Abstract
본 발명은 TGF-β-농축 유 단백질 분획물, 이의 제조 방법 및 만성 염증성 질환 및, 특히 건선 예방 및/또는 치료용 약제 및/또는 식품 조성물 제조에 이의 이용에 관계가 있다.
Description
본 발명의 목적은 신규 TGF-β-농축 유 단백질 분획물, 이의 제조 공정 및 만성 염증성 질환, 특히 건선 예방 및/또는 치료용 약제 및/또는 식품 조성물의 제조를 위한 이의 용도이다.
재발에 의해 건선은 팔꿈치, 무릎 및 두피에서 우세하게 발병하는 홍반성-편평 발진의 특성을 나타내는 만성 피부과 병이다.
생물학적 관점에서, 건선은 각질층에서 미세종기를 형성하는 T 림프구 및 다핵성 호중구에 의한 표피 및 진피의 침투와 관련된 각질세포의 비정상적인 증식 및 분화의 특성을 나타내는 만선 염증성 과정이다.
개체에서 건선 출현의 원인은 잘 알려져 있지 않다. 건선 출현을 촉진시키는 것으로 알려진 여러 인자를 인용하였다:
- 유전성 인자,
- 심리적 인자(스트레스, 호르몬 변화, ...),
- 면역 이상.
마지막으로, 특정 약제 및 세균성 또는 바이러스성 감염은 건선 발병을 촉진할 수 있다.
일반적으로 건선을 치료할 수 있는 치료법은 알려져 있지 않다. 지금까지 알려진 상기 치료법은 단지 건선 증상을 지연 및/또는 약화할 수 있는 것으로 알려져 있다.
소수의 플라크에 한정된 건선에 대해서, 비타민 D는 지역적으로 적용된 코르티코이드(corticoids)와 함께 선택적으로 처방된다. 또한 국소적으로 적용된 레티노이드를 처방하는 것도 가능하다.
대부분의 중증의 경우에서는, 광선요법이 처방되거나 일반적인 경로로 선택적으로 메토트렉사트 또는 레티노이드가 처방된다. 상기 후자의 치료법은 중대한 부작용과 결부된다.
그러므로 만족스러운 건선 치료법이 현재까지는 없다.
그 외의 만성 염증성 질환, 이를테면 류마티스성 관절염, 골관절염, 크론씨병, 다발성 경화증, 홍반성 루푸스는 의사들을 동일한 문제로 괴롭힌다: 상기 병리학적 상태의 치료가 가능한 치료법이 존재하지 않고, 상기 병리학적 상태의 징후를 치료하기 위한 현존하는 치료법들은 부적절하거나 매우 심각한 부작용을 수반한다. 만성 염증성 성분은 또한 자가면역질환에도 존재한다.
케모카인 및 싸이토카인과 같은 성장인자는 염증성 과정에 영향을 주는 것으로 알려져 있다. 상기 영향은 염증성 과정의 증폭에서부터 억제까지의 범위이다.
특히 유액(乳液) 내의 특정 성장인자는 염증성 과정에 대한 조절 활성을 가지는 것으로 알려져 있다.
예를 들면, 문헌 WO 96/34614호는 화학요법 또는 방사선 요법에 의해 발생한 소화관 점막 손상의 예방용 약제의 제조에 유액 산물의 이용을 기술한다.
바람직하게는 양이온-교환 크로마토그래피 컬럼에 우유을 통과시킴으로써 상기 추출물을 수득한다. 이것은 바람직하게는 락토페린 및/또는 락토퍼옥시다제 및 성장인자, 특히 TGF-β를 포함한다.
TGF-β(형질전환 성장인자)는 세포성장 및 분화를 조절하는 펩타이드 성장인자이다. 이것은 다양한 이성체로 존재할 수 있는 25 kD의 이량체 분자이다: TGFβ1, TGFβ2 및 TGFβ3. TGF-β는 면역 및 염증성 반응의 특정 단계를 조절 및 특히 감소시킬 수 있는 것으로 알려져 있다(G. Prud'homme et al., J. Autoimmun. (2000), 14, 23-42; M. Shull et al ., Nature (1992), 359, 693-699; J. Graycar et al ., Mol. Endocrinol. (1989), 3, 1977-1986; J. Letterio et al ., Annu. Rev. Immunol. (1998), 16, 137-161). 이것은 잠재적 형태(비-생물학적 활성) 또는 활성 형태로 존재한다.
