Abstract
본 발명은 월계수 알코올 추출물 및 헥산층 분획물, 클로로포름 분획물들의 전 처리로 H2O2와 Dopamine에 의한 세포사멸이 억제되었으며, Dopamine에 의한 염색체 응축이나 DNA 절편 등 세포사멸의 특징이 줄어드는 효과가 있고, 또한 Dopamine을 처리한 실험군에서는 а-synuclein 단백질의 양이 증가하였는데 월계수 분획물과의 동시 처리는 а-synuclein의 발현 양을 감소시켜주는 효과가 매우 좋음을 확인할 수 있고, SH-SY5Y cell과 흰쥐 대뇌피질 일차배양 신경세포에서 Dopamine 유도된 세포사멸에 대해 월계수 hexane 분획물이 신경세포 손실에 대해 보호 및 회복할 수 있어 퇴행성 뇌질환인 파킨슨병 예방 및 치료용 조성물의 유효성분으로 이용될 수 있는 월계수 잎의 알코올 추출물 및 헥산, 클로로포름 분획물을 함유하는 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물에 관한 것이다.
Description
본 발명은 월계수 잎의 알코올 추출물 및 헥산, 클로로포름 분획물을 함유하는 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물에 관한 것으로, 보다 상세하게는 신경세포 사멸에 대해 보호 효과가 있고, 세포 자멸사 염색에 효과가 있는 월계수 잎의 알코올 추출물, 헥산 분획물, 클로로포름 분획물 중 어느 하나의 성분을 선택적으로 사용하여 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물에 관한 것이다.
노인성치매, 뇌졸중, 파킨슨병 등의 퇴행성 신경계 뇌 질환은 신경세포 손상과 소실로 이어지는 세포죽음이 그 궁극적 원인이 되고 있다. 예를 들면 노인성 치매의 경우 기억, 인지 등에 관여하는 대뇌피질, 해마 등에서 신경세포들이 소실됨 으로써 기억상실을 포함한 주 증상들이 나타나는 것으로 여겨지고 있다. 비슷하게 파킨슨병의 경우에는 중뇌의 흑색질에 있는 도파민성 신경세포들이 선택적으로 소실됨으로써 운동기능조직에 관여하는 기저핵의 기능 이상을 초래하여 운동기능 장애를 유발하게 된다. 파킨슨병(Parkinson's disease)은 두 번째로 흔한 퇴행성 신경 뇌 질환으로 65세 이상 인구의 1%, 85세 이상 인구에서는 5%정도로 발병한다. 특징적인 임상적 증상으로는 지속적 떨림, 경직, 서동, 자세 불안정을 수반하며 특히 일부 환자들에서 알츠하이머병 혹은 루게릭 병이 동반되기도 하여 파킨슨병과 알츠하이머병간의 공통적인 병리 기작이 암시되어 있다.( Orly W, Silvia M, Tamar A, Moussa B H, Neurological mechanisms of green tea polyphenols in Alzheimer's and Parkinson's diseases. 파킨슨병의 병리적 소견은 흑색 치밀부(substantia nigra pars compactra)의 도파민성 신경세포의 손실과 세포 내 단백질 축적체인 루이소체(LB)의 형성이며( Inamdar N, Arulmozhi D, Tandon A, Bodhankar S, Parkinson's disease: genetics and beyond. 도파민(Dopamine, C8H11NO2)은 뇌신경 세포들간에 어떠한 신호를 전달하기 위해 분비되는 신경전달물질 중 하나이며, 중뇌의 복측에 위치해 있는 흑색질 부위에서 생성된다. 뇌의 많은 기능을 포함하는 중요한 행동과 역할에 영향을 준다. 특히 운동 조절이나 호르몬 조절, 감정, 동기 부여, 욕망, 쾌락, 의욕, 수면, 인식, 학습 등에 영향을 미친다. Dopamine 분비 조절에 이상이 발생하면 사람에게 다양한 질환이 발생한다. 현재로서는 파킨슨병의 치료법이 없고, 단지 Dopamine 전구물질인 L-dopa 혹은 Dopamine 수용체 촉진제가 증상 호전제로 알려져 있으며, 이러한 약물치료 약물들도 지속적인 도파민성 신경의 손실을 막아내지 못해 결국은 5~6년 내에 그 효과도 급격히 감소하며 운동장애를 포함하는 부작용이 증가하여 병의 후기에는 on-off 현상으로 알려진 L-dopa의 문제들이 급격히 증대되어 발병 후 10~15년 후에 사망에 이른다.(Vautier S, Milane A, Fernandez C, Buyse M, Chacun H, Farinotti R, Interactions between antiparkinsonian drugs and ABCB1/P-glycoprotein at the blood-brain barrier in a rat brain endothelial cell model. 따라서 파킨슨병의 정확한 분자 병리 기작에 의거해 병의 진행과정을 늦추거나 막을 수 있는 신경보호 치료제 개발이 시급한 실정이다.
