Claims (15)
- 핵산 분자(M)들을 분석하기 위한 장치(1)로,
- 비드(20)로, 상기 비드 상에 하나의 분자의 일 단부가 고정될 수 있는, 비드(20),
- 표면(520)으로, 상기 표면 상에 상기 분자의 타 단부가 고정될 수 있는, 표면(520),
- 비드(20)가 상기 표면에 대해 한 방향 운동으로 이동하도록 만들기에 적합한 액추에이터(30),
- 비드와 표면 사이의 거리를 측정하기에 적합한 센서(50)를 포함하는 핵산 분자 분석 장치에 있어서,
상기 장치는, 비드의 운동 방향을 따라 연장하는 축(X-X)과 상기 표면에 의해 형성되는 바닥부(110)를 구비하는 웰(well)(11)을 추가적으로 포함하고, 상기 웰(11)은 전기 전도성 용액(40)으로 채워지고, 비드(20)는 웰 내에 수용되며, 센서(50)는 비드(20)와 표면(520) 사이의 거리에 따라 달라지는 웰(11)의 임피던스를 측정하고 측정된 임피던스로부터 비드와 표면 사이의 거리를 결정하기에 적합한 것을 특징으로 하는, 핵산 분자 분석 장치. - 제1항에 있어서,
센서(50)는,
- 웰의 상부에 전기 전도성 용액(40)과 접촉하도록 배치되는 메인 전극(51)으로, 상기 전극에는 알려진 전위가 걸려지고,
- 웰(11)의 바닥부(110)에 있는 제2 전극(52)으로, 분자가 고정될 수 있는 표면(520)을 구비하는 제2 전극(52), 및
- 전극들 사이에 흐르는 전류를 측정하기에 적합한 전자 회로(53)를 포함하는 것을 특징으로 하는, 핵산 분자 분석 장치. - 제2항에 있어서,
전자 회로(53)는,
- 제2 전극(52)에 연결되는 전류-전압 증폭기(530),
- 전류-전압 증폭기(530)의 출력 전압을 측정하기에 적합한 전압계(531), 및
- 측정된 전압으로부터 웰의 임피던스를 연산하기에 적합한 연산 회로(532)를 포함하는 것을 특징으로 하는, 핵산 분자 분석 장치. - 제1항에 있어서,
웰(11)의 축(X-X)을 가로지르는 단면의 면적은 웰의 바닥부(110)로부터 상부까지 순증가(strictly increasing)하는 것을 특징으로 하는, 핵산 분자 분석 장치. - 제4항에 있어서,
웰(11)의 바닥부에서의 단면적은 비드(20)의 최대 단면적보다 큰 것을 특징으로 하는, 핵산 분자 분석 장치. - 제4항 또는 제5항에 있어서,
웰(11)은 원뿔대 형상을 갖는 것을 특징으로 하는, 핵산 분자 분석 장치. - 제4항 또는 제5항에 있어서,
웰(11)의 단면적은 웰의 바닥부(110)로부터의 거리에 따라 선형 증가하는 것을 특징으로 하는, 핵산 분자 분석 장치. - 제7항에 있어서,
웰(11)의 단면은 웰의 바닥부(110)에서는 원형이고, 웰의 바닥부로부터의 거리에 따라 웰(11)의 축(X-X)에 직각인 방향으로 선형으로 성장하는 것을 특징으로 하는, 핵산 분자 분석 장치. - 제1항에 있어서,
액추에이터는 웰의 축(X-X) 방향을 따라 변위 가능하게 장착되는 적어도 하나의 자석(31)을 포함하며, 비드(20)는 상자성 재료(paramagnetic material)로 제조되어, 웰의 바닥부(110)와 자석(31) 사이에 삽입되는 것을 특징으로 하는, 핵산 분자 분석 장치. - 제2항에 있어서,
복수의 동일한 비드(20)와, 각각이 하나의 비드를 수용하기에 적합한 복수의 동일한 웰(11)를 포함하는 것을 특징으로 하는, 핵산 분자 분석 장치. - 제10항에 있어서,
상기 장치는 복수의 제2 전극(52)들을 포함하고, 각각의 제2 전극은 각각의 웰 바닥부에 배치되어, 각각의 비드가 고정되는 표면을 형성하며,
전자 회로는 복수의 전류-전압 증폭기(530)를 포함하고, 각각의 전류-전압 증폭기(530)는 각각의 제2 전극에 연결되며, 연산 회로(532)는 동시에 전류-전압 증폭기들의 출력 전압들을 측정하여 각각의 웰의 임피던스를 연산하기에 적합한 것을 특징으로 하는, 핵산 분자 분석 장치. - 제10항 또는 제11항에 있어서,
상기 장치(1)는 전기 절연성 재료로 된 플레이트(10)를 포함하고, 플레이트(10) 내에 웰들이 형성되고, 모든 웰들은 플레이트(10)의 상부면(101)에서 개방되며, 액추에이터(30)는 자성 재료로 제조되는 복수의 막대(32)를 추가적으로 포함하고, 각각의 막대는 상부면(101)에 배치되어 인접한 두 웰(11)들 사이에서 연장하는 것을 특징으로 하는, 핵산 분자 분석 장치. - 제12항에 있어서,
각각의 막대(32)는 길이가 10㎛ 미만이고, 각각의 비드(20)는 직경이 1㎛ 이하인 것을 특징으로 하는, 핵산 분자 분석 장치. - 핵산 분자의 분석 방법으로,
상기 방법은 제1항에 따른 장치에 의해 수행되고, 상기 방법은 비드와 웰의 바닥부 사이의 거리를 측정하는 단계(300)를 적어도 하나 포함하며, 각각의 측정 단계는 웰 내 비드의 위치를 결정하는 것을 고려하여 웰의 임피던스를 측정하는 것을 특징으로 하는, 핵산 분자 분석 방법. - 제14항에 있어서,
상기 방법은 웰의 상부에 전기 전도성 용액(40)과 접촉하도록 배치되는 메인 전극(51)으로, 상기 전극에는 알려진 전위가 걸려지는, 메인 전극(51); 웰(11)의 바닥부(110)에 있는 제2 전극(52)으로, 분자가 고정될 수 있는 표면(520)을 구비하는 제2 전극(52); 및 전극들 사이에 흐르는 전류를 측정하기에 적합한 전자 회로(53)를 포함하는 장치에 의해 수행되고,
상기 방법은 적어도 하나의 분자를 비드와 웰의 바닥부에 고정시키는 예비 단계(100)를 포함하며, 상기 예비 단계는,
- 분자가 고정되는 적어도 하나의 비드를 용액(40) 내에 배치하는 단계(110),
- 적어도 하나의 비드가 웰의 바닥부에 접촉하도록 하기 위해 메인 전극(51)과 제2 전극(52) 사이에 제1 전위차를 가하는 단계(120), 및
- 웰의 바닥부와 결합하지 않은 비드들이 웰 밖으로 빠져 나오게 하기 위해, 전극들 간에 전위차를 수정하는 단계(130)를 포함하는 것을 특징으로 하는, 핵산 분자 분석 방법.
