Abstract
본 발명은 나노입자-유리체 기반 단백질 복합체에 관한 것으로서, 보다 구체적으로는 상기 복합체를 유효성분으로 포함하는 혈관신생억제용 조성물, 혈관신생 관련 질환 또는 망막질환 예방 또는 치료용 조성물에 관한 것이다.
본 발명에 따른 나노입자-유리체 기반 단백질 복합체는 유리체강 내 국소 주사 시, 혈관내피세포성장인자와 현저히 우수한 결합력을 나타내어 혈관신생을 억제할 수 있는바, 망막 및 맥락막 혈관신생 관련 질환의 예방, 개선 또는 치료를 위한 치료제 등의 제조에 용이하게 사용될 수 있다.
본 발명에 따른 나노입자-유리체 기반 단백질 복합체는 유리체강 내 국소 주사 시, 혈관내피세포성장인자와 현저히 우수한 결합력을 나타내어 혈관신생을 억제할 수 있는바, 망막 및 맥락막 혈관신생 관련 질환의 예방, 개선 또는 치료를 위한 치료제 등의 제조에 용이하게 사용될 수 있다.
Description
본 발명은 나노입자-유리체 기반 단백질 복합체에 관한 것으로서, 보다 구체적으로는 상기 복합체를 유효성분으로 포함하는 혈관신생억제용 조성물, 혈관신생 관련 질환 또는 망막질환 예방 또는 치료용 조성물에 관한 것이다.
혈관신생(angiogenesis)은 조직이나 장기에 신규의 혈관을 제공하는 생물학적 과정으로, 구체적으로는 기존의 미세혈관으로부터 새로운 모세혈관이 생성되는것을 의미하여 성장 후 체내에서 혈관이 생성되는 근본적인 과정이다. 신생혈관이 생성되는 과정은 매우 복잡하고 정교하나, 요약하면 다음과 같다. 첫째, 혈관 신생을 위한 자극이 기존의 혈관에 전달되면 혈관이 팽대하고 막투과도가 증가한다. 둘째, 팽대된 혈관을 통하여 피브린이 혈관 밖으로 빠져나와 혈관 주위의 세포질 기질에 침적된다. 셋째, 기존 혈관의 기저막을 분해하기 위한 효소가 활성화되며, 기저막이 파괴되어 그 사이로 내피세포가 혈관을 빠져나와 주위 세포의 기질에서 증식하고 이동한다. 마지막으로, 일렬로 배열한 내피세포들이 맥관을 이룸으로써 새로운 혈관을 생성하게 된다.
혈관이 신생되는 과정은 다양한 음성 및 양성 조절인자들에 의해 엄격히 조절되고 있는데 이러한 혈관 신생이 정상적으로 조절되지 못하면 암, 류마티스성 관절염, 당뇨병성 망막병증 등 여러 가지 질환들이 야기된다. 특히 이러한 병적 혈관신생(pathological angiogenesis)이 망막에서 발생하는 경우 이는 망막부종, 망막 또는 유리체 출혈, 그리고 망막박리를 일으키게 된다. 또한 망막에서의 혈관 신생은 미숙아 망막병증, 당뇨망막병증 및 노인성 황반변성의 주요 원인이 된다.
한편, 나노입자들(NPs)은 산업적으로 그리고 생의학적 목적으로 광범위하게 사용되어오고 있다. 특히, 약물 전달, 유전자 전달, 세포 내 영상, 광선치료와 같은 생체의학 분야에서 촉망되는 도구로서 사용되어 왔으며, 특히 금이나 실리카 나노소재는 합성과 작용의 용이성, 화학적 안정성, 생체 적합성, 및 조정 가능한 광학적 및 전기적 특성으로 인해 많은 관심을 받고 있다.
최근에는 금 또는 은 나노입자가 혈관내피세포성장인자(VEGF, vascular endothelial growth factor)에 의해 유도되는 혈관신생을 저해하는 것이 알려졌으며, 이를 이용한 혈관신생 억제제를 개발하려는 연구들이 시도되고 있다. 그러나 나노입자를 치료에 이용하기 위해서는 나노입자의 독성에 대한 세심한 평가와 독성을 최소로 하는 노력이 요청된다.
