Abstract
본 발명은 써머스 캘도필러스(Thermus caldophilus) GK24 유래의 내열성 알카라인 포스파타제에 관한 것이다. 좀 더 구체적으로, 본 발명은 써머스 캘도필러스 GK24로부터 유래하는 내열성 알카라인 포스파타제를 코딩하는 유전자 및 그로부터 유추되는 아미노산 서열에 관한 것이다. 본 발명자들은 써머스 캘도필러스 GK24로부터 APase(이하, 'TcaAPase'라 함)를 분리하여 일부 아미노산 서열을 결정하고, 이에 기초하여 올리고뉴클레오티드를 합성한 다음, 이를 프로브로 사용하여TcaAPase 유전자를 클로닝하였다. 전기 클로닝된 유전자의 염기서열 및 그로부터 유추되는 아미노산 서열을 결정한 결과,TcaAPase 유전자는 약 27개 아미노산의 신호펩타이드 및 성숙단백질을 포함하는 총 501개의 아미노산으로 구성된 단백질을 코딩하는 것으로 확인되었다. 본 발명의TcaAPase 유전자는 효소항체분석(enzyme immuno assay, EIA), 항체염색(immunostaining), 효소-항체결합 형광분석법(ELISA), 웨스턴블롯팅, 도트블롯팅 및 하이브리도마 선별(hybridoma screening) 등에 유용한 내열성 알카라인 포스파타아제의 대량생산을 실현시킬 수 있을 것이다.
Description
본 발명은 써머스 캘도필러스(Thermus caldophilus) GK24 유래의 내열성 알카라인 포스파타제에 관한 것이다. 좀 더 구체적으로, 본 발명은 써머스 캘도필러스 GK24로부터 유래하는 내열성 알카라인 포스파타제를 코딩하는 유전자 및 그로부터 유추되는 아미노산 서열에 관한 것이다.
알카라인 포스파타제(alkaline phosphatase, orthophosphoric-monoester phosphohydrolase, EC 3.1.3.1., 이하, 'APase'라 함)는 비 특이적인 인산 모노에스터(phosphate monoester)의 가수분해 및 전이인산화(transphosphorylation)를 촉매하는 효소이다. APase는 미생물로부터 포유류에 이르기까지 폭넓게 분포하는 아연을 포함하는 효소로서, 대부분 활성부위(active site)에 2개의 아연이온(Zn2+)과 1개의 마그네슘이온(Mg2+)이 작용한다. APase는 배지 내에 인산이 결핍되면 유도되고 과잉으로 존재하는 인산에 의해 저해되는 효소로서, 세포내의페리플라즘(periplasm)이나 세포외로 분비된다.
여러 APase들 중에서 대장균의 APase는 대표적인 세균유래의 APase로서,pho레귤론(phoregulon)에 속하는phoA유전자의 산물이다. 전기 APase는 세포질에서 신호펩티드(signal peptide)가 붙어 있는 형태로 합성된 후, 전위(translocation)되면서 펩티다제(peptidase)에 의해 신호펩티드가 절단되어 성숙된 형태(mature form)로 페리플라즘에 존재하면서 활성을 나타낸다. 세포내에서 APase는 인산의 결핍시 주위 환경으로부터 미량의 무기인산과 인산함유 영양분 예를 들면, 인산화 화합물이나 폴리인산을 이용할 수 있도록 유도된다.
이러한 APase는 분자생물학 연구에 있어서 매우 유용한 효소로서, 사용하는 기질에 따라 가용성 또는 불용성의 색소체를 생산하는 것이 가능하여, 세포 또는 조직체의 효소항체분석(enzyme immuno assay, EIA) 또는 항체염색(immunostaining)에 이용된다. 뿐만 아니라, 효소-항체결합 형광분석법(ELISA), 웨스턴블롯팅, 도트블롯팅 및 하이브리도마 선별(hybridoma screening)에도 이용하는 것이 가능하다. 그러나, 종래의 APase들은 내열성이 약해서 반응속도가 느리다는 단점을 지니고 있어서, 당업계에서는 보다 우수한 내열성 APase의 개발이 계속적으로 요구되어 왔다.