이것은 유액에 매주 적은 양으로 존재한다(30 ㎍/유액 ℓ). 그러나 TGF-β의 활성과 상이한 활성, 가능하게는 TGF-β와 반대의 활성을 갖는 다른 많은 인자가 유액에 존재하고, 이의 상기 결과는 유액에 포함된 TGF-β가 염증성 과정에 주목할 만한 활성을 갖지 않는다는 수 있다.
게다가 유액에서 TGF-β의 분리는 산업적 규모로는 거의 실행가능하지 않은데, 무엇보다도 비용 고려 및 또한 분리 공정에서 종종 표적 단백질을 변성시킬 수 있고, 이는 단백질이 포함한 모든 생물학적 특성이 소실된 유 단백질 분획물을 생산하게 하기 때문이다.
문헌 EP 0313515호는 유액에서 TGF-β 분리 공정을 기술한다. 그러나 상기 공정은 복잡한 특성의 매우 많은 단계로 구성되어 있고, 그러므로 경제적 관점에서 볼 때 거의 실용성이 없다.
염증성 과정에 생물학적 활성을 나타내는 즉, 염증성 과정을 감소 또는 억제하는 유 단백질 분획물을 수득하기 위해, 유액으로부터 시작하여 유액에 존재할 때에 TGF-β의 항-염증성 성질을 방해하는 TGF-β에 대한 길항체 인자의 적어도 일부를 제거하면서 다른 단백질에 비해 TGF-β 함량을 증가시킬 수 있는 것이 필요하다. 상기 시도의 어려움은 TGF-β의 정확한 길항체가 알려지지 않았기 때문에 엄청나다.
여러 저자들은 TGF-β-농축 유 단백질 분획물의 분리의 문제에 관심을 집중하였다. 그것은 출원 특허 및 특허 WO 96/34614호, EP 0545946호, WO 01/25276호, WO 03/008447호, FR 2827240호, EP 0869134호에 대한 경우이다.
상기 문헌 EP-0545946호는 유장으로부터 성장인자를 추출하는 공정을 기술한다; 상기 공정은 하기 단계를 포함한다: 불용성 물질을 제거하기 위해 유장을 여과하는 단계, pH 6.5 내지 8로 조정하는 단계, 주요 염기성 구성성분을 흡수하고 주요 산성 단백질을 용출하는 세파로오스-형태 양이온-교환 레진을 사용하는 단계, 완충용액을 사용하여 레진을 평형화하는 단계, 상기 레진에 여과액을 가하는 단계, 강 이온 세기의 완충용액으로 용출하는 단계, 염을 제거하기 위해 여과하는 단계, 농축하는 단계.
상기 문헌 EP 0869134호는 양이온 교환기에 흡착, 분획물 용출, 성장인자가 농축된 분획물의 생산 및 pH 3.5 내지 4.5로 처리하는 단계에 의해, 하나 이상의 성장인자를 유액 또는 유액 파생체로부터 회수하는 공정을 기술한다. 상기 공정은 바람직하게는 매우 높은 표면 속도 및 높은 액체 하중으로 적용된다.
상기 문헌 WO 01/25276호는 하기 단계를 포함하는 TGF-β 및 IGF-1 추출 고정을 기술한다: 양이온-교환 크로마토그래피로 유액으로부터 염기성 분획물을 회수하는 단계, 하이드록시아파타이트 컬럼에 회수된 분획물을 통과시키는 단계, 두 개의 분획물을 수득하도록 염 농도를 높이면서 적어도 두 개의 용출액을 이용하여 용출하는 단계:
- IGF-1/TGF-β > 10의 IGF-1-농축된 분획물
- TGF-β/IGF-1 > 5 및 전체 단백질량에 비례하여 30 내지 50%의 면역 글로불린을 포함하는 TGF-β-농축된 분획물.
상기 문헌 FR 2827240호는 유액 액상의 수용성 단백질이 풍부한 용액으로부터 활성형의 TGF-β-농축된 단백질 분획물을 하기의 방법에 의해 생산하기 위한 공정을 나타낸다: a) 수용성 단백질 농도를 5-30 g/ℓ로 조절하는 단계, b) 산처리에 의해 침전시키는 단계, c) 정밀여과-투석여과를 수행하는 단계, d) 정밀여과 농축물을 회수하는 단계, e) 건조하는 단계.