본 발명은 상기와 같은 종래 기술상의 문제점을 해결하기 위해 안출된 것으로서, 본 발명자들은 기관 특이적으로 망막 및 맥락막 혈관신생 억제에 좋은 효과를 가지면서, 낮은 독성을 나타내는 나노입자 치료제를 개발하기 위해 연구 노력한 결과, 나노입자-유리체 기반 단백질 복합체가 혈관내피세포성장인자와 현저히 우수한 결합력을 나타냄을 확인하고, 이에 기초하여 본 발명을 완성하게 되었다.
이에, 본 발명의 목적은 나노입자와 상기 나노입자의 표면을 둘러싼 유리체 기반 단백질로 이루어진 복합체를 유효성분으로 포함하는 혈관신생 억제용 약학적 조성물을 제공하는 것이다.
또한, 본 발명의 또 다른 목적은 나노입자와 상기 나노입자의 표면을 둘러싼 유리체 기반 단백질로 이루어진 복합체를 유효성분으로 포함하는 망막 질환 예방 또는 치료용 조성물을 제공하는 것이다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
상기 목적을 달성하기 위하여, 본 발명은 나노입자와 상기 나노입자의 표면을 둘러싼 유리체 기반 단백질로 이루어진 복합체를 유효성분으로 포함하는 혈관신생 억제용 약학적 조성물을 제공한다.
본 발명의 일 구현예로, 상기 나노입자는 금 또는 실리카일 수 있다.
본 발명의 다른 구현예로, 상기 나노입자는 20-100 nm의 직경을 가질 수 있다.
본 발명의 또 다른 구현예로, 상기 혈관신생은 미숙아 망막병증, 증식성 망막병증, 연령관련 황반변성(age-related macular degeneration), 당뇨병성 황반부종(DME, Diabetic Macular Edema), 당뇨병성 망막병증(diabetic retinopathy) 또는 중심성 장액 맥락망막병증 (Central serous (chorio)retinopathy)에 수반할 수 있다.
본 발명의 또 다른 구현예로, 상기 유리체 기반 단백질은 비트린(vitrin), secreted frizzled-related protein 2, 혈청 알부민(serum albumin), 레티놀결합단백질 3(retinol-binding protein 3), 및 alpha-crystallin A chain로 이루어진 군으로부터 하나 이상 선택될 수 있다.
본 발명은 상기 조성물을 유효성분으로 포함하는 망막 질환 예방 또는 치료용 약학적 조성물을 제공한다.
본 발명의 일 구현예로, 상기 망막질환은 미숙아 망막병증, 당뇨병성 황반부종(DME, Diabetic Macular Edema), 당뇨병성 망막병증(diabetic retinopathy), 중심성 장액 맥락망막병증 (Central serous (chorio)retinopathy), 연령관련 황반변성(Age-related macular degeneration), 및 증식성 망막병증으로 이루어진 군으로부터 선택될 수 있다.
본 발명은 하기 단계를 포함하는 망막 질환 치료에 적합한 단백질 스크리닝 방법을 제공한다:
(1) in vitro에서 유리체에 나노입자를 주입하는 단계;
(2) 상기 나노입자와 상기 유리체 내 단백질과의 복합체를 분리하는 단계;
(3) 상기 복합체와 혈관내피세포성장인자(VEGF)를 결합시키는 단계; 및
(4) 상기 혈관내피세포성장인자와 결합하는 복합체를 선별하는 단계.
본 발명은 상기 약학적 조성물의 약제학적 유효량을 개체에 투여하는 단계를 포함하는 혈관신생 억제방법을 제공한다.

본 발명에 따른 나노입자-유리체 기반 단백질 복합체는 유리체강 내 국소 주사 시, 혈관내피세포성장인자와 현저히 우수한 결합력을 나타내어 혈관신생을 억제할 수 있는바, 망막 및 맥락막 혈관신생 관련 질환의 예방, 개선 또는 치료를 위한 치료제 등의 제조에 용이하게 사용될 수 있다.