일반적으로, 내열성 효소의 생산에는 써머스속(Thermus sp.) 균주가 많이 사용되나, 현재까지 써머스속 유래의 APase는 써머스 아쿠아티쿠스(T. aquaticus) YT-1로부터 유래한 한가지만이 알려져 있다. 그러나, 전기 써머스속 유래의 APase조차 내열성이 그다지 강하지 못하므로, 보다 강한 내열성을 지니는 APase를 개발하여야 할 필요성이 계속적으로 대두되었다.
이에, 본 발명자들은 다른 써머스 속 균주로부터 보다 우수한 내열성 알칼라인 포스파타제 및 그를 코딩하는 유전자를 분리하고자 예의 연구노력한 결과, 써머스 캘도필러스(Thermus caldophilus) GK24로부터 내열성 APase 및 게노믹 유전자를 분리한 데 이어, 전기 유전자의 염기서열과 그로부터 유추되는 아미노산 서열을 결정하여 종래의 APase와는 상이한 아미노산 서열을 가지는 신규한 효소임을 확인하고, 본 발명을 완성하게 되었다.
결국, 본 발명의 주된 목적은 써머스 캘도필러스(Thermus caldophilus) GK24로부터 유래하는 내열성 APase를 코딩하는 유전자 서열을 제공하는 것이다.
본 발명의 다른 목적은 전기 유전자 서열로부터 유추되는 아미노산 서열을 제공하는 것이다.

본 발명자들은 써머스 캘도필러스(Thermus caldophilus) GK24(참조: Taguchi, H., et al.,J. Biochem.,91, 1343(1982))로부터 APase(이하, 'TcaAPase'라 함)를 분리하여 일부 아미노산 서열을 결정하고, 이에 기초하여 올리고뉴클레오티드를 합성한 다음, 이를 프로브로 사용하여TcaAPase 유전자를 클로닝하였다. 전기 클로닝된 유전자의 염기서열 및 그로부터 유추되는 아미노산 서열을 결정한 결과,TcaAPase 유전자는 약 27개 아미노산의 신호펩타이드 및 성숙단백질을 포함하는 총 501개의 아미노산으로 구성된 단백질을 코딩하는 것으로 확인되었다.
즉, 본 발명자들은 써머스 캘도필러스 GK24로부터TcaAPase를 분리한 다음, 친화크로마토그래피로 정제하여 분자량이 약 54,000달톤인TcaAPase를 수득하였다. 전기 정제된TcaAPase의 내열성을 조사한 결과, 50 mM Tris-HCl (pH 8.6) 완충용액을 사용한 경우에는 80℃ 및 85℃에서 2시간 방치한 후에도 각각 60% 및 80% 이상의 활성을 유지하였으나, 50 mM Glycine-NaOH (pH 11.0) 완충용액을 사용한 경우에는 80℃에서 2시간 방치한 후 50%, 85℃에서 10분간 방치 후 40%까지 활성이 감소하는 것으로 나타났다.
정제된TcaAPase로부터 펩티드 단편들을 제조하여 일부 펩티드의 아미노산 서열을 결정하고, 전기 아미노산 서열을 바탕으로 올리고뉴클레오티드들을 제조한 다음, 이들을 서던블롯시에 프로브로 사용하여 써머스 캘도필러스 게노믹 유전자로부터TcaAPase 유전자를 포함하는 유전자 절편을 선별하였다. 전기 유전자 절편을 프로브로 사용하여 전체TcaAPase 유전자를 클로닝하고, 염기서열을 결정한 다음, 피씨진(PCGENE) 및 NCBI-블라스트 서치(NCBI-blast search)로 분석하여, 신호펩티드 및TcaAPase를 코딩하는 총 1503bp(종지코돈 포함 1506bp)의 염기서열을 포함하는TcaAPase 유전자의 리딩프레임(reading frame)을 결정하였다.
