Abstract
Description
도 2a 내지 2d는 hCD39 안티센스 올리고뉴클레오타이드의 hCD39 mRNA 녹다운 효율을 보여주는 그림으로, 각각 인간 암세포주 HDLM-2(인간 호지킨 림프종)에서 1차 및 2차 스크리닝 시의 녹다운 효율[도 2a(파트 1과 2) 및 도 2b(파트 1과 2)]과, A-172 (인간 교아종)에서 1차 및 2차 스크리닝 시의 녹다운 효율[도 2c(파트 1과 2) 및 도 2d(파트 1과 2)]를 각각 나타낸다. HDLM-2 및 A-172 세포에 3일간 각각의 안티센스 올리고뉴클레오타이드 10μΜ를 처리하였다. 음성 대조군으로서, 세포들에 CGTTTAGGCTATGTACTT (W2014154843 Al 참조)의 서열을 가지는 안티센스 올리고뉴클레오타이드인 negl를 처리하였다. 처리하지 않은 세포와 비교한 상대적인 hCD39 mRNA 발현량을 나타내었다. 발현값은 하우스키핑 유전자인 HPRT1의 발현량에 대해 표준화하였다. 표시된 값은 3배수 웰 평균의 +/- 표준편차이다.

도 6은 3차 스크리닝을 통한 추가적인 안티센스 올리고뉴클레오타이드 설계를 보여주는 그림이다. 이들 안티센스 올리고뉴클레오타이드는 1차 및 2차 스크리닝에서 선정된 것들 중 효율적인 안티센스 올리고뉴클레오타이드에 기반한다. 따라서, hCD39 안티센스 올리고뉴클레오타이드들은 인간암세포주 HDLM-2(인간 호지킨 림프종)(도 6a) 및 A-172(인간 교아종)(도 6b)에서 시험하였다. 도 8은 A04040H(서열목록 제3서열) 및 A04045H(서열목록 제4서열)에 의한 농도 및 시간 의존적인 CD39 단백질 녹다운을 보여주는 그림이다. 각 안티센스 올리고뉴클레오타이드를 3, 4 및 6일간 처리 후 HDML-2 세포의 유세포분석을 통해 CD39 단백질 발현을 측정하였다. 대조군 처리 후, 세포들에 negl을 정해진 농도로 3, 4 및 6일간 처리하였다. 미처리 대조군 세포(=1)와 비교한 상대적 발현량을 표시하였다.
도 9는 항-CD3 항체의 존재 하에 10μΜ의 hCD39 특이적 ASO A04040H(검은색 컬럼) 또는 대조군 올리고뉴클레오타이드 S6(흰색 컬럼)를 1차 인간 CD8+ 및 CD4+ T 세포에 처리한 결과를 보여주는 그림이다. 대조군 세포는 항-CD3로 활성화되었으나 어떤 올리고뉴클레오타이드도 처리하지 않았다(줄무늬 컬럼). 이후, 올리고뉴클레오타이드 및 항-CD3를 제거하고 올리고뉴클레오타이드 제거 3, 6 및 11일 후 유세포분석으로 hCD39 단백질 발현을 측정하였다. 도 lOa-lOc는 hCD39 녹다운이 JIYOYE 세포의 생존성과 ATP 농도에 미치는 영향을 보여주는 그림이다. JIYOYE 세포에 안티센스 올리고뉴클레오타이드 A04040H(서열목록 제3서열) 또는 negl를 6일간 총 5μΜ를 처리하였다. 3일 후 배지를 신선한 올리고뉴클레오타이드-함유 배지로 교환하고 6일 째에 유세포분석으로 hCD39 단백질 녹다운 효능을 분석하였다. 잔여 hCD39 발현 및 올리고뉴클레오타이드-처리 세포의 생존성은 미처리 세포와 비교하여 표시하였다(도 10a 및 10b). 6일 후, 20μΜ of the CD39의 저분자 억제제인 ARL67156 트리소듐 염 20μΜ를 ASO 미처리 세포에 첨가하고 1시간 동안 37℃에서 배양하였다. 이후, 2μΜ ATP를 세포 또는 각 조건의 세포가 없는 세포 배양배지에 첨가하고 30분 후 ATP Bioluminescence 어세이 Kit(Roche)을 이용하여 세포 상층액 또는 세포 배양배지의 ATP 농도를 측정하였다(도 10c).
도 11a-11d는 hCD39 단백질의 녹다운(도 11a) 및 MACS로 말초혈에서 분리한 일차 인간 CD8+ T 세포의 생존성(도 11b)을 보여준다. CD8+ T 세포를 플레이트-결합 항-인간 CD3(OKT-3) 항체로 활성화시켰다. 활성화된 세포에 A04040H(서열목록 제3서열)이 보충된 RPMI-1640 배지 및 negl 5μΜ가 보충된 RPMI-1640 배지를 각각 총 6일 동안 처리하였다. 3일 후, 배지를 5μΜ A04040H(서열목록 제3서열) 및 neg1를 각각 함유하는 신선한 배지로 교체하고, 유세포분석으로 hCD39 단백질 녹다운 효율(도 11a) 및 세포 생존성(도 11b)을 측정하였다. 잔여 hCD39 발현 및 생존성(7-AAD 양성세포의 중앙값)은 미처리 세포와 비교하여 표시하였다(도 11a-11b). 같은 날, 세포를 수집하고 세척 후 다시 일정한 세포수(96-웰플레이드에 150.000 세포/웰)로 3배수로 플레이팅하였다. 이후, 2μΜ(도 11c) 또는 20μΜ(도 11d) ATP를 세포 또는 세포가 없는 세포 배양배지에 첨가하고 30분 후 ATP Bioluminescence 어세이 Kit(Roche)을 이용하여 세포 상층액 또는 세포 배양배지의 ATP 농도를 측정하였다.