본 발명자들은 망막 및 맥락막 혈관신생을 조절할 수 있는 방법에 대하여 연구 노력한 결과, 유리체강 내 주사를 통한 나노입자의 국소 주사 시, 유리체 기반 단백질과 복합체(corona)를 형성하고, 이러한 나노입자-유리체 기반 단백질 복합체가 혈관내피세포성장인자와 현저히 우수한 결합력을 나타냄을 확인하고, 이에 기초하여 본 발명을 완성하게 되었다.
이하, 본 발명을 상세히 설명한다.
본 발명은 나노입자와 상기 나노입자의 표면을 둘러싼 유리체 기반 단백질로 이루어진 복합체를 유효성분으로 포함하는 혈관신생 억제용 약학적 조성물을 제공한다.
본 발명에서 "혈관신생"은 혈관이 새로 형성되는 과정, 즉, 새로운 혈관이 세포, 조직 또는 기관 내로 발생되는 것을 지칭하는 것이며, "신생혈관"은 혈관신생 과정을 통해 새로 생성된 혈관을 의미한다. 본 발명에서 "혈관신생"과 "신생혈관"은 상호 호환적으로 기재할 수 있다.
또한, 본 발명에서, 상기 혈관신생은, 미숙아 망막병증, 증식성 망막병증, 연령관련 황반변성(age-related macular degeneration), 당뇨병성 황반부종(DME, Diabetic Macular Edema), 당뇨병성 망막병증(diabetic retinopathy), 중심성 장액 맥락망막병증 (Central serous (chorio)retinopathy) 또는 만성염증(chronic inflammation)에 수반하는 것일 수 있으나, 이에 한정되는 것은 아니며, 혈관신생이 질병을 유발하거나 진행시키는 임의의 질환에 수반되는 것일 수 있다.
본 발명에서 "나노입자(nanoparticle)"란, 나노 단위의 직경을 가지는 다양한 물질의 입자를 의미하며, 상기 나노입자는 나노크기를 갖는 입자라면 특별히 제한되지는 않으나, 직경이 100 nm 이상으로 커질 경우 나노 입자로서의 특성이 소멸될 수 있는바, 20-100nm의 직경을 갖는 것이 바람직하다. 또한, 나노입자의 종류로는 금 나노입자 또는 실리카 나노입자를 사용할 수 있지만, 이것으로 제한되는 것은 아니다.
본 발명에서 "유리체 기반 단백질"이란, 나노입자와 결합하여 복합체를 형성할 수 있는 유리체 내 단백질을 의미하며, 상기 유리체 기반 단백질의 종류에는 이에 제한되는 것은 아니지만, Vitrin, Secreted frizzled-related protein 2, Serum albumin, Retinol-binding protein 3, Alpha-crystallinA chain, Beta-crystallin S, Beta-crystallin B2, Latent-transforming growth factor beta-binding protein 2, Complement C4-A, Alpha-enolase, Beta-crystallin B1, Spondin-1, Calsyntenin-1, Gelsolin, Retinal dehydrogenase 1, Beta-crystallinA2, Collagen alpha-1(II) chain, Actin, cytoplasmic 1, EGF-containing fibulin-like extracellular matrix protein 1, Opticin 등이 있으며, 바람직하게는 비트린(vitrin), secreted frizzled-related protein 2, 혈청 알부민(serum albumin), 레티놀결합단백질 3(retinol-binding protein 3), 및 alpha-crystallin A chain로 이루어진 군으로부터 선택되는 하나 이상의 단백질을 포함할 수 있다. 한편, 상기 비트린(vitrin), secreted frizzled-related protein 2, 혈청 알부민(serum albumin), 레티놀결합단백질 3(retinol-binding protein 3), 및 alpha-crystallin A chain은 각각 서열번호 1 내지 서열번호 5의 아미노산 서열로 이루어질 수 있으며, 이에 한정되지 않고 상기 아미노산 서열과 70% 이상, 바람직하게는 80% 이상, 보다 바람직하게는 90% 이상, 가장 바람직하게는 95% 이상의 상동성을 갖는 아미노산 서열을 포함할 수 있다.
