Abstract
본 발명은, 글루타민 tRNA 5' 말단 유래의 단편(Gln 5' tRF)이 과발현된 형질전환 예쁜꼬마선충, 이의 제조방법, 및 상기 형질전환 예쁜꼬마선충을 이용한 노화 관련인자의 스크리닝 방법에 관한 것이다. 본 발명에서 제공하는 형질전환 예쁜꼬마선충 모델은, 글루타민 tRNA 5' 말단 유래의 단편(Gln 5' tRF)이 과발현되어 노화가 억제된 동물모델로서, 본 발명의 모델을 이용하면 항노화 메커니즘을 손쉽게 규명할 수 있고, 이를 통하여 항노화 신약 개발, 노화 유도 물질 스크리닝 등 다양한 연구 분야에서 크게 기여할 수 있을 것이다.
Description
본 발명은, 글루타민 tRNA 5' 말단 유래의 단편(Gln 5' tRF)이 과발현된 형질전환 예쁜꼬마선충, 이의 제조방법, 및 상기 형질전환 예쁜꼬마선충을 이용한 노화 관련인자의 스크리닝 방법에 관한 것이다.
약 20-50bp 정도의 small non-coding RNA는 단백질로 발현되는 mRNA와 달리 RNA 그 자체로 기능을 하는 RNA로서, 최근 다양한 small RNA들이 생체내에서 중요한 기능들을 한다는 것이 밝혀지고 있다. Small RNA에는 마이크로 RNA(miRNA), 짧은 간섭 RNA(siRNA), PIWI 결합 RNA(piRNA) 등이 있으며, 자신의 염기서열과 상보적인 염기서열을 가지는 RNA 타겟을 간섭 및 억제하는 기능을 한다.
tRNA 유래 단편(tRNA-derived fragments; tRFs)은 다양한 개체에서 새롭게 발견된 small non-coding RNA의 한 종류이다. 사람을 비롯하여 쥐, 꼬마선충, 애기장대 등 다양한 종에서 발견이 되는 것으로 보아 진화적으로 보존된 기능을 할 것이라고 추측되고 있다. 또한, tRFs는 이미 만들어진 mature tRNA에서 특정 부분이 절단됨으로써 생성되며, 임의의 염기서열이 아닌 특정한 염기서열로만 발견이 되므로, 단순히 tRNA가 랜덤하게 분해되어 생긴 단편이 아닌 특이적인 기능을 하는 단편으로 여겨진다. 이때, mature tRNA에서 절단되는 부위에 따라 4가지 종류의 단편이 생성될 수 있는데, 예를 들면 5' tRF, 3' CCA tRF, 3' U tRF, internal tRF 로 분류될 수 있다(도 1 참조).
tRFs는 암세포주, 바이러스 감염세포, 지방조직 유래 줄기세포, 마우스 혈청 등에서 발견된 바 있고, 스트레스 등 다양한 생리적 상황에서 단편들이 생성되며, small RNA와 유사한 조절기능이 보고된 바 있으나, 아직까지 항노화 작용과 관련된 기능은 밝혀진 바 없는 실정이다.
특히, tRNA 유전자의 특성상 tRFs의 서열분석에는 어려움이 따르는데, 예를 들면 tRNA 유전자는 게놈 전체에 다중 카피로 존재하고 이들 중 일부는 동일하나 일부는 서열에 차이가 있기 때문에, tRF가 유래된 정확한 게놈 origin을 확인하는 것이 어렵다. 또한, mature tRNA는 변형이 심하기 때문에 mature form으로부터 생성된 tRF와 상응하는 게놈 서열을 정확히 매칭하기도 쉽지 않다.
한편, 예쁜꼬마선충(Caenorhabditis elegans)은 알에서 부화되어 L1, L2, L3, L4의 4단계의 유충단계를 거쳐 성충이 되는데 약 3일 정도만이 소요되고 수명도 약 3주 정도 되어 발달실험이나 수명실험에 적합한 모델동물이다. 또한, 유전자 조작이 매우 쉽고, 크기가 작아 동시에 많은 수를 비교적 저렴하게 키울 수 있고, 몸이 투명해 현미경 등으로 꼬마선충의 내부를 직접 관찰하거나 형광을 관찰하기도 아주 쉽다.
