Abstract
Description
The present invention relates to an external preparation composition for skin containing hackberry extract, and more particularly, to an external preparation composition for skin having excellent anti-aging, whitening and moisturizing effects by containing hackberry extract.
Human skin is changed by a number of internal and external factors as it ages. That is, internally, the secretion of various hormones that regulate metabolism decreases, and the function of immune cells and the activity of cells decreases, thereby reducing the biosynthesis of immune proteins and constituent proteins necessary for living organisms. Due to the increase in the amount of ultraviolet rays that reach the surface of the sun's rays and intensifying environmental pollution, free radicals and free radicals increase, thereby reducing the thickness of the skin, increasing wrinkles, reducing elasticity, The color of the skin becomes dull, skin problems frequently occur, and there are various changes such as blemishes, freckles, and black mushrooms.
As aging progresses, symptoms of alteration or decrease in the content and arrangement of collagen, elastin, hyaluronic acid and glycoproteins, which are constituents of the skin, appear and are subject to oxidative stress caused by free radicals and active harmful oxygen. In addition, cyclooxygenase-2 (Cox-2, cyclooxygenase), an enzyme that produces proinflammatory cytokine, which is known to cause inflammation in most cells constituting the skin due to aging or ultraviolet rays. Biosynthesis of the enzyme increases the biosynthesis of matrix metalloproteinase (MMP), an enzyme that breaks down skin tissues by these inflammatory factors, and the production of nitric oxide (NO) by iNOS (inducible nitric oxide synthase). It is known to increase. In other words, the biosynthesis of the substrate material is reduced by the reduction of cellular activity and micro-inflammation due to naturally occurring endogenous aging, and by external factors such as the increase of stress caused by various harmful environments and the increase of active coral species caused by sunlight. As a result, decomposition and degeneration are accelerated, and the skin matrix is destroyed and thinned, and various symptoms of skin aging appear. Therefore, many studies have been conducted on active ingredients that can prevent and improve these aging phenomena.
Many factors are involved in determining the skin color of humans, including the activity of melanocytes that make melanin, the distribution of blood vessels, the thickness of the skin and the presence or absence of pigments in the body, such as carotenoids and bilirubin. It is important.
Especially, the most important factor is the black pigment called melanin which is produced by the action of various enzymes such as tyrosinase in melanocytes in the human body. The formation of melanin pigment affects genetic factors, hormonal secretion, physiological factors such as stress, and environmental factors such as ultraviolet irradiation.
The melanin pigment produced in melanocytes of body skin is a phenolic polymer substance having a complex form of black pigment and protein. It protects the subcutaneous skin organs by blocking ultraviolet rays irradiated from the sun, and at the same time, free radicals And protects proteins and genes in the skin.
As such, melanin produced by stress stimulation inside and outside the skin is a stable substance that does not disappear until it is released to the outside through skin keratinization even if the stress disappears. However, if the melanin is produced more than necessary to cause hyperpigmentation, such as blemishes, freckles, and spots, cosmetically bad results.
Today, Asian women prefer white and clean skin like white jade, and this is an important standard of beauty, increasing the demand for treatment and cosmetic problems for hyperpigmentation.
In response to this demand, ascorbic acid, kojic acid, arbutin, hydroquinone, glutathione or derivatives thereof, or substances having tyrosinase inhibitory activity have been used in combination with cosmetics or pharmaceuticals, but they are insufficient. Its use has been limited due to its whitening effect, safety issues on the skin, formulation and stability problems when formulated in cosmetics, and the like.