본 발명에 따른 조성물에 유효성분으로 포함되는 나노입자-유리체 기반 단백질 복합체는 혈관내피세포성장인자와의 결합력이 우수하여 혈관신생을 효과적으로 억제할 수 있다.
본 발명의 일 실시예에 따르면, 나노입자와 결합하는 상위 5개의 유리체 기반 단백질을 확인하고(실시예 1 참조), 상기 5개의 단백질과 나노입자를 배양하여 나노입자-유리체 기반 단백질 복합체를 형성한 후(실시예 2 참조), 이의 혈관신생억제 효과를 확인한 결과, 나노입자와 비교할 때, 물에서는 혈관내피세포성장인자와의 결합력이 큰 차이가 없는 반면, 유리체 내에서는 나노입자 단독 처리군에 비해 현저히 우수한 혈관내피세포성장인자와의 결합력을 나타냄을 확인하였다(실시예 3 참조).
본 발명의 다른 실시예에서는, in vitro 또는 in vivo에서 나노입자-유리체 기반 단백질 복합체의 혈관신생 억제 효과 확인한 결과, 나노입자-유리체 기반 단백질 복합체가 in vitro 또는 in vivo 모두에서 우수한 혈관신생 억제효과를 나타냄을 확인하였다(실시예 4 참조).
이러한 실험결과로부터, 본 발명에 따른 나노입자-유리체 기반 단백질 복합체는 혈관신생을 억제함으로써 혈관신생 관련 질환, 특히 망막 및 맥락막 혈관신생 관련 질환의 예방, 개선 또는 치료에 유용하게 이용할 수 있음을 알 수 있다.
이에, 본 발명의 다른 양태로서, 본 발명은 나노입자와 상기 나노입자의 표면을 둘러싼 유리체 기반 단백질로 이루어진 복합체를 유효성분으로 포함하는 혈관신생 관련 질환 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명의 또 다른 양태로서, 본 발명은 나노입자와 상기 나노입자의 표면을 둘러싼 유리체 기반 단백질로 이루어진 복합체를 유효성분으로 포함하는 망막 질환 예방 또는 치료용 약학적 조성물을 제공한다.
본 발명에서 사용되는 용어, "예방"이란 본 발명에 따른 약학적 조성물의 투여에 의해 혈관신생 관련 질환 또는 망막 질환을 억제시키거나 발병을 지연시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어, "치료"란 본 발명에 따른 약학적 조성물의 투여에 의해 혈관신생 관련 질환 또는 망막 질환에 의한 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명에서 "혈관신생 관련 질환"은 상기한 바와 같은 신생혈관 형성이 비정상적으로 진행되어 야기되는 질환을 의미하며, 미숙아 망막병증, 증식성 망막병증, 연령관련 황반변성(age-related macular degeneration), 당뇨병성 망막병증(diabetic retinopathy), 중심성 장액 맥락망막병증 (Central serous (chorio)retinopathy) 등일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에서 "망막 질환"은 망막에 병변이 발생하는 질환으로서, 미숙아 망막병증, 당뇨병성 황반부종(DME, Diabetic Macular Edema), 당뇨병성 망막병증(diabetic retinopathy), 중심성 장액 맥락망막병증 (Central serous (chorio)retinopathy), 연령관련 황반변성(Age-related macular degeneration), 및 증식성 망막병증 등일 수 있으나, 이에 한정되는 것은 아니다.
본 발명에 따른 약학적 조성물은 약제학적으로 유효한 양으로 투여한다. 본 발명에 있어서, "약제학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명에 따른 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명에 따른 약학적 조성물은 임상투여 시에 다양한 경구 또는 비경구 투여 형태로 제제화되어 투여될 수 있으나, 바람직하게는 유리체강 내 주사 방식으로 적용될 수 있고, 유리체강 내 주사에 적합한 약제학적 제형으로 제조될 수 있다.
본 발명의 또 다른 양태로서, 본 발명은 나노입자와 상기 나노입자의 표면을 둘러싼 유리체 기반 단백질로 이루어진 복합체를 개체에 투여하는 단계를 포함하는 혈관신생 관련 질환 또는 망막 질환 치료방법을 제공한다.
