococcus epidermidis is aerobic and grows in the middle of the outer capsule or hair follicle. Propionibacterium acnes is anaerobic and grows inside the hair follicle.

Until now, research on acne treatment has focused on the exfoliation of pores using salicylic acid and vitamin A derivatives, retinoic acid, and the enhancement of sterility against acne bacteria using antibiotics such as triclosan.

Triclosan has been reported to cause skin irritation when contacted with skin, and many recent studies are raising the interest that triclosan is likely to increase the resistance of bacteria to antibiotics. Since triclosan kills bacteria in a manner similar to antibiotics, there is a growing concern about the abuse of triclosan because bacteria already resistant to triclosan are more likely to be resistant to other antibiotics. In fact, a number of mutation strains resistant to triclosan and some antibiotics have been found in experimental studies.

On the other hand, the physiological activity of ginseng is due to saponin, polysaccharides and peptides, and many studies have been conducted centering on ginsenoside. Thus, the central nervous system control, memory, improvement of learning efficacy and antistress action, As the various pharmacological actions such as proliferation and transfer inhibition action, antioxidative action, antidiabetic action, liver function improvement, radiation protection action and inhibition of thrombogenesis are scientifically proved, the demand for medicine and functional supplements Is increasing. The efficacy associated with ginseng skin is to improve wrinkles of the skin or to protect against skin damage from exposure to ultraviolet light. Although it has been found that a mixture containing ginseng natural fermentation polysaccharide is effective for the treatment of acne (Patent Document 1), there has not been confirmed the effect of the extract of red ginseng, a ginseng processed product, on the killing action of acne.

The present inventors have studied a raw material which shows excellent antimicrobial effect to acne bacteria similar to triclosan but does not induce skin irritation and does not cause resistance to bacteria. The purified fraction of red ginseng extract has excellent antimicrobial activity against acne bacteria The present invention has been completed.

The term "acne " as used herein refers to any one selected from the group consisting of Propionibacterium acnes, Staphylococcus epidermidis and Pityrosporum ovalet; Or all other skin conditions such as purulent pimples, nodular acne and cystic acne caused by or partially induced by other microorganisms capable of producing acne, as well as closed and open skin.

The term " treatment "as used herein refers to the treatment or prevention of acne, which is a result of application of the pharmaceutical composition of the present invention to an acne site, as well as a cure for acne symptoms as well as a partial cure for acne symptoms, Prevention and " prevention "refers to the application of the pharmaceutical composition of the present invention to a body part to inhibit or prevent the infection or growth of acne bacteria such as Propionibacterium acnes and prevent acne symptoms from occurring in advance .

The term "cosmetic composition" referred to herein includes detergents and cosmetics as a composition used in the human body to cleanse and polish the body to brighten the appearance or to maintain or promote the health of skin and hair .

Hereinafter, the present invention will be described in detail.

One aspect of the present invention provides a cosmetic composition for improving acne comprising a column-purified fraction of an extract of red ginseng ethanol or aqueous ethanol solution.

The extract may be obtained by extracting from a raw material or a dried product thereof, and the raw material of the extract may be cultivated or commercially available.

When the extract is obtained by extracting from the raw material, any conventional extraction methods known in the art such as a solvent extraction method, an ultrasonic extraction method, a filtration method and a reflux extraction method may be used as the extraction method. Preferably, the extraction method or the reflux extraction method can do. The extraction process can be repeated several times, and then the step such as concentration or freeze-drying can be further roughened. Specifically, the obtained extract is concentrated under reduced pressure to obtain a concentrated liquid, the concentrated liquid is lyophilized, and a concentrated powder can be produced using a pulverizer.

The extract may be extracted using an aqueous solution containing ethanol or ethanol as an extraction solvent, and preferably extracted with an aqueous ethanol solution. The aqueous ethanol solution may be 50 to 95% (v / v), preferably 50 to 90% (v / v), more preferably 50 to 85% (v / v) But not limited thereto. When the concentration of the aqueous ethanol solution is less than 50% (v / v), the effect of the acne scavenging effect is relatively lower as compared with the case of extracting the ethanol with a proper amount.

A filler such as silica gel, activated alumina, synthetic polymer, magnesium silicate, activated carbon, cellulose, ion exchange resin and the like may be used as the column used in the production of the purified fraction, and it is preferable that aromatic synthetic resin is used as a filler, The column may be an aromatic type synthetic adsorbent column, and more preferably, but not exclusively, a Diaion HP-20 synthetic adsorbent is used as a filler. The separation using the column can be carried out once to several times until the fraction of the desired purity is purified, and concentration and recrystallization can be carried out if necessary.