전기TcaAPase 유전자는 G+C 함량이 약 69.3%이며, 라이보좀 결합부위로 유추되는 GGAG 서열이 ATG 코돈 직전에 나타나고 있으나, 다른 종에서 나타나는 -10 보존부위나 -40 보존부위는 나타나지 않았다.TcaAPase는 우선 총 501개의 아미노산으로 구성된 단백질로 발현된 다음, 그중 약 27개의 아미노산으로 구성된 신호핍티드가 전위(translocation) 과정에서 제거되는 것으로 보인다. 한편,TcaAPase의 아미노산 서열을 다른 종의 APase들의 아미노산 서열과 비교·분석한 결과,TcaAPase가 다른 종들로부터 유래한 APase들과는 상이한 아미노산 서열을 지니고 있는 신규한 APase임이 확인되었다.
이하, 본 발명을 실시예에 의하여 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 이들 실시예에 의하여 본 발명의 범위가 한정되지 않는다는 것은 본 발명이 속하는 기술분야에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예 1:TcaAPase의 정제
종균배양한 균주를 전배양배지[0.4% 박토트립톤, 0.2% 효모 추출물, 1 × 캐스텐홀즈 염(Castenholz salts)] 500 ㎖에 접종하여 72℃에서 180 rpm으로 진탕배양한 후, 인산이 제거된 APase 최적유도배지[0.3% 소듐글루타메이트, 0.2% 박토트립톤, 0.5% 글루코스, 10 mM Tris-HCl (pH 8.0), 1× 캐스텐홀즈 염] 2.5ℓ에 2% 접종하고, 72℃에서 180 rpm으로 16시간 진탕배양하였다. 이어,냉각삼투압충격법(cold osmotic shock)으로 균체를 파쇄하여 APase를 포함하는 조효소액을 얻었다. 전기 조효소액에 70% (w/v) 포화농도의 황산암모늄을 천천히 첨가하여 단백질을 침전시킨 후, 10 mM Tris-HCl (pH 7.4)/1 mM MgCl2완충용액을 사용하여 투석하였다. 투석에 의해 염이 제거된 조효소액은 4(4-아미노페닐아조)-페닐라소닉산-세파로스4B(4(4-aminophenylazo)-phenylarsonic acid-Sepharose 4B)를 충진재로 하는 친화크로마토그래피 칼럼을 통과시켜TcaAPase를 결합시켰다. 전기TcaAPase가 결합된 컬럼에 [1 M KCl/10 mM Tris-HCl (pH 7.4)] 및 [2 M guanidium-HCl/1 M KCl/10 mM Tris-HCl (pH 7.4)]로 조성된 완충용액을 농도구배를 형성시키면서 흘려TcaAPase를 유리시켜 정제된TcaAPase를 얻었다. 이를 SDS-PAGE 전기영동한 결과를 도 1에 나타내었다. 도 1에서 제 1레인은 정제된TcaAPase를 나타내며, M은 분자량 마커를 각각 나타낸다. 도 1에서 보듯이, 순수하게 정제된 APase를 얻을 수 있었으며,TcaAPase의 분자량은 약 54,000달톤이었다.