상기 문헌 WO 03/008447호는 상기 성장인자 TGF-β, IGF-1, 락토퍼옥시다제 및 면역 글로불린을 분리하기 위한 공정을 나타낸다. 유액의 염기성 단백질 분획물을 양이온-교환 크로마토그래피에 의해 회수한다. 상기 수득한 분획물을 소수성 상호반응 크로마토그래피에 통과시킨다.
그러나 상기 공정 중에 산업적으로 실용적인 조건에서 만성 염증성 병리학적 상태 및 이의 징후 또는 증상, 및 특히 건선에 실질적인 효능을 갖는 유 단백질 분획물을 수득할 수 있는 것은 없다.
그러므로 만성 염증성 과정 및 특히 건선에 작용하는 신규 유 단백질 분획물을 발견한 본 출원은 놀랄만하다.
본 발명의 상기 유 단백질 분획물은 하기 단계를 포함하는 공정에 의해 수득될 수 있다:
유액 또는 유장(whey)을 출발물질로서 사용한다.
(a) 상기 유액 또는 유장을 정밀여과 또는 열처리하는 단계;
(b) 상기 유액 또는 유장 또는 단계 (a)의 상기 산물을 양이온-교환 레진에 공급하는 단계;
(c) 상기 양이온-교환 레진을 탈염수로 세척하는 단계;
(d) 상기 레진을 염류 용액의 농도를 증가시키면서 용출하는 단계;
(e) 전도도 21.0 내지 22.0 mS/㎝의 염류 용액에 상응하는 용출액을 회수하는 단계.
본 발명의 상기 공정은 추가적으로 하나 이상의 하기 단계를 포함할 수 있다:
(f) 단계 (e)에서 수득한 상기 용출액은 한외여과로 농축하고 상기 농축물을 회수하는 단계;
(g) 단계 (f)에서 수득한 상기 농축물을 정밀여과로 멸균하고 상기 투과액을 회수하는 단계;
(h) 단계 (g)에서 수득한 상기 투과액을 분무 건조하는 단계.
본 발명에 따른 공정에서 출발물질로서 바람직하게는 소로부터 수득한 유액 또는 유장을 사용하는 것이 가능하다. 유장은 유액 또는 버터밀크로부터 단백질 및 지방을 추출한 후 수득한 잔여 액체이다. 유장의 세 가지 범주는 일반적으로 분리될 수 있다. 상기 첫 번째 두 개의 범주들은 1.8 g 젓산/ℓ보다 적거나 또는 많은 유장의 산도에 따라 분류된다; 조작된 또는 조작되지 않은 압축 치즈(에멍탈(Emmenthal), 쌩 뽈랭(Saint-Paulin) 등)의 제조에서 파생된 유청 및 카제인 또는 혼합 또는 젖산 응집에 의해 수득한 그 외의 치즈(연질치즈, 프로마즈 프레(fromages frais))의 산성 유장. 참조로써 상기 유청의 상기 평균 조성물은 유장 1 ㎏ 당 건조 물질 61 g, 유장 42 내지 48 g, 단백질 8 g, 지방 2 g, 미네랄 5 내지 7 g, 젖산 1 내지 5 g 및 미네랄 및 비타민과 같은 균형이다.
평균 다공성 0.1 ㎛의 지지체에서 유액의 정밀여과에 의해 수득한 이상적인 유장도 알려져 있다.
본 발명의 첫 번째 변형에 따라, 본 발명에 따른 공정에 의해 더욱 유리한 생물학적 특성을 갖는 단백질 분리물을 수득하는 것을 가능하게 하는 조성물인 유액 및 유리하게는 소의 우유(牛乳)를 출발물질로 사용한다. 상기 변형은 또한 출발물질로 유장을 사용한 변형보다 단백질 수율을 더 좋게 하는 것이 가능하다.
본 발명의 두 번째 변형에 따라, 카제인 제조에서 산성 유장을 출발물질로 사용한다. 상기 변형은 출발물질이 산업적 개발의 부산물이고 그러므로 저가이기 때문에 경제적 이점을 나타낸다.