또한, 예쁜꼬마선충은 사람의 질병에 관여하는 많은 유전자가 보존되어 있어 이를 이용한 유전자 발굴 및 분석, 이를 타겟으로 하는 신약 개발의 가능성이 타진되고 있다. 예를 들어, 포유류의 대사 및 노화 조절에 중요한 유전자들인 인슐린 신호전달 체계의 유전자들, 세포 사멸 조절 유전자들, 면역 조절에 중요한 MAP 인산화효소 신호전달체계 등이 예쁜꼬마선충에도 존재하며, 이들의 조절 기전 규명이 활발하게 연구되고 있다.
이에, 본 발명자들은, 예쁜꼬마선충이 유전자 조작이 용이하고 항노화 메커니즘 및 병리학적 역할을 밝히는데 좋은 동물모델이 될 수 있다는 전제하에, 또한 예쁜꼬마선충이 나이-의존적으로 특정 tRF의 레벨이 변화한다는 것을 발견하고, tRFs와 노화/수명과의 관련성에 주목하여, 특정의 tRNA 유래 단편(tRFs)을 과발현하는 예쁜꼬마선충 형질전환 동물을 완성하게 되었다.
따라서, 본 발명의 목적은, 글루타민 tRNA 5' 말단 유래의 단편(Gln 5' tRF)이 과발현된 형질전환 예쁜꼬마선충(C. elegans), 이의 제조방법, 및 상기 형질전환 예쁜꼬마선충을 이용하여 노화 관련 인자를 스크리닝하는 방법을 제공하는 것이다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
본 발명은, 글루타민 tRNA 5' 말단 유래의 단편(Gln 5' tRF)이 과발현된, 형질전환 예쁜꼬마선충(Caenorhabditis elegans)을 제공한다.
본 발명의 일 구현예에서, 상기 예쁜꼬마선충은 항노화 동물모델용인 것을 특징으로 한다.
본 발명의 다른 구현예에서, 상기 Gln 5' tRF는 서열번호 1의 염기서열로 이루어진 것을 특징으로 한다.
본 발명의 또 다른 구현예에서, 상기 예쁜꼬마선충은 미토콘드리아 활성화에 의하여 항노화 효과를 보이는 것을 특징으로 한다.
본 발명의 또 다른 구현예에서, 상기 예쁜꼬마선충은 DR(Dietary Restriction)/AMPK(AMP-dependent kinase) 경로에 의해 항노화 효과를 보이는 것을 특징으로 한다.
또한, 본 발명은, (a) U6 프로모터와 Gln 5' tRF DNA와 U6 3' UTR(untranslated region)을 포함하는 재조합 벡터를 제조하는 단계; 및 (b) 상기 재조합 벡터를 예쁜꼬마선충에 주입하는 단계를 포함하는, 상기 형질전환 예쁜꼬마선충(Caenorhabditis elegans)의 제조방법을 제공한다.
본 발명의 일 구현예에서, 상기 단계 (a)의 U6 프로모터는 small RNA 과발현에 특이적인 프로모터인 것을 특징으로 한다.
본 발명의 다른 구현예에서, 상기 단계 (b)의 재조합 벡터를 예쁜꼬마선충에 주입하는 방법은 미세주입법(microinjection)인 것을 특징으로 한다.
또한, 본 발명은, 상기 형질전환 예쁜꼬마선충을 이용한, 노화 유도 후보물질의 스크리닝 방법을 제공한다.
본 발명의 일 구현예에서, 상기 스크리닝 방법은, (a) 상기 형질전환 예쁜꼬마선충에 후보 물질을 처리하는 단계; (b) 상기 후보 물질 처리 후 미토콘드리아 활성을 측정하는 단계; 및 (c) 상기 미토콘드리아 활성이 후보 물질을 처리하지 않은 대조군에 비해 감소한 후보 물질을 선별하는 단계를 포함하는 것을 특징으로 한다.
본 발명의 다른 구현예에서, 상기 스크리닝 방법은, (a) 상기 형질전환 예쁜꼬마선충에 후보 물질을 처리하는 단계; (b) 상기 후보 물질 처리 후 AMPK(AMP-dependent kinase) 활성을 측정하는 단계; 및 (c) 상기 AMPK 활성이 후보 물질을 처리하지 않은 대조군에 비해 감소한 후보 물질을 선별하는 단계를 포함하는 것을 특징으로 한다.
본 발명에서 제공하는 형질전환 예쁜꼬마선충 모델은, 글루타민 tRNA 5' 말단 유래의 단편(Gln 5' tRF)이 과발현되어 노화가 억제된 동물모델로서, 본 발명의 모델을 이용하면 항노화 메커니즘을 손쉽게 규명할 수 있다.