The desorption solvent may be an ethanol or ethanol aqueous solution, and the desorption solvent may be an aqueous solution of 50 to 99% (v / v) ethanol, preferably 70 to 97% (v / v) , More preferably 85 to 95% (v / v) aqueous ethanol solution, but is not limited thereto. When the concentration of the aqueous ethanol solution used as the desorption solvent is less than the lower limit value, there is a problem that the obtained antimicrobial effect of the acne bacteria is lowered.

When adding the aqueous ethanol solution, which is a desorption solvent in the production of the purified fraction, an aqueous solution of ethanol is flowed into the column, for example, it is pressurized and flowed.

In one embodiment of the present invention, a red ginseng ethanol extract is prepared by adding 70% (v / v) ethanol aqueous solution to red ginseng, and the obtained extract is adsorbed in an HP-20 column to adsorb the components adsorbed to the column at various concentrations (V / v) aqueous ethanol solution as a desorbing solvent when the fraction was desorbed using an ethanol solvent (0 (purified water), 30, 50, 70, 85, 95% (Table 1). The extracts of the extracts showed higher antimicrobial activity against acne bacteria than the concentrate of red ginseng alcohol extract (see Table 1).

In one embodiment of the present invention, when the HP-20 ion resin column is repeatedly used five times or more in the preparation of the purified fraction of the red ginseng ethanol extract, when the fraction is recovered with a solvent having an ethanol concentration of 70% (v / v) When the fraction is recovered with a solvent having an ethanol concentration of 85% (v / v) or 95% (v / v) remarkably lowering the antibacterial effect, even if the HP-20 ion resin column is reused 5 times or more, The antimicrobial effect is maintained (see Table 1).

The purified fraction of the red ginseng ethanol extract may be contained in the cosmetic composition for improving acne in an amount of 0.01 to 30% by weight, preferably 0.01 to 20% by weight, more preferably 0.01 to 10% by weight , And even more preferably from 0.01 to 5 wt%. If the content is less than 0.01% by weight, the effect of killing an acne bacterium may be insignificant. If it is more than 30% by weight, the effect of killing an acne bacterium may be relatively low.

In the cosmetic composition of the present invention, the fraction may be prepared into a liquid or a solid form using bases, adjuvants and additives commonly used in the cosmetics field. Liquid or solid cosmetics may include, for example, but not limited to, lotions, creams, lotions, bath salts, and the like.

The base, adjuvant and additives commonly used in the cosmetics field are not particularly limited, and examples thereof include water, alcohol, propylene glycol, stearic acid, glycerol, cetyl alcohol, liquid paraffin and the like.

When the composition of the present invention is prepared with a cosmetic composition, the composition of the present invention may contain not only the purified fraction of the red ginseng ethanol extract, but also the components commonly used in cosmetic compositions such as antioxidants, stabilizers, Vitamins, pigments and flavoring agents, and carriers.

The cosmetic composition of the present invention may be prepared in any form conventionally produced in the art and may be in the form of a solution, suspension, emulsion, paste, gel, cream, lotion, powder, soap, surfactant- Oils, powder foundations, emulsion foundations, wax foundations and sprays, but are not limited thereto. More specifically, it can be manufactured in the form of a soft lotion, a nutritional lotion, a nutritional cream, a massage cream, an essence, an eye cream, a cleansing cream, a cleansing foam, a cleansing water, a pack, a spray or a powder.

Production of red ginseng extract

1-1. Manufacture of red ginseng water extract

1.5kg of purified water was added to 1kg of red ginseng, and the mixture was extracted three times at 70 ° C for 8 hours. The mixture was centrifuged at low temperature and concentrated under reduced pressure to obtain 50g or more of a red ginseng water extract concentrate having a moisture content of 40%. After the solvent was completely evaporated, purified water was added to the extract, frozen at -70 ° C, placed in a freeze dryer for 72 hours, and dried to obtain a solid.

1-2. Manufacture of red ginseng ethanol extract

70% (v / v) ethanol was added to 1 kg of red ginseng in 5.5 times and extracted three times at 70 ° C for 8 hours. The extract was extracted with 50 g or more of a red ginseng alcohol extract concentrate having a moisture content of 40% by using a reduced pressure concentrator. After the solvent was completely evaporated, purified water was added to the extract, frozen at -70 ° C, placed in a freeze dryer for 72 hours, and dried to obtain a solid.