출발물질로 사용된 상기 유액은 소, 염소, 양, 버팔로 유액일 수 있다. 이것은 원심분리에 의한 알려진 방식으로 지방이 제거될 수 있다.
상기 유액 또는 유장은 미생물 기원의 어떠한 오염도 제거하기에 가능하도록 처리될 수 있다. 상기 목적에 의해, 상기 유액은 유리하게 68℃를 넘지 않는 온도로 열처리 되거나 첫 번째 정밀여과 단계, 예를 들면 다공성이 대략 1.4 ㎛의 여과기로 여과될 수 있다.
다음으로, 상기 정밀여과 투과액, 또는 직접적으로 상기 유액 또는 상기 유장은 상기 레진 컬럼에 공급한다.
앞서 언급된 상기 공정에서 사용된 상기 레진은 다중음이온 나노다공성 합성 중합체로 구성되어 있다. 상기 중합체는 바람직하게는 삼차원의 구형 또는 타원형이다. 이것은 강산 유형의 작용기를 상기 강산의 음이온의 결합 염기의 형태로 운반한다. 이것은 바람직하게는 SO3 - 기로 기능화된다. 게다가, 상기 레진을 구성하는 상기 중합체는 바람직한 용출 매개변수로부터의 수압을 견디게 하는 기계적 저항성을 지니고 있어야 한다.

단계 (b)에서, 레진의 용적에 대비한 유액의 용적의 비율은 유리하게는 100 내지 200 이다.
상기 유액 또는 상기 유장을 상기 레진 컬럼에 공급한 후 탈염수, 바람직하게는 10 내지 15 ℓ 물/ℓ 레진을 사용하여 세척한다.
그 후 10 내지 14 g/ℓ의 염 농도, 바람직하게는 11 내지 13 g/ℓ를 포함하는 염류 용액을 사용한다.
염류의 유량은 1.5 내지 2 m3/h이고, 염류 용액의 선형 속도는 유리하게는 0.4 내지 0.6 m/h이다. 레진의 용적에 대비한 염류의 용적의 비율은 2.5 내지 3.5이다.
그러므로 회수된 상기 용출액은 하나 이상의 한외여과 단계 및 선택적으로 하나 이상의 투석여과 단계에 의해 농축된다. 상기 단계들은 유리하게는 저온(2 내지 6℃)에서 수행된다.
그러므로 수득한 상기 농축물은 미세여과되고 분무 건조된다.
그러나 당업자에게 알려져 있는 그 외의 탈염 방법 및 농도는 파악될 수 있다.
그러므로 유 단백질 분획물은 하기 특성이 나타내도록 수득된다:
- 전체 단백질 중량에 비례하여 0.010 내지 0.025% 중량의 TGF-β 함량;
- 전체 단백질 중량에 비례하여 < 25%의 IgG(면역 글로불린) 함량;
- TGF-β/IGF 1 비율 ≥5 중량/중량.
유리하게는, 이것들은 또한 하나 이상의 하기 특성에 부합한다:
- 전체 단백질 중량에 비례하여 35 내지 45% 중량의 락토퍼옥시다제 함량.
상기 유 단백질 분획물은 본 발명의 다른 개체들을 구성한다.
놀랍게도, 본 발명의 상기 유 단백질 분획물은 희생된 쥐의 비장 세포에서 측정된 것과 같이 림프구 증식의 감소의 원인이 된다: 염화 암모늄으로 처리되고 세척된 상기 세포를 배양 마지막에 세포 배양액에 5-브로모디옥시유리딘(5-bromodeoxyuridine)의 첨가와 함께 본 발명의 단백질 분획물 및 콘카나발린 A(concanavalin A)와 같은 유사분열 자극제(mitogenic agent)가 존재 하에서 48 시간 동안 인큐베이션 하였다. 상기 세포에 포함된 5-브로모디옥시유리딘의 함량에 의해 림프구 증식을 평가한다.
