Abstract
(1) 서열번호 1 에 기재된 아미노산 배열을 갖는 폴리펩티드, 및 (2) 서열번호 1 에 기재된 아미노산 배열에 있어서, 3번째의 아미노산이 다른 아미노산으로 치환된 아미노산 배열을 갖고, 또한 인슐린 모양 성장 인자 (1)(IGF-1) 활성을 갖는 폴리펩티드로 이루어지는 군에서 선택되는 폴리펩티드; 상기 폴리펩티드를 코드하는 DNA; 상기 DNA 를 함유하는 벡터; 상기 벡터를 함유하는 숙주 세포, 그리고 상기 폴리펩티드를 이용하여 재조합 단백질을 제조하는 방법.
Description
본 발명은 신규한 햄스터 IGF-1 폴리펩티드에 관한 것이다. 본 발명은 또한, 상기 신규한 IGF-1 폴리펩티드를 이용하여, 효율적이고 저렴하게 원하는 재조합 단백질을 제조하는 방법에 관한 것이다.
유전자 재조합 기술을 이용하여, 의약으로서 유용한 단백질을 생산할 때에, 동물 세포를 이용하면, 세균 세포가 행할 수 없는 복잡한 번역 후 수식이나 폴딩이 가능해지기 때문에, 동물 세포는 재조합 단백질 생산을 위한 숙주 세포로서 다용되고 있다.
종래, 동물 세포를 배양하여 그 동물 세포가 생성하는 천연형 단백질을 얻고자 하는 경우, 혹은 원하는 단백질을 코드하는 유전자를 도입한 동물 세포를 배양하여 원하는 단백질 등을 제조하는 경우에는, 염류, 당류, 아미노산류, 및 비타민류 등의 기초 영양물 외에, 그 동물 세포의 증식을 위하여, 통상 포유 동물 유래의 추출물, 구체적으로는 우태아 혈청 등의 혈청을 5∼20% 의 범위에서 배지에 첨가해야만 했다. 그러나, 1) 이러한 포유 동물 유래의 혈청은 고가인 점, 2) 품질에 로트간 차이가 있기 때문에 안정된 증식이 얻어지지 않는 점, 3) 바이러스 또는 미 코플라즈마 오염될 가능성이 있는 점, 및 4) 배양 배지로부터의 세포 산물인 원하는 단백질의 단리, 정제를 복잡화하는 점 등의 결점이 있었다. 따라서, 혈청을 함유하지 않는 배지에서 동물 세포를 배양하여, 저렴하게 재조합 단백질을 생산할 수 있는 기술이 요구되고 있었다.
단백질 제조를 무혈청 배지에서 행하기 위해서는, 혈청에 함유되는 증식 인자를 첨가할 필요가 있고, 예를 들어 CHO 세포의 배양시에는 통상 인간 ICF-1 (인슐린 모양 성장 인자 1: insulin-like growth factor 1) 을 첨가함으로써, 세포 증식 및 생존율을 개선하고, 장기 배양 혹은 대량 배양을 가능하게 하고 있다.
IGF-1 은 인슐린과 유사한 활성을 갖는 폴리펩티드로서 발견된 70 아미노산의 단쇄 폴리펩티드이며, 프로인슐린과 유사한 구조를 갖고 있다. IGF-1 은 세포 증식 작용과 함께 인슐린 모양의 대사 활성을 가지며, 개체의 성장에 가장 중요한 성장 인자의 하나이다. IGF-1 은 뇌하수체로부터 분비되는 성장 호르몬 (Growth Hormone; GH) 의 작용을 받아 간장에서 생성되어, 원격의 표적인 골조직의 성장을 촉진함과 함께, 근육이나 지방 세포에 있어서 대사 활성을 나타내는 것으로 생각되고 있다.
최근, 상기 기술한 바와 같은 내분비적 작용에 더하여, 여러 가지 세포 배양계에 있어서 IGF-1 이 생성되고, 또한 IGF-1 이 이들 세포에 대하여 증식 작용뿐만 아니라 여러 가지 생리 활성을 갖는 것이 보고되어 있다. 또한, IGF-1 이 갖는 세포 증식 촉진 작용에 주목하여, 재조합 단백질을 생산하는 동물 세포 배양계에 인간 IGF-1 등의 증식 촉진 작용을 갖는 폴리펩티드를 코드하는 DNA 를 도입하는 재조합 단백질의 제조 방법이 제안되어 있다 (US 2002/0102650 Al).
IGF-1 과 같은 각종 증식 인자는 인간 유래의 재조합체가 사용되지만, 매우 고가이므로, 제조 비용의 관점에서, IGF-1 의 첨가량을 감소시킬 것이 요망되고 있었다.
발명의 개시
발명이 해결하고자 하는 과제
본 발명은 제조 비용이 낮고, 또한 강력한 세포 증식 촉진 작용을 갖는 신규한 IGF-1 폴리펩티드를 얻는 것을 목적으로 한다. 본 발명은 또한 상기 신규한 IGF-1 폴리펩티드를 코드하는 DNA 를 이용하여, 효율적이고 저렴하게 재조합 단백질을 생산할 수 있는 숙주 동물 세포를 제작하는 것을 목적으로 한다. 본 발명은 또한, 이러한 숙주 동물 세포를 이용하여 재조합 단백질을 제조하는 방법의 제공을 목적으로 한다.
과제를 해결하기 위한 수단
상기 목적을 달성하기 위하여 예의 연구한 결과, 본 발명자들은 햄스터 유래의 신규한 인슐린 모양 성장 인자 (1)(IGF-1) 폴리펩티드를 발견하고, 그 신규한 IGF-1 폴리펩티드가 동물 세포의 배양에 있어서 현저한 세포 증식 촉진 작용을 갖는 것을 발견하여 본 발명을 완성하였다.
즉, 본 발명은 이하의 것을 제공한다.
1. (1) 서열번호 1 에 기재된 아미노산 배열을 갖는 폴리펩티드, 및
(2) 서열번호 1 에 기재된 아미노산 배열에 있어서, 3번째의 아미노산이 다른 아미노산으로 치환된 아미노산 배열을 갖고, 또한 인슐린 모양 성장 인자 (1)(IGF-1) 활성을 갖는 폴리펩티드로 이루어지는 군에서 선택되는 폴리펩티드.
2. 3번째의 아미노산이 세린으로 치환되어 있는 상기 1 에 기재된 폴리펩티드.
3. 3번째의 아미노산이 리신으로 치환되어 있는 상기 1 에 기재된 폴리펩티드.
4. 상기 1 에 기재된 폴리펩티드를 코드하는 DNA.
5. 상기 4 에 기재된 DNA 를 함유하는 벡터.
6. 상기 5 에 기재된 벡터를 함유하는 숙주 세포.
7. CHO 세포인 상기 6 에 기재된 숙주 세포.
8. 상기 6 또는 7 에 기재된 숙주 세포를 이용하여, 상기 1 에 기재된 폴리펩티드를 제조하는 방법.
9. 상기 1 에 기재된 폴리펩티드와 결합하는 항체.

발명을 실시하기
위한 최선의 형태
본 발명의 폴리펩티드는 도 1 및 서열번호 1 에 나타내는 아미노산 배열을 갖는 분비형 성숙 햄스터 IGF-1 이 바람직하지만, 전구형 햄스터 IGF-1 이어도 된다.
또한, 본 발명의 폴리펩티드는 서열번호 1 에 나타내는 햄스터 IGF-1 의 아미노산 배열에 있어서, 3번째의 아미노산이 다른 아미노산으로 치환된 아미노산 배열을 갖고, 또한 인슐린 모양 성장 인자 (1)(IGF-1) 활성을 갖는 폴리펩티드도 포함한다. 치환되는 아미노산은 특별히 한정되지 않고, 어떠한 아미노산으로 치환되어도 되지만, 세린, 리신, 또는 아스파르트산으로 치환되는 것이 바람직하고, 특히 세린 또는 리신으로 치환되는 것이 바람직하다.
본 발명에 있어서, 인슐린 모양 성장 인자 (1)(IGF-1) 활성을 갖는다는 용어는 동물 세포에 대한 증식 촉진 활성 및/또는 생존율 상승 활성을 갖는 것을 말한다.
본 발명에서는, 「폴리펩티드」를 「단백질」과 동일한 의미로 사용하는 경우가 있다.
본 발명의 폴리펩티드는 당업자에게 공지된 방법에 의해, 재조합 폴리펩티드로서, 또한 천연 폴리펩티드로서 조제하는 것이 가능하다. 재조합 폴리펩티드이면, 본 발명의 폴리펩티드를 코드하는 DNA 를 적당한 발현 벡터에 삽입하고, 이것을 적당한 숙주 세포에 도입하여 얻은 형질 전환체를 회수하고, 추출물을 얻은 후, 이온 교환, 역상, 겔 여과 등의 크로마토그래피, 혹은 본 발명의 폴리펩티드에 대한 항체를 칼럼에 고정시킨 어피니티 크로마토그래피를 실시함으로써, 또는 추가로 이들 칼럼을 복수 조합함으로써 정제하여, 조제하는 것이 가능하다.