본 발명에서 "개체"란 질병의 치료를 필요로 하는 대상을 의미하고, 보다 구체적으로는 인간 또는 비-인간인 영장류, 생쥐 (mouse), 쥐 (rat), 개, 고양이, 말 및 소 등의 포유류를 의미한다.
본 발명의 또 다른 양태로서, 본 발명은 하기 단계를 포함하는 망막 질환 치료에 적합한 단백질 스크리닝 방법을 제공한다:
(1) in vitro에서 유리체에 나노입자를 주입하는 단계;
(2) 상기 나노입자와 상기 유리체 내 단백질과의 복합체를 분리하는 단계;
(3) 상기 복합체와 혈관내피세포성장인자(VEGF)를 결합시키는 단계; 및
(4) 상기 혈관내피세포성장인자와 결합하는 복합체를 선별하는 단계.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실시예]
실시예 1. 나노입자에 결합하는 유리체 기반 단백질 확인
1-1. 나노입자 준비
유리체 단백질과의 결합을 위한 나노입자는 각각 20 및 100nm의 직경을 갖는 금 및 실리카 나노입자를 준비하였다. 상기 나노입자들의 구체적인 정보 및 TEM 이미지를 각각 표 1 및 도 1에 나타내었다.

1-2. 나노입자와 유리체 단백질 결합
상기 실시예 1-1을 통해 준비된 직경 20 nm의 금(Au20)과 실리카(Si20) 나노입자 및 직경 100 nm의 금(Au100)과 실리카(Si100) 나노입자(1 x 1011개) 각각을 170 μg의 단백질을 보유한 유리체(vitreous)와 microcentrifuge tube 안에서 4℃ 조건으로 20 rpm의 회전을 가하여 6시간 동안 배양하였다. 이후 15,000 rpm 조건에서 20분 간 원심분리하여 침전물을 획득하였고, 이후 증류수로 2회 세척하여 비특이적으로 결합한 단백질을 제거하였다. 이때 침전물은 유리 나노입자 및 단백질이 결합된 나노입자를 포함한다. 이를 30 μL의 Laemmli buffer에 부유시키고, 100℃에서 3분 간 가열하여 나노입자와 단백질의 분리를 유도하였다. 이후 15,000 rpm 조건에서 1분 간 원심분리하여 상층액을 사용하여 단백질체 분석하여, 각 나노입자와 코로나를 형성한 상위 20개의 유리체 기반 단백질을 확인하였고, 그 결과를 도 2 내지 도 5에 나타내었다. 더욱이, 상기 결과를 모두 종합하여 상위 20개의 유리체 기반 단백질을 확인하였고, 그 결과를 도 6에 나타내었다.
도 6에 나타낸 바와 같이, 나노입자와 코로나를 형성하는 상위 5개의 유리체 기반 단백질은 순서대로 vitrin, secreted frizzled-related protein 2, serum albumin, retinol-binding protein 3, alpha crystallin A chain임을 확인할 수 있었고, 각 상대적 양은 11.25, 6.80, 5.44, 4.85, 4.23%였다.
실시예
2. 나노입자-유리체 기반 단백질 복합체 형성
상기 실시예 1-2에 의해 확인된 상위 5개의 유리체 기반 단백질이 각각 50 ng, 25 ng, 25 ng, 25 ng 및 25 ng으로 이루어진 총 150 ng의 단백질 (SALVAR complex)을 준비하여 1 x 109개의 나노입자와 4℃에서 20 rpm의 회전을 가하여 1시간 동안 배양하여 나노입자-유리에 기반 단백질 복합체를 형성하였다.
실시예
3. 나노입자-유리체 기반 단백질 복합체의 혈관 신생 억제 효과 확인
금 및 실리카 나노입자의 경우, 물 또는 세포배양액에서 혈관내피세포성장인자와 결합하는 현상이 알려져 있는바, 이러한 나노입자와 비교할 때, 본 발명에 따른 나노입자-유리체 기반 단백질 복합체가 물 또는 세포배양액에서 혈관내피세포성장인자와 효과적으로 결합하는지를 확인하기 위해 하기와 같이 실험을 실시하였다.