상기 건선성 표피가 활성된 T 림프구 유입의 근원임을 알기에, 본 발명의 상기 단백질 분획물은 건선 예방 또는 치료용으로 사용될 것이나, 또한 만성 염증성 병리학적 상태 및/또는 이의 증상과 같은 다른 상태에도 사용될 수 있다. 상기 병리학적 상태들 중에서, 건선 뿐만 아니라 류마티스성 관절염, 골관절염, 크론씨병, 다발성 경화증, 홍반성 루푸스가 언급될 수 있다. 게다가 이것들은 자가면역질환의 염증성 성분에 영향을 주는 것이 가능하다. 그러므로 이것들은 자가면역질환의 예방 및/또는 치료, 특히 자가면역질환의 염증성 성분의 예방 및/또는 치료에 사용될 수 있다.
게다가 본 발명의 상기 공정은 산업적 규모에서 쉽게 수행될 수 있고, 오직 제한된 개수의 단계를 포함하여 실행하기 쉽다.
본 발명의 공정의 또 다른 이점은 산업적 중요성을 확신하는 다른 유 단백질 분획물의 회수를 가능하게 하는 사실에 있다. 선택적으로 단계 e)의 상기 분획물을 회수한 후, 다양한 유 단백질 분획물을 회수하는 것이 가능하도록 수용성 염류 용액의 농도를 증가하면서 상기 컬럼을 용출하는 것이 가능하다. 단계 e')에서 전도도 50.5 내지 51.5 mS/㎝ 범위의 염류 용액으로 용출된 상기 분획물은 락토페린-농축 분획물이다. 이것은 단일 크로마토그래피 단계로 락토페린이 풍부한 유 단백질 조성물을 회수하는 것이 가능하게 한다. 바람직하게는 상기 공정 단계를 하나 이상의 하기 조건을 이용하여 수행한다:
- 레진의 용적에 대비한 염류의 용적의 비율이 2.5 내지 3.5,
- 1.5 내지 2 m3/h의 염류 유량,
- 0.4 내지 0.6 m/h의 용출 선형 속도.
그것은 추가적으로 한외여과, 투석여과, 정밀여과 및/또는 건조, 특히 분무 건조의 수반되는 단계를 포함할 수 있다.
락토페린-농축 유 단백질 분획물 분리를 위한 상기 공정은 본 발명의 또 다른 개체를 구성한다.
본 발명의 또 다른 개체는 본 발명의 유 단백질 분획물 및 약제학적으로 허용가능한 담체를 포함하는 약제학적 조성물이다.
포함된 병리학적 상태 및 상기 병리학적 상태의 심각성에 따라, 당업자에 의해 투여 형태 및 부형제가 변경될 수 있다: 국소 적용(크림, 로션, 패취), 구강 투여(젤라틴 캡슐, 시럽, 정제, 수용액 또는 현탁액), 주사(주사용 액제).
또한 더욱 상세하게는 만성 염증성 병리학적 상태 및, 특히 건선 또는 자가면역질환에 걸린 사람을 위한 식품 조성물, 특히 식이 조성물의 제조에 본 발명의 상기 유 단백질 분획물을 사용하는 것이 가능할 수 있다. 상기 식품 조성물들은 보통 사용되는 상기 유 단백질을 대신하여 본 발명의 유 단백질 분획물을 포함한다. 이것들은 단백질 음료, 유액 식품 등일 수 있다. 이것들은 본 발명의 다른 개체들을 구성한다.

실시예
1: 유 단백질
분획물의
제조
68℃에서 15초 동안 미리 열처리한 탈지 유액 250 m3을 하기 특성을 나타내는 2000 ℓ의 양이온-교환 레진(BIOSEPRA 제공 SPEC 70)에 크로마토그래피 컬럼에 통과시켰다:
완전한 합성의 마크로다공성(macroporous) 다중음이온 중합체는 SO3 - 기로 기능화되었다; 상기 중합체는 삼차원의 구형 또는 타원형이다; 상기 입자 크기는 261 ㎛ 이상이다; 상기 지지체는 AMPS(2-acrylamido-2-methylpropanesulfonic acid; 2-아크릴아미노-2-메틸프로판술폰산)의 중합에 의해 제조된다.
유액을 상기 컬럼에 공급하는 유량은 14 m3/h이고 선형 속도는 3.2 m/h이다.
유액을 통과한 후, 상기 컬럼을 23,000 ℓ의 탈염수로 세척한다(하향으로)
이후에 12 g/ℓ의 염류용액을 사용하여 상기 레진으로부터 결합한 단백질을 추출한다. 사용된 유량은 1900 ℓ/h이고 선형 속도는 0.6 m/h이다; 수득한 용출액의 용적은 8000 ℓ정도이다.
