In order to evaluate the antioxidant efficacy of the genera, the scavenging ability of DPPH radical and the amount of GSH present in the cell were examined. The Blois method (using the reducibility of DPPH radicals) was used to measure the radical scavenging effect of the sample using DPPH (1,1-diphenyl-2-picrylhydrazyl). 0.1 mM DPPH dissolved in MeOH was added to the same amount of the raw material and reacted for 10 minutes. The DPPH radical scavenging ability of the sample was measured by measuring the absorbance at 565 nm using an absorber.

메탄올에 용해시킨 0.1mM DPPH 용액 1ml에 시료를 각 농도별로 희석하여 1ml씩 넣어주고 잘 혼합하여 주었다. 실온에서 10분간 반응시킨 후 565nm에서 흡광도를 측정하였다. [실험예 2. UVA 손상 방어능 평가]Experimental Example 2 Evaluation of UVA Damage Defense Capacity

피부 진피까지 침투하여 주된 피부 노화를 발생시키는 UVA를 섬유아세포에 직접적으로 조사함으로서 UVA에 의한 콜라겐 합성능이 저하됨을 확인하고 이에 대한 주안진의 콜라겐 합성 촉진능의 여부를 알아보았다. 콜라겐의 합성을 감소시켜줄 수 있는 양의 UVA(2J/㎠) 를 사람 유래의 섬유아세포 CCD-986sk에 처리하여 준 후 주안진을 농도별로 처리하여 주었다. 48시간 배양 후 ELISA 법을 이용하여 세포 배양 배지내에 존재하는 콜라겐의 발현 여부를 알아보았다. 사람 유래의 섬유아세포(CCD-986sk)를 배양접시의 바닥에 접종한 후, 페니린(100U/mL), 스트렙토마이신(100 μg/mL), 10% FBS (fetal bovine serum)를 함유하는 IMDM (Iscove's Modified Dulbecco's Medium) 배지를 넣고 37℃, 5% 이산 화탄소를 포함하는 배양기 내에서 배양하였다. 세포를 35mm 배양 접시에 2× 105/well의 농도가 되도록 접종한 후 24시간 추가 배양하였다. PBS 완충 용액으로 교체한 후 UVA 2J/m2을 조사하여 주었으며 주안진 0.05, 0.1% 농도가 되도록 처리하여 48시간 추가 배양 하였다. 세포 배양 배지를 이용하여 ELISA 법으로 콜라겐 양을 측정하였으며 단백질 양으로 보정 하여 주었다. [실험예 3. 피부 탄력도 측정]Experimental Example 3. Measurement of Skin Elasticity

시료의 피부 탄력도의 증가 정도를 확인하기 위하여 Cutometer MPA 580 (Courage Khazaka, Germany)을 이용하였다. Cutometer는 피부에 밀착시킨 probe 내부에 음압을 가해 피부가 잡아당겨지는 정도와 복원되는 정도의 차이를 표현하는 물리적인 지수를 이용하여 피부 탄력성을 측정하는 기기로 물리적 특성에 따라 R0에서 R9까지의 지수로 피부 탄력성을 표현하게 된다. 원료 농축액을 1%가 되도록 시험부위에 2주 동안에 걸쳐 하루에 2회 도포하고 마지막 시료 도포 24시간 후 Cutometer를 이용하여 피부 탄력성의 변화를 측정하였다. Cutometer MPA 580 (Courage Khazaka, Germany) was used to confirm the increase in skin elasticity of the sample. Cutometer measures the elasticity of skin using a physical index that expresses the difference between the degree of pulling and restoring the skin by applying a negative pressure inside the probe that is in close contact with the skin. 피부의 총 탄력도 (피부 재변형 능력) 를 나타내 주는 수치인 R2값을 시료 도포전과 도포 후 값을 비교하였다. R2값은 1에 가까울수록 음압을 이용하여 피부를 잡아당겼을 때 회복율이 좋음을 의미한다. 아래의 표 5와 같이 탄력 측정 결과 시료 처리 전과 시료 처리 후의 탄력도가 증가한 것을 확인할 수 있었다.R2 values, which represent the total elasticity of the skin (skin remodeling capacity), were compared before and after application of the sample. 4-2. pH에 대한 안정성4-2. stability to pH

진귀단 원료의 pH에 따른 안정성을 확인하고자 진귀단 원료를 이용하여 1, 3, 5% 수용액으로 희석하여 제조한 후 상온에서 pH 2.0에서 pH 10.0까지의 범위에서 측정하였다. 그 결과 표 7과 같이 pH에 대해서는 매우 안정한 것으로 판단된다.In order to confirm the stability according to the pH of the true raw material raw material was prepared by diluting with 1, 3, 5% aqueous solution using the raw material and measured in the range from pH 2.0 to pH 10.0 at room temperature.
