본 발명은 프로모터, 이러한 프로모터를 함유하는 DNA 단편 및 특히, G 단백질 수용체에 대한 활성화 또는 억제 효과를 나타내는 물질의 동정 방법에서 이의 용도에 관한 것이다.
Description
GPCR(G 단백질-커플링된 수용체)은 구조적으로 및 기능적으로 연관된 트랜스막 단백질의 유전자 패밀리를 형성한다. GPCR은 의학 조사 및 약리학적 활성 물질의 개발에 있어 매우 중요한 표적 분자이며 병리학의 다발성에서 중요한 위치를 차지하고 있다[참조문헌: Stadel et al., 1997]. 천식의 약리학적 치료시 GPCR의 중심적 위치의 통상적 예로는 β2AR(β2-아드레날린성 수용체) 또는 카포시 육종(KS) 발병기전에서의 카포시 육종 관련 헤르페스바이러스 GPCR의 관련성이 있다[참조문헌: Geras-Raaka et al., 1998]. 이러한 부류의 수용체는 예를 들어, 단백질 호르몬, 케모카인, 펩타이드 또는 2가 양이온과 같은 광범위한 스펙트럼의 리간드에 결합한다. 따라서, G 단백질-커플링된 수용체의 활성화제 및 억제제의 동정은 보다 우수한 지식 및 질병의 치료를 지향하는 가장 전도 유망한 접근법 중 하나이다[참조문헌: Wilson et al., 1998].
GPCR 신호전달 경로의 다수의 성분은 빵 효모인 사카로마이세스 세레비시애(Saccharomyces cerevisiae)의 GPCR 신호전달 경로의 공통성(orthologous) 성분을 갖는다. 교배 인자(페로몬 α 또는 페로몬a)에 의한 자극은 예를 들어, 페르몬 의존성 유사분열유발원-활성화 단백질 키나제 다단계반응(cascade)(이후에는 MAPK 다단계반응으로 지칭함)을 활성화시킨다[참조문헌: Frederickson, 1999]. 특히, 외래 종의 GPCR이 효모에서 기능적으로 발현되고, 도입된 GPCR이 특정 리간드에 의해 활성화되고 이 신호가 효모 MAPK 다단계반응을 통과할 경우 효모 MAPK 다단계반응을 통해 측정가능한 세포 반응을 유발할 수 있기 때문에, 관련 GPCR 또는 상응하는 신호전달 경로의 활성화제 또는 억제제를 조사하기 위해서 효모의 상기 특성을 효모 모델에서 포유동물 GPCR, 특히 사람 GPCR을 검정하는데 사용할 수 있다. 현재까지도 그 기능이 밝혀지지 않은 GPCR의 활성화제 또는 억제제인 "orphan GPCR"에 대한 조사가 특히 관심대상이다.
페로몬 α 및a는 에스. 세레비시애(S. cerevisiae)에서 내인성 G 단백질-커플링된 수용체 Ste2p 및 Ste3p를 통해 작용한다[참조문헌: Gustin et al., 1998]. 이와 관련하여, 페로몬 α는 페로몬 의존성 MAPK 다단계반응에 직접적으로 작용함으로써 특정 에스. 세레비시애 유전자의 발현을 조절한다. 물질이 에스.세레비시애에서 이종적으로 발현된 GPCR에 활성화 또는 억제 방식으로 작용하는지는 MAPK 다단계반응에 의해 조절되는 에스. 세레비시애 유전자(마커 유전자)를 발현시켜 검출할 수 있다. 이러한 마커 유전자의 프로모터는, 예를 들어, GPCR 활성화제 또는 억제제를 동정하기에 적합한 리포터 시스템과 함께 사용할 수 있다.

현재까지, GPCR의 억제제 또는 활성화제를 동정하기 위한 기능 검정은 주로 포유동물 세포에서 수행되어 왔다[참조문헌: Wilson et al. (1998) British Journal of Pharmacology 125, 1387-1392; Geras-Raaka et al. (1998) J. Exp. Med. 188 No.2, 405-408]. 에스. 세레비시애에서의 기능 검정을 위한 FUS1 및 FUS2 유전자의 프로모터의 용도는 문헌[참조문헌: Cisnowski et al. (1999) Nature 17, 878-883; Frederickson (1999) Nature Biotechnology A, 852-853]에 기술되어 있다. FUS1은 페로몬 α로 활성화된 후에 효모 사카로마이세스 세레비시애의 야생형 세포에서의 발현이 증가되는 유전자이다. US 5,063,153에서는 FUS1 및 FUS2 프로모터를 관심있는 단백질을 암호화하는 구조적 유전자를 대량으로 발현시키기 위해 사용하고 있다.
