Total antioxidant capacity assay는 시료 내에 존재하는 ascorbate, uric acid, GSH, vitamin E 등과 같은 소분자(Small molecules) 또는 albumin, transferrin과 같은 단백질에 의하여 Cu2+가 Cu+로 환원되는 정도를 측정함으로써 시료의 total 항산화능(antioxidant capacity)을 평가하는 것이다. 동애등에 A.C 및 D.W 추출물의 total antioxidant capacity는 Total Antioxidant Capacity Colormetric Assay Kit(Biovision, CA)를 구매하여 제조자로부터 제공되는 방법에 준하여 분석하였다. 각 시료(실시예 1, 2)의 추출물(50μg/mL, 100μg/mL, 500 μg/mL, 1000μg/mL) 100μL에 Cu2+ working solution 100μL를 혼합한 후 상온에서 빛을 차단하여 90분 동안 반응시킨 다음 570nm에서 흡광값을 측정하였다. 시료의 total antioxidant capacity는 표준물질인 trolox(수용성 tocopherol)를 동일한 조건으로 반응시켜 작성한 표준 곡선(standard curve)에 준하여 trolox equivalent capacity (nM TEC/mg sample)로 환산하여 나타내었다. α,α'-diphenyl-β-picrylhydrazine (DPPH)는 안정한 자유라디칼(free radical)을 포함하는 화합물로 시료내의 항산화물질로 부터 전자 혹은 수소를 제공받아 환원되는 정도를 측정함으로써 시료의 DPPH radical scavenging activity를 평가하는 것이다. 동애등에 A.C 및 D.W 추출물의 DPPH free radical scavenging activity를 평가하기 위하여, 상기 시료 100μL와 methanol에 0.2mM가 되도록 용해한 DPPH 용액 50μL를 혼합하여 상온에서 빛을 차단하여 30분 동안 반응시켰다. 양성대조구인 tocopherol의 농도는 반응액에서의 최종 농도가 20μM이 되도록 준비하여 사용하였다. 반응이 끝난 혼합액은 517 nm에서 흡광값을 측정하여 아래의 식을 이용하여 DPPH radical scavenging activity를 계산하였다.