또한, 본 발명에서 제공하는 항노화 형질전환 예쁜꼬마선충의 제조방법은, 통상의 단백질이 아닌 tRNA 조각 RNA를 과발현시킨 것으로서 small RNA를 과발현하는 데에 특이적인 프로모터를 사용하였는 바, 이와 같은 방법을 이용하면 다양한 small RNA를 과발현하는 예쁜꼬마선충의 제조도 용이하게 할 수 있을 것이다.
또한, 본 발명에 의하면, tRFs의 항노화 기능을 규명함으로써, 항노화 신약 개발, 노화 유도 물질을 포함하는 다양한 인자의 스크리닝 등 많은 연구분에야 크게 기여할 수 있을 것으로 기대된다.

본 발명에서 '예쁜꼬마선충'은 학명으로는 Caenorhabditis elegans 이며 흙에서 박테리아를 먹고사는 선충류로서 유전학, 분자생물학, 세포생물학 실험에서 널리 사용된다.

본 발명에서 'tRNA 유래 단편(tRNA-derived fragments; tRFs)'은 small non-coding RNA의 한 종류로서 mature tRNA에서 특정 부분이 절단됨으로써 생성된 특정 염기서열의 단편을 의미한다.
본 발명에서 'Gln 5' tRF'란 글루타민 tRNA의 5' 말단에서 유래된 단편으로서 특별한 제한은 없으나, 서열번호 1의 염기서열로 구성된 단편이거나, 이와 80% 이상의 상동성이 있는 것이 바람직하다. 이때 "상동성"은 공통된 조상 핵산 서열로부터 천연적으로 또는 인공적으로 유래된 것을 의미하며 임의의 천연 발생 핵산이 임의의 이용가능한 돌연변이유발에 의해 하나 이상의 선택자 코돈을 포함하도록 변형될 수 있다.
본 발명에서 '∼로부터 유래된'이라는 용어는 특정화된 분자 또는 유기체로부터 단리되거나, 그로부터의 정보를 이용하여 제조된 성분을 지칭한다.
본 발명에서 항노화 동물모델용 예쁜꼬마선충은, 메커니즘의 종류에 상관없이 항노화 형질을 발현하는 것이면 족하고 특별한 제한은 없으나, 미토콘드리아 활성화에 의하거나, AMPK(AMP-dependent kinase) 활성에 의한 것일 수 있다.
이때, 미토콘드리아는 생체내 에너지를 생성하는 세포 소기관으로서 각종 유전병/성인병 등에 중요한 역할을 하며, 미토콘드리아가 에너지를 잘 만들어 에너지 대사가 원활히 진행되도록 돕는 경우, 즉 미토콘드리아가 활성화되어 있을 경우 노화가 억제된다고 보고된 바 있다. 또한, AMPK는 체내 에너지 항상성을 조절하는 주요 효소로서 대부분의 동물 조직에서 발현되는 진화적으로 잘 보존된 효소이다. AMPK는 노화에도 연관되어 있는데 노화는 여러 장기들의 정상적인 기능 하락이 수반되는 증상으로 제2형 당뇨병, 신경 퇴행성질환, 암, 심혈관 질환과 같은 여러 질환들의 발생과 이로 인한 사망을 유도하고 있으므로, AMPK의 활성도를 증강시키거나 감소시키는 물질을 이용한 신약개발의 가능성은 매우 크고 다양하게 활용될 수 있다.
또한, 본 발명은, (a) U6 프로모터와 Gln 5' tRF DNA와 U6 3' UTR(untranslated region)을 포함하는 재조합 벡터를 제조하는 단계; 및 (b) 상기 재조합 벡터를 예쁜꼬마선충에 주입하는 단계를 포함하는, 상기 형질전환 예쁜꼬마선충(Caenorhabditis elegans)의 제조방법을 제공한다.
본 발명에 있어서, "재조합 벡터"란 유전공학에 이용되는 벡터로서, 플라스미드 벡터가 바람직하나 이에 한정되지 않고, 예를 들면 바이러스 벡터, 코스미드 벡터, 세균 인공 염색체(BAC), 효모 인공 염색체(YAC) 및 다른 비플라스미드 벡터도 이용될 수 있다.
본 발명에서 상기 재조합 벡터는 형질전환을 확인할 수 있는 표지 유전자를 더 포함할 수 있으며, 이때 표지 유전자에 특별한 제한은 없으나 녹색 형광단백질(GFP), 적색 형광단백질(RFP) 등과 같은 형광단백질 유전자인 것이 바람직하다.
