단백질 분해 관련 프로테아제 활성은 다음과 같은 방법으로 측정하였다. 검사를 위한 균주는 실험예 1에서의 조건에 맞춰 배양한 균주를 사용하였다. 각 시료의 10ㅅl를 이용하여 실험하였으며 형성된 clear zone에 의해 프로테아제 활성이 구분되었다. 프로테아제 활성을 확인하기 위해 식물성 단백질인 pea protein과 동물성 단백질인 skim milk를 이용한 배양배지를 사용하였다. 각 배양배지는 protein 28g/L, tryptone 5g/L, yeast extract 2.5g/L, glucose 1g/L, agar 15g/L로 만들었으며 단백질 응고를 막기 위해 protein + 50% D.W, 나머지 조성물 + 50% D.W로 만들어 멸균 후 혼합하여 사용하였다. 단백질 배양배지에 프로바이오틱스를 10ㅅl 떨어트린 후 37℃에서 48시간 배양하였고, 배양 후 프로바이오틱스 spot 주위에 단백질이 분해되어 생겨난 clear zone의 길이를 측정하여 프로테아제 활성을 평가하였다.Protease activity related to protein degradation was measured by the following method.
