섬유아세포에 대한 꼬리조팝나무 추출물의 세포 독성을 확인하기 위해, MTT assay를 실시하였다. 96 well plate에 섬유아세포(CCD-986sk)를 5Х104 cells/mL의 농도로 배양하고, 24시간 후에 무혈청 배지에 제조예의 각 추출물을 0.4 내지 100 μg/mL 농도로 희석하여 처리하였다. 24시간 후, 5 mg/mL MTT(M2128, sigma)를 20 μL 넣어 3시간 배양하였다. 이후, 배지를 모두 제거하고, 150 μL DMSO를 각 well에 넣어 교반한 후, Microplate reader로 570 nm에서 흡광도를 측정하였다. 하기 식으로 세포 생존율을 계산하였고, 세포 생존율이 80 % 이상인 경우, 세포 독성이 없는 것으로 판단하였다. 96 well plate에 섬유아세포(CCD-986sk)를 5Х104 cells/mL의 농도로 24시간 배양하였다. 배지를 제거하고 HBSS를 넣은 후, UV irradiation system(VILBER)을 이용하여 UVA 30 J/cm2 를 조사하였다. UVA 조사 종료 후, 무혈청 배지에 비타민 C, 제조예 및 비교제조예의 각 추출물을 100 μg/mL 농도로 희석하여 처리하였다. 21시간 후, 5 mg/mL MTT(M2128, sigma)를 10 μL 넣어 3시간 배양하였다. 이후, 배지를 모두 제거하고, 150 μL DMSO를 각 well에 넣어 교반한 후, Microplate reader로 570 nm에서 흡광도를 측정하고, 하기 식으로 세포 생존율을 계산하였다.Fibroblasts (CCD-986sk) were cultured in a 96 well plate at a concentration of 5Х10 4 cells/mL for 24 hours.