본 발명에서는 월계수(Laurus nobilis) 잎의 추출물을 이용하였다. 일반적으로 월계수 잎은 향신료나 간간히 민간 약재로 이용되고 있다. 현재 문헌에 보고된 바에 의하면 월계수 잎의 추출물은 혈중 알코올 농도의 저하, lipopolysaccharide (LPS)에 의해 활성화 된 nitric oxide(NO) 생성 억제 활성 등의 생물학적 활성이 보고되어 있다.(Yoshikawa M, Shimoda H, Uemura T, Morikawa T, Kawahara Y, Matsuda H, Alcohol absorption inhibitors from bay leaf (Laurus nobilis): structure-requirements of sesquiterpenes for the activity. Planta Med.5(1998) 454-5.) 하지만 뇌 관련 질환의 신경세포의 보호에는 연구된 바 없으며 파킨슨병 동물모델 실험에도 역시 처음이다.
따라서 월계수의 산화적 스트레스와 도파민유도성 신경세포사멸의 보호기전과 동물 모델에서의 도파민성 신경세포의 손실 후 치료효과를 통해 파킨슨병에 보호와 치료효과는 물론 파킨슨병과 공통적인 병리 기작을 갖는 알츠하이머병 혹은 루게릭 병과 Dopamine이 관여하는 조울증이나 정신 분열증, 우울증에 효과를 기대할 수 있으며 신경세포들이 소실됨으로써 발생하는 노인성 치매, 뇌졸중, 기억력 상실에 대한 효과를 기대할 수 있다.
본 발명은 상기와 같은 문제점을 해결하기 위해 안출된 것으로서, 그 목적은 신경세포의 사멸에 대한 보호 효과 및 파킨슨 동물 모델의 도파민손실에 대한 치료효과로 월계수 잎의 알코올 추출물, 헥산층 분획물, 클로로포름층 분획물이 효과가 있음을 증명하여 뇌신경 세포보호 및 파킨슨병을 비롯한 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물을 제공하는데 있다.
상기의 목적을 달성하기 위한 본 발명의 제 1 특징에 따르면, 음건 건조하여 세절한 월계수 잎에 100% 주정을 가하고 55℃에서 48시간 동안 추출하여 에탄올 층을 추출하는 제 1 단계; 제 1 단계를 통해 추출한 100% 에탄올 전체 추출물을 감압 농축하여 알코올 용매로 얻은 엑스를 분획하여 클로로포름 분획하는 제 2 단계; 건조된 월계수 잎을 절단한 뒤 메탄올에 냉침한 후 여과지로 여과하는 과정을 3회 반복하여 헥산을 분획하는 제 3 단계; 및 메탄올 추출물에 헥산을 가하여 진탕 반복 추출하고 분액 깔때기에서 분획하여 헥산층과 메탄올층을 분취한 후, 헥산층을 감압 농축하여 헥산층 분획물을 분획하는 제 4 단계를 포함하여 제조되는 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물을 제공하는데 있다.
이때, 본 발명의 부가적인 특징에 따르면, 제 1 항에 따른 월계수 에탄올 추 출물, 월계수 헥산층 분획물 또는 월계수 클로로포름층 분획물을 유효성분으로 포함하고 통상의 약제학적으로 허용가능한 담체와 혼합하여 이루어지는 것이 바람직하다.
또한, 본 발명의 부가적인 특징에 따르면, 약제학적으로 사용되는 제제 형태가 산제, 과립제, 정제, 캅셀제, 액제, 주사제 중에서 선택된 어느 하나로 이루어지는 것이 바람직하다.
또한, 본 발명의 부가적인 특징에 따르면, 상기 조성물은 월계수 잎 추출물을 1∼2,000㎎/㎏체중의 농도로 함유함을 특징으로 한다.
본 발명에 의한 월계수 잎의 에탄올 추출물 및 헥산, 클로로포름 분획물을 함유하는 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물은 월계수 에탄올 추출물 및 헥산층 분획물, 클로로포름 분획물들의 전 처리로 H2O2와 Dopamine에 의한 세포사멸이 억제되었으며, Dopamine에 의한 염색체 응축이나 DNA 절편 등 세포사멸의 특징이 줄어드는 효과가 있고, 또한 Dopamine을 처리한 실험군에서는 а-synuclein 단백질의 양이 증가하였는데 월계수 분획물과의 동시 처리는 а-synuclein의 발현 양을 감소시켜주는 효과가 매우 좋음을 확인할 수 있고, SH-SY5Y cell과 흰쥐 대뇌피질 일차배양 신경세포에서 Dopamine 유도된 세포사멸에 대해 월계수 hexane 분획물이 신경세포 손실에 대해 보호 및 회복할 수 있 어 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물의 유효성분으로 이용될 수 있다.
