실시예 1

시약
페노피브레이트는 Sigma (Cat# F6020)로부터 구매하였다. PPARα 길항물질은 Sigma (Cat# GS04S)로부터 구매하였다. IL-lβ(Mm00434228-ml) 및 PGC-lα RT-PCR 분석은, Life Technologies (Carlsbad, CA)로부터 구매하였다. 세포 배양 시약은 또한 Life Technologies로부터 수득되었다.
세포 배양물
마우스 중간뇌 세포주, MN9D는 페노피브레이트의 신경보호 효과를 평가하기 위해 사용되었다. MN9D 세포는, 5 % 이산화탄소, 37 ℃에서 3.7 g/L 탄산수소나트륨 및 10 % 소태아혈청으로 보충된 Dulbecco's 개량된 이글 배지(Dulbecco's Modified Eagle Medium, DMEM)에서 배양되었다. PGC-lα 상향-조절을 매개하는데 있어서 페노피브레이트의 신경보호 효과는, PD의 MPTP 독성물질 마우스 모델에서 평가되었다. MPTP는 모델 PD에 광범위하게 사용된 강력한 신경독성(neurotoxicant)이다. MPTP는 ROS를 유도하고, 세포 사멸을 유도하는, 미토콘드리아의 복합체 Ⅰ을 억제한다. 로타-로드 성능(Rota-Rod Performance)

