Abstract
Description
도 2는 본 발명의 전기화학 바이오 센서의 제작 방법에 따른 전극 오염(electrode fouling) 현상의 방지 효과를 확인한 도면이다. 구체적으로 검출 목적 물질에 특이적으로 결합하는 항체를 바이오 센서의 상판과 하판 조립 전에 상판에 결합시켜 제조하는 경우(A, 청색), 항체를 상판과 하판 조립 후에 결합시켜 제조하는 경우(B, 적색) 및 항체를 결합시키지 않고 PBS를 처리한 음성 대조군(C, 녹색)의 전기화학적 검출 실험의 결과를 확인하였다.
도 3은 실리카 나노 입자를 이용한 검출 신호를 증가시키는 효과를 확인한 도면이다. 구체적으로 실리카 나노 입자(SiNP/SBP-ProG/AP-anti-IgG 복합체)를 이용한 경우(A, 청색), 검출 목적 물질에 특이적으로 결합하는 항체(AP-anti-IgG)를 이용한 경우(B, 적색) 및 항체를 결합시키지 않고 PBS를 처리한 음성 대조군(C, 녹색)의 전기화학적 검출 실험의 결과를 확인하였다.
도 4는 본 발명의 고감도 전기화학 바이오 센서 키트를 이용하여, 다양한 농도(0, 0.01, 0.1, 1, 10 ng/mL)의 전립선암 생체 마커(prostate specific antigen, PSA) 단백질을 검출한 결과이다.
Claims (20)
2) 상기 상판을 전극이 형성되지 않은 하판과 부착시켜 마이크로 채널을 형성하는 단계;
3) 상기 마이크로 채널에 금 결합 단백질-단백질 G(GBP-ProG)를 포함하는 유체를 통과시키는 단계;
4) 상기 마이크로 채널에 검출 목적 물질에 특이적으로 결합하는 항체를 포함하는 유체를 통과시켜, GBP-ProG와의 결합을 통해 항체를 바이오 센서 상판에 고정하는 단계; 및
5) 상기 상판을 전극이 형성되지 않은 하판에서 분리하여 전극이 형성된 하판에 부착하여 바이오 센서를 제조하는 단계를 포함하는, 전기화학적 바이오 센서 키트의 제조 방법.
a) 실리카 나노 입자와 실리카 결합 단백질-단백질 G(SBP-ProG) 융합 단백질을 섞어, 실리카 나노 입자 표면에 SBP-ProG 융합 단백질을 부착시키는 단계; 및
b) 상기 a) 단계를 거친 실리카 나노 입자에 알칼라인 탈인산화 효소(Alkaline phosphatase, AP)로 표지된 검출 목적 물질에 특이적으로 결합하는 항체를 포함하는 액체를 섞어, SBP-ProG와의 결합을 통해 항체를 실리카 나노 입자에 고정하여 실리카 나노 입자를 제조하는 단계를 추가로 포함하는, 제조 방법.
a) 기판 상에 금을 박막한 마이크로 채널 상판;
기판 상에 전극을 포함하는 마이크로 채널 하판;
검출 목적 물질에 특이적으로 결합하는 항체; 및
상기 상판에 상기 항체를 고정하는 금 결합 단백질(Gold binding protein, GBP)-단백질 G(protein G) 융합 단백질; 을 포함하며, 상기 금 결합 단백질-단백질 G 융합 단백질을 구성하는 금 결합 단백질은 상기 상판에 박막된 금에 결합되고, 단백질 G는 항체에 결합되고, 상기 채널 상판과 채널 하판이 부착하여 마이크로 채널을 형성하는 것인 전기화학적 바이오 센서;와
b) 실리카 나노 입자;
산화 환원 효소로 표지된 검출 목적 물질에 특이적으로 결합하는 항체; 및
상기 나노 입자에 상기 검출 목적 물질에 특이적으로 결합하는 항체를 고정하는 실리카 결합 단백질(Silica binding protein, SBP)-단백질 G(protein G) 융합 단백질; 을 포함하며, 상기 실리카 결합 단백질-단백질 G 융합 단백질을 구성하는 실리카 결합 단백질은 실리카 나노 입자에 결합되고, 단백질 G는 항체에 결합되는 것인 검출용 실리카 나노 입자;를 포함하는 것을 특징으로 하며, 상기 전기화학적 바이오 센서 키트는 전극 오염(electode fouling) 현상이 방지되는 것인, 전기화학적 바이오 센서 키트.
1) 상기 전기화학적 바이오 센서 키트의 바이오 센서의 마이크로 채널 내로 시료를 통과시키는 단계;
2) 상기 마이크로 채널 내로 상기 전기화학적 바이오 센서 키트의 실리카 나노 입자를 처리하는 단계; 및
3) 상기 마이크로 채널 내로 알칼라인 탈인산화 효소의 기질인 ρ-Aminophenylphosphate monosodium salt (PAPP)를 통과시켜 얻어지는 산화 환원 그래프를 측정하는 단계를 포함하는, 검출 방법.
