Description
Claims (16)
- 다음의 단계를 포함하는 기질에 단백질을 고정화하기 위한, A-B-C 형태의 링커분자를 제조하는 방법:(a) 단백질의 비특이적 결합을 저해하는 단백질 고정화 리간드(B)의 일측 말단에, 기질표면에서 자기조립 단분자층을 형성하는 물질(A)을 결합하는 단계; 및(b) 상기 단백질 고정화 리간드의 타측 말단에 단백질과 결합하는 친화성 물질(C)을 결합하는 단계.
- 제1항에 있어서, 상기 단백질 고정화 리간드(B)는 양 말단이 아민으로 치환된 올리고에틸렌글리콜 또는 폴리에틸렌글리콜인 것을 특징으로 하는 방법.
- 제1항에 있어서, 상기 단백질은 단백질에 친화성인 물질로 태그된 단백질인 것을 특징으로 하는 방법.
- 제3항에 있어서, 상기 단백질에 친화성인 물질로 태그된 단백질은 글루타치온 S-전이효소로 태그된 단백질, 말토즈결합 단백질로 태그된 단백질 및 히스티딘 으로 태그된 단백질로 구성된 군에서 선택되는 것을 특징으로 하는 방법.
- 단백질의 비특이적 결합을 저해하는 단백질 고정화 리간드(B)의 양 말단에, 기질표면에서 자기조립 단분자층을 형성하는 물질(A) 및 단백질과 결합하는 친화성 물질(C)이 각각 결합되어 있는, 기질에 단백질을 고정화하기 위한, A-B-C 형태의 링커분자.
- 제5항에 있어서, 상기 단백질 고정화 리간드(B)는 올리고에틸렌글리콜 또는 폴리에틸렌글리콜인 것을 특징으로 하는 A-B-C 형태의 링커분자.
- 제7항에 있어서, 상기 Y는 L-환원 글루타치온, 이미노다이아세트산 (iminodiacetic acid), 니트로트리아세트산 (nitrilotriacetic acid), 말토즈 유도체, 갈락토즈, 칼모듈린, 바이오틴, 키틴, 셀룰로즈, 씨-믹 (C-myc), 티오레독신 (thioredoxine), 인테인 (intain), S-펩타이드, 및 DNA로 구성되는 군에서 선택되는 것을 특징으로 하는 링커분자.
- 제7항에 있어서, 화학식(I)의 구조식을 갖는 링커분자는 링커분자 1~5로 구성된 군에서 선택되는 것을 특징으로 하는 링커분자.
- 제10항에 있어서, 상기 Y는 L-환원 글루타치온, 이미노다이아세트산 (iminodiacetic acid), 니트로트리아세트산 (nitrilotriacetic acid), 말토즈 유도체, 갈락토즈, 칼모듈린, 바이오틴, 키틴, 셀룰로즈, 씨-믹 (C-myc), 티오레독신 (thioredoxine), 인테인 (intain), S-펩타이드, 및 DNA로 구성되는 군에서 선택되는 것을 특징으로 하는 링커분자.
- 다음 단계를 포함하는 단백질 칩의 제조방법:(a) 제5항 내지 제11항 중 어느 한 항에 따른 A-B-C 형태의 링커분자를 기질에 도입하여, 기질표면에서 자기조립 단분자층을 형성하는 물질(A)을 통해 기질상에 상기 링커분자의 자기조립 단분자층을 형성하고, 단백질과 결합하는 친화성 물질(C)을 노출시키는 단계; 및(b) 상기 노출된 단백질과 결합하는 친화성 물질(C)에 단백질을 결합시키는 단계.
- 제5항 내지 제11항 중 어느 한 항에 따른 A-B-C 형태의 링커분자를 매개로 기질상에 단백질이 결합되어 있는 단백질 칩.
- 제5항 내지 제11항 중 어느 한 항에 따른 A-B-C 형태의 링커분자를 매개로 단백질 고정용 담체에 단백질을 고정하는 것을 특징으로 하는 단백질의 고정화 방법.
- 제5항 내지 제11항 중 어느 한 항에 따른 A-B-C 형태의 링커분자를 매개로 단백질 고정용 담체에 단백질이 결합되어 있는 고정화 단백질.
- 제15항에 있어서, 단백질 고정용 담체는 멤브레인 또는 비드 형태인 것을 특징으로 하는 고정화 단백질.