또한, 본 발명의 폴리펩티드를 글루타티온 S트랜스페라제 단백질과의 융합 단백질로서, 혹은 히스티딘을 복수 부가시킨 재조합 단백질로서 숙주 세포 (예를 들어, 동물 세포나 대장균 등) 내에서 발현시킨 경우에는, 발현시킨 재조합 단백질은 글루타티온 칼럼 혹은 니켈 칼럼을 이용하여 정제할 수 있다.
융합 단백질의 정제 후, 필요에 따라 융합 단백질 중 목적의 폴리펩티드 이외의 영역을 트롬빈 또는 팩터-Xa 등에 의해 절단하여, 제거하는 것도 가능하다.
천연 폴리펩티드이면, 당업자에게 주지된 방법, 예를 들어 본 발명의 폴리펩티드를 발현하고 있는 조직이나 세포의 추출물에 대하여, 후술하는 햄스터 IGF-1 폴리펩티드에 결합하는 항체가 결합한 어피니티 칼럼을 작용시켜 정제함으로써 단리할 수 있다. 항체는 폴리클로날 항체이어도 되고, 모노클로날 항체이어도 된다.
또한, 본 발명은 본 발명의 햄스터 ICF-1 을 코드하는 DNA 를 제공한다. 본 발명의 DNA 는 상기 기술한 바와 같은 본 발명의 폴리펩티드의 in vivo 나 in vitro 에 있어서의 생산에 이용하는 것이 가능하다. 본 발명의 DNA 는 본 발명의 폴리펩티드를 코드할 수 있는 것이면 어떠한 형태이어도 된다. 즉, mRNA 로부터 합성된 cDNA 인지, 게놈 DNA 인지, 화학 합성 DNA 인지 등을 불문한다. 또한, 본 발명의 폴리펩티드를 코드할 수 있는 한, 유전 암호의 축중에 기초하는 임의의 염기 서열을 갖는 DNA 가 포함된다.
본 발명의 DNA 는 당업자에게 공지된 방법에 의해 조제할 수 있다. 예를 들어, 본 발명의 폴리펩티드를 발현하고 있는 세포로부터 cDNA 라이브러리를 제작하고, 본 발명의 DNA 의 배열 (예를 들어, 서열번호 2) 의 일부를 프로브로 하여 하이브리다이제이션을 실시함으로써 조제할 수 있다. cDNA 라이브러리는, 예를 들어 Sambrook, J. et al., Molecular Cloning, Cold Spring Harbor Laboratory Press (1989) 에 기재된 방법에 의해 조제해도 되고, 시판되는 DNA 라이브러리를 이용해도 된다. 또한, 본 발명의 폴리펩티드를 발현하고 있는 세포로부터 RNA 를 조제하고, 본 발명의 DNA 의 배열에 기초하여 올리고 DNA 를 합성하고, 이것을 프라이머로서 이용하여 PCR 반응을 실시하여, 본 발명의 폴리펩티드를 코드하는 cDNA 를 증폭시킴으로써 조제하는 것도 가능하다.
구체적으로는, 다음과 같이 하면 된다. 우선, 본 발명의 폴리펩티드를 발현하는 세포, 조직, 장기 등으로부터 mRNA 를 단리한다. mRNA 의 단리는 공지된 방법, 예를 들어 구아니딘 초원심법 (Chirgwin, J. M. et al., Biochemistry (1979) 18, 5294-5299), AGPC 법 (Chomczynski, P. and Sacchi, N., Anal. Biochem.(1987) 162, 156-159) 등에 의해 전체 RNA 를 조제하고, mRNA Purification Kit (Pharmacia) 등을 사용하여 전체 RNA 로부터 mRNA 를 정제한다. 또한, Quick Prep mRNA Purification Kit (Pharmacia) 를 이용함으로써 mRNA 를 직접 조제할 수도 있다.
얻어진 mRNA 로부터 역전사 효소를 이용하여 cDNA 를 합성한다. cDNA 의 합성은 AMV Reverse Transcriptase First-strand cDNA Synthesis Kit (생화학공업) 등을 이용하여 실시할 수도 있다. 또한, 본 명세서에 기재된 프라이머 등을 이용하여, 5'-Ampli FINDER RACE Kit (Clontech 제조) 및 폴리머라제 연쇄 반응 (polymerase chain reaction; PCR) 을 이용한 5'-RACE 법 (Frohman, M. A. et al., Proc. Natl. Acad. Sci. U.S.A. (1988) 85, 8998-9002; Belyavsky, A. et al., Nucleic Acids Res. (1989) 17, 2919-2932) 에 따라 cDNA 의 합성 및 증폭을 실시할 수 있다.
후술하는 본 발명의 실시예에 있어서는, 햄스터 IGF-1 폴리펩티드를 코드하는 cDNA 를 PCR 클로닝에 의해 얻었다. PCR 프라이머의 설계는 이미 알려진 래트/마우스/인간 IGF-1 사이에서 유전자 배열이 보존되어 있는 영역을 이용하여 행하였다.
얻어진 PCR 산물로부터 목적으로 하는 DNA 단편을 조제하여, 벡터 DNA 와 연결한다. 또한, 이로부터 재조합 벡터를 제작하고, 대장균 등에 도입하고 콜로니를 선택하여 원하는 재조합 벡터를 조제한다. 목적으로 하는 DNA 의 염기 서열은 공지된 방법, 예를 들어 디데옥시뉴클레오티드 체인 터미네이션법에 의해 확인할 수 있다.
또한, 얻어진 cDNA 를 프로브로 하여 게놈 DNA 라이브러리를 스크리닝함으로써 게놈 DNA 를 단리할 수 있다.
또한, 본 발명의 DNA 에 있어서는, 발현에 사용하는 숙주의 코돈 사용 빈도를 고려하여, 보다 발현 효율이 높은 염기 서열을 설계할 수 있다 (Grantham, R. et al., Nucleic Acids Research (1981) 9, p43-74). 또한, 본 발명의 DNA 는 시판되는 키트나 공지된 방법에 의해 개변할 수 있다. 개변으로서는, 예를 들어 제한 효소에 의한 소화, 합성 올리고뉴클레오티드나 적당한 DNA 프래그먼트의 삽입, 링커의 부가, 개시 코돈 (ATG) 및/또는 종지 코돈 (TAA, TGA 또는 TAG) 의 삽입 등을 들 수 있다.
또한, 본 발명은 본 발명의 DNA 가 삽입된 벡터를 제공한다. 본 발명의 벡터로서는, 숙주 세포내에 있어서 본 발명의 DNA 를 유지시키기 위해서나, 또한 본 발명의 폴리펩티드를 발현시키기 위하여 유용하다.
벡터로서는, 예를 들어 대장균을 숙주로 하는 경우에는, 벡터를 대장균 (예를 들어, JM109, DH5α, DH10α, HB101, XL1-blue) 등에서 대량으로 증폭시켜 대량 조제하기 위하여, 대장균에서 증폭되기 위한 「ori」를 갖고, 또한 형질 전환된 대장균의 선발 유전자 (예를 들어, 어떠한 약제 (암피실린이나 테트라사이클린, 카나마이신, 클로람페니콜) 에 의해 판별할 수 있는 약제 내성 유전자) 를 가지면 특별히 제한은 없다. 벡터의 예로서는, M13 계 벡터, pUC 계 벡터, pBR322, pBluescript, pCR-Script 등을 들 수 있다. 또한, cDNA 의 서브클로닝, 절취를 목적으로 했을 경우, 상기 벡터 외에, 예를 들어 pGEM-T, pDIRECT, pT7 등을 들 수 있다. 본 발명의 폴리펩티드를 생산하는 목적에 있어서 벡터를 사용하는 경우에는, 특히 발현 벡터가 유용하다. 발현 벡터로서는, 예를 들어 대장균에서의 발현을 목적으로 했을 경우는, 벡터가 대장균에서 증폭되는 것과 같은 상기 특징을 갖는 것 외에, 숙주를 JM109, DH5α, DH10α, HB101, XL1-blue 등의 대장균으로 한 경우에는, 대장균에서 효율적으로 발현할 수 있는 프로모터, 예를 들어 lacZ 프로 모터 (Ward 외, Nature (1989) 341, 544-546; FASEB J. (1992) 6, 2422-2427), araB 프로모터 (Better 외, Science (1988) 240, 1041-1043), 또는 T7 프로모터 등을 갖고 있는 것이 불가결하다. 이러한 벡터로서는, 상기 벡터 외에 pGEX-5X-1 (Pharmacia 사 제조), 「QIAexpress system」(Qiagen 사 제조), pEGFP, 또는 pET (이 경우, 숙주는 T7 RNA 폴리머라제를 발현하고 있는 BL21 이 바람직하다) 등을 들 수 있다.
또한, 벡터에는 폴리펩티드 분비를 위한 시그널 배열이 포함되어 있어도 된다. 폴리펩티드 분비를 위한 시그널 배열로서는, 대장균의 페리플라즘에 생성 시키는 경우, pelB 시그널 배열 (Lei, S. P. et al J. Bacteriol. (1987) 169, 4379) 을 사용하는 것이 가능하다. 숙주 세포로의 벡터의 도입은 예를 들어 염화 칼슘법, 일렉트로포레이션법을 이용하여 실시할 수 있다.