상기한 목적을 달성하기 위해서, 본 발명은 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물에 이용될 수 있는 물질로서 월계수 잎으로 건조하여 세절한 월계수 잎을 에탄올, 메탄올을 통해 분획된 월계수 추출물, 헥산 분획물, 클로로포름 분획물을 유효성분으로 함유하는 것을 특징으로 한다.
이하, 본 발명을 보다 구체적으로 설명한다.
본 발명의 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물은 총중량에 대하여 월계수 분획물을 0.5~ 50 중량%로 포함한다.
본 발명은 상기 월계수 분획물은 월계수 에탄올(EtOH) 분획물, 월계수 헥산(Hexane) 분획물 또는 월계수 클로로포름 분획물 중 어느 하나의 분획물을 선택적으로 사용할 수 있는 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물을 제공한다.
본 발명의 월계수 분획물을 포함하는 조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 및 희석제를 더 포함할 수 있다.
본 발명의 상기 분획물을 약제학적으로 사용되는 제제 형태가 산제, 과립제, 정제, 캅셀제, 액제, 주사제 중에서 선택된 어느 하나인 뇌신경 세포보호 및 파킨 슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물을 제공한다.
이하, 본 발명을 실시예들을 통하여 보다 구체적으로 설명하지만, 본 발명이 이에 의해서 한정되는 것은 아니고, 당업자에 의해서 용이하게 실시될 수 있는 삽입, 삭제 및 수정 등의 변형 등도 본 발명의 범위 내에 포함된다.
실험예
시료의 조제
본 실험에서 사용된 월계수는 터키산 월계수 잎으로 이를 알코올 용매로 얻은 엑스를 분획하여 클로로포름 분획을 얻었다.
상기 첨부된 도 1a 내지 도 1b를 참조하여 본 발명에 따른 월계수 잎에서 에탄올 추출물과 헥산, 클로로포름층의 분리 과정을 살펴보면, 월계수 잎 추출물은 통상적인 방법에 따라 제조될 수 있으며, 예를 들어, 음건 건조하여 세절한 월계수 잎에 100% 주정을 가하고 55℃에서 48시간 동안 추출하여 에탄올 층을 얻을 수 있다.
이렇게 얻은 100% 에탄올 전체 추출물은 감압 농축하여 알코올 용매로 얻은 엑스를 분획하여 클로로포름 분획을 얻었고 헥산층 역시 건조된 월계수 잎을 절단한 뒤 메탄올에 냉침한 후 여과지로 여과하는 과정을 3회 반복하였다.
이 메탄올 추출물에 헥산을 가하여 진탕 반복 추출하고 분액 깔때기에서 분 획하여 헥산층과 메탄올층(10% aqueous MeOH) (1:1,v/v)을 분취한 후, 헥산층을 감압 농축하여 헥산층 분획물을 얻었다.

인간 신경아세포종 SH-SY5Y cell (ATCC no. CRL-2266)은 10% fetal bovine serum(Gibco-BRL, USA) 및 antibiotics (Sigma-Aldrich, USA)가 함유된 dulbecco’s modified eagle’s medium (Sigma-Aldrich, USA) 배지 하에서 배양하였다. 배양기는 37℃ 온도를 유지했고 95% 공기와 5% CO2가 혼합된 기체가 계속 공급되어 세포 배양의 적절한 조건을 갖추었다. 세포는 6-well plate에 3x105 cells/well로 배양하였다. Dopamine의 농도는 세포 생존율을 평가한 후 600μM로 결정하였으며, 월계수 Ethanol추출물과 Hexane층, Chloroform층, 분획물들 (dissolved in dimethyl sulfoxide (DMSO, Sigma-Aldrich, USA), final concentration 0.25%)은 Dopamine (Sigma-Aldrich, USA) 처리 24시간 전에 선행하여 월계수 각층 분획물의 효과를 비교 관찰하였다.
일차 배양한 대뇌피질 신경세포
대뇌피질 일차배양 신경세포는 임신 18일의 암컷 흰쥐를 이용하였으며, 쥐 배자로부터 뇌를 적출한 후, 뇌막을 조심스럽게 제거하고 대뇌피질을 분리하였다. 끝의 크기를 줄인 파스퇴르 파이펫을 사용하여 개개 세포로 분리한 다음 PM (plating medium; Eagle’s minimum essential medium (Sigma-Aldrich, USA), 5% FBS, 5% horse serum, 2 M glutamine, and 20mM glucose )에 희석하여 plating 전에 미리 100 ㎕/㎖ poly-L-lysine과 4 ㎕/㎖ laminin (Sigma-Aldrich, USA)을 이용하여 코팅해둔 6-well plate에 세포를 분주한 후, 5% CO2, 37℃로 유지되는 세포배양기에서 배양하였다. 7일째에 성상세포(astrocyte)가 단일 층으로 자란 후 교세포(glia)의 과다분열을 억제하기 위해 10μM cytosine β-D-arabinofuranoside (Sigma-Aldrich, USA)를 처리한 GM (growth medium; Eagle’s minimum essential medium, 5% horse serum, 2 mM glutamine, and 20mM glucose)으로 갈아주었다. 일주일에 2번씩 GM을 갈아주면서 신경세포가 자라도록 하였다.