실시예 2:TcaAPase의 내열성 조사
상기 실시예 1에서 얻은TcaAPase의 최적 반응온도 및 내열성을 조사하였다. 반응액의 조성은 완충용액 185 ㎕,TcaAPase 5㎕ 및 10 mMpNPP 100 ㎕를 포함하도록 하였다.TcaAPase 활성은 반응결과 인산이 유리되어 형성된 발색단을 지니고 있는pNP를 410 nm에서 흡광도를 측정함으로써 결정하였다. 최적 반응온도를 결정하기 위하여 반응온도 40℃ 내지 100℃까지 매 10℃마다의 활성을 측정하였으며, 완충용액으로는 1mM MgCl2를 포함하는 50 mM Tris-HCl (pH 8.6) 또는 50 mM Glycine-NaOH (pH 11.0)을 사용하였다. 도 2는TcaAPase의 온도에 따른 상대적인 활성을 나타낸 그래프이다. 도 2에서 보듯이, 최적반응 온도는 약 85℃로 나타났다.
TcaAPase의 내열성을 조사하기 위하여, 80℃ 내지 100℃에서 5℃간격으로 완충용액과 효소액을 섞어 2시간까지 열처리한 후 얼음물에 담가 식히고 기질을 첨가하여 80℃에서 활성을 측정하였다. 완충용액으로는 50 mM Tris-HCl (pH 8.6) 및 50 mM Glycine-NaOH (pH 11.0) 완충용액을 사용하였고, 80℃ 와 85℃에서 방치한 다음, 활성을 측정한 결과를 도 3에 나타내었는데, 50 mM Tris-HCl (pH 8.6) 완충용액을 사용한 경우에는 80℃ 및 85℃에서 2시간 방치한 후에도 각각 60% 및 80% 이상의 활성을 유지하였으나, 50 mM Glycine-NaOH (pH 11.0) 완충용액을 사용한 경우에는 80℃에서 2시간 방치한 후 50% 및 85℃에서 10분간 방치 후 40%까지 감소하는 것으로 나타났다. 결과적으로,TcaAPase의 내열성에는 50 mM Glycine-NaOH (pH 11.0) 완충용액 보다는 50 mM Tris-HCl (pH 8.6)이 더 바람직하였다. 도 3에서 △는 80℃, Glycine-NaOH 완충용액에 방치한 경우; ◇는 85℃, Glycine-NaOH 완충용액에 방치한 경우; ○는 80℃, Tris-HCl 완충용액에 방치한 경우; 및, □는 85℃, Tris-HCl 완충용액에 방치한 경우의 내열성을 각각 나타낸다.
실시예 3:TcaAPase의 일부 아미노산 서열결정
TcaAPase의 아미노산 서열을 부분적으로 결정하기 위하여, 먼저 상기 실시예 1에서 수득한 정제된TcaAPase를 동결 건조시킨 다음, 각각 메타이오닌(methionine)과 트립토판(tryptophan)의 카복시말단을 절단하는 CNBr과 BNPS-skatole을 처리하였다. 즉, CNBr을 처리하기 위하여 우선TcaAPase의 시스테인(cystein) 잔기를 카복시메틸레이션(carboxymethylation)시켜 단백질을 하기의 방법으로 완전히 변성시켰다. 완전히 동결건조된TcaAPase 1 ㎎을 완충용액 200 ㎕에 녹인 후, 상층에 질소가스(N2 gas)를 15분간 처리하고, 4 mM DTT 200 ㎕를 첨가하여 질소가스를 1시간 동안 추가로 처리하였다. 전기 시료를 빛으로부터 차단하기 위하여 알루미늄 호일로 감싼 후, 천천히 교반하면서 500 mM 이디오아세트산(idioacetic acid, pH 8.5) 160 ㎕를 첨가한 다음, 다시 질소가스를 처리하고 완전히 봉합하여 암조건으로 37℃에서 30분간 반응시켰다. 그런 다음, 50 mM 암모늄 바이카보네이트 완충용액으로 하룻밤 동안 투석하고 동결건조시켰다. 동결건조된TcaAPase를 70% 포름산(formic acid)용액에 첨가하고, 첨가한TcaAPase 무게의 100배되게 CNBr을 첨가하여 질소가스를 처리한 후 25℃ 암조건에서 24시간 반응시켰다. 여기에 증류수를 첨가하고 원심진공건조(speed vacuum drying)시켜 CNBr처리된TcaAPase를 얻었다.