본 발명의 목적은 페로몬 α에 의해 활성화될 수 있으며 조절된 유전자의 강력한 발현을 달성하는데 사용할 수 있는 상이한 에스. 세레비시애 프로모터를 동정하는 것이다.
본 발명은 에스. 세레비시애 YNL279w 유전자의 프로모터에 관한 것이다. 본 발명은 서열 4의 서열을 갖는 프로모터에 관한 것이다.
본 발명은 또한 구조 유전자에 기능적으로 연결될 경우 이의 전사를 조절하는 사카로마이세스 세레비시애 YNL279w 유전자 프로모터를 함유하는 재조합 DNA 단편 및 서열 4에 기술된 DNA 서열을 갖는 프로모터를 함유하는 DNA 단편에 관한 것이다. 본 발명은 YNL279w 프로모터 및 구조 유전자를 함유하고 상기 프로모터가 상기 구조 유전자에 기능적으로 연결되어 있는 재조합 DNA 단편에 관한 것이다. 구조 유전자는, 예를 들어, 수용체 유전자를 암호화하거나 에스. 세레비시애에서 대량으로 생성시키고자 하는 단백질을 암호화한다.
본 발명은 이러한 재조합 DNA 단편을 함유하는 DNA 벡터 및 재조합 에스. 세레비시애 세포에 관한 것이다. 이러한 재조합 에스. 세레비시애 세포는 바람직하게는 기능적으로 활성인 내인성 수용체 Ste2p 및 Ste3p을 발현하지 않거나 단지 소량만을 발현한다.
또한, 본 발명은 GPCR을 기능적으로 특성화하고 GPCR 억제제 및/또는 활성화제를 선별하는 방법 및 에스. 세레비시애에서 단백질을 제조하는 방법에 관한 것이다.

재조합 에스. 세레비시애 세포에는 적절한 내인성 GPCR인 Step 2 및/또는 Step 3이 예를 들어 결실되었기 때문에 이들 유전자는 바람직하게는 발현되지 않는다. 이러한 결실은 예를 들어, 문헌[참조문헌: Broach Thorner (1996) Nature, 384, 14-16]에 기재되어 있는 바와 같이 수행할 수 있다.
다른 양태는 발현될 이종 단백질의 구조 유전자를 YNL279w 유전자의 프로모터에 기능적으로 연결시키고 이의 조절하에서 발현시켜 이종 단백질을 에스. 세레비시애에서 제조하는 방법에 관한 것이다.
본 발명은 프로모터, 이러한 프로모터를 함유하는 DNA 단편 및 특히 G 단백질-커플링된 수용체에서 활성화 또는 억제 작용을 나타내는 물질을 동정하는 방법에서 이의 용도에 관한 것이다.

공지된 유전자 및 특히 아직 특성화되지 않은 유전자가 페로몬 α에 의해 유도 또는 억제되는지를 측정하기 위해, 빵 효모 사카로마이세스 세레비시애의 배양물을 페로몬 α로 처리하거나 페로몬 α로 처리하지 않음으로써 페로몬-유도성 및 페로몬-억제성 ORF(개방 판독 프레임)을 동정한다. 6000개 이상의 동정된 효모 유전자의 발현 패턴을 평행하게 나타낼 수 있는 DNA 마이크로칩을 사용한다. 페르몬으로 처리하지 않았거나 상이한 방식으로 페르몬으로 처리한 2종의 세포 또는 세포들의 배양물의 유전자 발현 패턴의 비교는 상응하는 mRNA를 적절하게 제조한 후 DNA 미세정렬을 보조로 하여 특이적으로 활성화되거나 억제된 전자를 측정할 수 있도록 한다.