본 발명에서 "프로모터"란 특정한 숙주 세포에서 작동 가능하게 연결된 핵산 서열의 발현을 조절하는 DNA 서열을 의미하며, 발현용으로 사용되는 것이라면 특별히 한정되지 않으나, small RNA의 발현에 적합한 U6 프로모터인 것이 바람직하다. 또한, 상기 "작동 가능하게 연결(operably linked)"이란 하나의 핵산 단편이 다른 핵산 단편과 결합되어 그의 기능 또는 발현이 다른 핵산 단편에 의해 영향을 받는 것을 말한다.
아울러, 본 발명의 재조합 벡터에는 전사를 조절하기 위한 임의의 오퍼레이터 서열, 적합한 mRNA 리보좀 결합 부위를 코딩하는 서열 및 전사 및 해독의 종결을 조절하는 서열을 추가로 포함할 수 있다.
상기 재조합 벡터에 도입된 Gln 5' tRF의 DNA는 각각 프로모터의 활성에 의한 상기 각 유전자의 발현을 효과적으로 유도할 수 있도록 벡터상에 존재하는 프로모터의 전사 방향과 일치하도록 배열하는 것일 수 있다.
본 발명에서 재조합 벡터를 예쁜꼬마선충에 주입하는 방법은 당업계에 공지된 방법을 제한 없이 사용할 수 있으나, 마이크로피펫을 이용한 미세주입법(microinjection)인 것이 바람직하다.
또한, 본 발명은 상기 형질전환 예쁜꼬마선충을 이용한, 노화 유도 후보물질의 스크리닝 방법을 제공한다.
본 발명에서 노화 유도 후보물질의 스크리닝 방법에 제한은 없으나, 상기 형질전환 예쁜꼬마선충에 후보 물질을 처리한 후, 미토콘드리아 활성 또는 AMPK 활성을 측정하여, 이들의 활성이 후보 물질을 처리하지 않은 대조군에 비해 감소한 후보 물질을 선별함으로써 행하는 것이 바람직하다.
본 발명의 스크리닝 방법에 의하면, 노화와 관련된 인자(유전자나 약물 등)에 대한 탐색을 빠르고 쉽게 수행할 수 있는 바, 노화와 관련된 많은 질환을 연구하거나 신약을 개발하는 데 크게 기여할 수 있을 것이다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
실시예 1: 예쁜꼬마선충의 배양
예쁜꼬마선충은, 표준 야생형인 Bristol strain N2를 구입한 후, E. coli (OP50)으로 씨딩된 NGM(Nematode Growth Medium) agar 플레이트상에서 20℃로 배양하였다.

레벨 변화 확인
예쁜꼬마선충의 나이에 따른 small RNA 시퀀싱 결과를 이용하여, 나이에 따라 tRF의 레벨이 어떻게 변화하는지 분석하였다.
구체적으로, modENCODE (http://data.modencode.org/?Organism=C.%20elegans) 데이터베이스를 이용하여, 시퀀싱에서 얻은 RNA read를 tRNA 데이터베이스의 각 tRNA 염기서열에 매칭시켜 특정 tRNA에서 어떠한 부분의 리드만이 확인이 되는지 분석하였다.
그 결과, 도 2에 나타낸 바와 같이, 글루타민 tRNA 유래의 단편(Gln tRF), 특히 5' 말단 부분의 리드가 나이-의존적으로 증가됨을 확인하였다. 이후, 이 단편을 공지의 방법으로 시퀀싱한 결과, 하기 서열번호 1의 염기서열로 구성됨을 확인하였다.

나이-의존적으로 증가함을 재확인
데이터 베이스를 이용한 실시예 1의 결과를 재확인하기 위하여, 실제 예쁜꼬마선충 샘플에서도 노화될수록 글루타민 tRNA 유래의 단편이 증가하는지를 노던블랏을 통하여 검증하였다.
구체적으로, 0, 5, 8, 10일 나이의 예쁜꼬마선충 샘플 각각을 60 mm 플레이트 10장 분량을 모아서 total RNA를 추출한 후, 글루타민 tRNA의 5' 말단과 3' 말단에 특이적인 각 프로브를 이용하여 노던 블랏팅을 수행하였다. 이때 사용한 각 프로브 정보는 아래와 같다.

