Claims (18)
- (i) 단자엽 또는 쌍자엽 식물체의 종자를 발아시키는 단계;(ⅱ) 상기 발아된 종자의 자엽 절편 또는 배축을 식물 생장조절물질이 첨가된 캘러스 유도 배지에 접종하여 캘러스를 유도하는 단계;(ⅲ) 액체 현탁배양 배지를 이용하여 상기 캘러스를 액체배양함으로써 현탁배양 세포를 증식하는 단계;(ⅳ) 단계 (ⅱ)의 캘러스 또는 단계 (ⅲ)의 현탁배양 세포를 부정근 유도 배지에 접종하여 부정근 (adventitious root)을 유도하는 단계; 및(ⅴ) 생물반응기 (bioreactor)를 이용하여 상기 부정근을 대량으로 배양하는 단계를 포함하는 생물반응기를 이용한 단자엽 또는 쌍자엽 식물체의 부정근 대량 생산 방법.
- 제 1 항에 있어서, 상기 단계 (i)의 종자 발아에 사용된 배지는 MS (Murashige and Skoog), B5 (Gamberg), LS, N6 (Gupfa and Durzan) 및 White로 구성된 군으로부터 선택되며, 식물 생장조절물질을 포함하지 않는 것을 특징으로 하는 단자엽 또는 쌍자엽 식물체의 부정근 대량 생산 방법.
- 제 1 항에 있어서, 상기 단계 (ⅱ)의 캘러스 유도 배지는 MS 또는 N6 배지인 것을 특징으로 하는 단자엽 또는 쌍자엽 식물체의 부정근 대량 생산 방법.
- 제 1 항에 있어서, 상기 단계 (ⅱ)의 식물 생장조절물질은 IAA (Indoleacetic acid), IBA (Indolebutyric acid), NAA (Naphthalene acetic acid) 및 2,4-D (2,4-dichlorophenoxyacetic acid)로 구성되는 군으로부터 선택되고, 0.1-10 ppm의 농도로 처리하는 것을 특징으로 하는 단자엽 또는 쌍자엽 식물체의 부정근 대량 생산 방법.
- 제 1 항에 있어서, 상기 단계 (ⅲ)의 액체 현탁배양 배지는 MS, WPM (Lloyd and McCown) 또는 SH (Schenk and Hilderant) 배지인 것을 특징으로 하는 단자엽 또는 쌍자엽 식물체의 부정근 대량 생산 방법.
- 제 1 항에 있어서, 상기 단계 (ⅳ)의 부정근 유도 배지는 mB5 (modified Gamberg), MS, 1/2 MS, B5, SH, LS 및 N6로 구성된 군으로부터 선택되는 것을 특징으로 하는 단자엽 또는 쌍자엽 식물체의 부정근 대량 생산 방법.
- 제 1 항에 있어서, 상기 단계 (ⅳ)의 식물 생장조절물질은 IAA 또는 IBA이고, 0.1-10 ppm으로 첨가되는 것을 특징으로 하는 단자엽 또는 쌍자엽 식물체의 부정근 대량 생산 방법.
- 제 1 항에 있어서, 상기 단계 (ⅴ)의 부정근 대량 배양은 N6 또는 1/2 MS 배지에 0.1-5 ppm의 IAA를 첨가한 배지에서 배양하는 것을 특징으로 하는 단자엽 또는 쌍자엽 식물체의 부정근 대량 생산 방법.
- 제 1 항에 있어서, 상기 단계 (ⅴ)의 부정근 대량 배양은 0.5-2 cm 길이의 부정근을 배지 총량의 0.1-0.5%의 접종량으로 접종하여 7일 단위로 배지를 공급하는 방식으로 배양하는 것을 특징으로 하는 단자엽 또는 쌍자엽 식물체의 부정근 대량 생산 방법.