Dopamine의 농도는 세포 생존율을 평가한 후 300μM로 결정하였으며, 월계수 Hexane층 분획물(dissolved in DMSO, final concentration 0.25%)은 Dopamine 처리 24시간 전에 선행하여 효과를 관찰하였다.
DAPI 염색을 통한 핵의 관찰
Apoptosis에 의한 SH-SY5Y cell의 형태적 변화를 관찰하기 위하여 월계수 Hexane 분획물 및 Dopamine을 처리한 세포들을 PBS 세척하고, 4 % 포르말린 용액으로 상온에서 고정시킨 후, 0.1 % triton-X 용액을 10분간 가하였다. 그 후 형광 염색 물질인 DAPI를 1 ㎍/㎖의 농도로 30분간 암실에서 염색하였다. 핵의 형태를 관찰하기 위해 형광 현미경 (Eclipse TE 200, Nikon, Japan)을 이용하여 정상군과 비 교하였다.
유세포 분석
DNA content에 미친 영향을 알아보기 위해 유세포 분석을 실시하였다. 채취한 세포를 PBS로 3회 세척하였고 차가운 70% 에탄올로 -20℃에서 1일 고정시킨 후 1000 rpm에서 5분간 원심 분리하여 PBS로 씻어 주었다. 그 후 40㎍/㎖의 Propidium Iodide (PI, Sigma-Aldrich, USA)로 4℃에서 30분간 염색하고 유세포 분석기 (FACs, Coulter EPICS XL, USA)로 측정하였다. Apoptosis 세포는 PI 염색 강도가 감소하여 일반세포와 비교할 수 있었다.
Annexin Ⅴ-PI 염색
배양 종료 후 well에 trypsin-EDTA(SIGMA, USA)용액을 처리하고 차가운 PBS를 이용하여 세포를 모았다. 모아진 세포는 반응 용액 200μℓ에 다시 부유시키고 Annexin Ⅴ-PI 염색용 kit(Bender Medsytem, Austria)를 이용하여 염색하였다. Annexin Ⅴ-FITC항체와 PI용액을 10μℓ씩 첨가하고 15분간 반응하였다. 반응 후에는 유세포 분석기를 이용하여 Dopamine에 의한 세포사멸이 후기 Apoptosis로 진행됨을 확인하였고 이에 월계수 Hexane 분획물이 세포 사멸에 대한 보호효과 정도를 측정하였다.
Western blot 분석
SH-SY5Y cell을 배양한 후, PBS 세척을 하고 PRO-PREP protein solution (Intron biotechnology, Korea)를 이용하여 총세포 단백질을 추출하였으며 protein assay solution (Intron biotechnology, Korea)을 이용하여 단백질 농도를 측정하였다. 10μg/lane의 세포단백질을 SDS-polyacrylamide gel에서 전기영동 시킨 후 PVDF membrane (Amersham Pharmacia Biotech, UK)으로 transfer 시키고, 이 membrane을 5% skim milk in tris buffered-0.1% tween 20 (TBST)으로 실온에서 2시간 동안 blocking 시켰다. 그 다음 일차 항체 (α-synuclein, abcam, UK, β-actin, Santa cruz biotechnology, CA)와 4℃ 에서 24시간 반응시키고 TBST로 세척한 후 이차 항체 (anti-mouse HRP-conjugated IgG; Chemicon, USA)와 실온에서 1시간 반응시켰다. 세척 후 이미지 분석기 LAS1000 (Fuji photo film, Japan)으로 분석하였다. 모든 western blot 분석은 3회 반복 실험하였다.
실험동물과 파킨슨병의 유발
몸무게가 대략 250g의 수컷 Sprague-Dawley 흰쥐 (Samtako, Korea) 36마리를 5마리 이상씩 6군으로 나누어 사용하였다. 수컷 흰쥐를 ketamine 50mg/kg과 xylene 10mg/kg 혼합용액을 복강 주사하여 마취, 실험을 시행하였으며 초기 파킨슨병의 진행 정도에 해당하는 70~80% 뇌의 흑색질 부위의 도파민성 세포 소실을 만들기 위해 6-OHDA를 뇌의 직접 주사하는 수술방법을 사용했다.