The most important function of the outermost epidermis of the skin is the defense against various external stimuli (physical and chemical stimulating factors such as chemicals, air pollutants, dry environment, ultraviolet rays) and moisture in the body through the skin. It is a protective function to prevent excessive divergence of, and this protective function can maintain its function only when the stratum corneum composed of keratinocytes is normally formed. The outermost keratin layer (stratim corneum, horney layer) is formed from keratinocytes and consists of the differentiated keratinocytes and the surrounding lipid layer (J. Invest. Dermatol. 1983; 80: 44 ~ 49). Keratinocytes are the characteristic cells that the morphology and function of the basal cells that proliferate continuously in the lower epidermis gradually go up to the surface of the skin and rise up to the surface of the skin. Exfoliated and new keratinocytes take over the function. This repetitive process of change is called 'epidermal differentiation' or 'keratinization'. Keratinocytes during keratinization form the stratum corneum by generating natural moisturizing factor (NMF) and intercellular fat (ceramide, cholesterol, fatty acids), making the stratum corneum firm and flexible, making it a skin barrier. It has a function as).
However, the stratum corneum can easily lose its function due to excessive washing, lifestyle factors such as bathing, environmental factors such as dry air and pollutants, and endogenous diseases such as atopic or aged skin. In recent years, more and more people are complaining of dry skin symptoms and all kinds of obstacles due to various factors. Therefore, many studies have been conducted to supply moisture from the outside or to prevent water loss from the body in order to maintain proper skin moisture, and various types of moisturizers having moisture retention capabilities have been developed and mainly cosmetics. It is widely used in the field.
However, as the risk factors for humans increase and the aged population rapidly increases in the living environment, the turnover rate of the stratum corneum is slowed, the lipid synthesis ability of the keratinocytes is degraded, or the epidermis is normal in the epidermis. Insufficient cell division, maturation, and differentiation, which leads to a decrease in the amount of moisturizing factors and lipids in the stratum corneum, leading to a condition in which the skin is unable to maintain normal stratum corneum function, ie, skin barrier function is degraded. Is in the trend.
Due to the abnormal division and differentiation of epidermal cells, the skin causes various skin diseases such as dry skin, atopy and psoriasis, and these diseases can alleviate the symptoms slightly with a conventional moisturizer having only water retention function. However, it is difficult to expect radical healing.
Recently, in order to solve the above problems in the anti-aging, whitening or moisturizing effect on the skin while reducing side effects of various chemicals and the like, a lot of skin external preparation compositions such as cosmetics using natural products have been developed. In this regard, the present inventors studied the possibility of application as an external preparation for skin in various natural products which have not been widely known.
The inventors of the present invention, through repeated and various experiments, confirmed that the Sesame extract can provide a safe and sufficient anti-aging effect, whitening effect and moisturizing effect on the skin and completed the present invention.
Therefore, it is an object of the present invention to provide an external composition for skin containing hackberry extract as an active ingredient.
In order to achieve the above object, the present invention provides an anti-aging skin external composition comprising an extract of Sesame as an active ingredient.
The present invention also provides a whitening skin external composition comprising the extract of Sesame as an active ingredient.
The present invention provides a moisturizing skin external composition containing the extract as an active ingredient.
The composition of the present invention exhibits an anti-aging effect as well as a whitening effect and a moisturizing effect by containing the extract of Sesame, which can be variously used in cosmetics or medicine.
The skin external preparation composition according to the present invention ( Celtis) sinensis Pers . ) Contains the extract as an active ingredient.
Hackberry is a deciduous shrub of the elm family, scientific name Celtis sinensis Pers . It is also known as moon tree, plum tree, or maple. Stems stand straight, reaching 20m in height and 1m in diameter, with broad branches. The bark is gray with fine hairs on the branches. Leaves are alternate, egg-shaped, oval-shaped, and serrated at the top. Flowers bloom in May as a general flower, and male flowers hang in apical flowers in the bottom of newly grown branches, and 1-3 female flowers hang on the leaf axils in the upper part. Calyxes are 4 and male flowers have 4 stamens and 1 degenerate pistil. The female flower has a short stamen and one pistil divided into two pistils. Fruits are nucleus, round, 7mm diameter orange, ripen in October, and taste sweet. The surface has net-like wrinkles.