대장균 이외에도, 예를 들어 본 발명의 폴리펩티드를 제조하기 위하여 사용되는 벡터로서는, 포유 동물 유래의 발현 벡터 (예를 들어, pcDNA3 (Invitrogen 사 제조) 나, pEF-BOS (Nucleic Acids. Res. 1990, 18, p5322), pEF, pCDM8), 곤충 세포 유래의 발현 벡터 (예를 들어 「BAC-to-BACTM baculovairus expression system」(Invitrogen 사 제조, pBacPAK8), 식물 유래의 발현 벡터 (예를 들어 pMH1, pMH2), 동물 바이러스 유래의 발현 벡터 (예를 들어, pHSV, pMV, pAdexLcw), 레트로 바이러스 유래의 발현 벡터 (예를 들어, pZIpneo), 효모 유래의 발현 벡터 (예를 들어, 「Pichia Expression Kit」(Invitrogen 사 제조), pNV11, SP-Q01), 고초 균 유래의 발현 벡터 (예를 들어, pPL608, pKTH50) 등을 들 수 있다.
CHO 세포, COS 세포, NIH3T3 세포 등의 동물 세포에서의 발현을 목적으로 했을 경우에는, 세포내에서 발현시키기 위하여 필요한 프로모터, 예를 들어 SV40 프로모터 (Mulligan 외, Nature (1979) 277, 108), MMLV-LTR 프로모터, EF1α 프로모터 (Mizushima 외, Nucleic Acids Res. (1990) 18, 5322), RSV 프로모터, CMV 프로모터 등을 갖고 있는 것이 불가결하고, 세포로의 형질 전환을 선발하기 위한 유전자 (예를 들어, 약제 (네오마이신, G418, 하이그로마이신, 퓨로마이신 등) 에 의해 판별할 수 있는 약제 내성 유전자) 를 가지면 더욱 바람직하다. 이러한 특성을 갖는 벡터로서는, 예를 들어 pMAM, pDR2, pBK-RSV, pBK-CMV, pOPRSV, pOP13 등을 들 수 있다.
또한, 유전자를 안정적으로 발현시키고, 또한 세포내에서의 유전자의 카피수의 증폭을 목적으로 하는 경우에는, 핵산 합성 경로를 결손한 CHO 세포에 그것을 상보하는 DHFR 유전자를 갖는 벡터 (예를 들어, pCHOI 등) 를 도입하고, 메토트렉사트 (MTX) 에 의해 증폭시키는 방법을 들 수 있으며, 또한 유전자의 일과성의 발현을 목적으로 하는 경우에는, SV40 T 항원을 발현하는 유전자를 염색체 상에 갖는 COS 세포를 이용하여 SV40 의 복제 기점을 갖는 벡터 (pCD 등) 로 형질 전환하는 방법을 들 수 있다. 복제 개시점으로서는, 또한 폴리오머 바이러스, 아데노 바이러스, 소 파필로마 바이러스 (BPV) 등의 유래의 배열을 이용할 수도 있다. 또한, 숙주 세포계에서 유전자 카피수 증폭을 위하여, 발현 벡터는 선택 마커로서, 아미노글리코시드트랜스페라제 (APH) 유전자, 티미딘퀴나아제 (TK) 유전자, 대장균 크산틴구아닌포스포리보실트랜스페라제 (Ecogpt) 유전자, 디히드로 엽산 환원 효소 (dhfr) 유전자 등을 포함할 수 있다.
한편, 동물의 생체내에서 본 발명의 DNA 를 발현시키는 방법으로서는, 본 발명의 DNA 를 적당한 벡터에 삽입하고, 레트로 바이러스법, 리포솜법, 카티오닉 리포솜법, 아데노 바이러스법 등에 의해 생체내에 도입하는 방법 등을 들 수 있다. 사용되는 벡터로서는, 예를 들어 아데노 바이러스 벡터 (예를 들어 pAdexlcw) 나 레트로 바이러스 벡터 (예를 들어 pZIPneo) 등을 들 수 있지만, 이들에 제한되지 않는다. 벡터로의 본 발명의 DNA 의 삽입 등의 일반적인 유전자 조작은, 통상적인 방법에 따라 실시하는 것이 가능하다 (Molecular Cloning, 5.61-5.63). 생체내로의 투여는 ex vivo 법이어도 되고, in vivo 법이어도 된다.
또한, 본 발명은 본 발명의 벡터가 도입된 숙주 세포를 제공한다. 본 발명의 벡터가 도입되는 숙주 세포로서는 특별히 제한은 없고, 예를 들어 대장균이나 CHO 세포 등의 여러 가지의 동물 세포 등을 이용하는 것이 가능하다. 본 발명의 숙주 세포는, 예를 들어 본 발명의 폴리펩티드의 제조나 발현을 위한 생성계로서 사용할 수 있다. 폴리펩티드 제조를 위한 생성계는 in vitro 및 in vivo 의 생성계가 있다. in vitro 의 생성계로서는, 진핵 세포를 사용하는 생성계나 원핵 세포를 사용하는 생성계를 들 수 있다.
진핵 세포를 사용하는 경우, 예를 들어 동물 세포, 식물 세포, 진균 세포를 숙주에 이용할 수 있다. 동물 세포로서는, 포유류 세포, 예를 들어 CHO (J. Exp. Med. (1995) 108, 945), COS, 3T3, 미엘로마, BHK (baby hamster kidney), HeLa, Vero, 양서류 세포, 예를 들어 아프리카 발톱개구리 난모 세포 (Valle, et al., Nature (1981) 291, 358-340), 혹은 곤충 세포, 예를 들어 Sf9, Sf21, Tn5 가 알려져 있다. CHO 세포로서는, 특히 DHFR 유전자를 결손한 CHO 세포인 CHO dhfr- (Proc. Natl. Acad. Sci. USA (1980) 77, 4216-4220) 이나 CHO K-1 (Proc. Natl. Acad. Sci. USA (1968) 60, 1275) 을 바람직하게 사용할 수 있다. 동물 세포에 있어서, 대량 발현을 목적으로 하는 경우에는 특히 CHO 세포가 바람직하다. 숙주 세포로의 벡터의 도입은, 예를 들어 인산 칼슘법, DEAE 덱스트란법, 카티오닉 리보솜 DOTAP (베링거만하임사 제조) 를 이용한 방법, 일렉트로포레이션법, 리포펙션 등의 방법으로 실시하는 것이 가능하다.
식물 세포로서는, 예를 들어 니코티아나·타바컴 (Nicotiana tabacum) 유래의 세포가 폴리펩티드 생산계로서 알려져 있으며, 이것을 칼루스 배양하면 된다. 또한 물풀에 의한 배양 (Biolex 사) 도 유효하다. 진균 세포로서는, 효모, 예를 들어 사카로미세스 (Saccharomyces) 속, 예를 들어 사카로미세스 세레비시에 (Saccharomyces cerevisiae), 사상균, 예를 들어 아스페르길루스 (Aspergillus) 속, 예를 들어 아스페르길루스 니거 (Aspergillus niger) 가 알려져 있다.
원핵 세포를 사용하는 경우, 세균 세포를 이용하는 생성계가 있다. 세균 세포로서는, 대장균 (E. coli), 예를 들어 JM109, DH5α, DH10α, HB101 등을 들 수 있으며, 기타 고초균이 알려져 있다.
이들 세포를 목적으로 하는 DNA 에 의해 형질 전환하고, 형질 전환된 세포를 in vitro 로 배양함으로써 폴리펩티드를 얻을 수 있다. 배양은 공지된 방법에 따라 실시할 수 있다. 예를 들어, 동물 세포의 배양액으로서, 예를 들어 DMEM, MEM, RPMI1640, IMDM 을 사용할 수 있다. 그 때, 우태아 혈청 (FCS) 등의 혈청 보액을 병용할 수도 있고, 무혈청 배양해도 된다. 배양시의 pH 는 약 6∼8 인 것이 바람직하다. 배양은 통상, 약 30∼40℃ 에서 약 15∼240 시간 실시하고, 필요에 따라 배지의 교환, 환기, 교반을 추가한다.
한편, in vivo 로 폴리펩티드를 생성시키는 계로서는, 예를 들어 동물을 사용하는 생성계나 식물을 사용하는 생성계를 들 수 있다. 이들 동물 또는 식물에 목적으로 하는 DNA 를 도입하고, 동물 또는 식물의 체내에서 폴리펩티드를 생성시켜 회수한다. 본 발명에 있어서의 「숙주」란, 이들 동물, 식물을 포함한다.
동물을 사용하는 경우, 포유류 동물, 곤충을 이용하는 생성계가 있다. 포유류 동물로서는, 염소, 돼지, 양, 마우스, 소를 이용할 수 있다 (Vicki Glaser, SPECTRUM Biotechnology Applications, 1993). 또한, 포유류 동물을 이용하는 경우, 트랜스제닉 동물을 이용할 수 있다.
