Claims (19)
- 자이모모나스 모빌리스(Zymomonas mobilis)의 게놈으로부터 피루베이트 디카르복시라아제를 코딩하는 pdc 유전자(SEQ ID NO: 1) 및 락테이트 탈수소효소를 코딩하는 ldhA 유전자 (SEQ ID NO: 2)로 이루어진 군 중에서 선택된 하나 이상의 유전자를 제거하여, 자이모모나스 모빌리스에 의한 에탄올, 락테이트, 피루베이트, 시트레이트, 글루타메이트, 숙시네이트, 푸마레이트 및 말레이트로 이루어진 군 중에서 선택된 1종 이상의 일차 대사산물의 생산을 증가시키는 방법.
- 제1항에 있어서, pdc 유전자 (SEQ ID NO: 1)를 제거하여 숙시네이트 및 락테이트로 이루어진 군 중에서 선택된 1종 이상의 일차 대사산물의 생산을 증가시키는 방법.
- 제1항에 있어서, ldhA 유전자 (SEQ ID NO:2)를 제거하여 에탄올 및 숙시네이트로 이루어진 군 중에서 선택된 1종 이상의 일차 대사산물의 생산을 증가시키는 방법
- 제1항에 있어서, pdc 유전자 (SEQ ID NO: 1) 및 ldhA 유전자 (SEQ ID NO:2)를 모두 제거하여 숙시네이트의 생산을 증가시키는 방법.
- 게놈으로부터 pdc 유전자(SEQ ID NO: 1) 및 ldhA 유전자 (SEQ ID NO: 2)로 이루어진 군 중에서 선택된 하나 이상의 유전자가 제거되어, 에탄올, 락테이트, 피루베이트, 시트레이트, 글루타메이트, 숙시네이트, 푸마레이트 및 말레이트로 이루어진 군 중에서 선택된 1종 이상의 일차 대사산물의 대량 생산이 가능한 자이모모나스 모빌리스 (Zymomonas mobilis) 형질전환체.
- 제5항에 있어서, pdc 유전자 (SEQ ID NO: 1)가 제거되어, 숙시네이트 및 락테이트로 이루어진 군 중에서 선택된 1종 이상의 일차 대사산물의 대량 생산이 가능한 형질전환체.
- 제5항에 있어서, ldhA 유전자 (SEQ ID NO: 2)가 제거되어, 에탄올 및 숙시네이트로 이루어진 군 중에서 선택된 1종 이상의 일차 대사산물의 대량 생산이 가능한 형질전환체.
- 제5항에 있어서, pdc 유전자 (SEQ ID NO: 1)와 ldhA 유전자 (SEQ ID NO: 2)가 모두 제거되어 숙시네이트의 대량 생산이 가능한 형질전환체.
- 제5항에 있어서, KCTC 11012BP, KCTC 11013BP 및 KCTC 10908BP로 이루어진 군 중에서 선택된 형질전환체.
- 자이모모나스 모빌리스(Zymomonas mobilis)의 게놈으로부터 pdc 유전자 (SEQ ID NO: 1) 및 ldhA 유전자 (SEQ ID NO: 2)로 이루어진 군 중에서 선택된 1종 이상의 유전자를 제거하는 단계를 포함하는, 제5항에 따른 자이모모나스 모빌리스 형질전환체의 제조 방법.
- 제10항에 있어서,자이모모나스 모빌리스의 pdc 유전자 (SEQ ID NO: 1)를 갖는 유전자 단편을 플라스미드에 클로닝하고;상기 pdc 유전자가 클로닝된 플라스미드로부터 pdc 유전자를 제거하고;상기 pdc 유전자가 제거된 플라스미드를 이용하여 자이모모나스 모빌리스를 형질전환시키는 단계를 포함하는,자이모모나스 모빌리스 형질전환체의 제조 방법.
- 제11항에 있어서,상기 pdc 유전자를 갖는 유전자 단편은 자이모모나스 모빌리스의 pdc 유전자와 그 양쪽으로 위치하는 동성 재조합 (homologous recombination)을 위한 1500 내지 5000 bp의 상동 부위를 갖는 것인,자이모모나스 모빌리스 형질전환체의 제조 방법.
- 제10항에 있어서,자이모모나스 모빌리스의 ldhA 유전자(SEQ ID NO: 1)를 갖는 유전자 단편을 플라스미드에 클로닝하고;상기 ldhA 유전자가 클로닝된 플라스미드로부터 ldhA 유전자를 제거하고;상기 ldhA 유전자가 제거된 플라스미드를 이용하여 자이모모나스 모빌리스를 형질전환시키는 단계를 포함하는,자이모모나스 모빌리스 형질전환체의 제조 방법.
- 제13항에 있어서,상기 ldhA 유전자를 갖는 유전자 단편은 자이모모나스 모빌리스의 ldhA 유전자와 그 양쪽으로 위치하는 동성 재조합 (homologous recombination)을 위한 1500 내지 5000 bp의 상동 부위를 갖는 것인,자이모모나스 모빌리스 형질전환체의 제조 방법.
- 제10항에 있어서,자이모모나스 모빌리스의 pdc 유전자 (SEQ ID NO: 1)를 갖는 유전자 단편을 플라스미드에 클로닝하고;상기 pdc 유전자가 클로닝된 플라스미드로부터 pdc 유전자를 제거하고;상기 pdc 유전자가 제거된 플라스미드를 이용하여 자이모모나스 모빌리스를 형질전환시키는 단계; 및자이모모나스 모빌리스의 ldhA 유전자(SEQ ID NO: 1)를 갖는 유전자 단편을 플라스미드에 클로닝하고;상기 ldhA 유전자가 클로닝된 플라스미드로부터 ldhA 유전자를 제거하고;상기 ldhA 유전자가 제거된 플라스미드를 이용하여 자이모모나스 모빌리스를 형질전환시키는 단계를 연속하여 수행하는 것을 특징으로 하는,자이모모나스 모빌리스 형질전환체의 제조 방법.
- pdc 유전자 (SEQ ID NO: 1) 및 ldhA 유전자 (SEQ ID NO: 2)로 이루어진 군 중에서 선택된 하나 이상의 유전자가 제거된 자이모모나스 모빌리스 형질전환체를 제작하는 단계; 및상기 자이모모나스 모빌리스 형질전환체를 30 내지 34 ℃에서 10 내지 14 시간동안 배양하는 단계를 포함하는,에탄올, 락테이트, 피루베이트, 시트레이트, 글루타메이트, 숙시네이트, 푸마레이트 및 말레이트로 이루어진 군 중에서 선택된 1종 이상의 일차 대사산물의 생산 방법.
- 제16항에 있어서,상기 자이모모나스 모빌리스 형질전환체의 배양시에 CO2 기체를 0.2 내지 1 vvm의 양으로 첨가하거나, 배양 배지에 탄산염을 1 내지 50 mM의 농도 추가로 첨가 하여 배양하는 것을 특징으로 하는,일차 대사산물의 생산 방법.
- 제17항에 있어서,상기 탄산염이 NAHCO3, NA2CO3 및 CaCO3로 이루어진 군 중에서 선택된 것인,일차 대사산물의 생산 방법.
- 제16항 내지 제18항 중 어느 한 항에 있어서,상기 자이모모나스 모빌리스 형질전환체의 배양시에 수소 기체를 0.2 내지 1 vvm의 양으로 추가로 첨가하여 배양하는 것을 특징으로 하는,일차 대사산물의 생산 방법.
