Claims (6)
- (ⅰ) 형질전환체로부터 핵산 추출물을 수득하는 공정; 및,(ⅱ) 상기 핵산 추출물에 활성탄을 가하고, 교반한 다음, 이를 원심분리하여 얻은 상층액으로 부터 플라스미드 DNA를 수득하는 공정을 포함하는, 고순도 플라스미드 DNA를 분리정제하는 방법.
- 제 1항에 있어서,형질전환체는 분리정제하고자 하는 플라스미드 DNA에 의하여 숙주세포가 형질전환된 것인 것을 특징으로 하는고순도 플라스미드 DNA를 분리정제하는 방법.
- 제 2항에 있어서,숙주세포는 대장균인 것을 특징으로 하는고순도 플라스미드 DNA를 분리정제하는 방법.
- 제 1항에 있어서,핵산 추출물은 알칼리 용해(alkaline lysis) 방법에 의하여 수득되는 것을 특징으로 하는고순도 플라스미드 DNA를 분리정제하는 방법.
- 제 1항에 있어서,활성탄은 분말형태 또는 증류수에 현탁된 형태로 핵산 추출물에 가하는 것을 특징으로 하는고순도 플라스미드 DNA를 분리정제하는 방법.
- 제 1항에 있어서,핵산 추출물에 활성탄을 가함으로써, 500bp 이하의 핵산단편만을 제거하는 것을 특징으로 하는고순도 플라스미드 DNA를 분리정제하는 방법.