TaqMan 미세 유체 qRT-PCR(TaqMan micro fluidics qRT-PCR)
RNA는, 어떠한 오염시키는 게놈의 DNA를 분해시키기 위해, RQ1 DNase (Promega, Madison, WI)로 처리된 다음에, 페놀:클로로포름 추출하고, 에탄올/LiCl 침전을 받았다. 생화학적 검정
(1) STR DA 레벨 결정은 HPLC에 의해 측정된다. (2) 세포 사멸 검출 키트가 TUNEL 분석에 대해 사용된다. (3) RET 및 AKT의 인산화반응, 세포사멸의 단백질 레벨(SN 조직에서 분열된 카스파제-3, PARP, Bcl-2 및 Bax)는 웨스턴 블롯에 의해 측정된다. (4) BDNF, GDNF, 및 VIP는 ELISA에 의해 측정된다.
실시예 3
페노피브레이트는 PGC-1α 유전자 발현 미세아교세포주 BV2를 유도하고, 투여량-의존적인 방식으로 LPS-매개된 프로-염증성 사이토카인 IL-1β 생산을 저해한다. 게다가, PGC-1α의 siRNA 녹다운과 함께, BV1 세포에서 상기 페노피브레이트-매개된 항-염증성 효과는 PGC-1α를 필요로 하는 것을 나타낸 것이다. siRNA만이 PGC-1α 유전자 발현을 부분적으로 녹 다운(knocks down) 시키는 점을 고려하여, 동형의 및 이형의 PGC-1α 녹아웃 마우스(knockout mice)(PGC-1α-/- 및 PGC-1α-/+)로부터 유도된 주요한 CNS 세포의 사용은, 페노피브레이트 효과를 조정하는데 있어서 PGC-1α의 필요불가결한 역할의 보다 결정적인 증거를 제공할 것이다. 시약
배지, 인산염-완충된 염분(PBS) 및 소태아혈청(FBS)를 포함하는 조직 배양 물질은 Life Technologies로부터 수득되었다. Escherichia coli LPS로부터의 지질다당류는 Sigma (Cat# L6S29)로부터 구매되었다. 항체는 Santa Cruz Biotechnology로부터 구매되었다.
세포 배양
신생아 마우스(1-day old; PGC-lα+/+ 또는 PGC-lα+/-)의 대뇌피질은 뇌척수막을 벗겼고, 헤레스 평형 염류 용액(Hepes balanced salt solution)(HBSS; Mediatech Inc., Herndon, VA)에서 갈아졌다. 세포는, 이글'스 염(Earle's salts), I-글루타민, 0.01 % 피루브산염, 0.6 % 글루코스, 4 % 소태아 혈청, 및 6 % 말 혈청(완전 배지)를 함유하는 최소 필수 배지(MEM; Invitrogen, Frederick, MD)에서 분리되었고, 원심분리되고, 재현탁되고, T75 플라스크 당 하나의 뇌의 밀도에서 10 ml 완전 배지를 함유하는 플라스크 내로 평판된다. 배양물은 5 % CO2 하에서 37 ℃에서 성장되었다. 하루 후에, 상기 플라스크는 세포 파쇄물(cell debris)을 제거하기 위해 서서히 두드려졌고(tapped), 배지가 제거되고 신선한 완전 배지로 보충되었다. 배양물은, 상기 미세아교세포가 플라스크를 두드리고, 미세아교세포-풍부한 배지(microglia-enriched containing medium)를 수집함으로써 채취되는 시간인 대략 12 일 동안 상기와 같이 성장되었다. 미세아교세포는 원심분리(1,000 rpm, 5 min)에 의해 펫릿되고(pelleted), 0.01 % 피루브산염, 0.6 % 글루코스, 및 5 % 소태아 혈청을 함유하는 MEM에서 재현탁되고, 계산되었다(enumerated).
BV-2 세포(쥐과 미세아교세포)는 10 % 소태아 혈청 및 항생물질(페니실린, 100 U/mL, 스트렙토마이신 100 ㎍/mL)으로 보충된 Dulbecco's Modified Eagle Medium에서 유지되었다. 항생물질은 상기 페노피브레이트 평가 연구를 위해 제외되었다.
실시예 4
AMPK는 에너지 센서이고, PGC-lα를 인산화할 수 있다. AMPK가 PGC-lα를 인산화하고, 그 다음에 PGC-lα가 이의 자신의 유전자 발현을 증가시키기 위해 근세포 인핸서 인자(myocyte enhancer factor)(MEF)-결합 부위에 결합하는, 양성 피브백(positive feedback)을 통해 작용함이 타당한 것 같다. 따라서, AMPK 신호 전달이 CNS 세포에서의 페노피브레이트 효과를 조정하는데 포함되었는지 여부를 결정하기 위해, BV2 세포는 1 시간 동안 페노피브레이트의 다양한 투여량으로 처리된 다음에, 세포 용해물은 웨스턴 블롯팅 분석을 위해 수집되었다. AMPK 인산화는, 투여량-의존적인 방식으로 페노피브레이트 처리에 대한 대응으로 증가되었다(도 2a). 그 다음에, BV2 세포는 0.5 시간 동안 AMPK 억제제 화합물 C로 처리된 다음에 또 다른 1 시간 동안 페노피브레이트로 처리되었다. AMPK 인산화에서 화합물 C의 농도-의존적인 억제 효과가 관찰되었다(도 2b). BV2 세포에서 페노피브레이트 매개된 항-염증에서의 화합물 C의 억제 효과가 테스트되었다. 화합물 C는 0.5 시간 동안 BV2 세포 배양물에 첨가된 다음에, 밤새 페노피브레이트 처리되었다. 화합물 C는, 투여량 의존적인 방식으로 BV2 세포에서 페노피브레이트의 항-염증성 효과를 약화시켰다(도 2c). 이러한 데이터는, AMPK 활성이 CNS 세포에서 매개된 페노피브레이트 효과에서 중요한 역할을 함을 시사한다.

Claims (11)

상기 조성물은 상기 대상의 신경 세포 또는 신경 세포군에서 PGC-lα 발현을 유도하는 유효량의 페노피브레이트를 포함하고,
상기 대상은 신경변성 질병을 갖지 않는 대조군 대상과 비교해, 신경변성 질병 및 감소된 PGC-lα 발현을 갖는 것으로 결정된 것인, 약제학적 조성물.
- 제1항에 있어서,
상기 신경변성 질병은 파킨슨 질병, 파킨슨-플러스 증후군(Parkinson-plus syndrome), 가족성 치매, 알츠하이머 질병, 헌팅턴 질병, 다발성 경화증, 루이 소체 치매(dementia with Lewy bodies), 경도 인지 장애, 망막의 신경퇴화, 및 근위축성 측색 경화증(Amyotrophic Lateral Sclerosis)으로 이루어진 군으로부터 선택되는 것인, 약제학적 조성물.
- 제2항에 있어서,