동결 건조시킨TcaAPase 1.4 ㎎을 1.3 ㎍/㎕ BNPS-skatole이 용해되어있는 840 ㎕ 빙초산(glacial acetic acid) 용액에 첨가하고 증류수 280 ㎕를 가하여 암조건으로 47℃에서 1시간 동안 반응시키면서 BNPS-skatole 처리하였다. 전기액에 동량의 증류수를 첨가하고 원심분리하여 형성된 노란색 침전물인 BNPS-skatole은 제거하고, 상층액을 회수하여 증류수를 첨가한 후 동결건조하여 반응을 종결시켰다.
전기의 CNBr 또는 BNPS-skatole 처리하여 얻은 단편들은 펩티드분석용 전기영동을 실시하여 여러개의 밴드로 분리하고, 전기 분리된 밴드들을 PVDF 막에 10 mM CAPS 완충용액[2.213g CAPS, 15% MeOH, pH 11.0]을 사용하여 전기적 블롯팅(electroblotting)을 한 다음, 0.2% Ponceau S 과 1% 아세트산을 포함하는 액으로 염색하여, CNBr 처리에 의하여 생성된 1개의 펩티드 단편(AP-1)과 BNPS-skatole 처리에 의하여 생성된 3개의 펩티드 단편(Trp-1, Trp-2 및 Trp-3)을 선택하였다. 전기 총 4개의 펩티드 단편들의 아미노산 서열을 결정하고(참조: 도 4), 전기 아미노산 서열을 기초로 하여TcaAPase 유전자의 클로닝을 위한 서던블롯에 사용될 프로브를 합성하였다. 즉, 코돈 활용도(codon usage)를 고려하여 가능한한 코돈의 3번째 염기의 종류가 적은 아미노산 서열을 선택하고, 써머스 균주 유전자 코돈의 특징인 3번째 염기를 G 또는 C로 하여, 도 5에서와 같은 염기서열을 가지는 올리고뉴클레오티드 AP-2와 AP-3를 합성하고, 이들을 서던블롯에 프로브로 사용하였다.
실시예 4: 써머스 캘도필러스(Thermus caldophilus) 게노믹 유전자의 분리
다음과 같은 변형된 마머(Marmur)의 방법을 이용하여 써머스 캘도필러스(Thermus caldophilus)의 게노믹 유전자를 분리하였다(참조: Marmur, J.,J. Mol. Biol., 3:208-218(1961)): 먼저, 200 ㎖의 배지[[0.4% 박토트립톤, 0.2% 효모 추출물, 1 × 캐스텐홀즈 염(Castenholz salts)]를 사용하여 진탕배양한 써머스 캘도필러스 GK24 균체를 원심분리하여 회수하고, SETB (20% sucrose/ 50 mM Trsi-HCl (pH 7.6)/ 50 mM EDTA) 완충용액 4 ㎖로 현탁하고 5회의 동결(freezing) 및 해동(thawing)을 반복하여 세포막을 약화시킨 후, 10 ㎎/㎖의 라이소자임(lysozyme) 및 100 ㎍/㎖의 RNase A를 첨가하여 37℃에서 40분 동안 반응시켰다. 전기 반응액에 최종 부피의 1%가 되도록 SDS를 첨가하고 클로로포름:이소아밀알콜 (24:1, v/v)의 혼합액 5 ㎖을 첨가하여 하룻밤 동안 천천히 교반해 주었다. 전기 반응액에 페놀과 클로로포름을 첨가하고 10 ㎎/㎖ 프로티나아제 K 100 ㎕를 첨가하여 37℃에서 30분 반응시킨 다음, 55℃에서 20분 추가로 반응시키고, 다시 페놀과 클로로포름으로 추출하여 얻어진 상층액에 2배의 에탄올과 0.1배의 3 M 소듐아세테이트(sodium acetate, pH 5.2)를 가하여 유전자를 추출하였다. 전기 추출한 유전자를 70% 에탄올로 2회 세척한 후 진공건조시키고, TE(10 mM Tris-HCl, pH 7.6 / 1 mM EDTA, pH 8.0) 완충용액에 용해시킨 다음, 260nm에서의 흡광도를 측정하여 정량하였다.
