Claims (23)
- 미각 세포에서 단맛의 신호를 조절하는 제제를 동정하는 방법으로서,(i) 페릴라틴에 반응하는 T1R2 단일체 감미 수용체를 발현하는 세포를 하나의 제제와 접촉시키는 단계; 및(ii) 상기 세포에서 하나 이상의 기능적 반응에 의해 하나 이상의 제제가 상기 세포에서 상기 T1R2 감미 수용체의 기능적 활성에 영향을 주는지를 결정하는 단계를 포함하며, 이때상기 T1R2 감미 수용체는 서열 식별 번호: 2에 상동하는 폴리펩타이드, 서열 동일성에 의해 결정시 서열 식별 번호: 1에 상동하는 뉴클레오타이드에 의해 암호화된 폴리펩타이드, 및 하이브리드화에 의해 결정시 서열 식별 번호: 1에 상동하는 뉴클레오타이드에 의해 암호화된 폴리펩타이드로 구성되는 군으로부터 선택되며,상기 상동하는 폴리펩타이드는 서열 식별 번호: 2에 85% 이상의 서열 동일성을 가지며,서열 동일성에 의해 결정시 상동하는 뉴클레오타이드는 서열 식별 번호: 1에 85% 이상의 서열 동일성을 가지며,하이브리드화에 의해 결정시 상동하는 뉴클레오타이드는 50%의 포름아마이드, 5×SSC, 및 1% SDS로 이루어진 용액 중에서 42℃의 온도 하의 엄격한 하이브리드화 조건 하에서 하이브리드화되고 0.2×SSC 및 0.1% SDS로 이루어진 용액 중에서 65℃에서 세척되며,T1R2 단일체 감미 수용체 발현 세포들은 T1R3 수용체를 발현하지 않는, 방법.
- 제 1 항에 있어서,세포가 또한 G-단백질을 발현하는, 방법.
- 제 2 항에 있어서,G-단백질이 Gaq-구스트듀신(gustducin)에 기초한 키메라성 G-단백질인, 방법.
- 제 3 항에 있어서,G-단백질이 키메라성 G-단백질 G 알파 16-구스트듀신 44인, 방법.
- 제 1 항에 있어서,단계 (ii)가 세포내 메신저에서의 변화 또는 세포내 메신저에 의해 유발된 변화를 측정함으로써 수행되는, 방법.

- 삭제
- (i) 페릴라틴에 반응하는 T1R2 수용체 단일체를 발현하지만, T1R3 수용체를 발현하지 않는 재조합 세포, 및 (ii) 페릴라틴을 포함하는, T1R2 단일체의 조절물로서 시험 제제를 동정하기 위한 조합 용도용 키트로서,상기 T1R2 감미 수용체가, 서열 식별 번호: 2에 상동하는 폴리펩타이드, 서열 동일성에 의해 결정시 서열 식별 번호: 1에 상동하는 뉴클레오타이드에 의해 암호화된 폴리펩타이드, 하이브리드화에 의해 결정시 서열 식별 번호: 1에 상동하는 뉴클레오타이드에 의해 암호화된 폴리펩타이드로부터 선택되되, 상기 상동하는 폴리펩티드는 서열 식별 번호: 2에 85% 이상의 서열 동일성을 갖고, 서열 동일성에 의해 결정시 서열 식별 번호: 1에 85% 이상의 서열 동일성을 가지며, 하이브리드화에 의해 결정시 50%의 포름아마이드, 5×SSC, 및 1% SDS로 이루어진 용액 중에서 42℃의 온도 하의 엄격한 하이브리드화 조건하에서 하이브리드화되고 0.2×SSC 및 0.1% SDS로 이루어진 용액 중에서 65℃에서 세척되는, 키트.
- (i) 재조합 세포를 고체 지지체 상에서 성장시키는 단계,(ii) 작용물질의 존재 하에 한정된 플레이트 또는 웰의 배양 배지에 시험 제제를 첨가하는 단계, 및(iii) 세포의 기능적인 반응의 변화를 시험 제제의 존재 및 부재 하에서의 반응과 비교하여 측정하고, 그럼으로써 시험 제제를 T1R2 단일체 또는 이와 유사한 상동체의 조절물로서 동정하는 단계를 포함하는, 제 12 항의 키트를 사용하는 방법.
- 제 13 항에 있어서,작용물질의 존재 하에 한정된 플레이트 또는 웰의 배양 배지에 1nM 내지 100mM 이상의 양의 시험 제제를 첨가하는 단계를 포함하는, 방법.