예를 들어, 목적으로 하는 DNA 를 염소 β 카세인과 같은 유즙 중에 고유하게 생성되는 폴리펩티드를 코드하는 유전자와의 융합 유전자로서 조제한다. 다음에, 이 융합 유전자를 포함하는 DNA 단편을 염소의 배아에 주입하고, 이 배아를 암컷 염소에 이식한다. 배아를 수용한 염소에서 태어나는 트랜스제닉 염소 또는 그 자손이 생성하는 유즙으로부터 목적의 폴리펩티드를 얻을 수 있다. 트랜스제닉 염소로부터 생성되는 폴리펩티드를 함유하는 유즙량을 증가시키기 위하여, 적절히 호르몬을 트랜스제닉 염소에 사용해도 된다 (Ebert, K. M. et al., Bio/Technology (1994) 12,699-702).
또한, 곤충으로서는 예를 들어 누에를 이용할 수 있다. 누에를 이용하는 경우, 목적의 폴리펩티드를 코드하는 DNA 를 삽입한 바큘로 바이러스를 누에에 감염시킴으로써, 이 누에의 체액으로부터 목적의 폴리펩티드를 얻을 수 있다 (Susumu, M. et al., Nature (1985) 315, 592-594).
또한, 식물을 사용하는 경우, 예를 들어 담배를 이용할 수 있다. 담배를 이용하는 경우, 목적으로 하는 폴리펩티드를 코드하는 DNA 를 식물 발현용 벡터, 예를 들어 pMON530 에 삽입하고, 이 벡터를 아그로박테리움 튜머파시엔스 (Agrobacterium tumefaciens) 와 같은 박테리아에 도입한다. 이 박테리아를 담배, 예를 들어 니코티아나 타바컴 (Nicotiana tabacum) 에 감염시켜, 본 담배의 잎으로부터 원하는 폴리펩티드를 얻을 수 있다 (Julian K. -C. Ma et al., Eur. J. Immunol. (1994) 24, 131-138).
이로써 얻어진 본 발명의 폴리펩티드는 숙주 세포내 또는 세포외 (배지 등) 로부터 단리하여, 실질적으로 순수하고 균일한 폴리펩티드로서 정제할 수 있다. 폴리펩티드의 분리, 정제는 통상의 폴리펩티드의 정제에서 사용되고 있는 분리, 정제 방법을 사용하면 되고, 전혀 한정되는 것이 아니다. 예를 들어, 크로마토그래피 칼럼, 필터, 한계외 여과, 염석, 용매 침전, 용매 추출, 증류, 면역 침강, SDS-폴리아크릴아미드겔 전기 영동, 등전점 전기영동법, 투석, 재결정 등을 적절히 선택, 조합하면 폴리펩티드를 분리, 정제할 수 있다.
크로마토그래피로서는, 예를 들어 어피니티 크로마토그래피, 이온 교환 크로 마토그래피, 소수성 크로마토그래피, 겔 여과, 역상 크로마토그래피, 흡착 크로마토그래피 등을 들 수 있다 (Strategies for Protein Purification and Characterization: A Laboratory Course Manual. Ed Daniel R. Marshak et al., Cold Spring Harbor Laboratory Press, 1996). 이들 크로마토그래피는 액상 크로마토그래피, 예를 들어 HPLC, FPLC 등의 액상 크로마토그래피를 이용하여 실시할 수 있다. 본 발명은 이들 정제 방법을 이용하여 고도로 정제된 폴리펩티드도 포함한다.
또한, 폴리펩티드를 정제 전 또는 정제 후에 적당한 폴리펩티드 수식 효소를 작용시킴으로써, 임의로 수식을 가하여 부분적으로 펩티드를 제거할 수도 있다. 폴리펩티드 수식 효소로서는, 예를 들어 트립신, 키모트립신, 리실 엔도펩티다아제, 프로틴 퀴나아제, 글루코시다아제 등이 사용된다.
또한, 본 발명은 본 발명의 폴리펩티드와 결합하는 항체를 제공한다. 본 발명의 항체의 형태에는 특별히 제한은 없고, 폴리클로날 항체 외에, 모노클로날 항체도 포함된다. 또한, 토끼 등의 면역 동물에 본 발명의 폴리펩티드를 면역시켜 얻은 항 혈청, 모든 클래스의 폴리클로날 항체 및 모노클로날 항체, 또한 인간 항체나 유전자 재조합에 의한 인간화 항체도 포함된다.
항체 취득의 감작 항원으로서 사용되는 본 발명의 폴리펩티드는 본 명세서에 개시되는 유전자 배열 또는 아미노산 배열을 이용하여 얻을 수 있다.
본 발명에 있어서, 감작 항원으로서 사용되는 폴리펩티드는 완전한 폴리펩티드이어도 되고, 또한 폴리펩티드의 부분 펩티드이어도 된다.
본 발명의 폴리펩티드 또는 그 단편을 코드하는 유전자를 공지된 발현 벡터계에 삽입하고, 그 벡터에 의해 본 명세서에서 설명한 숙주 세포를 형질 전환시켜, 그 숙주 세포 내외로부터 목적의 폴리펩티드 또는 그 단편을 공지된 방법으로 얻어, 이들을 감작 항원으로서 사용하면 된다. 또한, 폴리펩티드를 발현하는 세포 또는 그 용해물 혹은 화학적으로 합성한 본 발명의 폴리펩티드를 감작 항원으로서 사용해도 된다.
감작 항원으로 면역되는 포유 동물로서는, 특별히 한정되는 것은 아니지만, 세포 융합에 사용하는 친세포와의 적합성을 고려하여 선택하는 것이 바람직하고, 일반적으로는 설치목, 토끼목, 영장목의 동물이 사용된다.
설치목의 동물로서는, 예를 들어 마우스, 래트, 햄스터 등이 사용된다. 토끼목의 동물로서는, 예를 들어 토끼가 사용된다. 영장목의 동물로서는, 예를 들어 원숭이가 사용된다. 원숭이로서는, 협비하목(狹鼻下目)의 원숭이 (구(舊) 세계원숭이), 예를 들어 게 먹는 원숭이, 붉은털 원숭이, 망토비비, 침팬지 등이 사용된다.
감작 항원을 동물에 면역시키려면, 공지된 방법에 따라 행해진다. 일반적 방법으로서는, 감작 항원을 포유 동물의 복강내 또는 피하에 주사한다. 구체적으로는, 감작 항원을 PBS (Phosphate-Buffered Saline) 나 생리 식염수 등으로 적당량으로 희석, 현탁시킨 것에 대하여, 소망에 의해 통상의 아쥬반트, 예를 들어 프로인트 완전 아쥬반트를 적량 혼합하고, 유화 후, 포유 동물에 투여한다. 또한, 그 후, 프로인트 불완전 아쥬반트에 적량 혼합한 감작 항원을 4∼21일마다 몇 차례 투여하는 것이 바람직하다. 또한, 감작 항원 면역시에 적당한 담체를 사용할 수 있다. 이와 같이 면역시켜, 혈청 중에 원하는 항체 레벨이 상승하는 것을 통상적인 방법에 의해 확인한다.
여기서, 본 발명의 폴리펩티드에 대한 폴리클로날 항체를 얻으려면, 혈청 중의 원하는 항체 레벨이 상승한 것을 확인한 후, 항원을 감작한 포유 동물의 혈액을 채출한다. 이 혈액으로부터 공지된 방법에 의해 혈청을 분리한다. 폴리클로날 항체로서는, 폴리클로날 항체를 함유하는 혈청을 사용해도 되고, 필요에 따라 이 혈청으로부터 폴리클로날 항체를 함유하는 획분을 추가로 단리하여, 이것을 사용해도 된다. 예를 들어, 본 발명의 폴리펩티드를 커플링시킨 어피니티 칼럼을 이용하여, 본 발명의 폴리펩티드만을 인식하는 획분을 얻고, 추가로 이 획분을 프로틴 A 혹은 프로틴 G 칼럼을 이용하여 정제함으로써, 면역 글로블린 G 혹은 M 을 조제할 수 있다.
모노클로날 항체를 얻으려면, 상기 항원을 감작한 포유 동물의 혈청 중에 원하는 항체 레벨이 상승하는 것을 확인한 후에, 포유 동물로부터 면역 세포를 채출하여, 세포 융합을 실시하면 된다. 이 때, 세포 융합에 사용되는 바람직한 면역 세포로서 특히 지라 세포를 들 수 있다. 상기 면역 세포와 융합되는 타방의 친세포로서는, 바람직하게는 포유 동물의 미엘로마 세포, 보다 바람직하게는, 약제에 의한 융합 세포 선별을 위한 특성을 획득한 미엘로마 세포를 들 수 있다.
상기 면역 세포와 미엘로마 세포의 세포 융합은 기본적으로는 공지된 방법, 예를 들어 밀스테인 외의 방법 (Galfre, G. and Milstein, C., Methods Enzymol. (1981) 73, 3-46) 등에 준하여 실시할 수 있다.
