Abstract
Description
도 2는 Lactobacillus paracasei IJH-SONE68주의 균체외 다당체의 음이온 교환 크로마토그래피(TOYOPEARL DEAE-650M 수지(토소 주식회사))에 의한 분리 프로파일이다. 0 mM~500 mM의 구배 농도의 NaCl(파선)로 균체외 다당체를 용출시키고, 각 획분 중의 균체외 다당을 페놀황산법에 의해 490 nm에서 모니터하였다(직선).
도 3은 Lactobacillus paracasei IJH-SONE68주의 균체외 다당체를 음이온 교환 칼럼 크로마토그래피에 의해 정제하여 얻어진 균체외 중성 다당을, 프로톤-NMR 및 카본-NMR 처리하여 얻어진 각각의 NMR 프로파일을 나타낸다. 도 3의 (A)가 프로톤-NMR의 NMR 프로파일이고, (B)가 카본-NMR의 NMR 프로파일이다.
도 4는 NMR 프로파일로부터의 균체외 중성 다당의 구조 해석 결과를 나타낸다. 이 구조 해석 결과로부터, Lactobacillus paracasei IJH-SONE68주의 균체외 중성 다당은, N-아세틸글루코사민이 α-1,6 결합에 의해 연결된 구조를 갖는 것이 분명해졌다.
도 5는 Lactobacillus paracasei IJH-SONE68주의 게놈 DNA의 pcel 클러스터 및 pce2 클러스터라고 명명한 균체외 다당 생합성 유전자 클러스터의 조직도이다.
도 6은 (A)가 Lactobacillus paracasei IJH-SONE68주, ATCC 334주 및 JCM 8130T주의 3개의 유산균 사이의 게놈 재편성 맵을 나타내고, (B)가 Lactobacillus paracasei IJH-SONE68주의 pce1 유전자 클러스터와 JCM 8130T주에서의 유전자 클러스터의 대응이다.
Claims (15)
- 락토바실러스·파라카제이(Lactobacillus paracasei) IJH-SONE68주(수탁 번호 NITE BP-02242)인 유산균의 N-아세틸글루코사민이 α-1,6 결합에 의해 연결된 구조를 갖는 중성 다당.
- 제1항에 있어서,
히알루로니다제 저해 활성을 갖는 중성 다당. - 제1항에 있어서,
락토바실러스·파라카제이(Lactobacillus paracasei) IJH-SONE68주(수탁 번호 NITE BP-02242)인 유산균의 배양물로부터 얻어지는 다당체를 이온 교환 크로마토그래피에 의해 분리 정제하여 얻을 수 있는 중성 다당. - 제1항에 있어서,
(1) 락토바실러스·파라카제이(Lactobacillus paracasei) IJH-SONE68주(수탁 번호 NITE BP-02242)인 유산균의 배양물로부터 원심 분리에 의해 균체를 제거하는 공정;
(2) 공정(1)에서 얻어진 배양물로부터, 에탄올 또는 아세톤에 의한 침전에 의해 다당체 및 단백질을 침전으로서 회수하는 공정;
(3) 회수한 침전으로부터 단백질을 제거하여 균체외 다당체를 회수하는 공정; 및
(4) 회수된 균체외 다당체를 음이온 교환 크로마토그래피에 의해 분리 정제하는 공정
에 의해 얻을 수 있는 중성 다당. - 제1항 내지 제4항 중 어느 한 항에 기재된 중성 다당을 함유하는 조성물.
- 제5항에 있어서,
락토바실러스·파라카제이(Lactobacillus paracasei) IJH-SONE68주(수탁 번호 NITE BP-02242)인 유산균의 산성 다당으로서, 글루코스, 만노스, 갈락토오스 및 람노스로 구성되는 산성 다당을 더 함유하는 조성물. - 제5항에 있어서,
조성물이 음식품 조성물인 조성물. - 제7항에 있어서,
음식품이, 기능성 식품, 발효 식품, 음료 또는 서플리먼트인 조성물. - 제5항에 있어서,