이후 상기 용출액은 상기 농축물의 상기 전도도가 15 mS/㎝ 이상으로 유지되도록 용적 농도 인자를 이용하여 4℃에서 UF(DSS, 400 m GR10D 막)에 의해 농축된다. 이후 수득한 상기 농축물의 전도도가 5 mS/㎝가 될 때까지 상기 농축물은 UF 및 투석여과(동일한 막이지만 표면 면적이 오직 34 m2인 막을 갖춘)에 의해 두 번 농축된다.
그러므로 수득한 상기 농축물은 1.4 ㎛ 다공성의 30℃의 17 m2 정밀여과 모 듈에 보내진다; 수득한 상기 투과액은 8% 정도의 건조 추출물을 갖고, 180℃의 공기 주입 온도 및 80℃의 타워 출구 온도로 단일-효과 노즐 타워에서 분무 건조된다.
유 단백질 분획물 1650 g을 분말 형태로 회수하였다. 상기 분획물은 하기 특징을 나타낸다:

실시예
2: 상기 유 단백질
분획물의
세포성장 효과
실시예 1에서 수득한 상기 유 단백질 분획물의 상기 효과를 유선암의 흉막 삼출로부터 유래한 세포, MCF7 배양액에서 검사되었다. 두 개의 실험이 수행되었다. 상기 MCF7 세포를 25 ㎝2 접시에 실험 1의 경우 700,000 세포/접시, 실험 2의 경우 200,000 세포/접시의 농도로 접종하였다. 24 시간 동안 대기한 후, 상기 배양액에 공기를 주입하고 실시예 1에서 수득한 상기 단백질 분획물을 218 ㎍/㎖, 109 ㎍/㎖ 또는 0 ㎍/㎖(대조군 배지)의 농도로 포함하는 배지 7 ㎖로 갈아주었다. 다음에, 상기 세포를 72 시간 동안 배양하면서 24 시간마다 배지를 교환해 주었다. 상기 결과는 각 분석에 대해 4 개의 접시로부터 얻은 값의 평균이다.
상기 실험은 실제로 실시예 1에서 수득한 상기 단백질 분획물의 MCF7 세포 성장에 대한 농도-의존적 억제 효과를 증명한다.

4: 상기 유 단백질

포함하는 영양첨가제의
건선
환자에 대한 효과
실시예 3에서 제조한 상기 영양첨가제의 효과를 검사하였다. 건선 환자 20명(23 내지 70세의 여성 8명 및 남성 12명)은 60일 동안 매일 실시예 3에서 제조한 상기 영양첨가제 젤라틴 캡슐 8개(아침에 4개, 오후에 4개)를 섭취하였다. 즉, 실 시예 1에서 수득한 상기 유 단백질 분획물의 일일 섭취량이 1 g 이었고, 이는 TGF-β 110 ㎍에 상응한다. 상기 징후 조절 및 상기 혈액 분석을 상기 영양첨가제 섭취 전 및 60일 후에 수행하였다.
결과:
- 건선성 증상: 상기 영양첨가제 섭취 60일 후 80%의 사람의 증상이 현저히 개선되었다(건선성 플라크, 염증, 표피박리 및 가려움증의 감소).

- 안전성: 상기 영양첨가제는 잘 허용되어 왔다. 60일간의 전체 섭취 기간 동안, 심각하게 바람직하지 못한 사건은 발견되지 않았다. 상기 영양첨가제의 안전성을 확인하는 간성 매개변수(ASAT, ALAT, GGT), 신장 매개변수(우레아, 크레아티닌(creatinine)) 및 혈액상 매개변수(백혈구, 다핵성, 림프구, 단핵세포, 혈소판)에서 발견된 이상은 없었다.