그 결과, 도 3에 나타낸 바와 같이, 실시예 1의 결과와 마찬가지로 나이에 따라 글루타민 tRNA 유래의 단편중에서도 5' 말단 부분의 단편이 증가함을 확인할 수 있었다.

제작
실시예 3의 결과를 바탕으로, in vivo 에서 Gln 5' tRF와 노화와의 관계를 규명하기 위하여, Gln 5' tRF를 과발현하는 형질전환 예쁜꼬마선충 모델을 제작하였다.
이를 위해 우선, 도 4에 나타낸 바와 같이, 예쁜꼬마선충의 small RNA 과발현에 특이적인 U6 프로모터와 Gln 5' tRF와 U6 3' UTR(untranslated region)을 포함하는 재조합 벡터를 제조하였다. 구체적으로는, U6 프로모터-Gln 5' tRF와, Gln 5' tRF-U6 3' UTR을 함께 PCR 증폭시키고, PCR fusion을 이용하여 U6 프로모터-Gln 5' tRF-U6 3' UTR construct를 제조한 후, 이를 EcoRI과 HindIII 제한효소를 이용하여 pUC57 벡터에 클로닝하였다. 이때 PCR에 사용한 각각의 프라이머 정보는 아래와 같다.

이후, 이 재조합 벡터를 GFP(Green Fluorescence Protein) 형광 마커와 함께 예쁜꼬마선충의 생식기관에 미세주입하여 과발현시킨 후, 형광현미경으로 녹색형광을 관찰하여 DNA 함유 여부를 확인하였다.
그 결과, 도 4에 나타낸 바와 같이, 예쁜꼬마선충에서 녹색형광이 밝게 나타남을 확인하였다.

수명 측정
실시예 4에서 제작된 Gln 5' tRF 과발현 예쁜꼬마선충을 이용하여, 실제 Gln 5' tRF가 노화에 있어 어떤 기능을 하는지 예쁜꼬마선충의 수명을 측정함으로써 평가하였다.
구체적으로, E. coli OP50를 streptomycin이 포함된 액체 LB에서 12-16 시간 동안 37℃에서 배양한 후 NGM 배지에 분주했다. 약 12-16 시간 후에 꺼내 FUdR (5-Fluoro-2'-deoxyuridine) 5μM 농도로 처리 후 Gln 5' tRF 과발현 예쁜꼬마선충 (Young-adult stage) 및 대조군(야생형)을 각각 하나의 플레이트에 30마리씩 총 4개에 플레이팅하였다. 이후 2-3일에 한번씩 살아있는 선충의 마릿수를 세는 방식으로 예쁜꼬마선충의 수명을 측정하였다.
그 결과, 도 5에 나타낸 바와 같이, 글루타민 tRNA 5' 말단 유래의 단편(Gln 5' tRF)이 과발현된(형질전환된) 예쁜꼬마선충 라인 3종 모두 대조군(야생형 WT)에 비하여 수명이 증가함을 확인하였다.
이러한 결과는, 특정 tRF의 과발현이 예쁜꼬마선충의 수명을 연장할 수 있음을 보여주는 것이기 때문에, 이 tRF가 단지 노화의 수동적인 부산물이 아니라 노화/수명을 능동적으로 조절하는 물질임을 의미하는 것이다.
또한, 본 실시예 5의 결과를 실시예 2 및 3의 데이터와 연관지어 보면, Gln 5' tRF는 노화를 지연시켜 수명을 연장하기 위한 보상 메커니즘으로서 예쁜꼬마선충에서 나이-의존적으로 증가됨을 알 수 있다.

항노화 메커니즘 규명
6-1. 유전자 스크리닝
실시예 4에서 제작된 Gln 5' tRF 과발현 예쁜꼬마선충을 이용하여, 실제 Gln 5' tRF의 과발현에 의해 어떠한 유전자에 변화가 있는지 확인함으로써, 항노화 메커니즘을 규명하고자 하였다.
우선, tRFs는 mRNA 전사를 조절한다는 것이 알려져 있기 때문에, RNA sequencing 분석을 통하여 Gln 5' tRF 과발현에 의해 유도된 mRNA 프로파일 변화를 관찰하였다.
구체적으로, day 1 adult 스테이지의 야생형 꼬마선충(대조군)과 Gln 5' tRF 과발현 꼬마선충을 대량으로 배양하여 3개의 독립적인 세트의 샘플을 준비하고, trizol base 방법으로 total RNA를 추출한 후, RNA 전기영동 및 qRT-PCR을 이용해 각 샘플들의 퀄리티를 확인하였다. 이후, RNA sequencing을 수행하여 얻는 raw data는 Cufflinks pipeline을 이용하여 분석하였다.