세포 융합에 의해 얻어진 하이브리도마는 통상의 선택 배양액, 예를 들어 HAT 배양액 (히폭산틴, 아미노프테린 및 티미딘을 함유하는 배양액) 에서 배양함으로써 선택된다. 당해 HAT 배양액에서의 배양은, 목적으로 하는 하이브리도마 이외의 세포 (비융합 세포) 가 사멸하기에 충분한 시간, 통상, 수 일·수 주간 계속하여 실시한다. 다음에, 통상의 한계 희석법을 실시하여, 목적으로 하는 항체를 생성하는 하이브리도마의 스크리닝 및 클로닝을 실시한다.
또한, 인간 이외의 동물에 항원을 면역시켜 상기 하이브리도마를 얻는 것 외에, 인간 임파구, 예를 들어 EB 바이러스에 감염된 인간 임파구를 in vitro 로 폴리펩티드, 폴리펩티드 발현 세포 또는 그 용해물로 감작하고, 감작 임파구를 인간 유래의 영구 분열능을 갖는 미엘로마 세포, 예를 들어 U266 과 융합시켜, 폴리펩티드에 대한 결합 활성을 갖는 원하는 인간 항체를 생성하는 하이브리도마를 얻을 수도 있다 (일본 공개특허공보 소63-17688호).
다음에, 얻어진 하이브리도마를 마우스 복강내에 이식하고, 동일 마우스로부터 복수를 회수하여, 얻어진 모노클로날 항체를, 예를 들어 황산 암모늄 침전, 프로틴 A 칼럼, 프로틴 G 칼럼, DEAE 이온 교환 크로마토그래피, 본 발명의 폴리펩티드를 커플링한 어피니티 칼럼 등에 의해 정제함으로써 조제하는 것이 가능하다. 본 발명의 항체는, 본 발명의 폴리펩티드의 정제, 검출에 사용되는 것 외에, 본 발명의 폴리펩티드의 아고니스트나 안타고니스트의 후보가 된다. 또한, 이 항체를 본 발명의 폴리펩티드가 관여하는 질환의 항체 치료에 응용하는 것도 고려된 다. 얻어진 항체를 인체에 투여하는 목적 (항체 치료) 으로 사용하는 경우에는, 면역 원성을 저하시키기 때문에, 인간 항체나 인간형 항체가 바람직하다.
예를 들어, 인간 항체 유전자의 레퍼토리를 갖는 트랜스제닉 동물에 항원이 되는 폴리펩티드, 폴리펩티드 발현 세포 또는 그 용해물을 면역시켜 항체 생성 세포를 취득하고, 이것을 미엘로마 세포와 융합시킨 하이브리도마를 이용하여 폴리펩티드에 대한 인간 항체를 취득할 수 있다 (국제공개공보 WO92-03918, WO93-2227, WO94-02602, WO94-25585, WO96-33735 및 WO96-34096 참조).
하이브리도마를 이용하여 항체를 생성하는 것 외에, 항체를 생성하는 감작 임파구 등의 면역 세포를 암 유전자 (oncogene) 에 의해 불사화시킨 세포를 이용해도 된다.
이와 같이 얻어진 모노클로날 항체는 또한, 유전자 재조합 기술을 이용하여 생성시킨 재조합형 항체로서 얻을 수 있다 (예를 들어, Borrebaeck, C.A.K. and Larrick, J. W., THERAPEUTIC MONOCLONAL ANTIBODIES, Published in the United Kingdom by MACMILLAN PUBLISHERS LTD, 1990 참조). 재조합형 항체는 그것을 코드하는 DNA 를 하이브리도마 또는 항체를 생성하는 감작 임파구 등의 면역 세포로부터 클로닝하여, 적당한 벡터에 삽입하고, 이것을 숙주에 도입하여 생성시킨다. 본 발명은 이 재조합형 항체를 포함한다.

항체 수식물로서 폴리에틸렌글리콜 (PEG) 등의 각종 분자와 결합한 항체를 사용할 수도 있다. 본 발명의 「항체」 에는 이들의 항체 수식물도 포함된다. 이러한 항체 수식물을 얻으려면, 얻어진 항체에 화학적인 수식을 실시함으로써 얻을 수 있다. 이들 방법은 이 분야에 있어서 이미 확립되어 있다.
또한, 본 발명의 항체는 공지된 기술을 사용하여 비인간 항체 유래의 가변 영역과 인간 항체 유래의 정상 영역으로 이루어지는 키메라 항체 또는 비인간 항체 유래의 CDR (상보성 결정 영역) 과 인간 항체 유래의 FR (프레임 워크 영역) 및 정상 영역으로 이루어지는 인간화 항체로서 얻을 수 있다.
상기와 같이 얻어진 항체는 균일하게까지 정제할 수 있다. 본 발명에서 사용되는 항체의 분리, 정제는 통상의 단백질에서 사용되고 있는 분리, 정제 방법 을 사용하면 된다. 예를 들어, 어피니티 크로마토그래피 등의 크로마토그래피 칼럼, 필터, 한계외 여과, 염석, 투석, SDS 폴리아크릴아미드겔 전기 영동, 등전점 전기 영동 등을 적절히 선택, 조합하면, 항체를 분리, 정제할 수 있는데 (Antibodies: A Laboratory Manual. Ed Harlow and David Lane, Cold Spring Harbor Laboratory, 1988), 이들에 한정되는 것은 아니다. 상기에서 얻어진 항체의 농도 측정은 흡광도의 측정 또는 효소 결합 면역 흡착 검정법 (Enzyme-linked immunosorbent assay; ELISA) 등에 의해 실시할 수 있다.
어피니티 크로마토그래피에 이용하는 칼럼으로서는, 프로틴 A 칼럼, 프로틴 G 칼럼을 들 수 있다. 예를 들어, 프로틴 A 칼럼을 이용한 칼럼으로서 Hyper D, POROS, Sepharose F.F. (Pharmacia) 등을 들 수 있다.
어피니티 크로마토그래피 이외의 크로마토그래피로서는, 예를 들어 이온 교환 크로마토그래피, 소수성 크로마토그래피, 겔 여과, 역상 크로마토그래피, 흡착 크로마토그래피 등을 들 수 있다 (Strategies for Protein Purification and Char acterization: A Laboratory Course Manual. Ed Daniel R. Marshak et al., Cold Spring Harbor Laboratory Press, 1996). 이들 크로마토그래피는 HPLC, FPLC 등의 액상 크로마토그래피를 이용하여 실시할 수 있다.
또한, 본 발명의 항체의 항원 결합 활성을 측정하는 방법으로서, 예를 들어 흡광도의 측정, 효소 결합 면역 흡착 검정법 (Enzyme-linked immunosorbent assay; ELISA), EIA (효소 면역 측정법), RIA (방사 면역 측정법) 혹은 형광 항체법을 이용할 수 있다. ELISA 를 이용하는 경우, 본 발명의 항체를 고상화한 플레이트 에 본 발명의 폴리펩티드를 첨가하고, 그 다음에 목적의 항체를 함유하는 시료, 예를 들어 항체 생성 세포의 배양 상청이나 정제 항체를 첨가한다. 효소, 예를 들어 알칼리 포스파타아제 등으로 표지한 항체를 인식하는 2 차 항체를 첨가하고, 플레이트를 인큐베이션하고, 다음에 세정한 후, p-니트로페닐인산 등의 효소 기질을 첨가하여 흡광도를 측정함으로써 항원 결합 활성을 평가할 수 있다. 폴리펩티드로서 폴리펩티드의 단편, 예를 들어 그 C 말단으로 이루어지는 단편 혹은 N 말단으로 이루어지는 단편을 사용해도 된다. 본 발명의 항체의 활성 평가에는 BIAcore (Pharmacia 제조) 를 사용할 수 있다.
본 발명은 본 발명의 폴리펩티드를 코드하는 DNA 및 원하는 단백질을 코드하는 DNA 가 도입된 원하는 단백질을 생산하기 위한 세포를 제공한다. 이 세포는 원하는 단백질의 제조에 유용하다.
본 발명에 있어서 원하는 단백질은 특별히 한정되지 않고, 항체 (천연 항체, 저분자화 항체, 키메라 항체, 인간화 항체, 등) 나 생리 활성 단백질 (과립구 콜로니 자극 인자 (G-CSF), 과립구 매크로퍼지 콜로니 자극 인자 (GM-CSF), 에리트로포에틴, 인터페론, IL-1 이나 IL-6 등의 인터루킨, t-PA, 우로키나아제, 혈청 알부민, 혈액 응고 인자 등) 등, 어떠한 단백질이어도 되지만, 특히 항체가 바람직하다.
본 발명의 폴리펩티드를 코드하는 DNA 및 원하는 단백질을 코드하는 DNA 의 도입은 단일 벡터에 의해 동시에 도입해도 되고, 복수의 벡터를 이용하여 따로 따로 도입되어도 된다.
본 발명의 폴리펩티드를 코드하는 DNA 및 원하는 단백질을 코드하는 DNA 가 도입되는 세포는 특별히 한정되지 않고 어떠한 세포이어도 되지만, CHO 세포, 특히 CHO dhfr-세포가 바람직하다.