6-OHDA는 ascorbic acid를 0.2%(w/v) 섞은 생리 식염수 1μl당 5.0 μg으로 녹여 사용하였고 이를 500μℓ씩 차광한 상태에서 냉동 보관한 후 필요에 따라 하 나씩 녹여서 사용하였다. 수컷 쥐를 동물용 뇌정위 고정수술틀(stereotaxic, Daejong, Korea)에 머리를 incisor bar를 이용하여 고정하였다.
두피에 약 10~15 mm의 수직으로 절개를 하고 정수리점(bregma)을 기준으로 후방 4.9 mm, 좌 측방 1.8 mm 되는 곳에 드릴로 구멍을 뚫고 그 부위에 10μl 용량의 Hamilton syringe(Daejong, Korea) 주사침을 경막으로부터 7.8 mm 깊이로 수직 주사 후 분당 1 μl의 속도로 총 4 μl의 6-OHDA을 주입하였다.
주입 후 15분을 기다린 후 서서히 주사침을 제거시키고 두피를 봉합하였다. 정상 대조군은 6-OHDA를 녹인 용매인 ascorbic acid를 0.2% (w/v) 넣은 생리 식염수를 같은 수술 부위에 주사하는 것 이외에 모든 마취와 수술 과정을 파킨슨병 유발 실험군과 동일하게 진행하였다.
약물투여
월계수 Hexane층 분획물을 올리브오일(Sigma-Aldrich, USA)에 녹인 후 파킨슨 동물모델 수술 1시간 후 1회 용량별로 복강 투여하였다. 대조군은 올리브오일만 복강 투여하였다.
도파민성 신경세포의 면역조직화학염색
쥐를 ether로 마취한 후 흉벽을 제거하고 왼쪽 심실에서 대동맥 방향으로 주사 바늘을 삽입하였다. 그 다음 오른쪽 심방 위를 가위로 절단한 뒤 연동펌프를 이용하여 0.9% 생리식염수를 관류시켜 혈액을 제거하였다. 그리고 나서 4% 포르말린 용액을 200㎖ 관류시켜 고정하였다. 제거된 뇌는 4℃에서 하루 동안 4% 포르말린 용액에 후 고정을 시행한 다음 4℃에서 30% sucrose (Sigma-Aldrich, USA) 용액에 뇌가 가라앉을 때까지 방치하였다. 적출된 뇌를 microtome (Leica, Germany)를 이용하여 -25℃에서 10 ㎛의 두께로 잘랐다. 도파민성 신경세포의 양을 확인하기 위하여 절편이 부착된 슬라이드를 실온에서 건조 후, PBS로 10분간 3회 세척 후 3% H2O2로 20분간 내인성 과산화효소의 활성도를 없애고, 다시 PBS로 3차례 세척하였다. 실온에서 normal blocking serum으로 1시간 배양하여 비특이적 반응을 제거한 후, 일차 Tyrosine Hydroxylase 항체(mouse anti-TH antibody, 1:1000, Chemicon, USA)를 4℃에서 18~24시간 배양하였다. PBS세척 후 이차 항체로 1시간 실온에서 배양한 후 세척하고 Vectastain ABC kit (Vector Lab., USA)와 Diaminobenzidined (DAB) (Vector Labortories, CA) 용액으로 5~10분간 발색하여 alcohol, xylene(DC chemical, Korea)으로 탈수 과정을 거친 후 Permount (Shandon, USA)로 cover slide(Muto-Glass, Japan)를 덮어 400 배율의 광학현미경(Olympus, Japan)으로 관찰하였다. TH 면역염색에 있어서 각 농도군 별로 1마리당 5~6개의 중뇌 흑색질 부위 절편을 취한 후 흑색질에서 6-OHDA를 주입하지 않은 좌측의 TH -염색 양성에 신경세포에 대한 병변 측의 도파민성 신경 세포의 평균 숫자를 퍼센트로 계산하여 각각의 대조군과 월계수 약물 처리군을 비교하였다.
통계적 처리
모든 측정값은 평균값±표준오차로 표시하였고, 각 실험군과의 통계학적 분석은 Sigma Plot 8.02 프로그램을 이용하여 t-test를 실시하였다. P < 0.05를 통계적으로 유의한 것으로 간주하였다.
실험결과 1
SH-SY5Y cell의 산화적 스트레스(H2O2)에 대한 월계수 Hexane 분획물의 영향
산화적 스트레스와 관련된 H2O2를 월계수 Hexane 분획물을 처리하고 24시간 뒤에 처리한 세포를 추출하여 apoptosis를 확인하기 위해 PI를 이용한 염색을 시행한 결과, 첨부된 도 2에서 도시된 바와 같이 정상 대조군에서는 3.7%만이 apoptosis를 나타내고, H2O2를 처리한 대조군은 44.1%의 apoptosis를 보였다.
반면 월계수 Hexane층 분획물은 0.16, 0.8, 4㎍/㎖을 전처리한 경우 각각 35.8, 30.4, 26.7%로 apopsosis를 감소시켰다..
