Superoxide dismutase(SOD)와 total antioxidant status(TAS) 시험 키트(test kits)는 RANDOX(영국)에서 구입하였다.Superoxide dismutase (SOD) and total antioxidant status (TAS) test kits were purchased from RANDOX (UK).

MDA(malondialdehyde)를 측정하기 위하여 TCA(15ml, 100%), 0.375g TBA, 25ml HCl(1N) 및 40mg BHT(butylated hydroxytoluene)/에탄올(ethanol)를 혼합하고, 100ml이 되게 재증류수(redistilled water)를 넣어 TBA 용액을 만들었다. 간과 뇌의 10% PBS 균질액을 1ml TBA 용액과 섞고, 0.5ml 시료와 50mM BHT 15㎕를 100℃에서 15분간 반응시키고 얼음물로 냉각시켰다. 3000rpm에서 30분간 원심분리하고 535nm에서 흡광도를 측정하여 말론다이알데히드(malondialdehyde)를 정량하였다. 피는 그대로 사용하였다. 모든 군에서 시각적 이상이 발견되지 않았고, 정상적 체중증가를 보였다. 혈액의 생화학적 지표로 malondialdehyde(MDA), superoxide dismutase(SOD) 및 total antioxidant status(TAS)가 측정된 결과를 표 1에 나타내었다. Aug 21 (2007) Temporal Gene Expression Profile in Hippocampus of Mice Treated with D -galactose). 하수오 추출물와 꾸지뽕잎 추출물이 혼합된 혼합물의 섭취가 5% D-galactose로 유도된 혈액 생화학 지표를 개선시켰다. 하수오 추출물군에서 SOD가 정상 대조군과 같은 수준으로 개선되었으며, 꾸지뽕잎 추출물 군에서는 TAS가 정상 대조군과 같은 수준으로 감소했다. 특히 중량 기준으로 50%의 하수오 추출물과 50%의 꾸지뽕잎 추출물이 배합된 실시예군에서는 MDA, SOD 및 TAS의 모든 측정치가 정상 대조군과 같은 수준으로 개선되어 두 추출물의 배합이 산화적 스트레스에 의한 생체의 노화를 효과적으로 억제시킬 수 있음을 보여주고 있다.Aging controls treated with 5% D-galactose increased serum MDA and TAS and decreased SOD compared to normal controls ( P < 0.05). 표 2는 하수오 추출물과 꾸지뽕잎 추출물의 배합비가 다른 실시예 2, 실시예 3, 실시예 4의 노화억제 배합물에 대한 생쥐 뇌 조직의 MDA와 SOD에 미치는 영향을 조사한 것이다. 노화 대조군에서 정상 대조군에 비하여 MDA가 증가하고 SOD가 감소했다 (P<0.05). 실시예 3의 노화억제 배합물(하수오 추출물과 꾸지뽕잎 추출물의 배합비가 60 : 40)일 때 노화억제 활성이 가장 우수하여 MDA와 SOD 모두 정상 대조군과 같은 수준으로 개선되었다. 실시예 2의 노화억제 배합물 및 실시예 4의 노화억제 배합물에서도 MDA와 SOD 수준이 개선되었으나 통계적 유의차는 없었다.