3-1. 물에서 혈관내피성장인자와의 결합력 비교
물에서 나노입자에 혈관내피세포 성장인자를 결합시키는 경우와 나노입자-유리체 기반 단백질 복합체에 혈관내피세포성장인자를 결합시키는 경우를 비교하였고, 해당 실험에 대한 개략적인 과정을 도 7에 나타내었다.
즉, 나노입자 또는 나노입자-유리체 기반 단백질 복합체를 혈관내피세포성장인자와 4℃에서 20 rpm의 회전을 가하여 6시간 동안 배양하였다. 배양 후 15,000 rpm 조건에서 20분 간 원심분리하여 나노입자 또는 나노입자-유리체 기반 단백질 복합체와 결합한 혈관내피세포성장인자를 침전시켜, 유리된 혈관내피세포성장인자를 상층액에서 효소결합면역흡착측정법(enzyme-linked immunosorbent assay)을 통해 측정하여 혈관내피세포성장인자의 결합력을 확인하였다.
그 결과, 도 9에 나타낸 바와 같이, 물에서 나노입자에 혈관내피세포성장인자를 결합시키는 경우("Bare" 그룹)와 나노입자-유리체 기반 단백질 복합체에 혈관내피세포성장인자를 결합시키는 경우("SALVAR" 그룹) 간의 혈관내피 세포성장인자와의 결합력은 큰 차이가 없음을 확인할 수 있었다.
3-2. 유리체 내에서 혈관내피성장인자와의 결합력 비교
유리체 내에서 나노입자에 혈관내피세포성장인자를 결합시키는 경우와 나노입자-유리체 기반 단백질 복합체에 혈관내피세포성장인자를 결합시키는 경우를 비교하였고, 해당 실험에 대한 개략적인 과정을 도 8에 나타내었다.
물 대신에 유리체를 사용하였다는 점을 제외하곤, 상기 실시예 3-1과 동일한 방법을 실시하였다.
그 결과, 도 9에 나타낸 바와 같이, 유리체 내에서 나노입자-유리체 기반 단백질 복합체에 혈관내피세포성장인자를 결합시키는 경우("SALVAR+Vitreous" 그룹)가 나노입자에 혈관내피세포성장인자를 결합시키는 경우("Vitreous" 그룹)에 비하여 현저히 우수한 혈관내피세포성장인자와의 결합력을 나타냄을 확인할 수 있었다.
상기 결과로부터, 본 발명에 따른 나노입자-유리체 기반 단백질 복합체가 생체 내에서 우수한 치료효과를 보일 수 있다는 것을 알 수 있었다.
실시예 4. In vitro에서 나노입자-유리체 기반 단백질 복합체의 생체 내 혈관신생 억제 효과 확인
20 ng/mL의 혈관내피세포성장인자를 혈관내피세포에 처리할 시 혈관신생 과정을 대표하는 혈관내피세포의 증식이나 관 형성을 촉진함이 알려져 있다. 나노입자-유리체 기반 단백질 복합체 투여가 혈관내피세포성장인자에 의한 in vitro 혈관신생 과정을 억제함을 확인하기 위하여 혈관내피세포 증식(proliferation) 및 관 형성(tube formation) 시험을 시행하였다. 혈관내피세포 증식 시험은 0.3% 젤라틴(gelatin)이 코팅된 plate의 각 well에 2,000개의 혈관내피세포를 1일 간 배양하고, 이후 혈관내피세포성장인자, 나노입자, 나노입자-유리체 기반 단백질 복합체, 및 bevacizumab을 조건에 따라 처리한 후 48시간 동안 혈관내피세포의 증식 정도를 비교하는 방식으로 진행하였다. 혈관내피세포의 증식 정도는 trypan blue 염색 후 직접 세포의 수를 측정하는 방식과, water soluble tetrazolium salt-1 처리 후 450 nm의 흡광도를 측정하는 방식으로 추정하였다. 관 형성 시험은 Matrigel이 코팅된 plate의 각 well에 100,000개의 혈관내피세포와 혈관내피세포성장인자, 나노입자, 나노입자-유리체 기반 단백질 복합체, 및 bevacizumab을 조건에 따라 처리한 후 12시간 후 혈관내피세포의 관 형성 정도를 비교하는 방식으로 진행하였다. 50배 확대된 화면에서 형성된 관(tube)의 숫자를 확인하여 정량적인 비교를 시행하였다.