본 발명은 또한 본 발명의 폴리펩티드를 함유하는 배지를 제공한다. 배지 중에 있어서의 본 발명의 폴리펩티드의 함유량은 사용하는 배지, 배양하는 세포 및 제조하고자 하는 원하는 단백질의 종류 등에 따라 적절히 결정할 수 있지만, 통상 0.1㎎/L∼5㎎/L, 바람직하게는 0.2㎎/L∼1㎎/L, 더욱 바람직하게는 0.5㎎/L 이다.
본 발명의 폴리펩티드를 함유하는 배지는 동물 세포, 특히 CHO 세포를 이용하여 단백질을 제조할 때에 유용하다.
본 발명의 배지는 본 발명의 폴리펩티드를 함유하면 되고, 본 발명의 폴리펩티드를 첨가해도 되고, 숙주 세포로부터 분비해도 된다.
본 발명에서 이용하는 배지의 기타 성분으로서는, 통상 세포 (바람직하게는, 동물 세포) 배양 배지에서 사용되고 있는 각 성분을 적절히 사용할 수 있지만, 이들에는 통상, 아미노산, 비타민류, 지질 인자, 에너지원, 침투압 조절제, 철원, pH 완충제를 포함한다. 이들 성분의 함량은, 통상 아미노산은 0.05-1500㎎/L, 비타민류는 0.001-10㎎/L, 지질 인자는 0-200㎎/L, 에너지원은 1-20g/L, 침투압 조절제는 0.1-10000㎎/L, 철원은 0.1-500㎎/L, pH 완충제는 1-10000㎎/L, 미량 금속 원소는 0.00001-200㎎/L, 계면 활성제는 0-5000㎎/L, 증식 보조 인자는 0.05-10000㎍/L 및 뉴클레오시드는 0.001-50㎎/L 의 범위가 적당하지만, 이들에 한정되지 않고, 배양하는 세포의 종류, 원하는 단백질의 종류 등에 따라 적절히 결정할 수 있다.
상기 성분 외에, 예를 들어 미량 금속 원소, 계면 활성제, 증식 보조 인자, 뉴클레오시드 등을 첨가해도 된다.

상기 성분 외, 예를 들어 황산 구리, 황산 망간, 황산 아연, 황산 마그네슘, 염화 니켈, 염화 주석, 염화 마그네슘, 아규산 나트륨 등, 바람직하게는 황산 구리, 황산 아연, 황산 마그네슘 등의 미량 금속 원소; Tween80, 플루로닉 F68 등의 계면 활성제; 및 재조합형 인슐린, 재조합형 IGF-1, 재조합형 EGF, 재조합형 FGF, 재조합형 PDGF, 재조합형 TGF-α, 염산 에탄올아민, 아셀렌산 나트륨, 레티노인산, 염산 푸트레신 등, 바람직하게는 아셀렌산 나트륨, 염산 에탄올아민, 재조합형 IGF-1, 염산 푸트레신 등의 증식 보조 인자; 데옥시아데노신, 데옥시시티딘, 데옥시구아노신, 아데노신, 시티딘, 구아노신, 우리딘 등의 뉴클레오시드 등을 첨가해도 된다. 또한 상기 본 발명의 적합예에 있어서는, 스트렙토마이신, 페니실린 G 칼륨 및 겐타마이신 등의 항생 물질이나, 페놀 레드 등의 pH 지시약을 포함하고 있어도 된다.
또한, 배지에 어육 추출물 또는 어육의 효소 분해물을 첨가해도 된다.
배지의 pH 는 배양하는 세포에 따라 달라지지만, 일반적으로는 pH 6.8∼7.6, 대부분의 경우 pH 7.0∼7.4 가 적당하다.
배지는 시판되는 동물 세포 배양용 배지, 예를 들어 D-MEM (Dulbecco's Modified Eagle Medium), D-MEM/F-12 1:1 Mixture (Dulbecco's Modined Eagle Medium: Nutrient Mixture F-12), RPMI1640, CHO-S-SFMII (Invitrogen 사), CHO-SF (Sigma-Aldrich 사), EX-CELL 301 (JRH biosciences 사), CD-CHO (invitrogen 사), IS CHO-V (Irvine Scientific 사), PF-ACF-CHO (Sigma-Aldrich 사) 등의 배지로부터 조제하는 것이 가능하다.
또한, 본 발명의 배지는 무혈청 배지, 혹은 본 발명의 폴리펩티드 이외에 포유 동물 유래의 성분을 함유하지 않는 배지인 것이 바람직하다.
본 발명은 본 발명의 폴리펩티드를 함유하는 배지를 이용하여 CHO 세포를 배양하는 것을 포함하는, 원하는 단백질의 제조 방법을 제공한다.
본 발명은 또한, 본 발명의 폴리펩티드를 코드하는 DNA 및 원하는 단백질을 코드하는 DNA 가 도입된 CHO 세포를 배양함으로써, 원하는 단백질을 제조하는 방법을 제공한다.
CHO 세포의 배양은 당업자에게 공지된 방법을 이용하여 실시할 수 있다. 예를 들어, 통상 기상의 CO2 농도가 0-40%, 바람직하게는 2-10% 의 분위기하, 30-39℃, 바람직하게는 37℃ 정도에서 1-28일간 배양하는 것이 가능하다.
또한, 동물 세포 배양용의 각종 배양 장치로서는, 예를 들어 발효조형 탱크 배양 장치, 에어 리프트형 배양 장치, 컬처 플라스크형 배양 장치, 스피너 플라스크형 배양 장치, 마이크로 캐리어형 배양 장치, 유동층형 배양 장치, 할로우 파이버형 배양 장치, 롤러 보틀형 배양 장치, 충전조형 배양 장치 등을 이용하여 배양할 수 있다.

상기의 여러 가지 원하는 단백질의 제조에 이용하는 세포의 제작 방법에 있어서는, IGF-1 이 본 발명의 폴리펩티드인 것이 바람직하다.

본 발명의 원하는 단백질의 제조에 이용하는 세포의 제작 방법 또는 원하는 단백질의 제조에 이용하는 세포의 스크리닝 방법에서 사용되는 IGF-1 을 코드하는 유전자는 특별히 한정되지 않고, 인간, 마우스, 래트, 햄스터 등 어떠한 동물 유래의 ICF-1 이어도 되지만, 햄스터 IGF-1 이 바람직하다. 또한, 햄스터 IGF-1 이외의 IGF-1 배열은 Nucleic Acids Res. 14(20), 7873-7882 (1986) 및 J. Biol. Chem. 262 (16), 7894-7900 (1987) 에 기재되어 있다.
본 발명자들은 후술하는 실시예에 나타내는 바와 같이, 항체를 생성시키기 위하여 확립한 세포에, 본 발명의 폴리펩티드를 코드하는 DNA 를 도입하고, 활성형 IGF-1 을 발현시킴으로써, IGF-1 무첨가 배양에 있어서 이상적인 배양 거동을 갖는 항체 생성용 세포를 확립할 수 있음을 확인하였다. 본 발명의 폴리펩티드를 도입한 항체 생성 세포는 증식능, 생존율이 우수하고, 또한 항체 생성량에 있어서도 매우 우수한 효과를 나타내었다. 본 발명은 모든 원하는 단백질의 생성 세포, 특히 항체 생성 세포에의 응용이 가능하여, 산업상의 이용 가치는 매우 크다.
본 발명을 실시예에 의해 더욱 자세하게 설명하지만, 본 발명은 이것에 한정되지 않는다. 여러 가지 변경, 수식이 당업자에게는 가능하고, 이들 변경, 수식도 본 발명에 포함된다.
실시예 1: 햄스터 IGF-1 을 코드하는 cDNA 의 클로닝
(1) cDNA 의 합성
CHO-DXB11 세포에 항 인터루킨 6 리셉터 (IL-6R) 항체 유전자를 도입한 항IL-6R 항체 생성 세포 (일본 공개특허공보 평8-99902호, 참고예 2) 로부터 total RNA 추출을 실시하였다. 얻어진 mRNA 로부터 cDNA 를 합성하였다.
