As shown in the above Table 3, the ALP activity was evaluated, and it was found that the Coleoptera extract was concentration-dependent and significantly increased ALP activity. Since ALP is a marking factor expressed in the differentiation of osteoblasts, it can be understood that the extract of Coleoptera promotes osteoblast differentiation.

<Experimental Example 4> Evaluation of bone calcification in osteoblasts

To determine the degree of calcification of osteoblast cells, the following evaluation was made according to the Alizarin-red staining method.

MC3T3-E1 cells were added to several 6-well plates at a concentration of 1 × 10 5 cells / well. After incubation for 24 hours in an incubator, the medium was removed and 50 μg ascorbic acid, 10 mM β-glycerol phosphate The culture medium containing 10 μg / ml of bark extract extract was dispensed into the culture medium and cultured in an incubator for 5 days, 10 days, and 20 days. After incubation, the medium was removed, washed with PBS, and fixed with 70 wt% ethanol for 1 hour at 4 ° C. The alizarin red (AR) solution was prepared by dissolving alizarin red in distilled water, adjusting the concentration to 40 mM and adjusting the pH to 4.2. The cells were fixed and stained with alizarin red (AR) solution (2 ㎖ / well) for 10 min. After washing 5 times with distilled water, the unstained portion was removed by washing with PBS and the degree of calcification was observed under a microscope.

Next, after confirming the formation of calcification, 2 ml of 10 mM sodium phosphate (pH 7.0) solution to which 10 wt% cetylpyridinium chloride was added was added to the wells for dissolving for 15 minutes. The degree of staining was measured using a microplate reader The absorbance at 570 nm was measured and the bone formation capacity was evaluated as a percentage based on the absorbance of the control group. The results are shown in Table 4 below.

As shown in the above Table 4, it can be seen that the extract of Coleoptera promotes bone calcification of osteoblasts. In addition, the ability to form bone calcification increased with time. Bone calcification is an important marker for osteoblast differentiation. Alizarin is stained in the matrix of mineralized cells, so the amount of calcification and the degree of dyeing are mutually in proportion. Therefore, it can promote bone calcification and formation of osteoblast, and promote osteoblast differentiation.

[Example 2]

Bee hair, soybean (seaweed), wool (roots) and safflower seeds were prepared, washed several times with water, and then thoroughly dried through hot air. The dried four kinds of raw materials were put into a pulverizer in an appropriate amount so as to have a content (based on dry weight) as shown in Table 5, and then mixed and pulverized. Next, the mixed pulverized product was put in a ventilator, and then subjected to compression molding to produce spherical pills (functional food). Thereafter, the prepared pills were sterilized.

The pills according to Example 4 and Comparative Example 1 were administered to a subject twice a day, ten times a day, and evaluated for improvement in tooth and periodontal condition two months after administration. The subject was a chronic periodontal disease patient, and 20 subjects (10 persons taking the sample of Example 4 and 10 people taking the sample of Comparative Example 1) aged 55 to 65 years were enrolled. The degree of improvement was evaluated on a 5-point scale according to the following evaluation criteria, and the average value of the results was shown in Table 6 below.