Claims (17)
- 하기 단계를 포함하는 것을 특징으로 하는 유 단백질 분획물 제조 공정:유액 또는 유장(whey)을 출발물질로서 사용하고;(a) 상기 유액 또는 유장을 대략 1.4 ㎛의 다공성을 갖는 여과기로 정밀여과하거나, 또는 미생물 기원의 오염을 제거하기 위해 68℃와 동등하거나 그 이하의 온도에서 열처리하는 단계;(b) 상기 유액 또는 유장 또는 단계 (a)의 산물을 강산 음이온의 결합 염기 형태의 작용기로 기능화된 나노다공성 합성 중합체로 구성되고, 선형 속도는 2.8 내지 4.5 m/h이며, 레진의 용적에 대비한 유액의 용적의 비율이 100 내지 200인 양이온-교환 레진에 공급하는 단계;(c) 상기 양이온-교환 레진을 탈염수로 세척하는 단계;(d) 상기 레진을 염류 용액의 농도를 증가시키면서 용출하는 단계;(e) 전도도 21.0 내지 22.0 mS/㎝의 염류 용액에 상응하는 용출액을 회수하는 단계.
- 제 1항에 있어서, 추가적으로 하나 이상의 하기 단계를 포함하는 것을 특징으로 하는 공정:(f) 단계 (e)에서 수득한 상기 용출액은 한외여과로 농축하고 농축물을 회수하는 단계;(g) 단계 (f)에서 수득한 상기 농축물을 정밀여과로 멸균하고 정밀여과액을 회수하는 단계;(h) 단계 (g)에서 수득한 상기 정밀여과액을 분무 건조하는 단계.
- 제 1항 또는 제 2항에 있어서, 출발물질은 소의 우유(牛乳)인 것을 특징으로 하는 공정.

- 제 1항에 있어서, 상기 중합체는 SO3 - 기로 기능화되는 것을 특징으로 하는 공정.
- 제 1항에 있어서, 단계 (b)에서 상기 양이온-교환 레진에 공급하는 유량은 10 내지 15 m3/h인 것을 특징으로 하는 공정.
- 제 1항에 있어서, 하기 조건의 하나 이상을 만족하는 것을 특징으로 하는 공정:- 상기 염류 용액은 10 내지 14 g/ℓ의 염 농도를 포함;- 상기 염류 유량은 1.5 내지 2 m3/h;- 상기 염류 용액의 선형 속도는 0.4 내지 0.6 m/h;- 레진의 용적에 대비한 염류의 용적의 비율은 2.5 내지 3.5 .
- 제 1항 또는 제 2항의 공정에 의해 수득될 수 있고 하기 특징을 만족하는 것을 특징으로 하는 유 단백질 분획물:- 전체 단백질 중량에 비례하여 0.010 내지 0.025% 중량의 TGF-β 함량;- 전체 단백질 중량에 비례하여 < 25%의 IgG(면역 글로불린) 함량;- TGF-β/IGF 1 비율 ≥5 중량/중량.

- 제 10항에 있어서, 상기 만성 염증성 병리학적 질환은 건선인 것을 특징으로 하는 만성 염증성 병리학적 질환의 약학적 조성물.
- 제 8항의 유 단백질 분획물 및 약제학적으로 허용가능한 담체를 포함하는 자가면역 질환의 예방 또는 치료용 약학적 조성물.
- 제 8항의 유 단백질 분획물 및 약제학적으로 허용가능한 담체를 포함하는 자가면역 질환의 염증성 징후의 예방 또는 치료용 약학적 조성물.
- 제 8항의 유 단백질 분획물을 포함하는 만성 염증성 병리학적 질환의 예방 또는 개선용 식품 조성물.
- 제 14항에 있어서, 상기 식품 조성물은 식이 조성물인 것을 특징으로 하는 만성 염증성 병리학적 질환의 예방 또는 개선용 식품 조성물.
- 제 1항 또는 제 2항에 있어서, 단계 (e) 다음에, 단계 (e)에서 획득한 용출액에서 전도도 50.5 내지 51.5 mS/㎝ 범위의 염류 용액에 상응하는 용출액에서 회수되는 락토페린-농축 유 단백질 분획물을 분리하는 단계 (e')를 추가적으로 포함하는 것을 특징으로 하는 공정.
- 제 16항에 있어서, 공정의 단계 (e')를 하기 조건의 하나 이상을 이용하여 수행하는 것을 특징으로 하는 공정:- 2.5 내지 3.5의 레진의 용적에 비례한 염류의 용적,- 1.5 내지 2 m3/h의 염류 유량,- 0.4 내지 0.6 m/h의 용출 선형 속도.