그 결과, 도 6에 나타낸 바와 같이, 야생형 예쁜꼬마선충에 비하여 1261종의 유전자가 상향조절되었고, 425종의 유전자가 하양조절되었음을 확인하였다.
이후, 상기 유전자들(DEG: differentially expressed genes)을 다른 알려진 longevity mutant 예쁜꼬마선충에서와 비교한 결과, 활성형 AMP-dependent kinase(AMPK) 과발현에 의해 상향조절된 유전자들, 그리고 daf -2(-)/insulin/insulin-like growth factor 1 (IGF-1) receptor mutations에 의해 상향조절된 유전자들과 상당히 오버랩됨을 알 수 있었다. 이러한 분석결과는, Gln 5' tRF가 AMPK 또는 insulin/IGF-1 signaling pathway와 함께 작용할 가능성이 있음을 의미하는 것이다.
또한, RNA sequencing 분석 결과, 도 7에 나타낸 바와 같이, Gln 5' tRF의 과발현에 의해 미토콘드리아 유전자들의 mRNA 수준이 야생형에 비해 매우 증가되었음을 확인하였다.
더욱이, qPCR(quantitative PCR)를 수행한 결과, 도 8에 나타난 바와 같이, 미토콘드리아 DNA의 카피수에 있어서도 증가가 관찰되었다. isp-1 돌연변이의 경우 미토콘드리아 DNA의 카피수가 증가해 있다고 보고된 바가 있으므로 양성 대조군으로 사용되었다.
DR은 다양한 종에서 미토콘드리아의 기능을 활성화시키는 것으로 알려져 있기 때문에, 이러한 결과는 Gln 5' tRF와 DR이 함께 작용함으로써 미토콘드리아의 기능을 촉진시켜 수명을 연장시키는 것을 이해된다.
6-2. 항노화 메커니즘 규명
종래 알려진 수명조절 경로(DR, insulin/IGF-1 pathway 등)와 Gln 5' tRF의 과발현 사이에 어떠한 유전적인 상호작용이 있는지 확인하기 위하여, DR mimetic eat-2(-), insulin/IGF-1 receptor mutant인 daf -2(-), catalytic alpha subunit of AMPK mutant인 aak -2(-), folkhead box O(FOXO) transcription factor mutant인 daf-16(-), heat-shock transcription factor 1(HSF-1) mutant인 hsf -1(-) backgrounds에서, Gln 5' tRF 과발현 예쁜꼬마선충의 수명 변화를 확인하였다.
구체적으로, 장수 꼬마선충 성체 5마리씩을 NGM 배지에 올리고 16시간동안 synchronize한 뒤, 알들이 자라서 young-adult 스테이지가 되었을 때 5 uM의 FUdR이 처리된 NGM 배지로 이틀 간격으로 두 번 옮긴다. 마지막 옮기는 과정에서 각 조건당 120마리로 마릿수를 맞춘 후 각 선충들이 모두 죽을때까지 2-3일 간격으로 살아있는 선충의 마릿수를 세어 수명 변화를 관찰하였다.
그 결과, 도 9A에 나타낸 바와 같이, Gln 5' tRF이 과발현된다고 하여 eat-2(-)의 장수(항노화) 효과를 더 증폭시키지는 않았다.
또한, 도 9B에 나타낸 바와 같이, Gln 5' tRF의 과발현에 의해 연장된 수명은, aak - 2이 변이됨으로써 완전히 억제됨을 알 수 있었으며, 상기 aak -2는 DR-매개 수명조절에 필요한 유전자로 알려져 있다.
이와 반대로, 도 9C 내지 9E에 나타낸 바와 같이, Gln 5' tRF이 과발현되면 daf -16 또는 hsf -1 변이를 갖는 예쁜꼬마선충의 수명을 독립적으로 증가시킨다는 것을 알 수 있었으며, 상기 daf - 16와 hsf - 1는 일반적으로 DR-매개 수명조절에 필요한 유전자는 아닌 것으로 알려져 있다.
따라서, 상기의 실험결과들을 종합해 보면, Gln 5' tRF에 의한 항노화 효과는 DR/AMPK pathway를 통하여 작용한다는 것을 알 수 있다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해되어야 한다.