실시예 5: 서던블롯
써머스 캘도필러스(Thermus caldophilus) GK24의 게노믹 유전자로부터TcaAPase 유전자를 선별하기 위한 서던블롯에 사용하고자, 상기 실시예 3에서 합성된 AP-2와 AP-3 올리고뉴클레오티드를 [γ-32P]dATP(3000Ci/mmol)와 T4 폴리뉴클레오티드 키나아제를 사용하여 표지하고, 세파덱스 G-25 미니 컬럼(Sephadex G-25 mini-column)으로 정제하였다. 상기 실시예 4로부터 수득한 써머스 캘도필러스 GK24의 게노믹 유전자를BamHⅠ,HindⅢ/PstⅠ,SacⅠ 및StuⅠ의 제한효소로 각각 처리하고 0.7% 아가로오스 젤 전기영동으로 분리한 다음, 전기 표지된 프로브를 사용하여 서던블롯한 결과,BamHⅠ으로 절단한 경우에는 약 2.1 kb에서 신호가 나타났고,HindⅢ/PstⅠ로 절단한 경우에는 약 7.0 kb에서 신호가 나타났다.
실시예 6:TcaAPase 유전자의 클로닝
전기BamHI으로 절단한 게노믹 DNA를 저융점 아가로오스 젤로 전기영동하고 약 2.1 kb 단편만을 회수하여, 동일한 제한효소로 처리한 pBluescript SK+에 클로닝하였다. 전기 조작된 pBluescript SK+ 플라스미드로 대장균을 형질전환시켜 약 700개의 콜로니를 얻었으며, 상기 실시예 5의 서던블롯에 사용한 것과 동일한 프로브를 사용하여 콜로니 하이브리다이제이션을 실시한 결과, 전기 700개의 콜로니 중에서 6개의 콜로니로부터 신호가 나타났다.
전기 6개의 콜로니로부터 플라스미드를 알카라인 변성법으로 분리하고, 상기 실시예 5의 서던블롯에 사용한 것과 동일한 프로브를 사용하여 플라스미드 하이브리다이제이션을 실시한 결과, 4개의 양성클론(positive clone)을 확보하였다. 이로부터 2.1kb의BamHⅠ 단편을 제조하여, 멀티-프라이밍 레이블링 킷트(multi-priming labelling kit)를 사용하여 [α-32P] dCTP(3000Ci/mmol)로 표지하고, 전기 표지된 2.1kbBamHⅠ 단편을 프로브로 사용하여 상기 실시예 5와 동일한 방법으로 써머스 캘도필러스 GK24의 게노믹 유전자에 대한 서던블롯을 실시하였다. 그 결과,SacⅠ으로 절단하여 얻어진 약 1.5 kb의 단편 및 1.8 kb의 단편과ApaⅠ으로 절단하여 얻어진 약 2.8 kb의 단편에서 신호가 나타났다. 이 중에서SacⅠ으로 절단한 결과 얻어진 약 1.5 kb 단편과ApaⅠ으로 절단한 결과 얻어진 약 2.8 kb의 단편을 동일한 제한 효소로 절단한 pBluescript SK+에 삽입하고, 대장균을 형질전환시켰다. 그 중, 약 2.8 kb의ApaⅠ단편을 포함하는 벡터로 형질전환시킨 경우에 5개의 양성클론을 얻었다. 전기 양성클론으로부터 플라스미드를 제조한 다음, 전체 약 3.1kb의 플라스미드 유전자를ApaⅠ,BamHⅠ,SacⅠ,SacⅡ, SmaⅠ 및XhoⅠ등의 제한효소로 절단하고 유전자마커와 함께 전기영동하여 각 크기를 비교함으로써 제한효소지도를 작성하였다. 그 결과, 도 6과 같은 제한효소 지도를 얻었다.