- (ⅰ) 식물세포의 게놈 DNA에 삽입될 수 있고, 유용 재조합단백질을 코딩하는 유전자 서열을 갖는 벡터가 내재되어 있는 아그로박테리움 튜머페이션스 (Agrobacterium tumefaciens)를 단자엽 또는 쌍자엽 식물체의 자엽 또는 배축에 감 염시키는 단계;(ⅱ) 감염된 식물체의 자엽 또는 배축을 식물 생장조절물질이 함유된 재분화 배지에서 재분화시켜 신초를 형성시키는 단계;(ⅲ) 재분화된 신초를 식물 생장조절물질 NAA가 함유된 발근 배지에서 발근시켜 형질전환 식물체를 형성시키는 단계;(ⅳ) 형질전환 식물체의 종자를 발아시키는 단계;(ⅴ) 캘러스 유도 배지 및 식물 생장조절물질을 첨가하여 발아된 형질전환 식물체의 종자로부터 캘러스를 유도하는 단계;(ⅵ) 캘러스를 액체배양함으로써 현탁배양 세포를 증식하는 단계;(ⅶ) 단계 (ⅴ)의 캘러스 또는 단계 (ⅵ)의 현탁배양 세포로부터 부정근을 유도하는 단계; 및(ⅷ) 생물반응기를 이용하여 형질전환 식물체의 부정근을 대량으로 배양하는 단계를 포함하는 형질전환 식물체의 부정근으로부터 유용 재조합단백질을 대량 생산하는 방법.
- 제 10 항에 있어서, 상기 단계 (i)의 벡터는 (a) 식물 세포내에서 작용하는 복제 원점; (b) 식물 세포 내에서 전사를 촉진할 수 있는 프로모터; (c) 상기 프로모터에 기능적으로 연결된 외래 단백질을 코딩하는 구조 유전자 서열; 및 (d) 식물 세포 내에서 작동하여 RNA의 3’ 말단에 폴리아데닐을 형성시키는 폴리아데닐 시그 널 서열을 포함하고, 추가적으로 항생제 내성 유전자 또는 리포터 물질을 인코딩하는 유전자를 포함하는 것을 특징으로 하는 형질전환 식물체의 부정근으로부터 유용 재조합단백질을 대량 생산하는 방법.
- 제 11 항에 있어서, 상기 단계 (ⅱ)의 식물 생장조절물질은 BA인 것을 특징으로 하는 형질전환 식물체의 부정근으로부터 유용 재조합단백질을 대량 생산하는 방법.
- 제 11 항에 있어서, 상기 단계 (ⅲ)의 0.01-5 mg/ℓ의 양으로 첨가되는 것을 특징으로 하는 형질전환 식물체의 부정근으로부터 유용 재조합단백질을 대량 생산하는 방법.
- 제 11 항에 있어서, 상기 단계 (ⅳ) 내지 (ⅷ)은 상기 제 1 항의 단계 (ⅱ) 내지 (ⅴ)와 동일하게 수행하는 것을 특징으로 하는 형질전환 식물체의 부정근으로부터 유용 재조합단백질을 대량 생산하는 방법.
- (ⅰ) 유용 재조합단백질을 코딩하는 유전자가 삽입된 Ri 플라스미드를 포함하는 아그로박테리움 리조게네스 (Agrobacterium rhizogenes)를 식물체의 뿌리 절편에 감염시키는 단계;(ⅱ) 감염된 뿌리 절편으로부터 모상근 (hairy root)을 유도하는 단계;(ⅲ) 캘러스 유도 배지 및 식물 생장조절물질을 이용하여 모상근으로부터 캘러스를 유도하는 단계;(ⅳ) 캘러스를 기내 배유도배지에 옮겨 배양함으로써 체세포배를 유도하는 단계;(ⅴ) 체세포배를 발아배지 및 재분화 배지에서 배양함으로써 형질전환 식물체를 유도하는 단계; 및(ⅵ) 형질전환 식물체의 모상근을 생물반응기에서 액체배양하는 단계를 포함하는 형질전환 식물체의 모상근으로부터 유용 재조합단백질을 대량 생산하는 방법.
- 제 15 항에 있어서, 상기 단계 (ⅲ)의 캘러스 유도 배지는 MS 또는 N6 배지이고, 식물 생장조절물질은 IAA, IBA, NAA 및 2,4-D로 구성된 군으로부터 선택되는 것을 특징으로 하는 형질전환 식물체의 모상근으로부터 유용 재조합단백질을 대량 생산하는 방법.
- 제 15 항에 있어서, 상기 단계 (ⅴ)는 BA 또는 NAA를 0.01-2 ppm 첨가하여 배양함으로써 형질전환 식물체로 유도시키는 것을 특징으로 하는 형질전환 식물체의 모상근으로부터 유용 재조합단백질을 대량 생산하는 방법.
- 제 1 항 내지 제 17 항에 있어서, 상기 식물체는 벼 (Oryza sativa), 보리 (Hordum sativum), 밀 (Triticum aestivum 또는 Triticum vulgare), 감자 (Solanum tuberosum), 고구마 (Ipomoea batatas), 오이 (Cucumis sativus) 및 참외 (Cucumis melo)로 구성된 군으로부터 선택되는 식물체인 것을 특징으로 하는 방법.