구체적으로는 이하의 방법으로 실시하였다. 계대 후 72시간 경과한 세포 증식기에 있는 약 5×106 개의 CHO 세포를 4℃, 1,000rpm 으로 5 분간 원심에 의해, 배양액을 포함하지 않는 원심관의 바닥에 팩된 상태에서 얻은 후, 미리 65℃ 로 데워 둔 세파졸 RNA Ⅰ 을 4㎖ 첨가하고, 피펫팅에 의해 재빨리 녹여, 실온에서 5 분간 방치하였다. 추가로 0.8㎖ 의 클로로포름을 첨가하고 Voltex 로 잘 교반한 후, 실온에서 3 분간 방치하였다. 냉각 원심기 AvantiTM J25 (BECKMAN 제조) 에 의해 4℃, 7,000rpm 으로 5 분간 원심 후, 상층을 신중하게 채취하여, 새로운 15㎖ 튜브로 옮겼다. 4㎖ 의 2-프로판올을 첨가하고 Voltex 로 잘 교반한 후, 실온 에서 10 분간 방치하였다. 4℃, 7,000rpm 으로 10 분간 원심 후, 상청을 완전히 제거하여, RNA 펠릿을 얻었다. 4㎖ 의 75% 에탄올로 원심관의 벽면을 잘 린스하고, 4℃, 7,000rpm 으로 5 분간 원심하였다. 상청을 완전히 제거한 후, 4㎖ 의 100% 에탄올을 첨가하고 다시 린스하였다. 4℃, 7,000rpm 으로 5 분간 원심에 의해 RNA 펠릿을 얻은 후, 실온에서 30 분간 방치하였다. 공기 건조시킨 RNA 펠릿은 100㎕ 의 DEPC-Water 를 첨가하고, 피펫팅에 의해 완전히 용해시켰다. 또한, RNeasy Mini Kit 의 프로토콜에 따라 total RNA 를 정제하였다. 얻어지는 30㎕ 의 total RNA 의 농도 측정은 분광 광도계 DU640 을 이용하여 행하고, 파장 260㎚ 의 흡광도 10D=40㎍/㎖ 로 하여 RNA 농도를 산출하였다. 고순도 total RNA 5㎍ 을 cDNA 합성에 이용하였다. 고순도의 total RNA 5㎍ 은 DEPC-Water 에서 10㎕ 로 조제한 후, 1㎕ 의 HPLC 그레이드 100pmol/㎕ T7-(T)24 프라이머를 피펫팅에 의해 첨가하고, 완전히 혼합한 후 스핀 다운하였다. 프라이머를 어닐시키기 위하여 70℃ 에서 10 분간 가열한 후, 재빨리 스핀 다운하여 빙상에 두었다. 빙상에서 2 분 이상 경과 후, 4㎕ 의 5×First-strand 버퍼, 2㎕ 의 0.1M DTT, 1㎕ 의 10mM dNTP Mix 를 피펫팅에 의해 첨가하고, 4℃ 에서 스핀 다운하여, 빙상에 두었다. 42℃ 의 히트 블록 상에서 인큐베이션을 개시하고, 2 분 경과한 시점에서 2㎕ 의 Super Script Ⅱ Reverse Transcriptase Mix 를 피펫팅에 의해 첨가하고, First-strand 합성 반응을 개시하였다. 60 분 경과하면, 재빨리 스핀 다운하고, 빙상에 두었다. 계속해서 Sccond-strand 합성 반응을 행 하기 위하여, 91㎕ 의 DEPC-Water, 30㎕ 의 5×Second-strand 버퍼, 3㎕ 의 10mM dNTP Mix, 1㎕ 의 E. coli DNA ligase, 4㎕ 의 E. coli DNA polymerase, 1㎕ 의 E. coli RNase H 를 피펫팅에 의해 순서대로 첨가한 후, 4℃ 에서 스핀 다운하고, 빙상에 두었다. 16℃ 에서 120 분간 인큐베이션하고, Second-strand 합성을 행하였다. 2 시간 후, 2㎕ 의 T4 DNA polymerase 를 첨가하고, 다시 5 분간 인큐베이션한 후, 재빨리 스핀 다운하고, 빙상에 두었다. 152㎕ 의 25:24:1 페놀:클로로포름:이소아밀알코올을 첨가하여 30 초간 볼텍스 후, 스핀 다운하고, 다시 피펫팅에 의해 혼합하고, 미리 1 분간 원심하여 겔을 팩해 둔 페이즈 록 겔 튜브에 어플라이하였다. 4℃ 에서 15,000rpm 2 분간 원심 후, 수층을 새로운 1.5㎖ 에펜도르프 튜브로 옮기고, 0.5㎕ 의 펠릿 페인트, 76㎕ 의 아세트산 암모늄 7.5M 용액의 순서로 첨가하고, 잘 볼텍스하였다. 600㎕ 의 99.5% 에탄올을 첨가하고 30 초간 볼텍스 후, 실온에서 5 분 방치하였다. 4℃ 에서 15,000rpm 10 분간 원심하고, 미반응 dNTP 를 제거한 cDNA 침전을 얻었다. cDNA 는 500㎕ 의 75% 에탄올을 첨가하여, 4℃ 에서 15,000rpm 3 분간 원심하였다. 세정을 2 회 반복함으로써 탈염시키고, 추가로 100㎕ 의 99.5% 에탄올을 첨가하고 4℃ 에서 15,000rpm 5 분간 원심한 후, 에탄올을 완전히 제거하였다. 약 5 분간의 공기 건조 후, 1.5㎕ 의 RNase free Water 에 녹여, cDNA 로서 -20℃ 에서 보존하였다. 50 분의 1 분량을 PCR 의 템플레이트로서 사용하였다.
(2) 햄스터 IGF-1 을 코드하는 cDNA 의 클로닝
햄스터 IGF-1 유전자는 PCR 클로닝에 의해 얻었다. PCR 프라이머의 설계 는 이미 알려진 래트/마우스/인간 IGF1 사이에서 유전자 배열이 보존되어 있는 영역을 이용하여 행하였다.
구체적으로는 이하의 방법에 의해 행하였다. 성숙형 래트 IGF-1 의 N 말단 13번째의 Ala 로부터 19번째의 Gly 를 코드하는 유전자 배열: 5'-gctcttcagttcgtgtgtgg (서열번호 3) 를 포워드 프라이머에, N 말단 50번째의 Arg 로부터 56번째의 Arg 를 코드하는 유전자의 상보 사슬 배열: 5'-ctcctcagatcacagctccg (서열번호 4) 를 리버스 프라이머에 이용하여, 상기 cDNA 를 주형으로 PCR 반응을 행하였다. 95℃, 5 분간의 전처리 후, 98℃, 5 초간의 디네이쳐, 53℃, 30 초간의 어닐링, 72℃, 30 초간의 신장 반응을 35 회 반복하여, 목적의 유전자를 증폭 후에, 72℃, 10 분간의 신장 반응을 추가하여, 131bp 의 햄스터 IGF-1 PCR 프래그먼트를 얻었다.
얻어진 PCR 단편의 염기 서열을 조사함으로써 햄스터 IGF-1 을 증폭시킨 DNA 인 것을 확인하고, 그 배열 정보를 이용하여 다시 PCR 반응을 계속하였다. N 말단 29번째의 Thr 로부터 35번째의 IlE 를 코드하는 유전자 배열: 5'-acaggctatggctccagcatt (서열번호 5) 를 포워드 프라이머에, N 말단 85번째의 Gln 으로부터 77번째의 Arg 를 코드하는 유전자의 상보 사슬 배열: tgagtcttgggcatgtcagtg (서열번호 6) 를 리버스 프라이머에 이용하여, 상기 cDNA 를 주형으로 PCR 반응을 행하였다. 95℃, 5 분간의 전처리 후, 98℃, 5 초간의 디네이쳐, 53℃, 40 초간의 어닐링, 72℃, 90 초간의 신장 반응을 35 회 반복하여, 목적의 유전자를 증폭 후에, 72℃, 10 분간의 신장 반응을 추가하여, 170bp 의 햄 스터 IGF-1 PCR 프래그먼트를 얻었다. 이상의 결과, 성숙형 햄스터 IGF-1 의 N 말단 13번째의 Ala 로부터 C 말단의 70번째 Ala 까지의 염기 서열을 얻었다.
또한, N 말단측의 배열을 얻기 위하여, 성숙형 래트 IGF-1 의 N 말단의 5' 에 상당하는 염기인 g 로부터 또한 5' 측 마이너스 33 염기의 t 로부터 14번째의 g 를 포함하는 배열: 5'-tgcttgctcacctttaccag (서열번호 7) 를 포워드 프라이머에, N 말단 50번째의 Arg 로부터 56번째의 Arg 를 코드하는 유전자의 상보 사슬 배열: 5'-ctcctcagatcacagctccg (서열번호 8) 를 리버스 프라이머에 이용하여, 상기 cDNA 를 주형으로 PCR 반응을 행하였다. 95℃, 5 분간의 전처리 후, 95℃, 30 초간의 디네이쳐, 44℃, 60 초간의 어닐링, 72℃, 90 초간의 신장 반응을 35 회 반복하여, 목적의 유전자를 증폭 후에, 72℃, 10 분간의 신장 반응을 추가하여, 185bp 의 햄스터 IGF-1 의 N 말단 영역을 포함하는 프래그먼트를 얻었다.
실시예 2: 햄스터 IGF-1 을 코드하는 cDNA 의 염기 서열 및 아미노산 배열의 결정
클로닝한 유전자는 DNA 시퀸서로 염기 서열을 결정하고, 또한 이로부터 아미노산 배열을 결정하고, 햄스터 IGF-1 을 코드하고 있는 것을 확인하였다. 얻어진 아미노산 배열을 도 1 및 서열번호 1 에, DNA 배열을 서열번호 2 에 나타낸다.
성숙 분비형 IGF-1 은 종을 넘어 70 아미노산으로 이루어지는데, 햄스터 1 GF-1 유전자는 특이적인 배열을 갖고 있어, 인간 IGF-1 과의 비교에서는 20번째의 P (Human:D), 35번째의 I (Human:S), 67번째의 T (Human:A) 의 3 아미노산이 인간 배열과 상이하다 (도 2).
