[실시예 2] 비타민 B9 정량분석[Example 2] Quantitative analysis of vitamin B9

준비된 시료는 액체 크로마토그래피(Nanospace SI-2 3023, SHISEIDO, Tokyo, Japan)를 사용하여 분석하였다. 물질의 분리와 정량을 위해 CAPCELL PAK UG 120 S5 column (4.6 mm id x 250 mm)을 사용하였고, 이동상은 PIC reagent 10 ml + 3th D.W 500 ml혼합액을 이동상 A, PIC reagent 10 ml + 60% Methanol 500 ml 혼합액을 이동상 B로 사용하였다. PIC reagent는 1-Heptanesulfonic acid 1 g + D.W 10 ml + Acetic acid 10 ml를 혼합하여 제조하였으며, 이동상은 0.5 ml/min의 유속으로 흘려주었다. 컬럼 온도는 40℃로 유지되었고, 20 ㎕의 시료를 주입하여 270 nm 파장의 UV 검출기를 이용하여 비타민 B9의 생성여부를 확인하였다. The PIC reagent was prepared by mixing 1 g of 1-Heptanesulfonic acid + 10 ml of D.W + 10 ml of Acetic acid, and the mobile phase was flowed at a flow rate of 0.5 ml/min. The column temperature was maintained at 40° C., and 20 μl of the sample was injected to confirm the production of vitamin B9 using a UV detector with a wavelength of 270 nm.

비타민 B9의 표준물질 농도는 1mg/ml로 준비하였으며, 도 1에서 나타난 바와 같이 검량선의 신뢰도는 99.9% (R2=0.9999)이상임을 확인하였다. 이를 토대로 프로바이오틱스 상등액 내의 비타민 B9 생산량을 정량하여 표 2에 나타내었으며, 생물자원센터에서 락토코커스 락티스 종에 속하는 여러 균주를 구매하여 비타민 B9 생성능을 비교하였다. 또한 장모사배지에서 비타민 B9는 검출되지 않았다.The standard concentration of vitamin B9 was prepared at 1 mg/ml, and as shown in FIG. 1, it was confirmed that the reliability of the calibration curve was 99.9% (R2=0.9999) or higher.