1-3. Purification fractionation of red ginseng extract

The purified water was diluted with 3 times of water to the red ginseng water extract concentrate and the red ginseng alcohol extract concentrate prepared in Examples 1-1 and 1-2, and the diluted solution was packed with 100 g of HP-20 ion exchange resin Column (Mitsubishi Chemical Industry). The column was washed with purified water corresponding to 5 times of the diluted solution, and then the column was subjected to various concentrations of aqueous ethanol solution (30, 50, 70, 85, 95% (v / v) The efficacious component fractions were recovered. After the solvent of the obtained fractions was completely evaporated, purified water was added to the fractions, and the mixture was frozen at -70 DEG C, and the mixture was put into a freeze dryer and dried to obtain a solid fraction.

1-4. Purification fractionation of red ginseng ethanol extract for mass production

For the mass production, 5.5 kg of 70% (v / v) ethanol was added to 25 kg of red ginseng and extracted once at 70 ° C for 8 hours to obtain an extract of about 135 kg. Extracts were extracted with 6.75 kg of red ginseng alcohol extract, which was sprayed with ethanol solvent using a vacuum concentrator. The concentrate was diluted by adding 3 times of purified water to make a diluted solution of about 27 kg. The diluted solution was passed through a column packed with 500 g of HP-20 ion exchange resin at a rate of about 5 kg at a time, (V / v) ethanol and purified water were passed through each 5 kg of the column. After the column was washed with 5 kg of diluted solution, The washed HP-20 ion exchange resin was passed through a column packed with a desorption solvent, and this procedure was repeated 5 times to fractionate all diluted solutions. After repeating 5 times, the solvent of the obtained fractions was completely evaporated. Then, purified water was added to the fractions, and the mixture was frozen at -70 ° C, and the mixture was put into a freeze dryer and dried to obtain a solid fraction.

Comparison of antimicrobial effect against acne bacterium according to alcohol concentration of desorbing solvent used in the preparation of purified fraction

The acne fungicidal effect of the red ginseng water extract, red ginseng ethanol extract, red ginseng ethanol extract, HP-20 column fraction obtained in Example 1, and the fraction subjected to the mass-production (hereinafter referred to as " . The solid content of each extract obtained in Example 1 was dissolved in purified water at 100, 50, 25, and 12.5 mg / ml, and the antimicrobial activity against acne bacteria was confirmed by the paper disk method. Propionibacterium acnes (ATCC 6919), a skin-borne bacterium that causes acne, was cultured in a Reinforced clostridial medium (difco, USA). Reinforced clostridial medium P. acne prepared on the plate medium was inoculated evenly with a sterile cotton swab. Clear zone, a range in which acne bacteria do not grow, was measured and the effect of each sample was compared. As a positive control, tea tree oil (100 mg / ml) and triclosan (3 mg / ml) used as acne antimicrobial agents were used.

Experimental results showed that when extracting red ginseng with water, there was no acne killing effect, but excellent acne antibacterial effect was obtained when extracting with high alcohol concentration. Particularly, when the alcohol extract was passed through HP-20 ion resin column and the fraction was recovered with ethanol at a high concentration of 85% (v / v) or more, it was 13 times higher than the concentration of the existing red ginseng alcohol concentrate / Ml), it showed a higher antimicrobial effect.

In addition, when the alcohol extract is passed through HP-20 ion resin column and the fraction is recovered with ethanol at a high concentration of 85% (v / v) or more, the amount of ethanol is lower than that of tea tree oil And showed a clear zone about 5 times larger. In addition, the purified fraction of the red ginseng alcohol extract of the present invention (using an aqueous solution of ethanol of 85% (v / v) or more as a desorption solvent) is now used as a possibility of disturbance of the endocrine system, The purified fraction of the present invention is expected to effectively replace triclosan because it exhibits an antibacterial effect against acne bacteria at a level of 95.5 to 46.4% as compared with triclosan, which is a component of triclosan.

On the other hand, when the HP-20 ion resin column is repeatedly used more than 5 times for mass production, the antibacterial effect is remarkably decreased when the fraction is recovered with a solvent having an ethanol concentration of 70% (v / v) , And when the fraction was recovered with a solvent having an ethanol concentration of 85% or 95% (v / v), it was found that the antibacterial effect against the acne bacterium was maintained even when the HP-20 ion resin column was reused five times or more .

Therefore, many of the components of the red ginseng alcohol extract that are highly effective against acne bacteria are adsorbed on the HP-20 column. When the adsorbed components are recovered using 85 to 95% (v / v) ethanol solvent, . Therefore, it was found that fractions having higher antimicrobial activity against acne were obtained. In addition, when the scale is up, that is, when HP-20 is repeatedly used to obtain purified fractions and the ethanol concentration is 85% or 95% (v / v), the column is reused, However, when the fraction was recovered with a solvent having an ethanol concentration of 70% (v / v) or less, it was found that when the column was reused, the antimicrobial effect of acne decreased and scale-up was difficult.