그 결과, 도 10에 나타낸 바와 같이, 나노입자-유리체 기반 단백질 복합체("SALVAR mixture")를 투여한 경우에, in vitro 혈관내피세포 증식 및 관 형성 시험에서 bevacizumab에 준하는 우수한 혈관신생 억제효과를 나타냄을 확인할 수 있었다.
실시예 5. In vivo에서 나노입자-유리체 기반 단백질 복합체의 혈관신생 억제 효과 확인
생쥐에서 레이저 유도 맥락막 신생혈관 모델을 만들고, 나노입자 또는 나노입자-유리체 기반 단백질 복합체를 유리체강 내 주사하여 효과를 확인하였다. 생쥐의 망막에 400 mW의 세기로 50 ms 지속시간의 diode 레이저를 조사하면 망막층과 맥락막층 사이의 Bruch 막의 파괴가 일어나는데, 레이저 조사 후 나노입자(109개/mL, 1 μL), 나노입자-유리체 기반 단백질 복합체(109개/mL, 1 μL), 항 혈관내피세포성장인자 항체(1 μg)를 유리체강 내로 주사하고, 레이저 조사 후 7일째에 맥락막 신생혈관의 형성 정도를 면역형광 염색을 통해 확인하였다.
그 결과, 도 11에 나타낸 바와 같이, 나노입자-유리체 기반 단백질 복합체("Au20+SALVAR mixture")를 투여한 경우에, 생체 내에서도 항 혈관내피세포성장인자 항체에 준하는 우수한 혈관신생 억제효과를 나타냄을 확인할 수 있었다.

Claims (8)
- 나노입자와 상기 나노입자의 표면을 둘러싼 유리체 기반 단백질로 이루어진 복합체를 유효성분으로 포함하는 혈관신생 억제용 약학적 조성물.
- 제1항에 있어서,
상기 나노입자는 금 또는 실리카인 것을 특징으로 하는, 조성물.
- 제1항에 있어서,
상기 나노입자는 20-100 nm의 직경을 가지는 것을 특징으로 하는, 조성물.
- 제1항에 있어서,
상기 혈관신생은 미숙아 망막병증, 당뇨병성 황반부종(DME, Diabetic Macular Edema), 당뇨병성 망막병증(diabetic retinopathy), 중심성 장액 맥락망막병증 (Central serous (chorio)retinopathy), 연령관련 황반변성(Age-related macular degeneration) 또는 증식성 망막병증에 수반하는 것임을 특징으로 하는 약학적 조성물.
- 제1항에 있어서,
상기 유리체 기반 단백질은 비트린(vitrin), secreted frizzled-related protein 2, 혈청 알부민(serum albumin), 레티놀결합단백질 3(retinol-binding protein 3), 및 alpha-crystallin A chain로 이루어진 군으로부터 하나 이상 선택되는 것을 특징으로 하는, 조성물.
- 제1항 내지 제5항 중 어느 항의 조성물을 유효성분으로 포함하는 망막 질환 예방 또는 치료용 약학적 조성물.
- 제6항에 있어서,
상기 망막질환은 미숙아 망막병증, 당뇨병성 황반부종(DME, Diabetic Macular Edema), 당뇨병성 망막병증(diabetic retinopathy), 중심성 장액 맥락망막병증 (Central serous (chorio)retinopathy), 연령관련 황반변성(Age-related macular degeneration), 및 증식성 망막병증으로 이루어진 군으로부터 선택되는 것을 특징으로 하는, 조성물.
- 하기 단계를 포함하는 망막 질환 치료에 적합한 단백질 스크리닝 방법:
(1) in vitro에서 유리체에 나노입자를 주입하는 단계;
(2) 상기 나노입자와 상기 유리체 내 단백질과의 복합체를 분리하는 단계;
(3) 상기 복합체와 혈관내피세포성장인자(VEGF)를 결합시키는 단계; 및
(4) 상기 혈관내피세포성장인자와 결합하는 복합체를 선별하는 단계.