페로몬 α를 첨가한 후, ORF YNL279w는 이의 발현이 강하게 시간 의존적으로 증가하는 것으로 나타났다(도 2 및 3). 유전자의 전사 및 발현은 일반적으로 전사 출발 지점의 5'에 위치한 조절 서열을 통해 조절되기 때문에, 이러한 조절 서열을 갖는 프로모터인 "프로모터"가 특히 관심대상이다. 약리학적으로 및 의학적으로 관심있는 GPCR의 활성화제 또는 억제제를 동정하기 위해서, 적합한 리포터 시스템을 나타내는 유전자 또는 구조 유전자 전방에 상기한 유도성 또는 억제성 프로모터를 동일한 방식으로 클로닝하여, 첨가 후에 당해 리포터 시스템에 대한 활성화 또는 억제 작용을 나타내는 물질을 동정할 수 있다. 적합한 리포터 시스템의 예로는 LacZ, 루시페라제, 에쿼린, 녹색 형광 단백질(GFP), DsRed, HIS3, URA3, TRP1 및 LEU2의 유전자 뿐만 아니라, 특정 항생제에 대한 내성 유전자, 예를 들어, 가나마이신이 있다. 이들 "리포터 유전자"는 예를 들어, MAPK 다단계반응에 대한 특정 물질의 작용이 세포 콜로니의 색상 변화를 통해 검출될 수 있는 lacZ와 같은 유전자일 수 있다. 리포터 유전자는 예를 들어, 세포가 특정한 선택 조건하에서 생장할 수 있도록 하는 유전자일 수도 있으며, 예로는 영양요구성 유전자가 있다.

이러한 리포터 유전자를 암호화하는 DNA를 효모 사카로마이세스 세레비시애의 ORF YNL279w의 페로몬-유도성 프로모터에 3' 방향으로 융합시키고 예를 들어, 고복사 벡터 또는 저복사 벡터내로 클로닝시킨다. 이어서 상기 벡터를 효모 세포를 형질전환시키는데 사용한다. 마찬가지로, 프로모터에 융합된 DNA를 효모 게놈내로 안정하게 삽입시킬 수 있다.
영양요구성 유전자(예: HIS3, URA3, TRP1, LEU2, ADE2 및 LYS1)의 그룹에 속하는 리포터 유전자로 유전적으로 조작한 효모 균주는 돌연변이시켜야 하는데, 즉 상응하는 유전자에 대해 기능적으로 불활성화시켜야 한다. 페로몬에 의한 효모 세포의 자극은 효모 프로모터 YNL279w를 활성화시고 이 프로모터에 의해 조절되는 리포터 유전자의 발현을 증가시킨다. GPCR 효능제를 조사할 경우에, 유전자 YNL279w의 프로모터를 예를 들어, HIS, ADE, TRP 또는 LEU와 같은 리포터 유전자의 전방에 클로닝시키는 것이 바람직한데, 그 이유는 이러한 조합이 적절하게 결실된 배지에서 세포 생장을 통해 판독을 가능케하기 때문이다. 연구할 물질에 의한 선택된 GPCR의 활성화는 MAPK 다단계반응의 기저에 위치한 프로모터 YNL279w를 활성화시키고 이로써 YNLw 프로모터에 의해 조절되는 리포터 유전자를 발현시킨다. LacZ, GFP 또는 EGFP를 사용할 경우, 판독은 비색 또는 발광 측정을 통해 수행할 수 있다. 이종 GPCR이 효능제에 의해 활성화된 세포만이 화학발광을 나타낸다.
사용되는 리포터 유전자가 CAN1일 경우, 세포는 카나바닌 함유 배지에서 생장한다. CAN1 유전자는 이종적으로 발현되는 GPCR의 활성화제(효능제)의 존재하에 발현시켜 세포가 카나바닌 함유 배지에서 생장을 중지하도록 한다. 길항제(억제제)를 첨가할 경우, 배양물은 상기 선택 배지에서 생장할 수 있다.
선별 방법은 프로모터-리포터 유전자 작제물(예: 표 1의 DNA 작제물)로 미리 형질전환시킨 효모 세포를 접종시킨 미세역가 플레이트 상에서 수행할 수 있다. 또한, 상기 세포를 효능제 또는 길항제를 밝히고자 하는 이종 GPCR로 형질전환시킨다. 예를 들어, 상기 방법에서는 사람 GPCR, 예를 들어, 아데노신 수용체, 소마토스타틴 수용체, 도파민 수용체, 브라디키닌 수용체, 리소지질(lysolipid) 수용체, β-아드레날린성 수용체 및 무스카린성 아세틸콜린 수용체를 사용할 수 있다.
페로몬 의존성 MAPK 다단계반응의 활성화제 및 억제제를 동정하기 위해 페로몬 α에 의해 유도되는 유전자로서, YNL279w 프로모터를 사용할 수도 있다. YNL279w 프로모터가 페로몬 α처리 후에 예를 들어 FUS1 프로모터에 비해 보다 민감하기 때문에, 본원에서 제안하는 바와 같은 YNL279w의 사용은, 예를 들어, FUS1 프로모터를 사용하는 다른 선별 방법과 비교하여 유리하다. 도 4는 YNL279w 프로모터/에쿼린 작제물(p415YNL279-AEQ)이 FUS1 유전자의 프로모터로부터의 영역에 상응하는 프로모터 요소(도 4에서 4PRE-AEQ로 지칭함)에서의 경우보다 사실상 우수하게 반응함을 나타낸 것이다.