SH-SY5Y cell의 Dopamine에 대한 월계수 분획물들의 영향
SH-SY5Y cell에 월계수 Ethanol 추출물과 Hexane층, Chloroform층 분획물들의 Dopamine에 대한 효과를 확인하기 위해, 첨부된 도 3a 내지 도 3c에 도시된 바와 같이 SH-SY5Y cell에 월계수 각 분획물들을 농도별로 각각 처리하고 24시간 후 600μM Dopamine을 처리하여 FACs analysis(유세포 분석)를 실시하여 그 결과가 도 3a 내지 도 3c의 그래프와 같이 도출되었다.
상기의 결과에 따라 월계수 Ethanol 추출물과 Hexane, Chloroform 분획물들은 [표 1]에 도시된 바와 같이 농도 의존적으로 세포 생존율을 증가시켰으며, 특히 월계수 Hexane 분획물은 EC50 값이 3.8±0.1 ㎍/㎖로 가장 효과가 있었다.
즉, 첨부된 도 3a 내지 도 3c는 월계수 Ethanol 추출물과 Hexane, Chloroform 분획물이 어느 농도에서 제일 잘 보호했는가를 측정한 실험결과로서 월계수 Hexane 분획물이 세포생존율을 50%로 생존시킨 효과농도(EC50)가 3.8±0.1 ㎍/㎖를 표시하고 있다.

또한, 도 4에 도시된 바와 같이 월계수 Hexane 분획물을 농도별로 처리한 24시간 뒤 600μM Dopamine을 처리한 세포를 추출하여 apoptosis를 확인하기 위해 PI 를 이용한 염색을 시행하였다. 그 결과 도 4에 도시된 바와 같이 정상 대조군에서는 6.0%만이 세포사멸(apoptosis)를 나타내는 반면, 600μM Dopamine을 처리한 대 조군은 63.1%의 apoptosis를 보였다. 월계수 Hexane 분획물 4, 0.8, 0.16 ㎍/㎖을 전처리한 경우 각각 32.1, 47.8, 53.7%로 세포사멸(apopsosis)를 감소시켰다.
SH-SY5Y cell에서 후기apoptosis 진행에 월계수 Hexane 분획물의 영향
SH-SY5Y cell에서 Dopamine에 의한 후기 apoptosis로 진행유발에 월계수가 어떤 영향을 주는지 확인하기 위해 Annexin Ⅴ-PI 염색 하였다. 그 결과 첨부된 도 5에서 도시된 바와 같이 정상 대조군에서는 1.3% 만이 후기 apoptosis로 진행된 반면, Dopamine만 처리한 실험군은 후기 apoptosis로 49.0% 진행이 되었고 월계수 Hexane 분획물을 0.16, 0.8, 4 ㎍/㎖을 처리한 실험군은 45.2, 42.2, 17.6% 후기 apoptosis가 진행되는 것을 농도 의존적으로 줄여주는 것을 알 수 있었다.
SH-SY5Y cell의 형태적 변화에 대한 월계수 Hexane 분획물의 영향
DAPI 염색에서 정상 대조군과 비교하였을 때, Dopamine만 처리한 실험군에서 apoptosis의 특징적인 응축된 핵과 분절된 핵(도 6에서 화살표 표시)을 많이 관찰할 수 있었다.
첨부된 도 6에 제시된 바와 같이 일차 배양한 대뇌피질의 신경세포에 월계수 Hexane 분획물을 전 처리한 24시간 후 300μM Dopamine을 처리하였더니 4㎍/㎖의 농도에서 크게 세포 사멸을 억제하는 것을 DAPI라고 하는 염색시약을 이용해서 확인할 수 있다.
즉, 월계수 Hexane 분획물을 전 처리한 실험군에서는 Dopamine만 처리한 실 험군에 비해서 응축된 핵의 세포 수가 감소하고 정상 대조군과 같은 형태를 나타내었다.
SH-SY5Y cell에서 월계수 분획물들의 α-synuclein 단백질 발현에 미치는 영향
첨부된 도 7에서 SH-SY5Y cell에 Dopamine 600μM을 투여한 후 단백질을 시간대별로 추출하여 α-synuclein의 단백질의 발현 양을 western blot 분석을 수행하여 시간 변화에 따른 Dopamine에 대한 과발현에 정도를 확인한 결과 α-synuclein의 단백질의 과발현은 9시간부터 15시간까지 시간 의존적으로 증가하였고 특히 15시간에 특징적으로 증가됨을 관찰할 수 있다.