실시예 7:TcaAPase 유전자 염기서열의 결정 및 아미노산 서열 비교
상기 실시예 6에서 결정된 제한효소 지도를 기초로 하여, 삽입 유전자의 크기가 약 0.5 kb가 되도록 서브클로닝하였다. T3 프라이머와 T7 프라이머를 사용하여 생거의 디데옥시 사슬 종결법(Sanger's dideoxy chain termination method)으로각 서브클론의 염기서열을 결정하고, 이를 컴퓨터프로그램인 피씨진(PCGENE)으로 분석한 다음, 인터넷프로그램인 NCBI-블라스트서치(NCBI-blast search)의 서열 분석결과와 전기 실시예 3에서 결정된TcaAPase의 내부 아미노산 서열을 바탕으로 하여, 신호펩티드 및 성숙단백질을 코딩하는 총 1503 bp의 염기서열을 포함하는TcaAPase 유전자의 리딩프레임 염기서열(참조: 서열번호 1)과 그로부터 유추되는 아미노산 서열(참조: 서열번호 2)을 결정하였다.
TcaAPase 유전자의 G+C 함량은 69.3%이며, 서열번호 1에서 보듯이, 라이보좀 결합부위로 유추되는 GGAG 서열이 ATG 코돈 직전에 나타나고 있으나, 다른 종의 APase에서 나타나는 -10 보존부위이나 -40 보존부위는 발견되지 않았다.TcaAPase는 우선 신호펩티드를 포함하는 총 501개의 아미노산으로 구성된 단백질로 발현되는 것으로 보이며, 대부분의 신호펩티드의 서열 특성을 고려할 때, 전위과정에서 아마도 Ala 27에서 절단되는 것으로 보인다. 한편, NCBI-블라스트 서치를 이용하여 대장균 APase 및 바실러스 서브틸리스(Bacillus subtilis) APase와의 아미노산 서열상동성을 비교분석한 결과, 각각 27%, 32%로 비교적 낮은 상동성을 가지는 것으로 확인되었다(참조: 도 7). 도 7에서, Tca는 써머스 캘도필러스 GK24의 APase, Bsu3는 바실러스 서브틸리스의 APaseⅢ, Bsu4는 바실러스 서브틸리스의 APaseⅣ, Eco는 대장균의 APase를 각각 나타내며, 음영처리된 부분은 종간의 서열 보존부위를 나타낸다.
이상에서 상세히 설명하고 입증하였듯이, 본 발명은 써머스 캘도필러스(Thermus caldophilus) GK24로부터 유래하는 내열성 알카라인 포스파타아제 유전자 및 그로부터 유추되는 아미노산 서열을 제공한다. 본 발명의 내열성 알카라인 포스파타아제 유전자는 효소항체분석(enzyme immuno assay, EIA), 항체염색(immunostaining), 효소-항체결합 형광분석법(ELISA), 웨스턴블롯팅, 도트블롯팅 및 하이브리도마 선별(hybridoma screening) 등에 유용한 내열성 알카라인 포스파타아제의 대량생산을 실현시킬 수 있을 것이다.

- 써머스 캘도필러스(Thermus caldophilus) GK24로부터 유래된 서열번호 1과 같은 염기서열을 가지는 내열성 알카라인 포스파타아제 유전자.
- 제 1항의 염기서열로부터 유추되는 써머스 캘도필러스(Thermus caldophilus) GK24로부터 유래된 서열번호 2와 같은 아미노산 서열을 가지는 내열성 알카라인 포스파타아제.
