- 삭제
- 효모의 골기체 단백질인 Ste13의 골기체 표적 신호 부위와 막횡단 부위를 포함한 부분, Kex2의 기질로 알려진 효모의 알파-팩터의 절단 부위 및 인버타아제를 포함하는 융합단백질을 코딩하는 재조합 cDNA를 ADH 프로모터 하부에 연결한 플라스미드 pADH-SteSubInv.
- kex2 유전자를 ADH 프로모터 하부에 연결한 플라스미드 pADH-kex2와 제 2항의 pADH-SteSubInv로 동시형질전환된 사카로마이세스 세레비지에 SEY6210/pSTE-KR-SUC, pCUP-Kex2(Saccharomyces cerevisiae SEY6210/pSTE-KR-SUC, pCUP-Kex2, KCTC 1024BP).
- (ⅰ) 단백질 분해효소를 코딩하는 유전자를 포함하는 벡터를 작제하는 단계;(ⅱ) 효모의 골기체에 존재하는 막단백질의 골기체 표적 신호 부위와 막횡단 부위를 코딩하는 유전자, 특정 아미노산 서열을 가지는 기질 부위를 코딩하는 유전자 및 효모의 인버타아제(invertase)를 코딩하는 유전자를 포함하는 벡터를 작제하는 단계;(ⅲ) 전기 두 가지 벡터를 이용하여suc2돌연변이종 효모를 동시에 형질전환시켜서 형질전환체를 제조하는 단계; 및,(ⅳ) 전기 형질전환체를 자당을 유일한 탄소원으로 포함한 배지에서 배양하여 생존율을 측정하는 단계를 포함하는, 단백질 분해효소의 기질특이성을 측정하는 방법.
- 제 4항에 있어서,단백질 분해효소를 코딩하는 유전자를 포함하는 벡터는 pADH-kex2인것을 특징으로 하는단백질 분해효소의 기질특이성을 측정하는 방법.
- 제 4항에 있어서,골기체 표적 신호 부위는 Ste13의 골기체 표적 신호 부위인 것을특징으로 하는단백질 분해효소의 기질특이성을 측정하는 방법.
- 제 4항에 있어서,효모의 골기체에 존재하는 막단백질의 골기체 표적 신호 부위와막횡단 부위를 코딩하는 유전자, 특정 아미노산 서열을 가지는기질 부위를 코딩하는 유전자 및 효모의 인버타아제(invertase)를코딩하는 유전자를 포함하는 벡터는 pADH-SteSubInv인 것을특징으로 하는단백질 분해효소의 기질특이성을 측정하는 방법.
- 제 4항에 있어서,형질전환체는 사카로마이세스 세레비지에 SEY6210/pSTE-KR-SUC, pCUP-Kex2(Saccharomyces cerevisiae SEY6210/pSTE-KR-SUC, pCUP-Kex2, KCTC1024BP)인 것을 특징으로 하는단백질 분해효소의 기질특이성을 측정하는 방법.
- 제 4항에 있어서,형질전환체의 배양은 25 내지 35℃에서 3 내지 10일간 배양하는것을 특징으로 하는단백질 분해효소의 기질특이성을 측정하는 방법.