실시예 3: 개변형 햄스터 IGF-1 유전자의 제작 및 그 IGF-1 활성
(1) 개변형 햄스터 IGF-1 유전자의 제작
분비형 햄스터 IGF-1 유전자는 N 말단으로부터 3번째에 마이너스 전하를 갖는 E (글루타민산) 를 갖는다. 이 부위의 아미노산을 플러스 전하측에 쉬프트하는 아미노산으로 변환한 개변형 햄스터 IGF-1 유전자를 제작하였다. IGF-1 유전자로의 변이 도입은 Site-directed Mutagenesis 법에 의해 행하였다.
천연형 햄스터 IGF-1 유전자 및 얻어진 개변형 햄스터 IGF-1 유전자를 각각, E3E (천연형), E3K (3번째의 글루타민산을 리신으로 변환), E3S (3번째의 글루타민산을 세린으로 변환) 라 칭한다 (도 3 참조).
(2) 천연형 및 개변형 햄스터 IGF-1 의 증식능 및 생존율에 미치는 효과
개변형 햄스터 IGF-1 유전자를 발현시키는 플라즈미드 (도 4) 를 구축하고, 친주인 항 IL-6R 항체 생성 CHO 세포에 도입하고, 하이그로마이신에 의한 약제 내성주를 선택, Stable 하게 발현하는 세포주를 얻었다. 얻어진 세포주는 추가로 Limiting Dilution 에 의해 싱글 셀 클로닝을 행하여, IGF-1 무첨가의 무혈청 배지에서 증식능이 높은 클론 세포를 얻었다.
각각의 상이한 전하를 갖는 E3E, E3K, E3S 로부터 얻어진 클론 세포 중에서 ME45(E3E), MK44(E3K), MS19(E3S) 를 선택하여, IGF-1 무첨가의 무혈청 배지에 있어서의, 100㎖ 스피너 플라스크에서의 세포의 증식능 및 생존율을 비교하였다. 친주인 항 IL-6R 항체 생성 세포에 대해서도, JRH 사부터 구입한 인간 GF-1 (LongTM R3IGF-1) 첨가 배지와 무첨가 배지에서 배양하여 비교하였다. S100 스피너 플라스크에 100㎖ 의 MRA 계대용 무혈청 배지를 첨가하고, 37℃ 의 CO2 인큐베이터내에서 1 시간 컨디셔닝한 후, 각각 3×106 개의 세포를 원심하여 첨가하였다. 1mL 의 배양액을 분취하여 초발 세포수 및 생존율을 측정하였다. 3일째 이후, 7일째까지의 세포수 및 생존율은 정시에 1mL 씩 분취하여 측정하였다.
얻어진 결과를 도 5 (증식능) 와 도 6 (생존율) 에 나타낸다. MS19(E3S) 는 증식능, 생존율이 모두 가장 우수한 것으로 판명되었다. MS19(E3S) 는 배양 7일째에 있어서도 80% 이상의 생존율을 나타내었다. 다음에, MK44(E3K) 가 양호한 결과를 나타내었다. 또한 친주인 항 IL-6R 항체 생성 세포는 IGF-1 무첨가 배양에서는 증식 불능이었다. MS19 의 증식능은 배양 3일째부터 6일째에 걸쳐 현저하고, 특히 4일째부터 5일째의 배가 시간 (MS19; 26.5hr, MK44; 33.0hr, ME45; 47.0hr, 항 IL-6R 친주; 86.7hr), 및 최고 도달 세포수 (MS19; 25.8×105cells/㎖, MK44; 20.3×105cells/㎖, ME45; 7.98×105cells/㎖, 항 IL-6 친주; 21.7×105cells/㎖) 로부터 MS19 의 우위성은 분명하였다. 세포 증식의 상승이 완만하고, 그 후에는 배양 후기까지 단번에 신장한다는 MS19 의 배양 거동은 항체 생성용 세포로서 이상적이었다. 또한, 생존율의 비교에 있어서 배양 7일째에 현저하도록 (MS19; 86.2%, MK44; 66.4%, ME45; 58.1%, 항 IL-6 친주; 76.0hr%), MS19 는 배양 후기까지 가장 안정적인 세포였다.
실시예 4: 햄스터 IGF-1(E3S) 도입 세포의 항체 생성량
햄스터 IGF-1 이 인간 IGF-1 이상의 항체 생성량 포텐셜을 갖는 것을 나타내기 위하여, 햄스터 IGF-1(E3S) 와 인간 ICF-1(E3S) 를 친주에 도입한 세포 중에서 가장 배양 거동이 양호한 싱글 클론의 항 IL-6R 항체 생성량을 비교하였다. 인간 IGF-1(E3S) 발현 플라즈미드는 햄스터 IGF-1(E3S) 를 주형으로 하여 P20D, I35S, T67A 변환을 위한 변이 도입을 순서대로 행함으로써 구축하였다. 실시예 1 과 마찬가지로 친주 항 IL-6R 항체 생성 CHO 세포에 도입하고, Hygromycin 내성 세포를 선택한 후, IGF-1(E3S) 를 안정적 발현으로 함으로써 IHF-1 무첨가 배지 중에 있어서의 증식능을 획득한 세포주를 선발하였다. 세포주를 한계 희석함으로써 싱글 클론을 얻고, 또한 증식이 우수한 IGF-1(E3S) 도입 세포를 이용한 Fed-Batch 배양을 행하여 항체 생성량을 비교하였다.
얻어진 결과를 도 7 에 나타낸다. 햄스터 IGF-1(E3S) 도입 세포의 항체 생성량은 배양 8일째에 973㎎/L 로서, 친주 (843㎎/L), 인간 IGF-1(E3S) 도입 세포 (821㎎/L) 보다 우위였다. 친주에 대하여 최적화된 배양 방법에도 불구하고 햄스터 도입 세포의 항체 생성량이 가장 높은 것이 확인되었다.
실시예 5: 햄스터 IGF-1(E3S) 도입 세포의 항체 생성량
실시예 4 에서 햄스터 IGF-1 이 인간 IGF-1 이상의 항체 생성량 포텐셜을 갖는 것을 나타낸 것으로부터, 추가로 마우스 IGF-1 을 첨가한 3 종의 IGF-1 의 항체 생성량 포텐셜을 비교하였다. 마우스 IGF-1(E3S) 발현 플라즈미드는 햄스터 IGF-1(E3S) 를 주형으로 하여 S69A 변환을 위한 변이를 도입함으로써 구축하였다. 이번에는 변이체 IGF-1(E3S) 에 더하여 천연형 IGF-1(E3E) 의 3 종간 비교도 함께 행하기 위하여, 햄스터, 인간, 마우스 IGF-1(E3S) 를 주형으로 E3E 로의 변이를 도입하여, 각각의 천연형 IGF-1(E3E) 발현 플라즈미드를 구축하였다.
얻어진 6 종의 IGF-1 발현 플라즈미드는 동시 조제한 친주에 일렉트로포레이션법으로 도입되어, Hygromycin 내성 세포를 선택한 후, IGF-1 을 안정적 발현으로 함으로써 IGF-1 무첨가 배지 중에 있어서의 증식능을 획득한 세포주 중에서 가장 증식이 양호한 세포를 3 주씩 선발하였다 (n=3). 확대 과정에서 인간 IGF-1(E3E) 의 1 주는 증식 불량이 되었으므로 인간 IGF-1(E3E) 만 n=2 에서 실험을 계속하는 것으로 하고, 합계 17 세포주는 100㎖ 스피너 배양으로 확대 후 Fed-Batch 배양에 의한 항체 생성량 비교를 행하였다.
도 8 은 햄스터, 마우스 및 인간 IGF-1(E3S) 도입 세포의 Fed-Batch 배양에 있어서의 항체 생성량을 나타낸다. 증식이 양호한 각종 3 주 (n=3) 의 평균 생성량은 햄스터>마우스>인간의 순서로 우위였다. 도 9 는 햄스터, 마우스 및 인간 IGF-1(E3S) 도입 세포의 3 주 중에서 최상위의 항체 생성량을 갖는 주의 비교이다. 평균 생성량에 더하여, 최상위 주의 생성량에 있어서도 햄스터 IGF-1(E3S) 도입주가 우위였다. 도 10 은 햄스터, 마우스 및 인간 IGF-1(E3E) 도입 세포의 Fed-Batch 배양에 있어서의 항체 생성량을 나타낸다. 증식이 양호한 각종 평균 생성량은 천연형 IGF-1(E3E) 도입주에 있어서도 햄스터>마우스>인간의 순서로 우위였다.

- 이하의 공정을 포함하는, 원하는 단백질의 제조에 이용하는 세포의 제작 방법:(a) CHO 세포에 원하는 단백질을 코드하는 DNA 를 도입하는 공정,(b) (a) 에서 제작한 세포에, IGF-1 을 코드하는 DNA 를 도입하는 공정.
- 이하의 공정을 포함하는, 원하는 단백질의 제조에 이용하는 세포의 제작 방법:(a) CHO 세포에 원하는 단백질을 코드하는 DNA 를 도입하는 공정,(b) (a) 에서 제작한 세포로부터, 원하는 단백질을 발현하는 세포를 선택하는 공정,(c) (b) 에서 선택한 세포에, IGF-1 을 코드하는 DNA 를 도입하는 공정.