It is said that the bark is called baksupi (朴樹 皮), and the leaves of the tree are called baksufolia (朴樹 葉), which has been medicinal for a long time. The method for producing the hackberry extract used in the present invention is not particularly limited, and the leachate obtained by leaching and transferring from the hackberry, or the concentrate obtained by partially or fully concentrating the leachate, or prepared by drying the concentrate again It contains the extraction extract and the chemical itself which exhibits the main effect contained in the extract.
For example, in the extraction process of the Sesame extract of the present invention, water or an organic solvent is put in a fine and dried Sesame, and reflux extraction is carried out, and the residue and the filtrate are separated through filtration and centrifugation, and the filtrate is separated. Concentrate under reduced pressure to obtain the hackberry extract. The organic solvent usable in the present invention may be selected from ethanol, methanol, butanol, ether, ethyl acetate, chloroform or a mixed solvent of these organic solvents and water, and considering the safety of the raw material, preferably 100% water, 70% ethanol Or butylene glycol (Butylene Glycol) is used. At this time, the extraction temperature is preferably 18 to 80 ℃ for water, 18 to 40 ℃ is preferable for 70% ethanol and butylene glycol, the extraction time is preferably 6 to 168 hours. If the extraction temperature and the extraction time is out of the extraction efficiency may be lowered or a change of components may occur. After obtaining the extract using the solvent in the above can be obtained by cooling, heating and filtration at room temperature in a conventional manner known in the art to obtain a liquid, or may further evaporate the solvent, spray drying or freeze drying.
The composition of the present invention preferably contains the hackberry extract in an amount of 0.1 to 20% by weight based on the total weight of the composition. This is because when the content of the active ingredient is less than 0.1% by weight, the efficacy and effect by the above ingredients are weak, and when the amount of the active ingredient exceeds 20% by weight, there is a problem of skin stability or formulation.
The composition of the present invention may be a topical skin composition, and more specifically, may be used as an anti-aging skin topical composition, which is excellent in enhancing skin elasticity and improving wrinkles. In addition, the composition of the present invention can be used as a skin external preparation composition for whitening, which can provide an excellent whitening effect by inhibiting the production of melanin. Furthermore, the composition of the present invention may be used as a moisturizing skin external preparation composition, which may enhance skin barrier function and induce differentiation of skin keratinocytes. Therefore, it can be usefully used as an external preparation composition for skin that prevents or improves skin dryness or psoriasis caused by incomplete epidermal differentiation.
When formulating the external composition for skin according to the present invention in the form of cosmetics, such as softening cosmetics, astringent cosmetics, nourishing cosmetics, eye cream, nutrition cream, massage cream, cleansing cream, cleansing foam, cleansing water, powder, essence or pack It may be formulated in a form, the formulation is not particularly limited. In addition, the composition according to the present invention can be used for fatty substances, organic solvents, solubilizers, thickeners, gelling agents, softeners, antioxidants, suspending agents, stabilizers, foaming agents, fragrances, surfactants, water, ionic or non- With ionic emulsifiers, fillers, metal ion sequestrants, chelating agents, preservatives, vitamins, blockers, wetting agents, essential oils, dyes, pigments, hydrophilic or lipophilic active agents, lipid vesicles or any other ingredients commonly used in cosmetics. It may contain adjuvants commonly used in the same cosmetic or dermatology field. The adjuvant is introduced in an amount generally used in the cosmetic or dermatological field. The compositions of the present invention may also contain a skin absorption promoting substance to increase the skin improving effect.
In addition, the composition according to the present invention may be a pharmaceutical composition. The pharmaceutical composition may further contain preservatives, stabilizers, hydrating or emulsifying accelerators, pharmaceutical adjuvants such as salts and / or buffers for controlling osmotic pressure and other therapeutically useful substances.
Hereinafter, the constitution and effects of the present invention will be described in more detail with reference to test examples and formulation examples. However, these test examples and formulation examples are provided for illustrative purposes only for the sake of understanding of the present invention, and the scope and scope of the present invention are not limited by the following examples.