실시예 1: ORF YN279w의 동정
유전형(MAT a sst1::LEU2;ade2;can1-100;his3-11,15;leu2-3,112;trp1-1;ura3-1)을 갖는 사카로마이세스 세레비시애 효모 균주의 세포 배양물을 1μM 페로몬 α(90% 메탄올 속에 용해시킴)로 자극하고, 대조 세포를 90% 메탄올로만 처리한다. 자극 후 20분째, 90분째 및 180분째에, 약 10 OD600nm에 상응하는 양의 세포를 분취한다. 전체 RNA를 표준 방법[참조문헌: Sambrock et al. Molecular Chemistry Press, (Cold Spring Harbor Laboratory 1989)]에 따라서 상기 세포로부터 분리하고, 각 경우에 전체 RNA 30㎍을 제1 쇄 합성을 위해 사용한다. 제1 및 제2 쇄 합성 및 또한 생성된 이본쇄 cDNA의 정제 단계, 시험관내 전사 및 하이브리드화용 cRNA의 제조를 위한 프로토콜은 아피메트릭 인코포레이티드(Affymetrix In., Santa Clara, CA, USA)의 프로토콜에 상응한다.
YNL279w ORF를 페로몬 α로 유도된 효모 세포 및 페로몬 α로 유도되지 않은 효모 세포의 발현 프로필의 비교를 통해 가장 강력하게 유도된 ORF 중 하나로서 동정한다. 적합한 리포터를 사용한 선별 시스템에서의 프로모터의 능력 및 유용성을 게놈 사카로마이세스 세레비시애 DNA로부터의 YNL279w 프로모터 영역을 증폭시켜 측정한다.
실시예 2: YNL279w 프로모터("프로모터")의 분리
YNL279w 유전자의 프로모터 영역을 프라이머 YNL279F1(서열 1: 5'-CCGAGTCCTACTCCTATGCTGTTTACAAGG-3') 및 YNL279R(서열 2: 5'-TGCTCTAGAATCATCAACGTTCACAAATTCG-3')을 사용하여 증폭시킨다. 수득되는 증폭체의 동일성을 표준 서열화 방법을 사용하여 측정한다. YNL279F1/YNL279R을 통해 증폭된 프로모터 영역의 길이는 제한 절단 부위를 제외하고 473bp이며, 프로모터 영역중 이 단편을 하기에서 "프로모터"로 지칭한다.
실시예 3: 기능성의 측정
제안된 선별 방법의 기능성은 YNL279w 유전자의 증폭된 프로모터 영역을 에쿼린 암호화 유전자 전방에서 벡터 p415(도 5 참조; 벡터 pRS415의 유도체, ATCC87520)내로 클로닝시켜 측정한다. 상기 작제물을 Li 아세테이트 방법[참조문헌: Ito et al., 1983]을 사용하여 사카로마이세스 세레비시애 균주(Mat a far1::hisG sst2::ura3FOAfus1::HIS3)내로 형질전환시킨다. 사카로마이세스 세레비시애 균주(Mat a far1::hisG sst2::ura3FOAfus1::HIS3)내의 MAPK 다단계반응을 페로몬 α로 자극한 후, 에쿼린을 다음과 같이 검출한다:
1. 세포 현탁액 25㎕(선택 배지, SC/글루코즈 류신 중 25㎕당 1.5×106세포)를 96-웰 플레이트에 첨가한다;
2. 일련의 희석액(10-6M 내지 10-8M) 및 1μM 코엘렌테라진(최종 농도 0.5μM) 중의 교배 인자 α를 함유하는 2×농축된 자극 혼합물 25㎕을 상기 세포에 첨가한다;
3. 96-웰 플레이트를 조심스럽게 교반한다:
4. 96-웰 플레이트를 암실에서 1.5시간 동안 30℃로 습한 대기중에서 배양한다.
5. 검출을 위해, 칼슘/용해 완충액 150㎕을 첨가한다; 당해 혼합물을 첨가한 직후 에쿼린은 광자를 방출한다; 생성되는 시그널을 15초 동안 통합한다; 시그널을 발광측정기(Luminoskan, LABSYSTEM)을 사용하여 검출한다.

코엘렌테라진 모액: 100% 메탄올중 1mM(MOLECULAR PROBES; ref C-2944)