또한, 월계수 Hexane과 Chloroform층 분획물의 α-synuclein의 단백질 과발현에 미치는 영향을 확인하기 위해 첨부된 도 8과 같이 월계수 Hexane과 Chloroform층 분획물 각각을 Dopamine을 600μM과 같이 15시간 동안 처리한 후 α-synuclein 단백질의 발현 정도를 western blot 분석하면, 그 결과 α-synuclein 단백질의 발현 정도는 두 번째 band가 제일 짙고 농도가 높아질수록 옅어지는 정도가 농도의존적 효과로 나타난다.
상기 도 8의 절차와 같이 SH-SY5Y cell에 Dopamine과 월계수 Chloroform 분획물을 농도별로 15시간 동시처리한 후 α-synuclein 단백질의 발현을 확인하면, 도 9에서 제시된 바와 같이 Dopamine이 α-synuclein 단백질 발현에 영향을 준 15시간 대를 기준으로 살펴본 결과, 월계수 Chloroform 분획물들은 농도 의존적으로 α-synuclein 단백질 과발현을 막는 효과가 있었다.
월계수 Hexane 분획물의 흰쥐 뇌의 흑색질 부위의 TH-양성반응 세포에 대한 영향
상기의 결과에 따라 월계수 Hexane 분획물의 흰쥐 뇌의 흑색질 부위의 TH-양성반응 세포를 확인하기 위해 Tyrosine Hydroxylase 항체로 TH-양성반응 신경세포의 소실 정도와 보호 정도를 Vectastain ABC 와 DAB 용액으로 염색하여 첨부된 도 10 내지 도 11에 도시된 바와 같이 sham, 파킨슨병 유도군, positive control인 apomorphine을 처리한 실험군과 월계수 Hexane층 분획물 투여 실험군의 Tyrosine Hydroxylase양성반응 신경세포를 조직 면역 염색한 결과와 그 결과를 광학현미경으로 확인하면 파킨슨병 유발 동물군에서는 흑색질 부위에서 현격하게 감소하였던 다수의 Tyrosine Hydroxylase 양성세포(도10 참조)를 현재 효과가 있다고 보고되어 있는 Apomorphine에 비해서 월계수 실험군에서 효과가 더 좋았으며 농도 의존적으로 그 수가 유의성 있게 증가함을 또한 관찰할 수 있었다 (도 11 참조).
즉, 하기에 [표 2]에 도시된 흰쥐의 뇌 흑색질 부위의 TH 양성적인 신경세포를 조직면역염색을 실험군 별로 정량화한 표와 같이 월계수 분획물들이 뇌 신경세포 손상에 미치는 영향을 확인하기 위해 흰쥐의 흑색질 부위에 6-OHDA를 직접 주사하는 수술방법을 사용했다. 뇌정위 고정틀에서 파킨슨병 동물모델 수술 후 7일 뒤 흑색질 부위의 도파민성 신경세포의 손상 정도를 조직면역염색을 통해 관찰하였다.
대조군은 파킨슨병 동물모델 1시간 후에 vehicle을 복강 투여하였고 실험군은 월계수 Hexane 분획물을 0.2, 1, 4mg/kg 용량으로 복강 투여하였다.
염색 결과, 뇌에 흑색질 부위에 6-OHDA를 직접 주사한 실험 군은 주사한 뇌의 반구는 주사하지 않은 쪽의 뇌에 비해 Dopamine을 생성시켜주는 Tyrosine Hydroxylase의 양이 현저하게 줄어드는 것을 확인하였다.
또한, 한쪽 반구에 6-OHDA를 주사하여 도파민성 신경세포를 손실시킨 흰쥐에 수술 1시간 후 월계수 Hexane 분획물을 복강 투여한 실험 군에서는 Tyrosine Hydroxylase의 양이 농도 의존성 있게 증가함을 보여 도파민성 신경세포의 손실에 월계수 Hexane 분획물이 신경세포를 손실을 회복하는 효과가 있음을 확인할 수 있었다.

실험결과 2
산화적 스트레스와 과도한 Dopamine의 처리는 신경세포의 독성이 있으며 뇌조직의 신경세포를 손상시키는 것으로 알려져 왔다. 특히 이로 인한 신경세포의 apoptosis와 뇌 조직에서의 손실은 중요한 역할을 하며 실험을 통해 SH-SY5Y cell 에서 H2O2 200μM 과 Dopamine 600μM이 그리고 일차 배양한 대뇌피질 신경세포에는 Dopamine 300μM이 사멸을 일으키는 것으로 밝혀졌다.
월계수 분획물들은 산화적 스트레스에 관여하는 H2O2와 Dopamine에 의한 SH-SY5Y cell과 일차 배양한 대뇌피질 신경세포의 사멸에 대해 보호효과를 나타내었다.
FACs analysis(도 2, 3a, 3b, 3c, 4 참조) 와 Annexin Ⅴ-PI 염색을 이용한 후기 세포 자멸사 염색(도 5 참조), DAPI를 이용한 핵 응축과 분절염색(도 6 참조)을 통해 월계수 Ethanol 추출물 및 Hexane층, Chloroform층 분획물들이 산화적 스트레스와 도파민유도성 세포사멸에 대해 보호 효과가 있음이 나타났다.