Example 1 Preparation of Hackberry Extract
The dried stems, leaves and bark of the dried hackberry were refluxed three times for 24 hours with 70% (ethanol aqueous solution), cooled, and filtered through Whatman # 5 filter paper. The filtered extract was concentrated under reduced pressure and freeze dried at 45 ° C or lower.
A hackberry extract was prepared by dispersing / dissolving using at least one solvent selected from purified water, ethanol, butylene glycol, and propylene glycol so as to contain 0.1 to 20% by weight of the vacuum concentrate and the lyophilisate.
[Test Example 1] melanin production inhibitory effect
MEM containing 10% fetal bovine serum in human melanoma cells (Y. Funasaka, Department of dermatology, Kobe university school of medicine, 5-1 Kusunoki-cho 7-chrome, Chuo-ku, Kobe 650, Japan) (Minimum Essential Medium) was incubated under 37 ℃, 5% CO 2 conditions. The cultured cells were spread over 75 flasks so that the number of cells was 3 × 10 5 per flask, waited overnight for the cells to adhere to the wall, and after confirming that the cells adhered well, the medium was tested. Change to fresh medium containing ppm. As a control, a medium containing DMSO was used. In this way, the cells were inoculated with fresh medium every two to three days and cultured until the cells were filled in the flask. When the cells were grown, the cells were collected and compared with those of the control and the treated cells. In addition, the culture medium was removed, washed with PBS and dissolved with 1N sodium hydroxide to measure the absorbance at 500 nm and calculated the inhibition rate of melanin production according to Equation 1 below, the results are shown in Table 1 below.
From the results of Table 1, it was found that the use of Sesame extract, the melanin production inhibition rate is higher than kojic acid known to have a very good whitening effect.
Production Example 1 and Comparative Production Examples 1 to 3
According to the composition shown in Table 2 below, the cosmetics of Preparation Example 1 and Comparative Preparation Examples 1 to 3. Looking at the manufacturing process in detail, the raw materials 1 ~ 8 were mixed and dissolved at 70 ℃ to give a water part. On the other hand, raw materials 9-15 were melt | dissolved at 70 degreeC, and it was set as the oil part. An oil part was added to the above water part, stirred with a homomixer (Tokushu Kika Co., Ltd., Japan) and emulsified first, and the raw materials 15 and 16 were added to increase. After removing the bubbles, the mixture was cooled to room temperature to prepare a cosmetic composition of Preparation Example 1 and Comparative Preparation Examples 1 to 3 (unit: wt%).
Test Example 2 Whitening Effect on Human Skin
To examine the whitening effect and skin irritation on human skin, an opaque tape with 6 holes of 1.5 cm diameter was attached to the upper forearm area of 30 healthy men and women aged 35.2 years old. Ultraviolet light (UVB) of 1.5 ~ 2 times the minimum Erythema Dose of the subject was irradiated to induce blackening of the skin, and after 2 months of application of test substances, the contrast of the skin was measured using a colorimeter. . To these subjects, the cosmetics of Preparation Example 1 and Comparative Preparation Examples 1 to 3 were applied twice daily in the morning and evening.
The determination of the effect was determined by obtaining a "L" value representing the contrast of the skin. For reference, Korean skin color that is not artificially burned generally shows a value of 50-70. The effect was determined by measuring the degree of black and white of the skin using a color difference meter (Minolta CR2002). L * a * b * colorimetric system is used for displaying the color, but in the present invention, the L * value (brightness) is mainly used as an index. The L * value was calibrated with a standard whiteboard, and the pigmentation part was measured evenly by repeating the measurement five times or more at one site. The difference in skin color (ΔL *) at the start and completion of the application was calculated according to Equation 2 below, which is shown in Table 3 below. The whitening effect is determined by comparing ΔL * between the sample application site and the control site. When the value of ΔL * is about 2, the whitening effect of the deposited pigment is clear. .
When the test substance is applied, the L value is gradually increased. The comparison between the test substances is based on the difference between the skin color (ΔL) at the start of application and completion (two months after application). It was calculated according to 2, the results are shown in Table 3 below.
Test Example 3 Investigation of Skin Irritability on Human Skin
For the test subjects of Example 2, the cosmetic preparations of Preparation Example 1 and Comparative Preparation Examples 1 to 3 were used for two months, and then skin irritation was compared. At this time, the criterion is compared by giving the subjects a score of 1 to 5 according to the degree of stimulus, the closer to 1 means no stimulus, the closer to 5 means more stimulus, each average score is shown in the following table 3 is shown.
As can be seen from the results of Table 3, the cosmetics of Preparation Example 1 containing the extract of Sesame according to the present invention is superior in whitening effect than Comparative Preparation Examples 1 to 3 and at the same time the skin irritation is lower than when using the same amount of koji acid I could confirm it.
Test Example 4 Change in Angular Mass
In order to examine the angular mass reduction effect of the whitening cosmetic composition according to the present invention, the skin angular mass change of the cosmetics prepared in Preparation Example 1 and Comparative Production Examples 1 to 3 was examined.
After applying the cosmetic composition of Preparation Example 1 and Comparative Preparation Examples 1 to 34 to the inner half of 50 males and females in their 20s to 30s without skin diseases, Charm view (Moritex, Japan) The amount of keratin decrease was measured using. The initial skin angular mass was measured and used as the basic value at the constant temperature and humidity conditions (24 ° C., humidity 40%) before starting the coating, and the change after 24 hours was measured, and the results are shown in Table 4 below. It was.
From the results of Table 4, it was found that Preparation Example 1, which is a whitening cosmetic composition according to the present invention, has a superior skin exfoliation effect as compared to Comparative Preparation Examples 1-3. Therefore, the cosmetic containing the extract of Sesame according to the present invention was able to provide excellent skin safety by minimizing skin irritation with excellent whitening effect while being safe for the skin.
[Test Example 5] Skin moisturizing effect test
In order to find out the moisturizing sustainability of the skin at the time of application, the cosmetic composition of Preparation Example 1, Comparative Preparation Example 1 and Comparative Preparation Example 3 After application to the inside of the face and forearms, the skin moisture content was measured using a corneometer (CM820 courage Khazaka electronic GmbH, Germany) for 24 hours. The conniometer measures the moisture level by measuring the amount of moisture by measuring the amount of moisture in the amount of water present in the epidermis of the skin using a sensor and quantifying the amount of water, and the measuring method is as follows.
1) A coneometer probe was placed on the skin area to be measured.
2) When the probe is pressed onto the skin, the electrical conductivity of the skin is quantified through the sensor and displayed on the screen.
3) Measurement was repeated with different measurement sites.
4) After measuring once, the sensor was wiped with a tissue such as Kimwipe and then measured again.
In the above method, the skin moisture content was measured using a conniometer at a constant temperature and a constant humidity condition (24 ° C. and a humidity of 40%) before the start of application in advance, and the change after 12 hours and 24 hours was measured. The results are shown in Table 5 below.
In the results of Table 5, when Comparative Preparation Examples 1 and 3 were applied, the skin moisture content increased to some extent after 12 hours after application, but after 24 hours, it was found to return to the state before application. On the other hand, when applying the skin preparation composition 1, the extract containing the hackberry extract to the skin, the moisturizing power lasted for more than 24 hours, it was confirmed that the effect of increasing the skin moisture content is also significantly higher than Comparative Preparation Examples 1 and 3.
Test Example 6 Evaluation of the Dry Skin Improvement Effect
At the end of Test Example 5, subjects were subjected to a subjective evaluation of efficacy through a questionnaire survey on improvement of dry skin after using Preparation Example 1, Comparative Preparation Example 1, and Comparative Preparation Example 3. The questionnaire survey was to indicate the degree of satisfaction of the skin dryness improvement effect is divided into very good, good, normal and poor, the results are shown in Table 6 below.