또한, 파킨슨병 임상환자들에게서 발견되는 파킨슨병의 병리적 소견인 루이소체를 구성하고 있는 주요 단백질인 α-synuclein은 Dopamine에 의해 그 발현 양이 증가하게 되는데(도 7 참조) 이 과발현되는 α-synuclein 단백질을 월계수의 Hexane층, Chloroform층의 분획물들이 모두 감소되는(도 8 참조)것을 확인함으로 특히 파킨슨병에 주요 병인이 되는 이 기전에 보호 및 치료의 효과가 있음을 밝혔다.
또한, 흰쥐 뇌의 흑색질 부위에 선택적으로 도파민성 신경세포를 손실시킨 파킨슨병 동물실험 결과에서 월계수 Hexane층의 분획물은 Dopamine을 생성하게 하는 효소인 Tyrosine Hydroxylase의 감소된 양을 다시 증가시키는 것으로서 도파민성 신경세포의 손실에 치료 효과가 있음을 나타내었다.(도 9, 10 참조)
이상의 결과로부터 월계수의 분획물들은 산화적 스트레스와 도파민유도성 신경세포 사멸에 뇌의 선택적 도파민성 신경세포를 손실시킨 파킨슨병 동물모델에 기존의 약물보다 신경보호 및 치료의 효과 모두 좋음을 규명하였다.
상기의 결과로부터 월계수 잎의 추출물, 헥산층 분획물, 클로로포름층 분획물은 도파민 유도성 신경세포 사멸 및 파킨슨병을 유발한 흰쥐에 대한 신경세포 보호 효과가 있다.
하기에 본 발명에 따른 신경세포 사멸 및 파킨슨병 예방 및 보호를 위한 약학조성물의 제제예를 설명하나 본 발명은 이를 한정하고자 함이 아닌 단지 구체적으로 설명하고자 함이다.

약전 제제 총칙중 주사제의 제조방법에 따라 1 앰플당(2㎖) 하기의 성분 함량으로 제조한다.
월계수 분획물ㆍㆍㆍㆍ ㆍㆍㆍㆍㆍㆍㆍ 300 mg
주사용 멸균 증류수 ㆍㆍㆍㆍㆍㆍㆍㆍ 적량
pH 조절제 ㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍ 적량

약전 제제 총칙중 액제제의 제조방법에 따라 액제 100㎖당 하기의 성분 함량으로 제조한다.
월계수 분획물ㆍㆍㆍㆍ ㆍㆍㆍㆍㆍㆍㆍㆍ 1 g
이성화당ㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍ 10 g
만니톨 ㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍ 5 g
정제수 ㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍㆍ 적량
상기 조성비는 비교적 약제 제조에 적합한 성분을 바람직한 실시예로 혼합 조성하였지만 수요계층이나, 사용 용도 등에 따라서 그 배합비를 임의로 변형 실시할 수 있다.

Claims (4)
- 음건 건조하여 세절한 월계수 잎에 100% 주정을 가하고 55℃에서 48시간 동안 추출하여 에탄올 층을 추출하는 제 1 단계;제 1 단계를 통해 추출한 100% 에탄올 전체 추출물을 감압 농축하여 알코올 용매로 얻은 엑스를 분획하여 클로로포름 분획하는 제 2 단계;건조된 월계수 잎을 절단한 뒤 메탄올에 냉침한 후 여과지로 여과하는 과정을 3회 반복하여 헥산을 분획하는 제 3 단계; 및메탄올 추출물에 헥산을 가하여 진탕 반복 추출하고 분액 깔때기에서 분획하여 헥산층과 메탄올층을 분취한 후, 헥산층을 감압 농축하여 헥산층 분획물을 분획하는 제 4 단계를 포함하여 제조되는 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물.
- 제 1 항에 따른 월계수 에탄올 추출물, 월계수 헥산층 분획물 또는 월계수 클로로포름층 분획물을 유효성분으로 포함하고 통상의 약제학적으로 허용가능한 담체와 혼합하여 이루어지는 것을 특징으로 하는 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물.
- 제 2 항에 있어서,약제학적으로 사용되는 제제 형태가 산제, 과립제, 정제, 캅셀제, 액제, 주사제 중에서 선택된 어느 하나인 것을 특징으로 하는 특징으로 하는 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물.
- 제 1 항 내지 제 3항 중 어느 한 항에 있어서,상기 조성물은 월계수 잎 추출물을 1∼2,000㎎/㎏체중의 농도로 함유함을 특징으로 하는 뇌신경 세포보호 및 파킨슨병과 퇴행성 신경계 뇌질환의 예방 및 치료용 조성물.
